Objective To Observe the effects of tumor necrosis factor ? (TNF ?) on cell proliferation and Intracellular free calcium concentraction ([Ca 2+ ]i) in endothelium of human umbilical vein endothelial cells (HUVEC) and investigate the pathogenesis of pregnancy induced hypertension syndrome (PIH). Methods Confluent monolayer of HUVEC was directly incubated with TNF ? at following final concentrations: 500, 1 000, 2 000 U/ml for 24 hours. The percentages of different cellcycles and [Ca 2+ ]i were measured by flow cytometry and fluorospectrophotometry. Results Incubated with TNF ?, the endothelial cells were elongated and transformed into fibroblast like cells. Border of nucleus was sharp, clarity, and cells were in regular shape. But there were abnormal granules in cytoplasma and some cells detached at the concentrotion of 2 000 U/ml of TNF ?. Stimulated by TNF ?, the percentage of cellcycles from phase G 0+G 1 to S and G 2+M decreased significantly and it was dose dependent. [Ca 2+ ]i increased significantly and dose dependent as well. Conclusion TNF ? may injure endothelium directly and inhibit its proliferation and repair through the changes of [Ca 2+ ]i level. It may play an important role in the pathogenesis of PIH.

In order to investigate the affinity of aptamers to Bacillus anthracis spore, a custom synthesized 78 mer random DNA library was subjected to 15 rounds of selection against spores of vaccine strain A.16R by using SELEX method. The selected aptamers were cloned and sequenced. Macaw 2.05 and DNAsis 2.5 package were employed to analyze the conserved sequences and second structure of the aptamers, respectively. Affinities of aptamers to the spores were visualized by biotin streptavidin horseradish peroxidase system. The results showed that affinities of the aptamers were different. The highest OD at 450nm was 1.2, and the lowest was 0.25. The second structure analysis revealed possible stem loops for binding to the spores. The conserved sequences, AGGGG, CCCCG, GGGTT and ACACT, were found and the aptamers having same conserved sequence demonstrated similar affinity to the spores.

This paper described the existing problems of transformation benefit from a target,two ideas,three questions,four reasons,and five measures.Through establishing clear objectives,combing the status of the achievement trasformation some problems were raised.Also,it Was analyzed for the reasons of deficient transformation benefit and suggested the taken measures.

Objective To study the expression of RhoA protein in colorectal carcinoma. Methods The expression of RhoA protein in 92 cases of colorectal ccarcinoma and 30 cases of normal rectal tissue was detected by SP immunohistochemical technique. Results RhoA protein were negative in 40 cases of normal colorectal tissue. In 92 cases of colorectal carcinoma, the positive rate of RhoA protein was 70.7%. It was significantly higher than that in the normal colorectal tissue. The positive rate of RhoA protein in high-differentiated 、moderate-differentiated and lowdifferentiated rectal carcinoma were 43.5% 、74.4% and 86.7% ,respectively(P ＜0.05). The positive rates of RhoA in phase Ⅰ、phase Ⅱ、phase Ⅲ and phase Ⅳ were 48.6% 、77.3% 、83.3% and 94.1%, respectively (P ＜ 0.05).The positive rate of RhoA protein in patients with lymphnode mestastasis and non-lymphnode mestastasis were 82% and 57.1% (P ＜ 0.05). Conclusion RhoA protein could be related with the differentiation、invasion、metastatsis and prognosis of colorectal carcinoma.

Objective　To monitor KBD prevalence rate in Changdu of Tibet.Methods　The aged 7～12 year children are tested with X- ray and epidemiological investigation.Results　Xizang are still high yet,the 3 of 4 porints X-rates are more than 20% and the highest one is 55.34%.Conclusions　The Changdu is the severest point of KBD in our country.

We here systematically summarize our practice and its effectiveness in the creation and development of the discipline proteomics in Academy of Military Medical Sciences,and the establishment of a system for identifying and training talented researchers in proteomics.The methods we used to train the researchers and to develop a vigorous team are discussed.These practices contributed much to the development of proteomics in China.

·AIM: To investigate the efficacy and security of the silicone membrane implant through observing intraocular pressure, filtering blebs and histopathologic results.·METHODS; Forty rabbits were randomly divided into 4 groups. Each group consisted of 10 rabbits. Trabecu-lectomy and silicone membrane implantation were performed in one eye of each rabbit, while the other eye was only performed trabeculectomy as control. The change of intraocular pressure, filtering blebs were observed postoperatively. And each eye had undergone histopathologic examination.·RESULTS:The duration of low intraocular pressure and existence of filtering blebs in implanted eyes was longer than that in controlled eyes. Light microscopy revealed that patent drainage tract and biting site could be seen in silicone membrane implanted eye. The activity and regularity of fibroblast proliferation in implanted eyes and controlled eyes were similar. Excessive expression of fibroblast proliferation was not induced by silicone membrane.·CONCLUSION: Characterized by safety, effectiveness and simplicity, silicone membrane implantation can act as a new drainage surgery.KEYWORDS: silicone membrane; drainage surgery;intraocular pressure

This paper is to report our results of the observation on the changes of the pulmonary water content of the rats and mice after their exposure to a simulated altitude of 6000 meters above sea level for seven days.It was found that the changes of the pulmonary water content varied with the duration of exposing to the high altitude. It was lower than the control value on the first day of exposure, and then it increased approaching or even being a little higher than the control value on the second and third day. But it decreased and was below the control value again from the fourth day to the seventh day. The lung weight was increasing continuously in the same period.In addition, there were progressive increase of both the wet-lung/body and dry-lung/body indices, progressive decrease of left/right ventricles ratio, and gradual rising of hemoglobin in the animals studied.
