1.Design of an interpersonal climate questionnaire for education research teams

Objective To provide a valid tool for assessment of the interpersonal climate of research teams by develo -ping a team interpersonal climate questionnaire .Methods Based on literature research and panel study , a pilot question-naire was constructed and handed out to a sample of 378 research graduate students in research teams .The analysis of va-lidity and reliability was conducted by SPSS 16.0 and Amos 17.0 for the questionnaire.Results The interpersonal climate in research teams is a two-facet concept , including mentor-mentee dyadic interpersonal relationship .The peer interpersonal relationship.The mentor-mentee dyadic interpersonal relationship consists of five dimensions: identification, support, developmental direction, individualized care , effectiveness of relationship , with the alpha coefficient ranging fron 0.931 to 0.959, while peer interpersonal relationship has only one dimension .Conclusion ①The interpersonal climate is a two-facet, six-dimension concept .②The self-developed interpersonal climate questionnaire achieves satisfactory validity and reliability .
2.Dynamic study and screening of new markers of spermatogonial stem cells by iTRAQ protein mass spectrometry.

OBJECTIVETo study the dynamic changes in the protein marker expression in the spermatogonial stem cells (SSCs) of mice at different ages by iTRAQ protein mass spectrometry and to screen new markers using the bioinformatic proteome database.
METHODSBased on the postnatal weeks, we divided 80 healthy male C57BL/6 mice into eight age groups of equal number, harvested their testicular tissues, extracted proteins following purification of the SSCs by compound enzyme digestion and magnetic-activated cell sorting. Then we analyzed and identified proteins using two-dimensional electrophoresis, protein mass spectrometry, and protein database information.
RESULTSTotally, 248,510 mass spectra were obtained from the MS experiment and 1132 proteins were identified. By the criteria of >1.2-fold for protein abundance difference and P value <0.05, we identified 298 differentially expressed proteins and 9 currently known makers of SSCs (PCNA, GFRalpha1, CDH1, Annexin A7, UCHL1, VASA, CD49f, CD29, and PLZf). Compara- tive analysis showed different expressions of the proteins in the SSCs of the mice of different ages, and the differences in the expressions of GFRalpha1, CD49f, and CD29 were consistent with the findings in other published literature. Ten proteins (P63, CD71, CD98, K19, ACE, K18, K15, K17, SH2, and SH3) were selected as SSC markers to be further studied.
CONCLUSIONThe proteins in SSCs are differentially expressed in mice of different ages. The technology of iTRAQ protein mass spectrometry can be used to analyze and compare the proteome information of mouse SSCs, obtain differentially expressed proteins in mice of different ages, and thus offers a new ap- proach to further analysis and study of the function and roles of these differential proteins.

3.High-efficiency gene transfer into rabbit smooth muscle cells by pseudotyped retroviral vector

Objective To construct pseudotyped retroviral vector MuLV/VSV-G and transfer it into rabbit smooth muscle cells (SMC) in order to provide a high-efficiency vector for SMC gene transfer. Methods We constructed pseudotyped retroviral vector MuLV/VSV-G containing the previously reported gene lacZ, determined the titer, and determined the efficiency of gene transfer into SMC mediated by pseudotyped retroviral vector MuLV/VSV-G. Finally the transfer efficiency was compared with that by MuLV. Results MuLV/VSV-G vector was constructed. The titer of the vector was 6-7.8×10~6CFU, the transfer efficiency was (92±12)% by using MuLV/VSV-G vector and (24±5)% by MuLV vector. Conclusion Pseudotyped retroviral vector MuLV/VSV-G which was constructed successfully is a kind of high-efficiency gene transfer vector in smooth muscle cells.
4.Clinical study on 27 children with Epstein-Barr virus encephalitis

Objective To explore the clinical characteristics and diagnosis of Epstein-Barr(EB) virus encephalitis(EBE) in children. Methods The verification of EBE was based on detection of EBV DNA in cerebrospinal fluid (CSF) by fluorogenic quantitative PCR(FQ-PCR) and Nest-PCR. The clinical and CSF changes of 27 EBE cases and 26 controls were analyzed and compared. Results EB-DNA in CSF by FQ-PCR of 13 cases of EBE was (2.82?2.03)?10 3copies.There was no significant difference in clinical manifestations between EBE and HSE groups, except that WBC in CSF of EBE was lower than those of HSE.Conclusions EBE is not infrequent, about 10 % of encephalitis in children. EBE is always present independently, which is not a complication of infectious mononucleosis(IM).Detection of EBV-DNA in CSF is a sensitive and specific test for diagnosing of EBE. Early treatment may be beneficial to the prognosis of EBE.
5.Screening of differential expression protein from human colorectal carcinoma and normal colorectal tissues by two-dimensional difference gel electrophoresis and mass spectrometry

Objective To find early diagnostic biomarkers for colorectal carcinoma by comparing differential(expressing) proteins from colorectal carcinoma and normal colorectal tissues.Methods Colorectal carcinoma tissues and paired normal tumor-adjacent colorectal tissues were collected,and tissue total protein was (extracted);differential proteome profiles were established and analysed by means of immobilized pH(gradient-based) two-dimesional polyacrylamide gel electrophoresis(2D-PAGE) and matrix-assisted laser(desorption)/ionization time of flight mass spectrometry(MALDI-TOF-MS).Results Well-resolved,(reproducible) 2-DE profiles of human colorectal carcinoma tissues and paired normal tumor adjacent colorectal tissues were obtained.For tumor tissue,a total of 1098?28 spots were detected,and for normal tissue,760?45 spots were detected.For normal tissue,The average deviation of spot position was(0.542?(0.12))mm in IEF direction and(0.933?0.098)mm in SDS-PGE direction for tumor tissue.The average deviation of spot position was(0.745?0.130)mm in IEF direction and(1.233?0.272)mm in(SDS-PGE) direction.30 differential expressing proteins were analysed by mass spectrometry and bioinformation,16 of them were well characterized including Apolipoprotein A1(apoA1),calreticulin precursor,glutathione(S-transferase),hepatic fatty acid-binding protein、heat shock protein 27 ect.Conclusions Differential expression proteins can be candidate biomarkers for early diagnosis of colorectal carcinoma;and proteomic technique is valuable for screening the diagnostic biomarkers.
6.Proteomic identification of chemosensitivity-associated proteins in human colorectal carcinomas

Objective:The study aims to screen chemosensitivity-associated proteins in colorectal carcinoma tissues by using two-dimensional gel electrophoresis (2-DE) and mass spectrometry,then identify some differentially-expressed proteins. Methods:The patients with advanced colorectal carcinoma were confirmed by clinical diagnosis. Fresh carcinoma specimens were collected by biopsy and preserved in liquid N2. The tissues were classified into two groups: high sensitivity group (HS) and low sensitivity group (LS) based on drug sensitivity test. The total proteins were extracted and separated by 2-DE. The images were composed, compared, and differentially analyzed to identify the proteins with differential expression in HS and LS groups. Then the differentially-expressed protein spots were incised from the gels and digested by trypsin. The peptide mass fingerprintings (PMF) was acquired after matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) and the proteins were identified by data searching in the Mascot database. Two proteins with differential expression were detected by Western blotting.Results:The 2-DE spectrum of HS and LS groups were established. Most protein spots were distributed in the area with pH 4-8 and relative molecular weight of (20-100)×10~3. The average number of the protein spots was 842±23 in HS group and 793±19 in LS group,respectively. The mean matching rate was 90.7%. The number of differentially-expressed dots between HS and LS group was 79.00±13.56. Thirty protein dots were selected for mass spectrum and bioinformatic analysis, and 9 proteins were identified. Conclusion:Colorectal carcinoma with different chemosensitivity had differential protein expression profiles. The differentially expressed proteins may be associated with chemosensitivity and could be used for prediction of chemosensitivity of colorectal carcinoma.
7.Repair effect of human acellular dermal matrix on filtering blebs leakage in rabbit eye

Background Package and tissue patch implantation are common methods for repair of filtering bleb leaking after anti-glaucoma surgery.But the scarring or re-leakage of filtering bleb probably occur again.Objective This study was to investigate the repair effect of acellular dermal matrix (ADM) on filtering bleb leaking in rabbit model and compare the effectiveness among ADM,amniotic membrane and conjunctival overlap.Methods Trabeculectomy was performed on 48 eyes of 24 New Zealand rabbits,and models of filtering bleb leaking were established.The models were randomized into ADM group,amniotic membrane group and conjunctival covering group based on randomized number table.ADM patches with 4 mm×4 mm were implanted across lamellar cornea and sclera at a bridge in the ADM group,and the same size of amniotic membranes were used in the amniotic membrane group,and conjunctiva was sutured to limbus in the conjunctiva overlap group.The intraocular pressure (IOP) was measured before surgery and 1 day,1 week,1 month,3 months and 6 months after surgery.The biocompatibility of materials above was assessed under the slit lamp microscope,and the status of filtering bleb was evaluated and compared with anterior segment optical coherent tomography (AS-OCT) 1 day,1 week,1 month,3 months and 6 months after surgery.Results Before surgery and 1 day,1 week,1 month,3 months and 6 months after surgery,the IOP was (26.9±4.3),(16.6±5.1),(22.1 ±6.2),(18.3±6.5),(22.7±2.5),(23.4±1.4) mmHg in the AMD group,(29.9±5.4),(14.9 ± 6.4),(21.6 ± 7.8),(26.3 ± 4.1),(26.0 ± 4.2) and (23.0 ± 5.3) mmHg in the amniotic membrane group,and (28.7 ±4.3),(15.7 ±7.0),(22.0±6.3),(28.2±4.1),(24.7 ±4.1),(23.0±2.7) mmHg in the conjunctival overlap group,showing significant differences among different groups and various time points (Fgroup =8.419,P=0.011 ;Ftme =15.543,P=0.000).The IOPs were significantly lower from 1 day through 3 months after operation than those before operation in the AMD group (P =0.000,0.000,0.006,0.045) ; while the IOPs were reduced only from 1 day through 1 week after operation in comparison with before operation in the amniotic membrane group and the conjunctival overlap group (P =0.000,0.001).One month after surgery,the IOPs were significantly declined in the ADM group compared with the amniotic membrane group and the conjunctival overlap group (P =0.001,0.000).The grafts were clear under the slit lamp microscope and exhibited the valid filtering bleb until 3 months after operation under the AS-OCT in the ADM group.However,the valid filtering bleb remained only 1 month after surgery in the amniotic membrane group and the conjunctival overlap group.Neovascularization on the filtering bleb was found 3 months in the AMD group but 1 month in the amniotic membrane group and the conjunctival overlap group.Conclusions Compared with amniotic membrane and conjunctival tissue,ADM patch for the repair of filtering bleb leakage can increase the survival duration of filtering bleb and remain lower IOP.

9.CT perfusion imaging study of perihematomal cerebral blood flow in experimental intracerebral hemorrhage

ObjectiveTo study the mechanism of hemodynamic alternation surrounding the hematoma in the acute stage of intracerebral hemorrhage (ICH) in rats.MethodsSeventy male Sprague Dawley rats were randomly divided into ICH group and sham operated group, which were microinjected with 40 μl fresh autologous blood or saline into the right caudatum respectively. The each group was divided into 7 subgroups at 1h,3h,6h,12h,24h,48h and 72h after the ICH. CT perfusion imaging in measurements of regional cerebral blood flow adjacent to hematomas was performed. The ratios of side to side were measured at the regions around the hematomas by personal computer aided mapping. So the parameters of regional cerebral blood flow(rCBF), regional cerebral blood volume(rCBV) and mean transit time(MTT) were calculated respectively.ResultsThe rCBF and rCBV adjacent to the hematomas were lower than those of the outer region pronouncedly. The alternation of rCBF around the hematoma were fluctuated, which reduced to the valley at 1h after ICH, and then gradually returned to the peaks at 6h and 24h after ICH. In the meantime, the rCBV around the hematoma reduced to the valley at 1h after ICH, and then gradually increased to the peak at 24h after ICH.ConclusionThe abnormal hemodynamic changes can be found in the perihematomal region after ICH. The alternation of rCBF around the hematomas are fluctuated, but the changes of rCBV remain continuous increase. The mass effect of hematoma, intracranial hypertension caused by the mass effect of hematoma, and the autoregulation of cerebral blood flow motivated by the initial depression of cerebral blood flow play a very important role in the changes of cerebral blood flow.
10.Clinical Observation of Puncturing Sifeng Points (EX-UE10) plus Pricking Back-Shu Points for Dyspepsia Due to Chemotherapy for Triple-negative Breast Cancer

Objective To assess the effectiveness of puncturing Sifeng points (EX-UE10) and pricking Back-Shu points in treating dyspepsia due to chemotherapy for triple-negative breast cancer (TNBC). Method Sixty patients were randomized into an observation group (30 cases) and a control group (30 cases). The observation group was intervened by puncturing Sifeng points and pricking Back-Shu points, once a week. The selected Back-Shu points included bilateral Pishu (BL20), Weishu (BL21) and Geshu (BL17). The control group was treated by promoting gastrointestinal motility (itopride hydrochloride 50 mg) and supplementing digestive enzymes (compound azintamide tablets). The two groups were observed before and after treatment in terms of traditional Chinese medicine (TCM) symptom score, nutritional status score and Karnofsky Performance Score (KPS). The therapeutic efficacies were also assessed. Result The total effective rate was 93.3% (28/30) in the observation group versus 70.0% (21/30) in the control group, and the between-group difference was statistically significant (P<0.05). The TCM symptom score showed significant improvement in both groups after treatment (P<0.01), and the improvement in the observation group was more significant than that in the control group (P<0.01). After treatment, the score of Patient Generated-Subjective Global Assessment (PG-SGA) decreased significantly in both groups (P<0.01), while there was no significant difference in the score between the two groups (P>0.05). The KPS score increased significantly in both groups after treatment (P<0.01), and there was a significant difference between the two groups (P<0.05), indicating a more significant improvement of KPS score in the observation group. Conclusion Puncturing Sifeng plus pricking Back-Shu points is effective in treating dyspepsia due to chemotherapy for TNBC. It can improve patient's appetite and quality of life.
