2.Histology research about nervalization of tissue engineered bones

[Objective]To study the ossification effect by histology research on tissue engineered bone by transplanting nerval bundles of different types.[Method]The Newzeland rabbit BMSCs(bone marrow stromal cells,BMSCs) were induced to osteoblast and compounded with ?-TCP(?-tricalcium phosphate,?-TCP)to make tissue engineered bones,and different kinds of nerval bundles were transplanted into these tissue engineered bone.All Newzeland rabbits were divided into 5 groups according to different types of nerval bundles:Group A,tissue engineered bones without nerval bundles transplantation.Group B,tissue engineered bones transplanted with sensory nerval bundles.Group C,tissue engineered bones transplantation with motor nerval bundles.Group D,tissue engineered bones transplanted with vascular bundles(autonomic nerves contained).Group E,tissue engineered bones transplantation with both sensory and motor nerval bundles.Then,1.5 cm bone defect in femur of Newzeland rabbits were repaired.At 12 week,gross specimen observation,histology studies and immunohistochemistry were used to compare the results of bone repair in different groups.[Result]During the observation period,tissue engineered bones transplanted with sensory and vascular bundles effectively accelerated the ossification of the tissue engineered bones,while those transplanted with motor nerval bundles did not.[Conclusion]Sensory nerval bundles and vascular bundles can effectively accelerate the ossification of tissue engineered bones.They can be used as a construction method for nervalization after tissue engineered bone.
3.Protective effects of propofol preconditioning on myocardium against hypothermia ischemia normothermia reperfusion injury on isolated rat hearts

Objective To investigate the protective effects of propofol preconditioning on myocardium against hypothermia ischemia normothermia reperfusion injury on isolated rat hearts. Methods The Langendorff apparatus was used.Sixty SD rat hearts were divided randomly into 5 groups after 20-minute equilibrium(n=12): control(Con) group,hearts were continually perfused with K-H buffer for 175 min;ischemia/reperfusion(I/R) group,hearts were perfused with K-H buffer for 40 min,then subjected to global ischemia at 27 ℃ for 75 min,and followed reperfusion at 37 ℃ for 60 min;propofol preconditioning group 1(P1),group 2(P2),and group 3(P3),hearts were perfused with K-H buffer including 50,100,and 150 ?mol/L propofol for 10 min and followed reperfusion like I/R group,respectively.Heart rate(HR),left ventricular end-diastolic pressure(LVEDP), left ventricular developed pressure(LVDP) and ?dp/dtmax at the end of equilibration,pre-ischemia and at the end of reperfusion were recorded.The contents of creatine kinase(CK) and lactate dehydrogenase(LDH) in coronary effluent were measured at the end of equilibration and 1,10,20,30,and 60 min during reperfusion.The activity of superoxide dismutase(SOD) and the contents of maleic dialdehyde(MDA) were measured at the end of reperfusion.The area of infarct region was determined at the end of reperfusion. 4.Effect of danhong injection on ET-1, sP-sel, and hs-CRP in patients with acute coronary syndrome undergoing percutaneous coronary intervention.

OBJECTIVETo observe the effect and explore the action mechanism of Danhong Injection (DHI) on serum levels of endothelin-1 (ET-1), soluble P-selectin (sP-sel) and high sensitivity C-reactive protein (hs-CRP) in patients with acute coronary syndrome (ACS) undergoing percutaneous coronary intervention (PCI).
METHODSSeventy ACS patients scheduled to receive PCI were assigned to two groups. Around PCI, the 34 patients in the control group were treated with conventional therapy and the 36 in the DHI group treated with conventional therapy combined with DHI treatment given from the day of PCI to the 14th day after operation at the dose of 40 mL per day by dissolving in 250 mL of 5% glucose for venous dripping. Plasma levels of ET-1, sP-sel and hs-CRP were detected using ELISA at various time points: before PCI (T0), after PCI (T1), 24 h (T2) and 2 weeks (T3) after PCI. The outcomes were compared between the two groups, and compared with those obtained from 20 healthy persons set as the normal control.
RESULTSAll the three indices in the two patients' group at T0, T1 and T2 were higher than those in the normal control respectively (P < 0.01), while at T3, the indices in the DHI group were lower than the normal control (P > 0.05). Comparisons between the two patients' group showed that the indices were not different at TO, T1 and T2, but at T3, all were lower in the DHI group than in the control group (P < 0.05, P < 0.01).
CONCLUSIONSCombined conventional therapy with DHI for 2 weeks can significantly reduce the plasma levels of ET-1, sP-sel and hs-CRP in ACS patients after PCI, suggesting that DHI has certain effects in protecting the endothelial function, inhibiting platelet activation and suppressing inflammatory reaction.

6.Analgesia and Anti-inflammatory Effect of Total Flavonoids Extracted from Euphorbia prolifera

Objective The study aimed to investigate the best extraction process and the analgesia and anti-inflammatory properties of total flavonoids from Euphorbia prolifera. Methods Through single factor experiment and orthogonal test,factors that affect the extraction yield of total flavonoids were studied,including the extraction time,solid to liquid ratio, ethanol concentration and extraction times. The mice models of ear edema induced by xylene and twisting induced by acetic acid were used to evaluate the analgesia and anti-inflammatory effects of different extractions from Euphorbia prolifera. Results Total flavonoids were extracted by the refluxing method,and the optimum conditions were extracting for 3 hours,solid to liquid ratio of 1:30,ethanol content of 60% in the solvent,and processed for 2 times. The highest extraction yield of total flavonoids from Euphorbia prolifera was 5. 63%. The ethanol,ethyl acetate and n-butanol extracts decreased twisting and ear swelling in mice. Conclusion The extraction for total flavonoids from Euphorbia prolifera is simple,efficient and reproducible. The ethanol,ethyl acetate and n-butanol extracts of total flavonoids have obvious analgesia and anti-inflammatory effects.
7.Effects of simultaneous bilateral central venous intubation on chest drainage for bilateral malignant pleural effusion induced by primary non-small lung cancer

Objective To observe the effects of simultaneous bilateral central venous intubation on chest drainage for bilateral malignant pleural effusion induced by primary non-small lung cancer and summarize the nursing measures. Methods Totally 65 patients with bilateral malignant pleural effusion were randomized into experiment group and control group. In the former group, chest drainage was done using bilateral central venous intubation and in the latter, unilateral chest drainage was used. The two groups were compared in terms of effectiveness rate, time for intubation, remission duration and hospitalization. Results The time for remission duration, hospital stay and time for intubation were all significantly shorter than those of the control group (all P<0.05). There were no significances in curative effect and adverse reactions between the two groups. Conclusion Chest drainage using simultaneous bilateral intubation for bilateral malignant pleural effusion induced by primary non-small lung cancer can be effective in shortening the time for remission, intubation and hospitalization. But it causes no more adverse reaction.
8.Preparation of andrographolide-loaded solid dispersions

AIM To prepare the andrographolide-loaded solid dispersions.METHODS Differential scanning calorimetry (DSC) and X-ray powder diffraction (XRPD) were used for the analysis of solid dispersions previously prepared from the mixture of andrographolide and PVPK30 (1 ∶ 9) in autoclave.In addition to the evaluation index of in vitro dissolution rate,the influencing factors of pressure,temperature and reaction time were taken into consideration for the preparation optimization by Box-Behnken method.RESULTS Andrographolide was totally dispersed in solid dispersions in an amorphous state,manifesting an inhibited crystal diffraction peaks' formulation.Under the optimal conditions of 21.26 MPa for pressure,40.76 ℃ for temperature,and 1.13 h for reaction time,the in vitro dissolution rate was 85.49%.CONCLUSION After andrographolide is made into solid dispersions,its in vitro dissolution rate is obviously increased.
9.Effects of ?-lipoic acid on bone metabolism in lead-poisoned juvenile rats

BACKGROUND:Previous study has proved that implantation of the tissue-engineering bone with vascular bundles or sensory nerve tracts can promote bone formation, which may be related to the expression of sensory neuropeptide receptors. OBJECTIVE:To investigate the effect of implantation of tissue-engineered bone containing vascular bundles and sensory nerve tracts on the middle-and long-term expression of calcitonin gene-related peptide type Ⅰ receptor (CGRP1R) in the repair of rabbit large femoral defects. METHODS:Thirty-six New Zealand white rabbits were enrol ed and modeled into 1.5 cm femoral defects, and then randomized into three groups. Bone marrow mesenchymal stem cells (BMSCs) were subjected to osteoblastic induction for 7 days, and then seeded onto the β-calcium phosphate scaffold to construct the tissue-engineered scaffold.In sensory nerve group, the tissue-engineered scaffold was implanted into the defect region, and autologous sensory nerve bundles were implanted into the lateral groove of the tissue-engineered bone;in vascular bundle group, the tissue-engineered scaffold was implanted, and autologous femoral blood bundles were implanted into the lateral groove of the tissue-engineered bone;in blank control group, only the tissue-engineered scaffold was implanted. X-ray examination, immunohistochemistry and real-time PCR were performed at 24 and 48 weeks postoperatively. RESULTS AND CONCLUSION:The X-ray scores in the sensory nerve and vascular bundle groups were significantly higher than that in the control group (P<0.05), but no significant difference was found between sensory nerve and vascular bundle groups. Real-time PCR found that the expression level of CGRP1R mRNA in the vascular bundle group was significantly higher than that in the other two groups (P<0.05), but showed no significant difference between sensory nerve and blank control groups.Immunohistochemistry findings showed that CGRP1R positive expression rate in the sensory nerve and vascular bundle groups was higher than that in the blank control group. These results reveal that implantation of the tissue-engineered bone containing vascular bundles can promote the CGRP1R expression.
