1.Research of three-dimensional localization for sentinel lymph node of breast cancer

Objective To investigate three-dimensional localization for sentinel lymph node (SLN) of breast cancer,and by which we can remove the SLNs directly.Methods The ipsilateral axillary lymph nodes of 40 patients were inspected by B-ultrasound and axillary artery and subscapular artery bifurcation point and its trend with Doppler B-ultrasound in the preoperation,then located them in the surface.We found SLNs using methylene blue as the mapping agent with endoscope during the operation,determined which lymphatic group the sentinel lymph node belonged and the spatial location and the surface projection according to the anatomical location.Results We found the three-dimensional location of SLNs in the group of 39 in 40 patients with endoscope,of which 34 cases located in central group,accounting for 87.18% ;while 4 cases located in the subscapular group,accounting for 10.26%,and their spatial location was as follows:set the root of subscapular artery in this location as a starting point,the subscapular artery as a diameter,and made a diameter of 5cm circle to the bottom,then let the latissimus dorsi as the end,and made a quasi-cylinder through the circle to the axillary central.The height of the quasi-cylinder got up to the surface of the intercostal brachial nerve.Then set the nerve as the diameter of circle of quasi-cylinder,and the centre of circle was crosspoint of subscapnlar artery's surface projection with intercostal brachial nerve.The height of quasi-cylinder varies with somatotypes of the patients,its height was less than or equal to 5cm.What's more,the fiften enlarged lymph nodes located by B-ultrasound in the preoperation were all in the quasi-cylinder,and they were SLNs.Conclusion SLN lies in quasi-cylinder consisting of spatial location of subscapular group and central group lymph nodes.If the enlarged lymph nodes found by B-ultrasound are in above mentioned quasi-cylinder,they can be considered as the SLNs.Make a 5cm-incision parallelling the intercostal brachial nerve and intersecting the surface projection of subscapular artery in the surface of quasi-cylinder,then dissect toward the origin of the subscapular artery,you can find SLNs.
2.Reliability and validity of 3D CT imaging in preoperative evaluation of tibial plateau fractures

Objective To compare the diagnostic values of 3D CT and 2D CT imaging for tibial plateau fractures. Methods A retrospective study was performed to evaluate 21 tibial plateau fractures. Four experienced orthopaedists were asked To make assessmem of fracture classifications and characteristics using 3D CT and 2D CT images.Agreement test Was performed to evaluate interobserver and intraobserver reliability for fracture classification and criteria validity for fracture characteristics(intraoperative findings were taken as the golden standard).Furthermore,MeNemar test was used to compare the agreement of 3D CT results and 2D CT results respectively with the golden standard for fracture characteristics.Results When 3D CT images were used.interobserver reliability and intraobserver reliability for fracture elassiftcation increased to"almost perfect agreement".The agreement of golden standard with 3D CT results for fracture characteristics Was significantly higher than that with 2D CT results. Condmiou Since 3D CT imaging can improve the reliability and accuracy of preoperative assessment oftibial plateau fractures,it is helpful and valuable for tibial plateau fractures.
3.Protective Effects of Salvia Yunnanensis Extract on Hypoxia/Reoxygenation Injury in Cultured Rat H9c2 Cardiomyocytes

Objective: To study the protective effects and the mechanism of Salvia yunnanensis extract on hypoxia/reoxygenation injury in cultured rat H9c2 cardiomyocytes.Methods: The hypoxia/reoxygen (H/R) injury model was established in H9c2 cell strain with or without the extract of Salvia yunnanensis.The cultured H9c2 cardiomyocytes were randomly divided into 6 groups: the normal control (C) group, H/R group, H/R+verapamil (H/R+V) group, H/R+Salvia yunnanensis extract at low dose (H/R+L, 0.01 mg·L-1) group, medium dose (H/R+M, 0.1 mg·L-1) group and high dose (H/R+H, 1.0 mg·L-1) group.The cell viability was measured by MTT assay and the activity of malondialdehyde (MDA) and lactate dehydrogenase (LDH) was measured by a detection kit.Fluorescence absorbance (A) value was measured by a fluoroscopy to show the intracellular reactive oxygen species (ROS) levels.Results: Compared with that in the model group, the survival rate of myocardial cells was significantly higher in Salvia yunnanensis extract at low, medium and high dose groups (P<0.05 or P<0.01), and the intracellular LDH leakage (P<0.05 or P<0.01), the content of MDA in cytoplasm (P<0.01) and the intracellular ROS levels significantly decreased in Salvia yunnanensis extract at high dose group (P<0.05).Conclusion: The extract of Salvia yunnanensis has protective effect on hypoxia/reoxygenation injury in cultured rat H9c2 cardiomyocytes, and the mechanism may be related to the reduction of lipid peroxides and removal of cell oxygen free radicals.
4.Interaction of butylphthalide with rat and human liver CYP450 isoenzymes.

The work aims to study the drug metabolizing enzymes involved in the metabolism of butylphthalide and evaluate the induction and inhibition activities of butylphthalide on CYP450 isoenzymes by using in vitro (liver microsome incubation system of rats and human) and in vivo (CYP induced model of rats) method. Butylphthalide was incubated with selective inhibitors of CYP450, and its metabolic rate was determined to identify the metabolizing isoenzymes of NBP in rat (normal and induced rats) and human liver microsomes. The in vitro inhibition effect of butylphthalide on 6 main liver microsomal CYP450 isoenzymes was evaluated by using probe drugs; the induction and inhibition activities in vivo of butylphthalide on CYP450 isoenzymes were evaluated by NBP ig dosing (160 mg x kg(-1)) and iv dosing (20 mg x kg(-1)) in rats. After adding the specific inhibitors of CYP2C11, 2E1 and 3A 1/2 for rat, CYP2C19, 2E1 and 3A4/5 for human, the metabolism of NBP in rat and human liver microsomes were reduced 38.8%, 86.2%, 78.4% and 51.0%, 92.0%, 58.9% of control, respectively. The metabolic rates of NBP in CYP2E1 and 3A 1/2 induced rat liver microsomes were increased 25.5% and 68.9%. High concentration of NBP (≥ 200 μmol x L(-1), in vitro) could inhibit the activities of CYP1A2, 2C6, 2C11 and 2D2 in rats, and high concentration of NBP ( ≥ 15 μmol x L(-1), in vitro) could inhibit the activity of CYP2C19 in human. All the results indicated that NBP should be mainly metabolized by CYP2E1, 2C11 and 3A 1/2 in rats and CYP2E1, 2C19 and 3A4/5 in human. High concentration of NBP could inhibit human CYP2C19 in vitro. No significant induction/inhibition effects of NBP were observed on rat liver CYP450 isoforms after ig 160 mg x kg(-1) NBP or iv 20 mg x kg(-1) NBP.

5.Reversal of resistance to adriamycin in human breast cancer cell line MCF-7/ADM by beta-elemene.

OBJECTIVETo study the reversal mechanism of adriamycin (ADM) resistance in human breast cancer cell line MCF-7/ADM by beta-elemene (beta-ELE), a wide spectrum anticancer drug derived from the Chinese herb Curcuma chaeocaulis.
METHODSSensitivity to ADM of MCF-7/ADM cells was studied by MTT assay. Intracellular accumulation of ADM in MCF-7/ADM cells was observed by fluorescent-spectrophotometry. Expression of bcl-2 protein was detected by flow cytometry.
RESULTSA non-cytotoxic dose (6 micro g/ml) and a weakly cytotoxic dose (13 micro g/ml) of beta-ELE could significantly enhance the cytotoxic effects of ADM on MCF-7/ADM cells to 1.4 and 2.2 fold as compared to the beta-ELE untreated control cells. The intracellular concentration of ADM in MCF-7/ADM cells was significantly increased after treatment with beta-ELE (P < 0.01). The expression of bcl-2 protein in MCF-7/ADM cells was reduced from 90.2% to 70.0% (P < 0.05).
CONCLUSIONbeta-ELE could partially reverse the drug resistance to ADM in MCF-7/ADM, which is related to the increased accumulation of intracellular ADM and the decreased expression of bcl-2.

6.Cellular compatibility of poly (lactic-co-glycolic acid)scaffold

BACKGROUND:The poly-lactic acid and its ramifications have many advantages, such as eligible biocompatibility,nontoxicity of degradation product, easy procession and suitable intensity. Thus, they have been widely used in bone tissue engineering.OBJECTIVE: To study the cellular biocompatibility and in vitro adhesion of poly(lactic-co-glycolic acid) (PLGA) scaffold and bone marrow stromal stem cells (BMSCs) so as to provide a basis for preparation a PLGA scaffold that can load many cytokines.DESIGN: Contrast observation.SETTING: Department of Orthopaedics and Traumatology in Nanfang Hospital of Southern Medical University.MATERIALS: The experiment was carried out in Key Laboratory of Tissue Construction and Detection of Guangdong Province from September 2004 to June 2005. One New Zealand healthy male rabbit (2 months old, 1.5-2.0 kg weight)was adopted in this study. Experimental materials: PLGA scaffold was obtained from Institute of Polymer Science in Sun Yat-sen University); beta-tricalcium phosphate (β-TCP) was supplied by AO Company (Switzerland).METHODS: Bone marrow was aspirated from the New Zealand rabbit. Mononuclear cells were harvested using whole bone marrow culturing, then were induced and amplified in the conditional culture medium. BMSCs were inoculated onto the PLGA and β-TCP at a concentration of 1 ×109 L-1, while those in the medium without materials were taken as blank control group. The development of implanted cells and the adhesion between cells and materials were observed with phase contrast microscope and scanning electron microscope. The proliferation and cycle of cells were tested with MTT method and flow cytometry.MATN OUTCOME MEASURES: ①Phase contrast microscope was used to observe the development of cells and the adherence between cells and materials at a fixed time every day. ②Cellular development on days 1, 3, 6 was observed by scanning electron microscope. ③Cellular proliferation was detected by MTT method. ④Alkaline phosphatase (ALP) activity was determined by chemical colorimetry.⑤Flow cytometer test: The effects of PLGA and β-TCP on the cellular cycle, content of DNA and polyploid levels of BMSCs were investigated. The DNA index of the candidate cells was also calculated.RESULTS: ①Phase contrast microscope observation: In the blank control group, cells culture for 7-10 days presented a larger number of shuttle-shaped, and no contact inhibition effect was observed. The time of cells beginning adherence in PLGA group was obviously later than that in β-TCP group. Cells began to develop on the circumambience and surface of the materials, with the prolonging of culture time. Most of the cells were multangular. The cells in both PLGA and β-TCP groups were similar to those in control group in items of cellular development and shape. ②Scanning electron microscope observation: On the seventh day of culture, the cells of control group remained a monolayer and amalgamated to be a patch with multangular-shaped ones increasing. There were substances in granule shape on the surface of the cells and micro-silk links between cells. In PLGA group, the cells After 7 days' cultivation proliferated sharply, and the compressed-shaped ones were inosculated to be a patch through integrations among cells, resulting in a large number of matrixes. While in β-TCP group, the number of cells increased from the 7th day. The cells were combined together to be a monolayer and moved to pores with matrixes creating out of cells.③Cellular proliferation: The number of the cells in each group all increased to some extents. However, there was no significant difference between the PLGA group and the control group, as well as the PLGA group and the β-TCP group (P > 0.05).④ALP activity: The content of ALP in all the groups enhanced without exception, while the activities between the PLGA group and the Both control group, as well as the PLGA group and the β-TCP group had no significant difference (P > 0.05).⑤Cellular cycle:Both PLGA and β-TCP had slight effects on cellular cycle of BMSCs. The cells in each group were all normal diploid,and no heteroploid cells were discovered.CONCLUSION: This type of PLGA scaffold possesses good cellular biocompatibility, and can be used as a carrier of many factors in bone tissue engineering.
7.Study on the changing status of morphological development among minority students in China, from 1985 to 2005

Objective To identify the changes of morphological development status on minority students in China from 1985 to 2005. Methods We selected a total of 15 groups of the Chinese minority students as subjects of the study, including Mongolian, Hui, Uygur, Zhuang, Korean, Tibetan, Yao, Li, Qiang, Buyi, Dong, Miao, Tu, Salar, Kirgiz, with data from the Chinese national survey on students' physical fitness and health condition in 1985, 1995, 2000 and 2005. Height, weight and waist of the subjects were calculated and analyzed. Results From 1985 to 2005, the growth and characteristics of height in the Chinese minority students had a similar increase when comparing to the Han students, but with different degrees. However the growth rate was gradually decreasing. The average heights of Kirgiz, Korean, Salar and Mongolian schoolboys aged 18 years old were 170 cm, being 170.91 cm, 170.47 cm, 170.29 cm and 170.27 cm, respectively, which were close to that of the Hart students. Some minority students had a substantial increase of body weight. However, the waist of some minority students decreased. Only a few groups of minority students had increasing waist, such as Mongolian and Korean rural boys, Mongolian, Zhuang, and Korean rural girls, with the growth being 0.101 cm, 0.095 cm, 0.126 cm, 0.163 cm and 0.107 cm, respectively. Uygur, Mongolian, Kirgiz and Korean students had the morphological development similar to Han urban students, especially Uighur boys and girls. Conclusion From 1985 to 2005, The height, weight and waist of Chinese minority students had an overall increase at different degrees. In order to improve the physical fitness of minority students, awareness on nutrition and health education of both students and parents should be strengthened. Surveillance and programs on growth, development and health status of the minority children and adolescents should also be carried out continuously.
8.Changes on secular growth among minority girls in China from 1985 to 2005

Objective To analyze the trend of secular growth among the minority girls in China from 1985 to 2005 in order to develop national and local intervention strategies and measures to improve their physical fitness and health.Methods A total of 19 Chinese minoritt girls,including Mongolian,Hui,Zhuang,Korean,Tibetan,Miao,Buyi,Dong,Yao,Bai,Tujia,Hani,Dai,Lisu,Wa,Nakhi,Tu,Qiang and Salar were sampled from the 1985 and 2005 Chinese national surveys to study the students' physical fitness and health.Probit analyses were used to calculate age at menarche (AAM),and the heights and weights of 17-years-old girls in various minority groups.Results From 1985 to 2005,the increments of adult heights and weights increased significantly in many minority groups.The average height of girls aged 17 years old in each minority group had an increase of 150 cm in 2005.Ethnic groups with height growth rates of more than 1 cm per decade in girls aged 17 years old were Korean ( 1.47 cm/decade),Tujia( 1.38 crn/decade),Qiang ( 1.32 cm/decade),Bai ( 1.25 cm/decade),Hui ( 1.13 cm/decade) and Buyi ( 1.07 cm/decade).And the mionority ethnics with weight growth rates of more than 1 kg per decade in girls aged 17 years old were Mongolian ( 1.79 kg/decade),Korean ( 1.69 kg/decade),Tibetan ( 1.66 kg/decade),Nakhi ( 1.39 kg/decade),Qiang ( 1.28 kg/decade) and Hui ( 1.10 kg/decade).The overall AAM showed a downward rate in all the 19 minority groups,but with different degrees.Tujia,Yao and Nakhi showed the largest reductions,with AAMs as 2.15,1.76 and 1.38 years earlier in 2005 than in 1985,respectively.Conclusion Our data suggested that the downward secular trend in AAMs and the increments of adult heights might reflect the secular growth change in the major minority groups during the past 20 years,but there was an obvious disequilibrium of puberty development and increments of heights in adults between the minority groups.

Objective To establish a RP-HPLC method for the determination of the blood concentration of paeoniflorin which was produced by seven kinds of warming-inside drugs (WID) being used with the promoting-blood drugs (PBD) individually and to explore the mechanism of PBD and WID compound prescription. Objective An HPLC method was established for determination of paeoniflorin in plasma after ig compound decoction of Radix Paeoniae Rubra with Fructus Piperis to mice. Methods The conditions of chromatography: Kromasil C 18 column (250 mm ? 4.6 mm, 7 ?m) was used with a mobile phase of CH 3OH-H 2O ( 38∶ 62); flow rate was 0.5 mL/min; the detecting wavelength was 230 nm; external standard method was quantitative analysis method. Results Paeoniflorin was fully separated from the other components in plasma. The linear range was 5.0—250.0 ng/?L (r= 0.999 9 ), the lowest detectability was 1.49 ng/?L, the average recovery was higher than 90%. Conclusion This method specially provides an accurate and sensitive way in detecting the in vivo blood concentration of paeoniflorin in plasma.
