Aim To observe the influence of haloperidol on QT interval and effects on L-type calcium channel mRNA expression.Methods By surface ECG technique and RT-PCR technique observe the influence of Various doses haloperidol to New Zealand rabbits ECG.And L-type calcium channel mRNA.Results Haloperidol elongated the QT interval.Haloperidol influence QT interval mainly happened after from 0 minutes to 120 minutes.360 minutes later,QT interval turned to normal standard.RT-PCR result display haloperidol obviously increase L-type calcium channel mRNA expression.Conclusion Haloperidol can prolong the QT interval.and it's mechanism concerned with increase L-type calcium channel mRNA expression.So,it can increase the risk of ventricular arrhythmias occur.

Objective To explore the experimental condition for hepatocellular steatosis models of Changliver cell induced by o‐leic acid (lleic acid ,OA) .Methods Changliver cells were induced by different concentration of oleic acid for different periods .MTT was used to detect hepatic cell activity ,oil red 0 staining was used to observe intracellular lipid droplets acumulation ,glycerin 3 phosphate oxidase method was applied to detect the contents of triglyceride (TG) in the Changliver cell .Results Hepatocellular steatosis models of Changliver cell can be established successfully by 0 .2 mmol/L OA inducing for 24 hours .TG content in model cells was (379 .98 ± 23 .19)mg/g ,however ,it was (185 .03 ± 12 .68)mg/g in control cells ,the difference was statistically significant (P< 0 .01) .Conclusion The proper condition for establishing hepatocellular steatosis models is 0 .2 mmol/L OA inducing Changliver cells for 24 h .This model is the reliable choice for nonalcoholic fatty liver disease research .
4. Comparative study of different extract methods for protein and peptides from Chinese medicine bovine bone marrow

Objective: To study the extraction method, structure information, and antimicrobial activity of the protein and polypeptide components from Chinese medicine bovine bone marrow (BBM) for improvement of its medicinal value. Methods: Ten kinds of extraction and three kinds of preparation methods were used to take the protein content, extraction rate, degree of hydrolysis, antimicrobial activity, and molecular weight as investigation index to screen optimal extraction ability and preparation parts of bovine bone marrow protein (BBMP) and polypeptide. Results: The comprehensive evaluation index of protein extracted in water, NaCl, and Tris-HCl solutions was high, their contents were (52.20 ± 0.58), (50.19 ± 0.21), and (43.21 ± 0.05) mg/mL, respectively. The antimicrobial ability was related to protein content. The SDS-PAGE analysis showed that the proteins from BBM contained approximately secondary subunits which related to protein and peptide with molecular weight in the range of 66 000 and 4 100- 10 000. The LC/MS data showed that the molecular weight of peptide extracted in water, 0.5 mol/L sodium chloride, and 30% (NH4)2SO4 solutions were 1 027.84-9 916.32, 1 131.17-4 989.95, and 1 150.46-9 917.00, respectively, and the results were basically consistent with 1-DE electrophoresis bands. Conclusion: Water extraction method is simple, and fast with relatively low cost, which is suitable for industrial production of BBMP. The SDS-PAGE and LC/MS method is adopted to analyze and provide a theoretical basis for construction of BBMP fingerprint.
5.Effects of two different extracts of total saponins from Cicer arietinum on kidney of T2DM rats.

Protective effects of two different extracts of TSCA (total saponins from Cicer arietinum) were studied on kidney of T2DM rats. The diabetic model group was established with high calorie feeding and small dose injection of streptozotocin (STZ, 45 mg x kg(-1)). DM rats were randomly assigned to model group (feed with propylene glycol 1 mL/100 g), TSCA high dose group (300 mg x kg(-1)), TSCA low dose group (100 mg x kg(-1)) and normal control group (feed with propylene glycol 1 mL/100 g). After four weeks treatment with TSCA I and II, the levels of FPG FIns, BUN, Scr, ATII, ET-1, TXB2 and 6-keto-PGF1alpha in blood and the activities of SOD, GSH-PX and MDA in kidney were analyzed by biochemical methods. After four weeks treatment with TSCA II, the levels of FPG FIns, BUN, Scr, ATII and ET-1 were reduced significantly; and the ratios of TXB2 to 6-keto-PGF1alpha and SOD were effectively alleviated in TSCA II group. While there is no significant change on FPG and BUN in comparison to the rats treated with TSCA I, Scr, ATII, ET-I, GSH-PX and SOD were alleviated. The results suggest that TSCA II could be used to reduce FPG and FIns. According to the result of vasoactive substances index, TSCA II is more effective than TSCA I on renal protection of DM rats.

8.Effects of pomegranate flowers polyphenols on liver PON expression of diabetes combining non-alcoholic fat liver diseases rats.

To investigate the protective effects and possible mechanism of pomegranate flowers polyphenols (PFP) on liver function of rats with diabetes combining non-alcoholic fat liver diseases, diabetes combining nonalcoholic fat liver disease model rats were established with high calorie feeding and small dose intraperitoneal injection of streptozotocin (STZ). Model rats were randomly divided into: model group, metformin group, pomegranate flowers polyphenols low, medium and high dose group (75, 150 and 300 mg x kg(-1)). After four weeks treatment, the levels of FPG, blood fat profiles and serum insulin, ALT, AST levels, SOD and MDA in the liver and serum separately were analyzed with biochemical methods. Paraoxonase (PON1 and PON3) mRNA and protein expression in liver were checked by RT-PCR and immunohistochemical method. Pathological changes of the liver were observed. FPG, IRI, non-HDL-C and transaminase significantly reduced and HDL-C raised in the each PFP dose group; Furthermore, compared with model group, fat drops in liver cells significantly reduced, antioxidant ability enhanced, PON1 mRNA and protein expression level in liver increased significantly. The protective effects of PFP against diabetes combining non-alcoholic fat liver diseases rats might through the increase liver PON1 mRNA and protein expression further enhanced the body antioxidant capacity and reduced IRI so as to ameliorate the rat hepatic steatosis.

9.Value of CT grayscale histogram features in the differential diagnosis of brucella spondylitis and pyogenic spondylitis

Objectives:To evaluate the values of sagittal CT image histogram features in the differential diagnosis of brucella spondylitis(BS)and pyogenic spondylitis(PS).Methods:The data of 40 BS patients[25 males,15 females;age:51.6±13.0 years old;body mass index(BMI):23(20,28)kg/m2,the BS group]and 33 PS patients[13 males,20 females;age:50.8±16.7 years old;BMI:23(20,26)kg/m2,the PS group]who underwent CT examination of the spine in our hospital and were confirmed through pathology and/or etiology were collected.The region of interest(ROI)was delineated on each level of the sagittal CT images of the two groups of patients by using the 3D Slicer platform and grayscale global histogram analysis was performed.The clinical data were compared using chi square test,independent sample t-test,and Mann Whitney U test between the two groups of patients;Univariate analysis,correlation analysis,and multivariate analysis were used in sequence to identify the histogram features with significant differences between the two groups(including 10%percentile,1%percentile,25%percentile,5%percentile,median,minimum,skewness,and variance);Logistic regression and the screened features were combined for modeling,and receiver operating characteristic(ROC)curves were drawn and areas under the curve(AUC)were calculated to compare the discriminative ability of each histogram feature.Results:There was no statistically significant difference in age,gender,and BMI between the two groups of patients(P>0.05).Among the histogram parameters,10%percentile value,1%percentile value,25%percentile value,5%percentile value,median,minimum value,skewness,and variance were statistically different between the two groups(P<0.05).The 10%percentile value displayed the best diagnostic performance,with an AUC value of 0.824 and a specificity of 0.893.The combined model had an AUC value of 0.860 and a specificity of 0.946.Conclusions:Based on 10%percentile value of CT grayscale histogram and joint model,PS and BS can be distinguished effectively,providing a basis for accurately distinguishing the two diseases in clinical practice.
