2.Prefabrication of vascularized grafts based on pre-differentiated adipose derived stem cells, fibrin sealant and porous calcium phosphate cement scaffold

BACKGROUND:Construction of vascularized bone substitutes to mimic free vascularized fibular grafting in treating large bone defects stil remains chal enges to researchers during the past years.
OBJECTIVE:To design and construct a new vascularized tissue-engineered bone graft using pre-differentiated adipose derived stem cells, fibrin sealant and porous calcium phosphate cement scaffold.
METHODS:Adipose derived stem cells isolated from rats were directly differentiated to endothelial cells and then incorporated in fibrin sealant and porous calcium phosphate cement scaffolds in vitro. Subsequently, the different composites of the three groups including vascularized tissue-engineered bone scaffold (group A), fibrin sealant and porous calcium phosphate cement scaffold (group B) and porous calcium phosphate cement scaffold (group C) were directly embedded into the quadriceps of the rats. Histological quantitative analysis and western blot assay were conducted 2 and 4 weeks after implantation.
RESULTS AND CONCLUSION:The pre-differentiated adipose derived stem cells were demonstrated in good condition after 7 days co-culturing with the fibrin sealant and porous calcium phosphate cement scaffold. The results of in vivo experiments showed that the scaffolds were in-grown together with fibrous connective tissues and blood vessels. Newly formed vessels and immature capil aries were observed in the group A. The vessel density, vessel diameter and vascular endothelial growth factor C expression in the group A were significantly higher than those in the groups B and C. Our findings demonstrated that compared with simply fibrin sealant and porous calcium phosphate cement scaffold, the combination of pre-differentiated adipose derived stem cells (endothelial differentiation) and fibrin sealant can achieve rapid angiogenesis of porous calcium phosphate cement scaffold.
3.The effects of Ortho-K contact lens by day and overnight wear on wave-front aberrations

Objective To investigate the change of wave-front aberrations after Ortho-K contact lens(CL)by day and overnight wear.Design Retrospective case series.Participant 180 eyes of 92 myopic patients wearing Ortho-K CL.Method Wave-front aberra- tions of 180 eyes of 92 myopic patients wearing Ortho-K CL were analyzed.Patients were divided into by day wear and overnight wear groups.Wave-front aberrations were measured with WFA 1000B subjective aberrometer before and 1 month after Ortho-K CL wear. Wave-front aberrations at initial,after one month of CL fitting and without CL were measured in same condition,and RMS values for overall wave-front aberrations and each order of the Zernike aberrations were analyzed with Matlab software.Main Outcome Measures RMS value of Zernike coefficient of each order.Result Compared with initial aberration in wear by day group,total RMS and each or- der aberrations(except 2nd order)with lens fitting significantly increased(P0.05).Compared with initial aberration in overnight wear group,overall wavefront aberrations and the RMS(except 2nd,3rd and 6th order Zernike aberrations)with lens fitting significantly increased(P0.05).Conclusion The high order aberrations with lens fitting and without lens fit- ting increased in each group.The aberration with lens fitting in wear by day group was higher than that in overnight wear group,while the aberration without lens fitting in overnight wear group was higher than that in wear by day group.(Ophthalmol CHN,2007,16: 351-354)

Objective To investigate the effects of the gene expressions of FAS、FASL、FADD and caspase-8 in esophageal carsinogenesis. Methods Immunohistochemical method was applied to detect the expression of FAS、FASL、FADD and caspase-8 proteins in esophageal epithelium. In situ hybridization method was applied to detect the expression of FADD and caspase-8 mRNA in esophageal epithelium. Results The positive rates of FAS, FADD and caspase-8 proteins and FADD、caspase-8 mRNA were decreased from normal epithelium to dysplasia and carcinoma tissues gradually. OBJECTIVETo summarize the recent findings of dysregulation of signaling pathways and miRNAs in chronic lymphocytic leukemia (CLL).
DATA SOURCESWe searched PubMed database with the keywords "chronic lymphocytic leukemia", "signal pathway", or "miRNA" for relevant articles in recent years.
STUDY SELECTIONResearch articles and reviews about signaling pathways and miRNAs in CLL were chosen for review.
RESULTSDysregulation of signaling pathways, such as B cell receptor , toll-like receptor, PI3K, nuclear factor κB, notch signaling pathway, Wnt/Fzd signaling pathway, and Hedgehog and Janus kinases/signal transducers and activators of transcription signaling pathway, as the terminal events of the aberrant gene expression and the pro-survival effects of microenvironment, plays a crucial role in the process of CLL. miRNAs, a novel found noncoding RNA, which regulate gene expression at transcription or post-transcription level and correlate with pathogenesis of CLL provide us new avenues to better evaluating prognosis and therapy of it.
CONCLUSIONFurther investigation of the dysregulation of signaling pathways and miRNAs and their relationship may provide us a new prospective to understand the pathogenesis of CLL and may provide us new strategies to resolve the clinical nodi in treatment of CLL.

8.Preliminary study on the possible correlation between anxiety and proteomic fingerprint

Objective A preliminary study on a new discovered proteomic fingerprint correlated with anxiety disorders, which M/Z range from 15000 to 16800. Methods 221 cases of neoplasm patients from April of 2004 to now were divided into anxious group and non-anxious group according to their scores using self-rating anxiety scale (SAS), and all the patients' sera were detected utilizing SELDI-TOF-MS. The data were analyzed with supported softwares. Results Compared the mass spectra of anxious group(n=49) and non-anxious group (n=172), 2 clusters were captured which M/Z range from 15 000 to 15 380 and 15 700 to 16 800 respectively. The coincidence with SAS was 75.51%. The M/Z range from 15 700 to 16 800 was the most frequent, then the second range from 15 000 to 15 380 and 15 700 to 16 800 which was doublet cluster, the least range from 15 000 to 15 380. The same fingerprints were captured in non-anxious group and their intensity was different. Conclusion The M/Z range from 15 000 to 16 800 was protein fingerprint correlated with anxiety disorders.
9.Analysis of 262 Cases of Seizures Detected with Video-Electroencephalogram

Objective To evaluate the clinical significance of video-electroencephalogram(VEEG) in the diagnosis of paroxysmal events in children.Methods The VEEG and routine electroencephalogram(EEG) were used to inspect 262 cases of seizures respectively collected of outpatient clinic and admission department from Apr.2006 to Dec.2007,and VEEG monitoring results were analyzed and compared with primary diagnoses retrospectively.According to the characteristics of EEG and clinical observation to decide whether it be epilepsy or not and compare the difference between the results of EEG and VEEG to evaluate the application value of the 2 examination methods in children with NES.SPSS 13.0 software was used to analyze the data.Results Of all 262 cases,113 cases of clinical outbreak were recorded,meanwhile the diagnose of 4 patients of epileptic seizures(ES) group were changed to non-epileptic seizures(NES) and 3 patients of NES were rediagnosed to ES,69 cases couldn′t be confirmed according to VEEG or EEG and there was no outbreak recorded in the other 149 cases.In the period onset,79 children′s characteristics of EEG were captured which included 76 children in ES group,and 3 children in NES group.Thirty-six children′s characteristics of EEG were not captured which included 4 children in ES group and 30 children in NES group.The differences of the ratio of total detect and abnormity between EEG and VEEG were significant,improving the diagniosis rate and the control rate of symptom.VEEG had a clinical significance in differential diagnosis of ES and NES.Conclusions Compared with EEG,VEEG will be more helpful in diagnosing and differentiating seizures diseases,improving the diagnosis rate and the control rate of symptom.VEEG has a clinical in diffe-rential diagnosis for ES and NES.
10.The relationship between thyroid dysfunction and pregnancy outcomes in pregnant women

Objective The purpose of this research is to calculate the prevalence of thyroid dysfunction during pregnancy and to discuss the relationship between gestational thyroid diseases and pregnancy outcomes.Methods There were 3 745 pregnant women who took antenatal care at Sun Yat-sen Memorial Hospital,Sun Yat-sen University were chosen as the research objects during March 2012 to February 2014.All of them were given close monitoring and tracking till the termination of pregnancy.These subjects took thyroid function testing (TT4,FT4,TSH,thyroid peroxidase antibody),if the result was abnormal,they were followed up periodically.Results The prevalence of overt hyperthyroidism is 1.90%,1.20% for subclinical hyperthyroidism,1.17% for hypothyroidism,6.78% for subclinical hypothyroidism,and 3.82% for isolated hypothyroxinemia.Compared with normal thyroid function group,the morbidity of placental abruption increased in overt hyperthyroidism group (5.8% vs 1.0%,P< 0.01) ; the incidence of gestational diabetes mellitus was increased in subclinical hyperthyroidism group (37.5 % vs 16.6%,P<0.01) ; and the morbidity of preterm birth increased in subclinical hypothyroidism group (14.3% vs 7.7%,P =0.002).Conclusion Gestational thyroid diseases may increase the incidence of adverse pregnancy outcomes ; such as placental abruption,gestational diabetes mellitus,and premature birth.
