1.Observation of TCM Syndromes of 200 Cases of Upper Airway Cough Syndrome

Objective To research the pattern distribution of TCM syndromes in upper airway cough syndrome(UACS)(source of the nose).Methods Two hundred cases of UACS(source of the nose) were observed to analyze the pattern distribution of TCM syndromes.Result The pattern distribution of TCM syndromes was as follows:wind-phlegm obstruction lung syndrome 76 cases(38%),wind-phlegm and stagnant heat syndrome 58 cases(29%),wind-cold and dampness syndrome 34 cases(17%),lung dry and Yin deficiency syndorme 32 cases(16%).Analysis of nose diseases history and TCM syndrome type showed that allergic rhinitis,chronic nasosinusitis and non-allergic rhinitis were main causes of UACS(source of the nose),account for 46.0%,25.5% and 14.5% respectively.Conclusion Wind-phlegm obstruction lung syndrome and wind-phlegm and stagnant heat syndrome were the most common syndromes of UACS.
2.Advancement of research on the role and signal transduction pathway of gastrin in gastroenteric tumor

Studies have proved that gastrin plays key roles in the development and progression of gastroenteric tumor. After gastrin combined with the tumor cell surface receptors, some genetic transcription and expression were adjusted through certain signal transduction pathways, resulting in the development and growth of gastrointestinal carcinoma. Effective treatment method would been founded about gastroenteric tumor through the studies of mechanism of action and signal transduction pathways of gastrin in gastroenteric tumor.
3.Expression of osteopontin and collagen type Ⅰ in rat myocardial fibroblasts infected by coxsackievirus B3

Objective To study the expression of osteopontin (OPN) and collagen type Ⅰ in the rat myocardial fibroblasts infected by coxsackievirus B3 (CVB3) and to explore the possible pathogenesis of OPN on viral myocarditis.Methods Cardiac fibroblasts were isolated from neonatal rat and cultured with an traditional method.The primary cultured rat myocardial fibroblasts were infected by CVB3 which multiplicity of infection (MOI) was 0.5 PFU/cell.The myocardial fibroblasts were divided into control group 12 h and CVB3 groups (infected after 12 h,1 d,2 d,3 d).The expression of OPN and collagen type Ⅰ were detected by RT-PCR,Western Blotting and immunohistochemistry.And the linear correlation between the expression of OPN and the expression of collagen type Ⅰ was analyzed.Results ( 1 ) The gray scale values of the OPN/β-actin in control group(12 h)and viral groups (12 h,1 d,2 d,3 d) were 0.38 ± 0.06,0.56 ± 0.06,0.72 ± 0.05,0.98 ± 0.06,0.86 ± 0.02 respectively with RT-PCR,and were 0.26 ± 0.03,0.36 ± 0.03,0.52 ± 0.04,0.76 ± 0.05,0.62 ± 0.02 respectively with Western Blotting.The expression of OPN was found to be increased after 12 h infection,reached to the maximum after 2 d infection and displayed a decreased tendency after 3 d infection.There was significant difference on the gray scale values of the OPN/β3-actin ( F=74.965,53.004,respectively,P < 0.05 ).(2) The gray scale values of the collagen type Ⅰ/β-actin in control group (12 h)and viral groups (12 h,1 d,2d,3 d) were 1.12 ± 0.03,1.18 ± 0.01,1.22 ± 0.02,1.33 ± 0.02,1.28 ± 0.03,respectively with RTPCR.These results suggested that the collagen type Ⅰ expression started to increase at 12 h when infected by CVB3,reached to the maximum at 2 d,and then decreased after 3 d infection,the difference was significant ( F =38.241,P < 0.05).(3) The expression of OPN was positively correlated with the expression of collagen type Ⅰ ( r=0.948,P < 0.001 ).Conclusion CVB3 can induce the expression of OPN and collagen type Ⅰ in the rat myocardial fibroblasts and the expression of OPN and collagen type Ⅰ displays positive correlation.It suggests that OPN can promote the expression of collagen type Ⅰ in myocardial fibroblasts,and may play an important role in the pathogenesis of viral myocarditis.
4.Effect of Kanglaite injection on immune function of rats with Lewis lung carcinoma

Objective To investigate the effect of Kanglaite injection (KLT) on immunological function of rat models with Lewis lung carcinoma. Methods Forty C57BL/6 mice were used to establish Lewis lung carcinoma models and divided randomly into the high dose(25 mL/kg), middle dose (12.5 mL/kg) and low dose (6.25 mL/kg) of KLT groups and model group(n=10). The mice in the KLT groups were sacrificed after injecting corresponding dose of KLT with intraperitoneal injection for 14 d. No treatment was performed on the rats in model group. The body weight, tumor and spleen weight was weighed, then the ratio of tumor restriction and the index of spleen was calculated. MTT colorimetric method and ELISA were used to detected activity of T cell proliferation and expression of IL-2 in spleen. The expression of NF-κB and IκBα protein was detected by Western blot. Results The ratio of tumor restriction in the high, middle, low dose of KLT groups decreased gradually. The indexes of spleen of the high and middle dose of KLT groups were higher than those in the model group (P<0.05 or P<0.01).Compared with the model group, the activity of T cell proliferation in the high, middle, low dose of KLT groups and the expression of IL-2 in the high and middle dose of KLT groups was increased significantly (P<0.05 or P<0.01). The expression of NF-κB protein in the nuclei of high, middle, low dose of KLT groups increased dose-dependently, and the expression of NF-κB and IκBα protein in the cytoplasm decreased dose-dependently. ConclusionKLT could enhance immunological function by effecting T cell proliferation, expression of IL-2, NF-κB and IκBα, while restricting tumor growth in Lewis lung carcinoma models.

7.Effects of Kanglaite injection on antitumor and expression of EGFR in Lewis lung carcinoma

AIM:To research the effects of Kanglaite(KLT)injection on antitumor and expression of epidermal growth factor receptor(EGFR)in Lewis lung carcinoma(LLC).METHODS:A total of 40 C57BL/6 mice were randomly divided into four groups,including model group,KLT 6.25,12.5,25 mL/kg groups.All the mice were injected with 2?106 LLC cells at 1st day.At the next day,mice in the KLT groups were peritoneally injected with KLT for 14 days.And at the 16th day,all the mice were sacrificed and the tumors were harvested.The tumor weight was measured and the tumor growth inhibition rate was analyzed.The expression of EGFR was detected by westernblot.RESULTS:The growth of LLC was significantly inhibited by KLT,the changes of tumor appearance and histology were affected and the expression level of EGFR was decreased.CONCLUSION:KLT injection could obviously inhibit the growth of LLC,whose mechanism maybe related to the inhibition of EGFR expression.
8.Protective effects of penehyclidine hydrochloride on A549 cells against oxidative injury

Objective To explore the effects of penehyclidine hydrochloride (PHC) on lung epithelial type Ⅱcells against oxidative stress damage. Methods A549 cells treated with H2O2 were used as oxidative stress damage cell model. A549 cells were divided into 3 groups: control group (C group), H2O2 treated group (H group) and PHC treated group (P group). The viability of A549 cells was measured by MTT assay. The apoptotic rate was measured by TUNEL assay. The lev?els of malonicdialed (MDA), reactive oxygen species (SOD), reduced glutathione hormone (GSH) and nicotinamide adenine dinucleotide phosphate (NADPH) of cells were detected by biochemistry colorimetry. Results Compared with group C, the viability of A549 and the contents of SOD, GSH and NADPH were significantly decreased in group H, while MDA content and apoptotic rate were increased (P<0.05). Compared with group H, the viability of A549, the contents of SOD, GSH and NADPH were significantly increased in group P, while MDA content and apoptotic rate were reduced (P<0.05). Conclu?sion Penehyclidine hydrochloride shows protective effects on A549 cells through reducing the oxidative damage induced by H2O2.
9.Docetaxel combined with cisplatin in patients with advanced non-small-cell Lung carcinoma

Objective To evaluate the efficacy and toxicity of weekly docetaxel and cisplatin in previously untreated patients with advanced non-small-cell lung carcinoma. Methods Between January 2002 and December 2003 ,34 patients with pathologically comfirmed advanced non-small-cell lung carcinoma who had not received treatment were enrolled. The mean age was under 66 years. The patients received intravenous infusions of docetaxel(25 mg/m2,dayl ,8,15) with dexamethasone premedication and cisplatin(25 mg/m2,dayl ,8,15) ,followed by a week of rest. The remedies which were less than 6 regimens lasted to disease progression or severe toxicity. Therapeutic effect was evaluated by CT scan every two courses . The patients were followed up for 24 months. Descriptive statistics and SPSSIO. 0 software were used to analyse the results. Results 34 patients finished 90 courses. The mean was 2. 6 courses. All patients were followed up. Two patients achieved complete responses, ten patients achieved partial responses, ten patients achieved stable disease. An objective response rate of 35. 29% (95% confidence interval 19. 25%-51. 33% )was obtained. Patients life quality was significantly improved. The median time to progression was 4. 1 months, and median overall survival was 11 months. The 1-year survival rate was 47. 06% , the 2-year survival rate was 11.76% . Toxicities were mild. Grade 3 to 4 neutropenia (11.76%), anemia (5.88%), hyponatremia (5.88%), alopecie (17.64%) and nausea/vomiting (5. 88% ) were observed. Conclusion Weekly Cisplatin plus docetaxel is an effective and well-tolerated regimen in chemo-naive patients with advanced NSCLC. Well-designed clinical trials should be conducted.
