Objective To establish a method performance verification project and experimental method for the clinical chemiluminescence immunoassay.Methods Referring to CLSI evaluation protocols and pertinent literature,and by combining our actual works,we designed a verification procedure and experimental method.By Using these above,the precision,accuracy,analytical sensitivity,analytical measurement range,clinical reportable range and biotic interval of AFP on the Bayer Centaur 240 chemiluminescence immunoassay system were verificated.Results would be compared with the declaration of the manufacturer or desirable specifications derived from biologic variation.Results The results showed that the between-day inaccuracy on AFP levels at 77.4 ng/ml and 168.0 ng/ml was 5.70% and 4.84% respectively,these were consistent with manufacturer's inaccuracy claimed.The relative bias between the results measured for calibrator at four levels and target value was less 5.0%,and the relative bias between the results measured for EQA control sample at five levels and target value was-3.4% to 11.9%.Lower limit of detection was 1.04 ng/ml,lower slightly manufacturer's analytical sensitivity claimed.Biologic limit of detection was 2.65 ng/ml-3.53 ng/ml,functional sensitivity was 3.53 ng/ml.Analytical measurement range was 3.53-912.00 ng/ml,within manufacturer's liner range claimed.Clinical reportable range was 3.53-182 400.00 ng/ml.Reference interval was 0.6-7.7 ng/ml,within manufacturer' s claimed.Conclusions The main performances of the detection system are accorded with the declaration of the manufacturer.The performance verification procedure and experimental method of our research ars simple and practical,which has important significations for building medical laboratory and laboratory accreditation, improving quality of the chemiluminescence immunoassay.

Objective To learn the population and infestation rates of cockroaches from 2017 to 2019 in Jiading District of Shanghai, to evaluate the effect of cockroach termination in household, and to provide information for cockroach control. Methods Cockroaches were controlled by dinotefuran baits and clean-up in households.Sticky trap and visual method were employed for density monitoring in farmers markets, supermarkets, hotels, restaurants, hospitals, and residential areas.Visual method was used in households before and after using the insecticide. Results Sticky trap result showed the room infestation rate was 3.24%, mean adhesion rate was 3.29%, the density was 0.06 per board, and the density peak appeared in May.Rate of invasion and density decreased year by year.Blattella germanica was the dominant species, counting for 71.88%.The density, and rate of infestation, as determined by sticky trap method, were the highest in the farmers markets, followed by hospitals and residential areas.Determined by visual method, room infestation rate was 1.16%, and the infestation rate was 4.44%.The peak appeared in January.Infestation rate of the farmers markets was the highest, followed by hospitals and residential areas.By visual method, the room infestation rate was 59.01%, and 48.45% for nymphs.The room infestation and ootheca rates were 54.04% and 17.39%.The rate decreased more than 80% in 30 days after use of the insecticide. Conclusion Infestation rate of cockroach remains at low level in Jiading District.The effect of bait combined with environmental cleaning is remarkable.Future work should strengthen monitoring and control in farmers markets, hospitals and residential areas.

OBJECTIVETo compare the adaptation of porcelain fused-to-metal (PFM) restorations made from Ni-Cr alloy, precious alloy and galvanized forming copings after cementation and to provide a theory guidance for their application.

METHODSThree kinds of crowns (Ni-Cr alloy, precious alloy and galvanized forming) were manufactured and cleaned by ultrasonic vibrate with alcoholic solution for 5 minutes, and cemented on their dies as their order. All the crowns were cemented by polycarboxylate zinc-cement and maintained 10 minutes. After coated in the center of methyl acrylic resins, all the samples were cut vertically along buccolingual direction. The cement thickness of PFM was measured by scanning electron microscope and the data were analyzed by multivariate ANOVA.

RESULTSNo significant difference was found between the cement thickness of precious alloy crown and galvanized forming crown (P>0.05), while both of these two kinds of crown had significant differences in cement thickness with Ni-Cr crown (P<0.05).

CONCLUSIONThe adaptation of precious alloy crown and galvanized forming crown are superior to Ni-Cr crown.

OBJECTIVETo investigate the expression and significance of Cyclin D1 in oral squamous cell ma (OSCC).

METHODSA immunohistochemistry method, Envosion, was employed to test the manifesting Cyclin D1 in pathological slices of 50 OSCC cases and 10 normal cases, and the results was treated with statistical lysis.

RESULTSIn 50 OSCC cases, Cyclin D1 mainly manifested in karyon, and a little in cytoplasm. manifesting rates of Cyclin D1 in the samples was 80.0%, which was significantly higher than the manifesting of 20.0% in normal oral mucous membrane (P < 0.01). The manifestation of Cyclin D1 was correlated with rent pathological grades, clinical phases and lymph node metastasis (P < 0.05).

CONCLUSIONThe abnormal tation of Cyclin D1 is closely related with the occurrence and development of OSCC. Therefore, it can subsidiary index for OSCC treatment and prognosis.

OBJECTIVETo investigate the effect of vascular endothelial growth factor 165 (VEGF165) gene transfer on the proliferation and metabolism of human bone marrow stromal cells (hBMSCs) in vitro.

METHODShBMSCs were divided into 3 groups and subjected to adenovirus mediated VEGF165 gene transfection, transfection with empty adenoviral vector, or left untreated (control). MTT assay and flow cytometry were performed to analyze the proliferation of the cells after corresponding treatments. The third passage of hBMSCs (2x10(4)/ml), after corresponding transfection procedures, were cultured in conditional medium and tested for ALP content 2, 4 and 6 days after the transfection. Also at 3, 5 and 7 days after the transfection, the cells were examined for osteocalcin (C) and laminin (LN) contents.

RESULTSThe number of cells in each group increased with the culture time without obvious differences in the optical density. No significant differences were noted between the 3 groups in the percentage of G1 phase cells or in the proliferation index (PrI) (P>0.05), but compared with the nontransfected and the empty vector-transfected cells, the cells with VEGF165 gene transfection had significantly higher ALP, OC and LN contents (P<0.05).

CONCLUSIONVEGf165 gene transfer does not obviously affect the proliferation of cultured hBMSCs, but can increase the cellular secretion of AIP, C and LN, suggesting that VEGF165 promotes the differentiation of hBMSCs into osteoblasts in vitro.

OBJECTIVETo study the treatment and therapeutic effects in the shearing-type and comminuted talar body fractures.

METHODSFrom October 1988 to September 2005,34 patients with shearing-type or comminuted talar body fractures were followed up. There were 19 males and 15 females ranged from 13 to 55 years (averaged 28.8 years). The disease course ranged from 3 to 14 days (averaged 6.0 days). Ten patients with a displacement of no more than 3 mm were treated with plastic cast. Eighteen patients were treated with open reduction and internal fixation, 6 patients were treated with joint fusion. The assessment of clinical efforts depend on patients' ache, active range of the joint and limp.

RESULTSThe mean follow up was 5.04 years (ranged from 3 to 19 years). All the patients were healed. The clinical outcomes were evaluated according to Hawkins evaluation score in which ache, active range of the joint and limp was respectively acssessed. There were 6 patients reached an excellent result, 9 good, 11 fair and 8 poor. Fifteen patients had osteonecrosis, 18 patients had traumatic arthritis of ankle joint, and 14 patients had traumatic arthritis of subtalar joint.

CONCLUSIONPatients whose displacement of fracture is not more than 3 mm should be treated with plastic cast. Operation and internal fixation should be performed in patients whose displacement of fracture is more than 3 mm after close reduction. Joint fusion should be performed in patients whose talar body fracture is comminuted severely and the surface of joint can not be repaired. The patients of talar body scissored fracture or comminuted fracture has bad prognosis.

OBJECTIVETo observe the clinical curative effect of Chinese herbal medicine combined with acitretin capsule in treating psoriasis of blood-heat syndrome (P-BH).

METHODSEighty patients of P-BH were randomly assigned to two groups, 39 in Group A and 41 in Group B. Both was treated with Chinese herbal medicines for clearing heat, cooling blood and removing toxic substance, and acitretin capsule was given to Group A additionally, with 8 weeks as one therapeutic course. The clinical curative effect was compared between groups, and the change of psoriasis activity severe index (PASI) scores before and after treatment was observed.

RESULTSThe total effective rate in Group A was 84.2% and that in Group B 68.2%, also showing significance between groups (P<0.01). PASI score lowered significantly after treatment in both groups, showing statistical significance (P<0.01), but no significant difference between groups. Little adverse reaction was found in Group B, while in Group A, the adverse reaction was of even milder degree, which could be alleviated by adjusting the herbal medicine and symptomatic treatment administration.

CONCLUSIONSThe effect of Chinese herbal medicine combined with acitretin capsule was superior to Chinese herbal medicine alone in treating P-BH, but the adverse reaction of acitretin capsule could be alleviated by adjusting the herbs used. However, the result is waiting to be verified further by larger samples.

Objective To understand the current status of the prevention and control of endemic fluorosis in the southwestern area of Shandong province. Methods In 2007, the progress of water-improving defluoridation, the operating state and water fluoride content of the water-improving project, which was determined by fluorosis selective ion electrode, and the inhabitant related indexes of endemic fluorosis were extensively surveyed in the three main fluorosis counties-Jiaxiang, Yuncbeng and Liangshan of the southwestern area of Shandong province. Results Among 1371 fluorosis villages in the 3 counties, 53.61%(735/1371) of which had undergone water-improving defluoridation, the rate in Jiaxiang, Yuncheng and Liangshan being 38.0% (220/579),65.51% (378/577) and 63.72% ( 137/215 ) respectively; the normally functioning rate of this project was 76.73% (564/735), projects out of order accounted for 23.27% (171/735). Among 263 well-functioning projects from the three counties, the rate with water fluoride higher than 1.0 mg/L was 35.36%(93/263), the maximum value being 4.17 mg/L. The urine fluoride content of 440 children aged 8 - 12 years and 484 adults over 30 years old were examined in 13 fluorosis villages of the three counties, the geometric mean was 2.98,3.06 mg/L respectively; the individual maximum was 12.83,14.49 mg/L respectively; the detectable rate of dental fluorosis among children aged 8 - 12 was 84.28% (649/770) ,17.66%(136/770) had defect and the index of dental fluorosis was 1.89; the rates of the clinical and X-ray skeletal fluorosis of the adults aged more than 30 were 44.40%(234/527) and 24.67%(130/527) respectively, and the abnormal electrocardiography rate was 32.43% (168/518) in the adult, mostly T-wave abnormality. Conclusions The progress of the water-improving defluoridation in the southwestern area of Shandong province was relatively slow, the water fluoride content of the water-improving projects seriously exceeded standard, and the condition of the fluorosis had not been effectively controlled.
