1.Effect comparison of statins on myocardial injure and inflammatory factors in patients with unstable angina pectoris undergoing percutaneous coronary intervention

Objective To compare the effect of atorvastatin and rosuvastatin on myocardial injure and inflammatory factors in patients with unstable angina pectoris(UAP)undergoing percutaneous coronary intervention(PCI).Methods Ninety six UAP pa-tients undergoing PCI were enrolled in the study and divided into two groups,patients were given atorvastatin(20 mg/d)or rosuv-astatin(20 mg/d)besides conventional treatment for 1 week before PCI.Datas were collected before medication,PCI and Twenty-four hours after PCI,including CK-MB,cTnI,hs-CRP,TNF-α,IL-6 and IL-10.Results Twenty-four hours after surgery,the PCI, CK-MB,cTnI,hs-CRP,TNF-α,IL-6 of rosuvastatin group were lower than those of atorvastatin group(P <0.05),but IL-10 of ro-suvastatin group was higher than that of atorvastatin group(P <0.05).Conclusion Rosuvastatin (20 mg/d)is better than atorvas-tatin (20 mg/d)in the efficacy of UAP patients undergoing PCI,which can reduce the level of myocardial injure and inflammatory factors effectively.
2.Experimental Study of the Effect of Fire Needling on Pain Threshold, and IL-1 and TNF-α in Rheumatoid Arthritis Rats

Objective To compare the analgesic effects of fire needling, conventional acupuncture and acupoint catgut embedding in rats with rheumatoid arthritis.Methods Fifty healthy male SD rats were randomized into control, model, acupuncture, fire needling and catgut embedding groups, 10 rats each. A SD rat model of rheumatoid arthritis was made by intracutaneous injection of complete Freund’s adjuvant into the right hind foot pad. Right Kunlun and Zusanli were selected as therapeutic points in the three groups and given fire needling, acupuncture or catgut embedding. The right paw mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) were used as observation indices and measured in every group of rats at one day before and four days after model making and at one, seven and fourteen days after the beginning of treatment. Rat paw inflammatory tissues were separated after the completion of treatment. The IL-1 and TNF-α contents of them were measured. Results There were statistically significant differences in MWT and TWL between the model, acupuncture, fire needling or catgut embedding group and the control group after model making, and one, seven and fourteen days of treatment (P<0.01). There were statistically significant differences in MWT and TWL between the fire needling group and the model, acupuncture or catgut embedding group after one, seven and fourteen days of treatment (P<0.01,P<0.05). There was a statistically significant difference in MWT between the acupuncture or catgut embedding group and the model group after one, seven and fourteen days of treatment (P<0.01). There was a statistically significant difference in TWL between the acupuncture or catgut embedding group and the model group after seven and fourteen days of treatment (P<0.01,P<0.05). There were statistically significant differences in IL-1 and TNF-α contents between the model, acupuncture, catgut embedding or fire needling group and the control group after model making (P<0.01). after treatment, there were statistically significant differences in IL-1 and TNF-α contents between the acupuncture, catgut embedding or fire needling group and the model group and between the acupuncture or catgut embedding group and the fire needling group (P<0.05).Conclusions Fire needling can raise pain threshold in rats with rheumatoid arthritis. It alleviates rheumatoid arthritis-induced chronic pain by reducing the inflammatory factor content of affected muscular tissues.
3.Analysis of risk factors and prognosis on the post-resuscitation multiple organ dysfunction syndrome after successful cardiopulmonary resuscitation

Objective To investigate the risk factors and prognosis on the post-resuscitation multiple organ dysfunction syndrome(MODS) after successful cardiopulmonary resuscitation(CPR).Methods Clinical data of 53 patients who were suffered with cardiac arrest(CA) and undergone successful CPR with return of spontaneous circulation(ROSC) were analyzed. Results There were 48 patients accompanied with MODS(90.6%),and among them,35 patients died in hospital(66.0%).All the 10 patients with CPR interval ≥6min were accompanied with MODS and they died in hospital.There were 43 patients who underwent CA immediately and of them,38 patients were accompanied with MODS.The incidence and mortality of MODS in the patients with CA-ROSC interval 0.05).Conclusion The risk factors such as ROSC interval ≥6 min,AC-ROSC interval ≥10min and the SIRS after ROSC are significantly associated with the incidence of MODS.The organic function of the patients should be evaluated promptly.
4.THE NUTRITIONAL VALUE OF A FLOUR MILLING BYPRODUCT VITAMIN-MINERAL ENRICHED FLOUR ON RATS

The vitamin-mineral enriched flour (VMF) was a byproduct obtained from a new processing technique of flour milling, and rich in vitamin B group, minerals and dietary fibers.The addition of 10 or 20% VMF to the stock diet composed mainly of white flour failed to improve PER and growth rate of young rats. When the rats were fed the tested diet for 6 weeks, in which the VMF served as the only diet source ot riboflavin, blood glutathione reductase activity coefficient was 1.13 and 1.09 respectively. This indicated that the amount of riboflavin supplied by VMF could maintain an adequate level in the body of the rats.Adult rats were fed high cholesterol plus 10, 20 and 30% VMF, the contents of serum cholesterol, liver lipids and liver cholesterol were similar to those of the normal diet group, these contents, however, in control group were increased obviously. The results suggested that VMF could protect the rats from the increment of serum cholesterol, liver lipids and cholesterol.
5.The preliminary efficacy of interferon-alpha and ribavirin combination treatment of chronic hepatitis C in HIV-infected patients.

BACKGROUNDIt is internationally accepted that in drug-naïve individuals with hepatitis C virus (HCV) and human immunodeficiency virus (HIV) co-infection, chronic hepatitis C should be treated first if the CD4 cell count does not require the initiation of anti-retroviral therapy. Present paper evaluated the clinical effect and side-effect of interferon-alpha (IFN-alpha) and ribavirin (RBV) combination therapy for Chinese patients with HCV-HIV co-infection, and compared with them for HIV infection alone.
METHODSTen patients with HCV-HIV and 17 patients with HCV received 5 million unit IFNalpha-2b every other day intramuscularly, and 300 mg RBV triple daily by oral. Dynamic observations were made for HCV RNA and HIV RNA loads, CD4+ and CD8+ T lymphocyte counts, liver function and blood cell measurement, and the medicine side-effects.
RESULTSAfter 12-week and 24-week treatments of IFN-alpha and RBV combination therapy, mean HCV RNA levels reduced 1.14 logs and 1.56 logs from the baseline at week 0 in HCV-HIV co-infection, and reduced 1.48 logs and 1.75 logs in HCV infection, respectively. The HIV RNA levels decreased 1.22 logs and 1.32 logs from the base line; however, there were no obvious different changes at T lymphocyte counts of HCV-HIV and HCV patients through 24-week treatments. Whole 27 patients showed satisfactory biochemical response to therapy. There were some mild or mediate influence-like symptoms, intestinal uncomfortable and depressed blood cell counts in early stage of the treatments. No neuropsychiatric and auto-immune disorders were found.
CONCLUSIONSIFN-alpha and RBV combination therapy had similar anti-HCV effects during 24-week treatment for HCV-HIV and HCV infected Chinese patients, and some anti-HIV effect. There were no obvious different biochemical responses and side-effects between two groups above.

Meanwhile, the HIV RNA levels decreased 1.22 log (t = 3.662, P < 0.01) and 1.73 log (t = 6.119, P < 0.01) from the base line. However, there were no obvious different changes among T lymphocyte counts of HCV-HIV and HCV patients at week 0, week 12 and week 24. All 27 patients showed satisfactory biochemical response to therapy. There were some mild or moderate influenza-like symptoms, intestinal discomfort and decreased blood cell counts in the early stages of the treatments. No neuropsychic and auto-immune disorders were found.
CONCLUSIONSIFNalpha-2b and RBV combination therapy showed similar anti-HCV effects during the 24 week treatment for HCV-HIV and HCV infected patients, and some anti-HIV effect was also observed. No obvious different biochemical responses and side-effects were found between the above two groups.

7.Tension-force induced cyclooxygenase-2 expression mediated by microfilament in human periodontal ligament fibroblast.

OBJECTIVETo study the role of microfilament polymerization in menchanotransduction by human periodontal ligament fibroblast (hPDLFs).
METHODSIn tension-force group, hPDLFs were treated by tension-force values of 18% for 8 h, 16 h, 24 h. In tension-force and inhibitor group, the sample was treated with 5 microg/mL cytochalasin B before using tension-forece. Each sample was collected and the expression of cyclooxygenase-2 was measured by using immunohistoche staining.
RESULTSIn tension-force group, the expression of cyclooxygenase-2 enhanced with the extension of loading time. In tension-force and inhibitor group, cyclooxygenase-2 expression was depressed and had no relation with loading time.
CONCLUSIONTension-force induced cyclooxygenase-2 expression is mediated by microfilament, disruption of the microfilament polymerization will destroy mechanotransduction in hPDLFs.

9.Effect of a desensitizing polishing paste on bonding strength of two self-etching adhesives to dentin.

OBJECTIVETo evaluate the effect of an 8% arginine-CaCO3 containing desensitizing polishing paste on bonding strength of two self-etching adhesives to dentin.
METHODSThirty-six intact human premolars extracted for orthodontic reasons were collected within 1 month after extraction and randomly assigned into three groups using a table of random numbers (n = 12): specimens without any treatment served as control. In the polishing powder group specimens were polished with a slurry of pumice, and in the desensitizing polishing paste group dentin surfaces of the sample teeth were treated with 8% arginine-CaCO3 containing desensitizing polishing paste. Then each group was divided into two sub-groups using a table of random numbers in order to evaluate the bonding strength of two self-etching adhesive agents (G-Bond, GC; Fl-Bond II, Shofu). Microtensile bond strength test was conducted immediately and after 5000 thermocycling (n = 15). Scanning electron microscope (SEM) was used to evaluate the occluding effect of the desensitizing polishing paste.
RESULTSIn the pre-thermocycling stage, there were no significant differences in Fl-Bond II bonding strength among the three groups [control: (30.34 ± 5.42) MPa, polishing powder group: (29.72 ± 5.16) MPa, desensitizing polishing paste group: (31.53 ± 4.86) MPa] (P > 0.05). However there were significant differences among the three groups in G-Bond bonding strength [control: (38.19 ± 4.42) MPa, polishing powder group: (36.47 ± 4.72) MPa, desensitizing polishing paste group: (46.88 ± 7.83) MPa] (P < 0.05). After thermocycling process, there were no significant differences in bonding strength among the three groups in both G-Bond groups and Fl-Bond II groups. SEM observation showed that the desensitizing polishing paste could occlude open dentinal tubules effectively, and the application of self-etching adhesives could re-open the dentinal tubular orifices. An even layer can be seen on the dentin surface treated with self-etching adhesive containing functional monomers.
CONCLUSIONSThe 8% arginine-CaCO3 containing desensitizing polishing paste could effectively occlude dentinal tubules, thus may have potential benefits in preventing post-operative sensitivity. Additionally, it had no adverse effect on bonding strength of self-etching adhesives to dentin.

10.Expression, purification and activity analysis of anti-human transferrin receptor scFv.

Human transferrin receptor (TfR) was isolated from homogenates of placental tissues by affinity chromatography on transferrin-Sepharose, and then used to screen human scFv against it from a fully-synthesized phage scFv library. After verifying the specificity, gene fragment of one of the selected scFv was inserted into the plasmid pET22b(+) and transformed into E. coli BL21(DE3) . Expression of scFv in transformant was induced with 0.5mmol/L IPTG. ELISA assay on HeLa cells showed that scFv protein could recognize and bind to TfR on the surface of HeLa cells. The scFv was purified by one-step affinity chromatography with Ni+ -NTA agarose, and injected into Kunming mouse via tail veins. This scFv was detected in brain tissues 1h later by capillary depletion method, which indicates that scFv protein can permeate through the blood brain barrier by mediation of the TfR receptor. Our works lay the foundation for the treatment of tumors and central nervous system diseases.
