1.Effects of immunosuppressor against kidney disease in treatment of 21 patients with cytomegalovirus pneumonia

Objective To study the clinical feature,treatment,and prognosis of the cytomegalovirus (CMV)pneumonia patients treated with immunosuppressor against kidney disease.Mlethod The patients received immunosuppressor against kidney disease in The First Affiliated Hospital of Sun Yat-sen University from June 1999 to December 2006.CMV antigen of leucocyte in the peripheral blood and/or bronchoalveolar lavage fluid of these patients were detected with immunocytochemical methods,and 21 patients were found suffering from CMV pneumonia.The 21 patients were introvenously injected with ganciclovir 5~10 mg/(kg?d),and the immunosuppressive agent treatment suspended.Their clinical feature and prognosis were retrospectively analyzed. Results The 21 patients received corticosteroids before CMV pneumonia contracted,of them,13 patients had been intensively treated with Methyllprednisolone with mean total dose(3.2?0.6)g.Of them,15 had been treated with cyclophosphamide with mean total dose(3.8?1.3)g.The median time from the beginning of using immunosuppressor to the onset of CMV pneumonia was 25(13~92)days.All patients had fever,cough, shortness of breath and X-ray showed interstitial pneumonia,of them,19 patients developed hypoxemia,and 11 patients' CMV antigen was positive in the leucocyte from bronchial lavage fluid.The result showed 9 patients survived and 12 died.The average duration of treatment with ganciclovir was(26.2?6.3)days. CMV pneumonia is a serious complication in patients who were treated with immunosuppressor against kidney disease.The mortality is high.Ganciclovir is a medicine of choice to treat CMV pneumonia.
2.Identification of EST-SSRs in Taxus cuspidata based on high-throughput sequencing.

OBJECTIVETaxus species are highly valued for the production of taxol. Based on high-throughput sequenceing, EST-SSRs were explored and studied in the transcriptome of Taxus cuspidata.
METHODT cuspidata leaf cDNA was extracted and sequenced by 454 GS FLX Titanium. High-quality sequences were assembled using Newbler Assembler Software, which produced unique sequences. SSRs motif was explored using simple sequence repeat identification tool (Perl Script). Primers were designed using PRIMER3.
RESULTA total of 81 148 high-quality reads from the needles of T. cuspidata were produced using the Roche GS FLX Titanium system. A total of 20 557 unique sequences were obtained. There were 753 simple sequence repeat motifs identified. Primers of PCR were obtained for 519 EST-SSRs, randomly selected cloning sequencing revealed that 87.5% of ESTs were the same as the results of Sanger sequencing.
CONCLUSIONThe results provide the first EST-SSRs collection in Taxus and are essential for future efforts of gene discovery, functional genomics, and genome annotation in related species.

3.Effect of vitamin E on the lag time of low-density lipoprotein oxidation in vitro in patients with cerebral infarction

Objective To investigate the effect of vitamin E on the lag time of serum low-density lipoprotein (LDL) oxidation in vitro in patients with acute cerebral infarction (ACI). Methods Twenty-two patients with ACI received vitamin E treatment at the dose of 300 mg/day, and another 22 ACI patients matched for age, gender ratio, blood pressure, blood lipid level, location and degree of the infarction served as the control group in which no vitamin E was given. Serum samples were collected from the patients within 24 h and at 2 weeks after the onset to test the lag time of LDL oxidation in vitro and measure the levels of vitamin E, total cholesterol, triglyceride and free cholesterol. Results At two weeks after the onset of the infarction, the patients receiving vitamin E treatment had significantly prolonged lag time of LDL oxidation in comparison with that of the control patients (71.84± 9.75 vs 60.95±9.33 min, P<0.05), and showed also significantly higher vitamin E level in the serum Vitamin E and stroke index. The two groups of patients showed no significant differences in total cholesterol (5.08cholesterol (1.62oxidation in vitro is prolonged in ACI patients with vitamin E treatment, suggesting that vitamin E can increase the resistance of the patients against anti-oxidative stress.
4.Estimation of oxidative stress in patients with Parkinson's disease

Objective To measure the changes in urinary 8-iso-prostaglandin-F2α (8-iso-PGF2α) and oxidized low-density lipoprotein (ox-LDL) lag time in patients with Parkinson's disease (PD). Methods A case-control study was performed involving 31 male patients with PD (mean age of 59.7 years) diagnosed according to the Calne criteria and 31 age-matched healthy male subjects with comparable status of smoking and life style. For each subject, urinary 8-iso-PGF2α was measured quantitatively using enzyme-linked immunosorbent assay, and LDL oxidizability was measured by determining LDL oxidation lag time in conjugated diene product at 234 nm using Cu-stimulated oxidation. Results Compared to the levels in the control subjects, the PD patients showed significantly increased urinary 8-iso-PGF2α (81.1±1.6 vs 46.9±1.1 ng/mmol creatinine, P<0.05) and significantly reduced LDL oxidation lag time (63.5±6.0 vs 84.4±8.8 min, P<0.05). Conclusion Increased 8-iso-PGF2α and decreased anti-oxidant ability are implicated in the pathogenesis of PD, suggesting the value of appropriate antioxidant therapy in controlling the progression of PD.

7.Development and clinical application of the tractor for the fracture reduction.

8.Cell suspension culture and flavonoids production in Saussurea involucrate.

OBJECTIVETo establish a system of cell suspension culture for Saussurea involucrate.
METHODThe effects of different factors on cell growth and flavonoids production of S. involucrate were systematically studied including the media, initial pH values of the medium, carbon sources, inoculum quantity, and plant growth substance.
RESULTThe optimum medium was N6, initial pH values of the medium was 5.8, sucrose concentration was 50 g x L(-1), inoculum quantity was 60-80 g x L(-1) FW. Medium supplemented with BA (0.5 mg x L(-1)) and NAA (3 mg x L(-1)) was suitable for cell growth, but medium containing BA (0.2 mg x L(-1)) and NAA (2 mg x L(-1)) was suitable for flavonoids production.
CONCLUSIONCell growth and flavonoids production in the suspention of S. involucrate culture cell should be optimized.

9.Tissue culture and plantlet regeneration from embryo of Saussurea involucrate.

OBJECTIVETo establish a protocol of rapid clonal propagation of Saussurea involucrate by using embryo as explants.
METHODMS medium was used as basal medium and BA and NAA were supplemented to find out the optimal hormone combinations for adventitious buds initiation, adventitious bud multiplication and rooting.
RESULTAll embryo explants started to grow adventitious buds within 45 days when they were cultured on the media supplemented with 0.02-0.1 mg x L(-1) NAA and 2 mg x L(-1) BA; The adventitious buds multiplicated within 30 days when they were transferred to the media containing 0.1-0.2 mg x L(-1) NAA and 2-3 mg x L(-1) BA; 92% of the adventitious buds rooted well after they were planted on the MS medium containing macroelements at half strength and 0.4 mg x L(-1) NAA for 25 days. The regenerated plantlets grew well after they were transplanted and the survival rate was up to 70%.
CONCLUSIONPlantlets of S. invducrate regenerated high frequence through adventitious bud.

10.Antibody levels against diphtheria monitored in Jiading District, Shanghai

Objective To investigate the antibody levels against diphtheria among healthy population in Jiading District of Shanghai, and to make evaluation of the effect of diphtheria toxoid vaccination strategy. Methods Serum samples were collected from healthy population in Jiading District of Shanghai in 2010-2013.Antibody against diphtheria was tested by the enzyme-linked immunosorbent assay (ELISA).Geometric mean titer (GMT) of diphtheria antibody was compared between different population characteristics. Results A total of 1 029 serum samples were collected from the healthy population.With 1 003 of 1 029 diphtheria antibody titer of IgG (over 0.01 IU/mL), the positive rate was 97.47%, reaching the level of security protection (over 0.1 IU/mL) that accounted for 70.36%, and achieving the long-term protection level (over 1 IU/mL) that accounted for 35.57%;IgG GMT 0.325 IU/mL.There was significant difference in the positive rates and antibody levels between city residents and floating population in the field of diphtheria antibodies (
