Journal of Guangzhou University of Traditional Chinese Medicine 2017;34(5):729-733
Objective To observe the hypoglycemic, hypolipidemic and antioxidant effects of Zhuye Shigao Decoction (ZSD) on experimental type 2 diabetes mellitus rats. Methods Type 2 diabetes mellitus rat model was induced by intragastric administration of high fat diet(10 mL/kg) and intraperitoneal administration of streptozotocin (30 mg·kg-1). Sixty male Sprague-Dawley rats were randomized into normal group, model group, low-, moderate-and high-dose ZSD groups (in the dosage of 3.6, 7.2, 14.4 g·kg-1·d-1 respectively), and Metformin group (0.15 g·kg-1·d-1) , 10 rats in each group. With fasting blood glucose, urine glucose, glycosylated hemoglobin, total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol(LDL-C), serum insulin, serum superoxide dismutase(SOD) and malondialdehyde (MDA) as the main indexes, we evaluated the influence of ZSD on the blood glucose and lipid levels as well as oxidative stress in type 2 diabetes mellitus rats. Results Compared with the model group, the levels of blood glucose, urine glucose and glycosylated hemoglobin, and the contents of TC, TG, MDA were decreased (P <0.05), whereas SOD activity and serum insulin level were obviously increased (P < 0.05) in ZSD groups. ZSD showed dose-related effectiveness in the treatment of diabetic rats. Conclusion ZSD has certain hypoglycemic, hypolipidemic and antioxidant effects in treating type 2 diabetes mellitus.

Objective To analyze the risk factors for traumatic cerebral venous sinus occlusion (CVSO)and to investigate the strategies of early diagnosis of traumatic CVSO. Methods The clinical data of 212 patients with moderate to severe closed traumatic brain injury from January 2012 to December 2015 were analyzed retrospectively. Logistic regression analysis was used to evaluate the risk factors for traumatic CVSO. Results Of the 212 patients with traumatic brain injury, 16.5%(35/212) patients had CVSO. Ten patients had CVSO of thrombotic type (typeⅠ), 16 patients had CVSO of compression type (typeⅡ), and 9 patients had CVSO of mixed type (typeⅢ). Logistic regression analysis showed that skull fracture (OR = 8.141; 95%CI: 3.224-20.840) and epidural hematoma of crossing venous sinus (OR = 3.179; 95%CI: 1.470-7.037) were the risk factors for CVSO, and the former was more significantly correlated with CVSO. Female gender was the risk factor for CVSO typeⅠ(OR =10.425; 95%CI: 1.831-30.053), epidural hematoma of crossing venous sinus was the risk factor for CVSO typeⅡ(OR = 5.766; 95%CI: 1.885-18.197), and skull fracture, epidural hematoma of crossing venous sinus, and the previous history of vein thrombosis was the risk factors for CVSO type Ⅲ(OR =18.005, 4.596, 11.394; 95%CI: 2.021-58.836, 1.144-19.525, 1.436-46.558). Conclusions In the early diagnosis of traumatic CVSO, the crossing venous sinus fracture line and epidural hematoma should be given attention. Attention should be paid to the history of venous thrombosis. MR venography and CT venography contributes to early diagnosis of CVSO.

Objective To investigate the survival, differentiation and action of neural stem cell transplanted into striatum of Parkinson disease rat model. Methods Neural stem cells were isolated and cultured. The character of self-renewing was confirmed by technique of single cell clone culture. The ability of multi-directional differentiation and specific antigen-nestin was showed by immunocytochemistry(ICC). PD rat models were established, and the DAPI-labelled neural stem cells were transplanted into striatum. The 6-HHDA induced rotary behavior was measured, distribution and migration of the cell were checked by fluoresce activating. ICC and HPLC are used to see its differentiation and action of secrete neurotransmitter dopamine(DA) and it’s metabolites. Results The cultured stem cell express nestin and have the ability of self-renewing and multi-directional differentiation. 6-HHDA induced rotation decreased after transplantation of NSC into the PD rat striatum(P

Objective To explore the clinical manifestations and the characteristics of images of indocyanine green angiography (ICGA) and fundus fluorescein angiography(FFA) of multifocal choroiditis. Methods Eight patients (10 eyes) with multifocal choroiditis were gathered. The clinical manifestations and the images of ICGA and FFA were analyzed. Results Foci of multifocal choroiditis were found in posterior pole and peripheral areas of ocular fundi of all of the 10 eyes. The images of ICGA revealed hypofluorescence in focal area. The images of FFA showed hypofluorescence at the early phase and fluorescein leakage at the late phase in the active focus, and fluorescein staining and window defect fluorescence in the inactive focus. Conclusions The clinical manifestations of multifocal choroiditis varied with disease course, location and numbers of the lesions. ICGA and FFA can show the development of the disease clearly, which may guide the treatment.

To investigate the changes in the activated T-lymphocyte CD3/HLA-DR and CD3/CD(16+56) populations in peripheral blood of the patients with allogeneic hand transplantation, lymphocytes from peripheral blood of the patients at different time points were immunologically labeled with dual color fluoresecent monoclonal antibodies CD3/CD(16+56) and CD3/HLA-DR, mono-color fluoresecent monoclonal antibody CD25. CD25, CD3/CD(16+56), and CD3/HLA-DR were determined with flow cytometry (FCM). The levels of activated T-lymphocyte (CD25 +,CD3 +/HLA-DR + ), silent T-lymphocyte [CD3 +/CD(16+56) -,CD3 +/HLA-DA - ] decreased significantly during the first week after transplantation and then increased gradually to the pre-operafive level. Nature killer cells [CD3 -/CD(16+56) +] increased significantly at the first day after transplantation, then decreased sharply and maintained a lower level. The results suggest that immunosuppressive agents have significantly effects on lymphocyte subsets in allogenaic hand transplanted patients, and dynamic determination of HLA-DR, CD3 /CD(16+56) could be valuable in immunomonitoring after allogeneichand transplantation.

Medical Journal of Chinese People's Liberation Army 2001;0(07):-
To design and manufacture a new bone matrix material (NBM) composed of both organic and inorganic materials for bone tissue engineering, the osteogenic potential of NBM combined with osteoblasts was studied in vitro and in vivo. Osteoblasts from bone marrow stromal cells of New Zealand rabbit were cultured with NBM in vitro , then the materials combined with osteoblasts were implanted into the skeletal muscles of rabbits. The osteogenic potential of NBM was evaluated using contrast microscope, scan electromicroscope and histological examination. Osteoblasts could attach and proliferate well in the NBM, secrecting lots of extracellular matrix, while in vivo experiment of the NBM in osteoblasts group showed that a large number of lymphacytes and phagocytes invaded into the inner of the material in the rabbit skeletal muscle beds after 4 weeks of implantation, no new bone formation was observed after 8 weeks. The different osteogenic potential expressed by NBM between in vitro and in vivo may be due to the immunogenity of NBM which causes cellular immunoreaction to destroy the osteogenic environment. More attention should be paid to the immunoreaction problem in tissue engineering between the host and organic inorganic composite materials.
