OBJECTIVE To observe the increased susceptibility to metabolic syndrome (MS) in offspring adult female rats which experienced prenatal caffeine exposure (PCE) and underwent early postnatal catch-up growth and late chronic stress, and to explore the underlying mechanism. METHODS Starting from gestational day 11, pregnant Wistar rats were intragastrically administered with caffeine at the dose of 120 mg·kg-1 per day until delivery. The female offspring rats were fed a high-fat diet from postnatal week 4 (PW4) to PW24, and then exposed to two weeks of unpredictable chronic stress at PW38-PW40. Blood glucose and serum adrenocorticotropic hormone, corticosterone, insulin, triglycer? ides, total cholesterol, low-density lipoprotein-cholesterol (LDL-C) and high-density lipoprotein-cholesterol (HDL-C) levels were detected. Meanwhile, the mRNA expression of adrenal steroidogenic acute regulatory protein, P450 side- chain cleavage enzyme, 3β- hydroxysteroid dehydrogenase, steroid 11β- hydroxy? lase, steroid 21β- hydroxylase, hepatic insulin receptor, insulin receptor substrate 2 and glucose trans? porter 2 (GLUT2) were examined by real- time quantitative PCR. The morphological changes of the adrenal gland, pancreas and liver were observed using hematoxylin-eosin staining. RESULTS Compared with control group〔(13.9±2.8) g〕, the body mass of the PCE offspring female rats at PW1〔(10.5±1.0) g〕 was significantly lower (P<0.01), which lasted until PW40 (P<0.05, P<0.01). However, the gain rate of body mass was higher in the PCE group at PW4- PW16 (P<0.05, P<0.01). Levels of blood glucose〔(5.9±0.3) mmol·L- 1〕and serum insulin〔(100±31) mU·L-1〕, the insulin resistance index (26.3±5.7), LDL-C〔(0.55±0.05) mmol·L-1〕level and LDL-C/HDL-C ratio (0.87±0.11) were increased compared with (4.3±0.3) mmol·L-1, (45±4) mU·L-1, 8.3±0.9, (0.38±0.04) mmol·L-1 and 0.66±0.07 in the control group, respectively (P<0.05, P<0.01). The mRNA expression of hepatic GLUT2 in the PCE group was lower than in the control group (P<0.05). Furthermore, the thicknesses of the adrenal zona fasciculata was re?duced and the area of pancreatic islets became smaller, but there was no significant change in liver morphology. CONCLUSION PCE offspring adult female rats display high susceptibility to MS, which is mainly manifested as insulin resistance, characterized by hyperglycemia and hyperinsulinemia, lipid metabolism disorder and structural and functional abnormalities of multiple organs. The mechanism is possibly related to the disorder of hypothalamic-pituitary-adrenal axis-associated neuroendocrine meta? bolic programming.

The way of "extracting-salting-chromatography" was used to purify the phycoerythrin and phycocyanin from Porphyra yezoensis in process scale-up.First,by comprehensive comparison of efficiency, the Sephadex G-25 was selected from four resins (Sephadex G-25、G-100、S-300 and CL-6B) as the best choice used in crude extract desalting of phycobiliprotein. Then the preparation process of phycobiliprotein was scaled-up with raw material(Porphyra yezoensis) increased from 1g to 20g,and finally to 400g.The results indicated that the yields of purified phycoerythrin and phycocyanin (absorption spectra purity above 3.2) increased during according to process scale-up,with 0.323% phycoerythrin and 0.148% phycocyanin obtained from 400g frozen Porphyra yezoensis blades respectively. It is no doubt that the process involved in the experiment is a potential way for large scale preparation of phycobiliproteins of high purity.

Objective:To study the effect of mindfulness-based training intervention on clinical efficacy in patients with nitrous oxide(laughing gas) addiction. Methods:From June 2019 to June 2020, sixty-six patients with nitrous oxide addiction in Beijing Gaoxin Hospital were selected and randomly divided into experimental group( n=33) and control group( n=33). The control group received Taijiquan training and physical training, while the experimental group added mindfulness-based training intervention on the basis of Taijiquan training and physical training.Symptom checklist 90 (SCL-90) scores and visual analog scales (VAS) craving scores were compared between the two groups at admission and 8 weeks after treatment.SPSS 22.0 software was used for statistical analysis.Independent sample t test and paired sample t test were used to compare the differences between groups and within groups. Results:(1)Before treatment, there were no significant differences in subscale scores of SCL-90 between the two groups except for depression factor((2.45±0.86), (2.03±0.46), t=2.474, P<0.05). After treatment, the subscale scores of somatization((1.38±0.35), (1.68±0.34), t=-3.656, P<0.05), phobic anxiety((1.49±0.37), (1.81±0.30), t=-3.993, P<0.05), paranoid ideation((1.50±0.47), (1.88±0.31), t=-3.898, P<0.05) and psychotism((1.34±0.54), (1.55±0.27), t=-3.094, P<0.05) of SCL-90 in the experimental group were significantly lower than those in the control group.(2)Before treatment, there was no significant difference in VAS craving score between the two groups( t=0.857, P=0.395). After treatment, the score of VAS in the experimental group was significantly lower than that in the control group( t=27.427, P<0.05). Conclusion:Mindfulness training intervention can effectively improve the clinical symptoms of patients with nitrous oxide addiction, which is worthy of clinical application.

This study aimed to assess the relationship of OAS2 rs739901 5'-flanking C/A polymorphisms with the susceptibility to Enterovirus-71 (EV71) infection. We investigated 294 hand-foot-mouth disease (HFMD) Chinese children with EV71 infection (165 mild cases and 129 encephalitis cases). The improved multiplex ligation detection reaction (iMLDR) technique was used to test the genotypes. In EV71-infected patients, the CA genotype distribution (P=0.007),A allele frequency (OR 1.32,95% CI 1.0-1.7,P=0.034)and CA+AA carriage frequency (P=0.003) of OAS2 rs739901 5'-flanking were obviously elevated as compared with controls, but there were no statistically significant differences between mild cases and encephalitis cases. In EV71-infected patients, the counts of white blood cells (P=0.034) and blood glucose concentrations (P=0.042) were raised in A carriers (CA+AA).Among different genotypes of encephalitis cases, the contents of cerebrospinal fluid (CSF) showed no significant differences.IFN-γ levels in EV71-infected patients were higher than those in controls (mild group vs.control group,P<0.01;encephalitis group vs.control group,P<0.001).In encephalitis cases,IFN-γ levels were reduced (P<0.05) in A carriers compared to CC genotype, however, there were no significant differences between genotypes CA and AA (P=0.226).These findings suggest that OAS2 rs739901 5'-flanking C/A genetic polymorphisms involve the susceptibility to EV71 infection, and A allele might be a risk factor of the susceptibility to EV-71 infection.
