This paper discusses the mechanism of cellobiose in fungal cellulase induction a nd repression, and its inhibition of cellulases hydrolytic activity. Depending on the research result of cellulose binding domain, our hypothesis is that the main function of Exo-1,4-?-glucanase is to destroy th e crystal structure of cellulose to facilitaty hydrolyzing of ?-1,4 glucosidic bonds. A new strategy for the efficient transformation of cellulose material is advanced at t he end.
2.Study on the Optimal Fermentation Process for Production Chitinase of Streptomyces sp. A048

Streptomyces sp. A048 was cultured in a complete medium to the last stage of log phase,the hyphae were washed and collected by centrifugation. Then the hyphae were inoculated in liquid medium for chitinase production using two-step fermentation. Activity of chitinase produced by two-step fermentation was 1.1 times higher than that from one-step fermentation,and ferment cycle was for 54 hours,which was 66 hours shorter than that of one-step fermentation. The hyphae and the powder of chitin were co-immobilizated and cultured in liquid medium for 36 hours,activity of chitinase was 1.8 times higher than that from one-step fermentation,and ferment cycle was 54h shorter than that of one-step fermentation. By adding 0.4% cellulose to two-step fermentation,activity of chitinase was 18.52 U/mL that was 4 times higher than that from the control and 10 times higher than that from one-step fermentation. Two step fermentation with chitin and cellulose may be the optimal fermentation process to produce Chitinase from Streptomyces sp. A048.
3.Study of intraocular pressure diurnal variation curves in patients with open-angle glaucoma and normal subjects

Objective To observe the diurnal variation of intraocular pressure(IOP)measured with Goldmann applanation tonomoter(GAT)in patients with primary open angle glaucoma(POAG),normal tension glaucoma(NTG)and healthy controls,and to compare the differences of diurnal variation curves between two eyes of each subject and define the distribution of the peak of IOP. Design Prospective case series.Participants POAG patients,NTG patients and healthy controls.Methods The diurnal variation of IOP in the two eyes of each of 30 POAG patients,30 NTG patients and 30 normal controls were investigated and the distributions of the peak IOP were observed.Main Outcome Measure Intraocular pressure.Results Measured with GAT,6.7% right eyes and 10.0% left eyes of normal people,20.0% right eyes and 23.3% left eyes of NTG patients,and 23.3% right eyes and 20.0% left eyes of POAG patients showed peak IOP at the time points of out-office period.Conclusions POAG patients,NTG patients and normal controls have asymmetric IOP diurnal variation.The two eyes of one individual cannot always be treated as the same.The IOP peak can occur at the time point during the out-office time.Measure IOP in office-time only may miss the dynamic information of IOP.
4.Identification of peucedani radix, peucedani decursivi radix and its adulterants using ITS2 sequence.

In order to identify Peucedani Radix, Peucedani Decursivi Radix and their adulterants, the internal transcribed spacer 2 (ITS2) regions of Peucedani Radix, Peucedani Decursivi Radix and their adulterants were amplified and bidirectionally sequenced based on the Principles for Molecular Identification of Traditional Chinese Materia Medica Using DNA Barcoding, which has been promulgated by Chinese Pharmacopoeia Commission. Sequences were analyzed and assembled by Codon Code Aligner V3. 7.1. The relevant data were analyzed by MEGA 5. 0. Species identification analyses were performed by using the nearest distance methods and neighbor-joining (NJ) methods. The result showed that the ITS2 sequence lengths of Peucedani Radix were 229-230 bp and the average intra-specific genetic distances were 0.005. The ITS2 sequence lengths of Peucedani Decursivi Radix were 227 bp and the sequences contained no variation site. The average inter-specific K2P genetic distance of Peucedani Radix, Peucedani Decursivi Radix and their adulterants species were 0.044 and 0.065 respectively. The minimum inter-specific divergence is larger than the maximum intra-specific divergence of Peucedani Decursivi Radix. The nearest distance methods and NJ trees results indicated that Peucedani Radix, Peucedani Decursivi Radix and their adulterants species could be identification clearly. The ITS2 regions can stably and accurately distinguish Peucedani Radix, Peucedani Decursivi Radix and their adulterants.

6.Study on novel colon position pulsatile capsule and its release in vitro.

OBJECTIVETo develop a colon position pulsatile capsule drug delivery system of which the lag-time is controlled by a plug tablet mainly made of konjac glucomannan and evaluate its release profile in vitro.
METHODImpermeable capsule body was prepared by filling method, the plug tablet was pressed by direct compression, and the rapid-disintegrable drug tablets was made by wet granulation. The pulsatile capsules were prepared by putting the drug tablet into the impermeable body and sealed it with the plug tablet The factors affecting the lag-time were investigated by dissolution testing.
RESULTThe formulation of the rapid-disintegrable drug tablets influenced the pulsatile release of the drug, the composition of the plug tablet significantly influenced the lag-time; the lag-time was extended with the higher viscosity of HPMC and the increased proportion of konjac glucomannan.
CONCLUSIONThe pulsatile capsule with a suitable lag-time and colon position characteristics can be achieved by adjusting the composition of the plug tablet.

7.Clinical efficacy and safety of losartan potassium in the treatment of aged patients with essential hypertension and hyperuricemia

Objective To evaluate the clinical efficacy and safety of domestic produced losartan potassium in aged patients with essential hypertension and hyperuricemia.Methods Eighty patients diagnosed with hypertension with hyperuricemia were randomly divided into treat-ment group ( n=40 ) and control group ( n=40 ).Patients in treatment group were given losartan potassium 50 mg· d -1 , and patients in control group given candesartan 4 mg · d-1.The treatment lasted for 8 weeks.Clinical efficiency , serum uric acid levels and blood pressure of the two groups were measured after treatment.Results After treatment , the total effective rate was equal 95.0%in treatment group and control group and the markedly effective rate were 55.0% and 57.5% respectively (P>0.05).After treatment, the reduction of systolic blood pressure ( SBP ) and diastolic blood pressure ( DBP ) were ( 19.65 ±12.23 )/( 11.80 ±8.21 ) mmHg vs ( 24.78 ±15.38 )/( 13.93 ±10.60 ) mmHg in treatment group and control group ( P<0.05 ) , there was no significant difference between the two groups ( P>0.05 ).After treatment , the level of uric acid decreased apparently in treatment group was (435.04 ±53.57 )μmol L-1 , and (483.68 ±63.50 ) μmol· L-1 in control group and treatment group was significantly lower than control group and before treatment ( P<0.05 ).There was no adverse reaction in treatment group , 1 case in control group ( 2.5%) .There was no significant change in heart rate in two groups before and after treatment.Conclusion Domes-tic produced losartan potassium was proved to be a stable and effective antihypertensive medication with little adverse drug reactions.
8.Changeable expressions of glutamate neurotransmitter and NR2B in neuroanatomical circuit of ventral temental area, nucleus accumbens and prefrontal cortex in morphine-psychic dependent rats

Objective To explore the mechanism of opioid-psychic dependence involving the aspects of pre-receptor and receptor by observing the changeable expressions of glutamate neurotransmitter and NR2B of N-methyl-D-aspartic acid (NMDA) receptor in the neuroanatomical circuit of ventral temental area, nucleus accumbens and prefrontal cortex (VTA-Nac-PFC) of rats subjected to morphine-induced conditioned place preference (CPP). Methods The models of CPP were validated by escalating doses of morphine in rats (n=16). The colorimetry and immunohistochemistry ways were applied to detect the contents of glutamic acid and the expression level of NR2B in VTA, Nac and PFC. Results As compared with those in the control group physiological saline), the prolonged detention time of white compartment in the model group was notably observed (P<0.05), and increased content of glutamic acid and expression level of NR2B in fields of VTA, Nac and PFC in the model group were significantly detected (P<0.05). Conclusion Increased level of giutamic acid and expression level of NR2B in nuroanatomieal circuit of VTA, Nac and PFC could play key roles in inducing morphine-psychic dependent rats.

10.Study on the characteristics of major mosquito-borne infectious diseases in Three Gorges Reservoir Area from 1997 to 2008

Objective Characteristics and tendency of mosquito and major mosquito-borne diseases (including epidemic encephalitis B and malaria) were analyzed in the Three Gorges Reservoir Area from 1997 to 2008. Methods Information on surveillance data related to mosquito-borne infectious diseases and on mosquito density and category, from 1997 to 2008 was collected from Health Surveillance System in Three Gorges Reservoir Area. Data regarding meteorological factors and construction of Three Gorges Reservoir was also collected. Pearson and Poisson models were used. Results From 1997 to 2008, in the Three Gorges Reservoir Area, the incidence rates of epidemic encephalitis B and malaria were decreasing. Positive correlations were shown between indoor and outdoor mosquito density as well with temperature with coefficient as 0.281 and 0.355 respectively. Correlations of mosquito-borne diseases with indoor and outdoor mosquito density were positive, with correlation coefficient as 0.340 and 0.328 respectively. Conclusion There seemed lack of evidence to prove that negative influences had occurred on the incidence of mosquito-borne infectious diseases or the mosquito density after the Three Gorges Reservoir water storage came into being in 2003, however, long-time surveillance program needs to be carried out to gather information on this issue.
