1.Network pharmacology study on major active compounds of siwu decoction analogous formulae for treating primary dysmenorrhea of gynecology blood stasis syndrome.

OBJECTIVETo explore the molecular mechanisms of main active ingredients of Siwu decoction analogous formulae for treating primary dysmenorrhea of gynecology blood stasis syndrome by network pharmacology study, and to investigate the correlations between multi-compounds, multi targets and multi pathways.
METHODMajor active compounds from Siwu decoction analogous formulae, including ligustilide, butylidene phthalide, senkyunolide, ferulic acid, gallic acid, peoniflorin, jioglutin A, catalpol, transanethole, zingiberone, commiphoric acid, eugenol, isorhamnetin-3-O-neohesperidoside, wulingzhic acid, alpha-cyperone, cyperene, costunolide, costic acid, tetrahydropalmatine, protopine, amygdalin, 24-methylene cycloartanol, oleic acid, linoleic acid, 3-p-coumaroylquinic acid, hydroxysafflor yellow A, coptisine, berberine, jatrorrhizine, baicalein, baicalin, wogonin were collected to build component-protein networks based on PharmMapper database. The targets information access was used to construct and visualize components-targets-pathways network model using the kyoto encyclopedia of genes and genomes (KEGG) pathway database and Cytoscape software.
RESULT AND CONCLUSIONSerine threonine protein kinases play an important role in the process of cells. They were potential targets in the effect of Siwu decoction analogous formulae. The effect of main active ingredients involved 51 the pathway. Besides the same ones, Shaofu Zhuyu decoction had more effect on lipid metabolism, Xiangfu Siwu decoction on amino acid metabolism pathways, Taohong Siwu decoction on carbohydrate metabolism, while, Qinlian Siwu decoction on ErbB, VEGF signal transduction pathway. Siwu decoction and its derived formulae not only had common targets and pathways, but also had their own emphasis. This reflected the formulae effect mode of multi-ingredients, multi-targets and multi-pathways. It may provide clues to deeper study of molecular mechanism of Siwu decoction analogous formulae action.

2.Anterolateral thigh flap transferred with iliotibial tract for coverage of complicated forearm wounds

Objective To introduce the transfer of anterolateral thigh flap with iliotibial tract in repair of massive skin and soft tissue defects of the forearm and reconstruction of extension or flexion of the forearm. Methods Eight cases of complicated raw wounds of the forearm were repaired with transfer of anterolateral thigh flap with iliotibial tract. Flexor tendons of 3 cases and extensor tendons of 5 cases were repaired with iliotibial tract. The axial vessel of the flap was used to rebuild blood supply of the hand. Results All the flaps survived completely. A follow-up ranging from 3 months to 2.5 years re- vealed that the reconstructed forearms were good in appearance and soft in texture and restored protective sensation. Affected limbs could perform extension or flexion. Total range of motion (TRM) of the hands was excellent in 6 cases and poor in 2. Conclusion Anterolateral thigh flap with iliotibial tract can not only repair soft tissue defects of the forearm, but also reconstruct the main extension or flexion of the forearm simultaneously.

4.A study on the influence of parental history of allergy on the level of cytokine in umbilical cord blood

Objective To explore the influence of parental history of allergy on the cytokine level of umbilical cord blood of neonates.Methods A prospective study was conducted which selected the pregnant women attending Yiwu maternal and child healthcare hospital from July to December in 2012 as objects.The umbilical cord blood sample were collected to detect the level of IgE,eosinophile granulocyte,eotaxin,IL -9,IL -6,IL -4,IL -5,IFN -γand IL -10. Results A total of 136 subjects were recruited and divided into three groups according to the status of parental allergy histories.There was no significant difference in social and demographic characteristics among the three groups (P>0.05). Three groups have significantly different median concentration of IL-13 (1.82pg/mL vs.3.24pg/mL vs.6.12pg/mL), eotaxin (29.33pg/mL vs.50.71pg/mL vs.60.15pg/mL),IL-9 (43.75pg/mL vs.111.17pg/mL vs.183.19pg/mL), IL-6 (11.49pg/mL vs.19.35pg/mL vs.26.09pg/mL),IL-4 (3.67pg/mL vs.21.27pg/mL vs.49.51pg/mL),IL-5 (4.26pg/mL vs.7.69pg/mL vs.10.66pg/mL),IFN-γ(338.65pg/mL vs.649.17pg/mL vs.834.14pg/mL),IL-10 (7.13pg/mL vs.10.56pg/mL vs.14.64pg/mL),P <0.05.Conclusion Parental history of allergy could have influence on cytokine level of umbilical cord blood.Early screening and surveillance in neonates whose parents have history of allergy should be taken to guide early interventions.
5.Dietary iodine intake level of adult population in coastal and inland areas of Zhejiang province

Objective To evaluate the dietary iodine intake level of population in coastal and inland areas of Zhejiang province.Methods The cluster sampling method was applied to select Zhoushan,Ningbo and Taizhou cities from Zhejiang province as coastal areas,to select Jinhua,Quzhou and Lishui cities from Zhejiang province as inland areas,and two counties(districts) were randomly selected in each chosen city,three sub-districts(towns) were randomly selected in each chosen counties (districts),then one community (village) was randomly selected in each chosen sub-districts(towns).Adult residents aged greater than 18 were selected as investigation subjects,their dietary iodine intake in the past 24 hours was investigated,and the Food composition table” was inquired to get the data of dietary iodine intake.One hundred copies of residential edible salt samples and drinking water samples in each of the selected community (village) were collected to detect salt iodine and water iodine by direct titrimetric and spectrophotometric method,respectively.Results The mean of adult residents' dietary iodine intake in Zhejiang coastal area was 267.76 μg/d,which was less than that in inland area (429.05 μg/d,t =- 6.90,P <0.05),and the means of adult residents' dietary iodine intake from drinking water,laver and fish(5.75,69.72,5.61 μg/d,respectively) in coastal area were higher than those in inland areas(3.25,35.27,3.43 μg/d,respectively,t =21.73,3.92,4.08,all P < 0.05),however,the means of adult residents' dietary iodine intake from salt,kelp and other food (166.81,3.04,16.82 μg/d,respectively) in coastal areas were less than those in inland areas (355.15,6.14,25.81 μg/d,respectively,t =- 8.76,- 5.49,- 18.56,all P < 0.05).In coastal areas,the proportion which was less than estimated average requirement of iodine(EAR,120 μg/d) was 46.48% (1029/2214),the ratio which was higher than the maximum tolerable intake of iodine(UL,1000 μg/d) was 3.34% (74/2214),and the average contribution rate of dietary intake of iodine in salt was 62.30%(166.81/267.76).While in inland areas,the corresponding proportions were 7.61% (171/2246),2.80% (63/2246) and 82.78% (355.15/429.05),respectively.Conclusions The dietary iodine intake in Zhejiang inland areas has reached the recommended nutrient intake levels of the Chinese Nutrition Society,but there is a certain degree of insufficient iodine intake in population of the coastal areas.
6.Curative effect of decompressive craniectomy with reserved superficial temporal artery for treatment of severe traumatic brain injury

Objective:To investigate the curative effect of decompression with reserved superficial temporal artery for treatment of severe traumatic brain injury (sTBI).Methods:A retrospective case-control study was conducted to analyze the clinical data of 265 patients with sTBI who underwent decompressive craniectomy in Shaanxi Provincial People's Hospital from July 2016 to December 2019. Among them, 152 were males and 113 were females, with the age of 33-72 years [(50.1±12.8)years]. On admission, the Glasgow Coma Score (GCS) was 3-8 points [(5.7±1.4)points]. A total of 135 patients were treated with reserved superficial temporal artery decompression after admission (observation group) and 130 patients were treated with conventional decompression without deliberately preserving the superficial temporal artery (control group). The amount of intraoperative scalp bleeding, scalp healing time, cerebrospinal fluid incision leakage, healing degree and postoperative craniocerebral CT angiography (CTA) were compared between the two groups.Results:All patients were followed up for 7-15 days [(12.4±2.5)days]. The amount of intraoperative scalp bleeding was (15.4±4.5)ml in observation group and (65.2±4.7)ml in control group ( P<0.01). The healing time was (7.1±1.1)days in observation group and (9.5±2.3)days in control group ( P<0.01). There were 5 patients (3.7%) with cerebrospinal fluid incision leakage in observation group compared to 21 patients (16.2%) in control group ( P<0.01). There were 114 patients with Grade A healing in observation group compared to 91 patients in control group ( P<0.05). Based on the postoperative craniocerebral CTA, the superficial temporal artery was intact and the blood supply was normal in observation group, while the main trunk of the superficial temporal artery was disconnected from the zygomatic arch segment and the blood supply was incomplete in control group. Conclusions:For patients with sTBI, decompressive craniectomy with reserved superficial temporal artery can promote the healing of the scalp. Moreover, keeping the superficial temporal artery can significantly reduce the amount of bleeding during operation, reduce postoperative scalp healing time and incidence of cerebrospinal fluid incision leakage and assist cerebral blood perfusion.
7.A primary study on the relationship between amino acid mutations in clinical isolates of Neisseria gonorrhoeae and their resistance to antibiotics.

OBJECTIVETo identify the relationship between amino acid mutations in Neisseria gonorrhoeae isolates and their antibiotic resistance.
METHODSPI gene fragments of Neisseria gonorrhoeae from 17 clinical isolates were obtained with PCR amplification. They were cloned into the PCR cloning vector pBS-T to form pBS-T-PI and sequenced. The sequences of PI genes were analyzed. At the same time, minimum inhibitory concentration (MIC) of penicillin and tetracycline to these 17 isolates were measured and contrasted with the corresponding PI sequence.
RESULTSThe recombinants of PI gene from 17 clinical isolates of Neisseria gonorrhoeae were successfully constructed and sequenced. They were divided into PIA and PIB subtypes according to the results from blastn software by comparing the sequences with the GenBank. Mutations were found at the sites of 120 and 121. There were only some of the sequences having an aspartic acid (D) mutation on 120 and 121 sites, which was not the same as reported. On the other hand,there were two PI sequences,5-9 and 6-1, whose mutations on No. 120 were lysine, similar to those documented.
CONCLUSIONSome relationship between PI amino acids mutations at sites 120 and 121 in Neisseria gonorrhoeae isolates from Chengdu, China and their resistance to penicillin and tetracycline were found. However,further studies need to be done in the future to confirm this hypothesis.

8.Chemical constituents of Kadsura oblongifolia and evaluation of their toxicity.

To study the chemical constituents of K. oblongifolia, silica gel column chromatography, MCI and Sephadex LH-20 were used to separate the 70% acetone extract of the stems of K. oblongifolia. The structures of the isolated compounds have been established on the basis of physicochemical and NMR spectroscopic evidence as well as ESI-MS in some cases. Twenty compounds were obtained and identified as heteroclitalignan A (1), kadsulignan F (2), kadoblongifolin C (3), schizanrin F (4), heteroclitalignan C (5), kadsurarin (6), kadsulignan O (7), eburicol (8), meso-dihydroguaiaretic acid (9), kadsufolin A (10), tiegusanin M (11), heteroclitin B (12), (7'S)-parabenzlactone (13), angeloylbinankadsurin B (14), propinquain H (15), quercetin (16), kadsulignan P (17), schizanrin G (18), micrandilactone C (19) and (-)-shikimic acid (20). Compouds 1, 5, 8, 11-15, 18 and 20 were isolated from this plant for the first time. Toxicity of compounds 1-10 were evaluated with zebrafish model to observe the effect on its embryonic development and heart function. The results showed that compounds 7, 9 and 10 caused edema of zebrafish embryo and decreased the heart rate of zebrafish, which exhibited interference effect on heart development of zebrafish.

Objective To explore the effect of maternal history of allergy on the cytokine level in umbilical cord blood. Methods A prospective study was conducted among pregnant women in Yiwu City,China.136 participants were selected from the Yiwu maternal and child healthcare hospital in 2012.A questionnaire interview was conducted.The umbilical cord blood sample were collected to detect the level of IgE,eosinophile granulocyte,eotaxin,IL -9,IL -6,IL -4,IL -5, IFN -γ,IL -10.The association between allergy history and cytokine levels was analyzed.Results All subjects were recruited and divided into two groups(pregnant women with allergy history and without allergy history).Compared with the pregnant women with allergy history,these subjects without allergy history had higher IL -13 (5.52 pg/ml vs 2.84 pg/ml) and IL4 (43.33 pg/ml vs 8.23 pg/ml)(P <0.05).Besides,there is a correlation between IL -13 and IL -4 (rs =0.87,P <0.01).Partial correlation analysis showed the correlaiots among IL -4,IL -5,IL -9,IL -10,IFN -γand eotaxin.Conclusion The levels of IL -13 and IL -4 are significantly associated with the history of allergy in pregnant women.It could be considered the indexes are sensitive for early screening and surveillance of allergy for babies.
10.Antisense DNMT1 gene fragment in the sensitivity change of SMMC-7721 cells to tumor necrosis factor related apoptosis inducing ligand and its mechanism.

OBJECTIVETo observe the sensitivity change of SMMC-7721 cells transfected with antisense DNMT1 gene fragment to tumor necrosis factor related apoptosis inducing ligand (TRAIL) and its mechanism.
METHODSCell survival rate was measured by trypan blue, apoptosis rate by TUNEL method and the expression of bcl-2, bax and bad by flow cytometry.
RESULTSCell survival rate of SMMC-7721 cells transfected with antisense DNMT1 gene fragment was markedly lower than that transfected with sense DNMT1 gene fragment or empty vector (P < 0.05 and 0.01), but the apoptosis rate was on the contrary (P < 0.05 or 0.01). The expression of bax and bad (especially the former), but not bcl-2 of SMMC-7721 cells transfected with antisense DNMT1 gene fragment was markedly higher than those of SMMC-7721 cells transfected with sense DNMT1 gene fragment or empty vector.
CONCLUSIONThe sensitivity of SMMC-7721 cells to TRAIL can be enhanced by the transfection of antisense DNMT1 gene fragment, which may be related to the increase of bax and bad expression.
