1.ROS-mediated mechanisms:an anticancer strategy

Under the influence of oncogenic signals and abnormal metabolism, the redox status of cancer cells often differs from that of the normal cells, manifesting as elevated generation of reactive oxygen species( ROS) and oxidative stress.Many signaling pathways involved in tumorigenesis can directly or indirectly regulate ROS metabolism.Currently, the biological significance of increased ROS in cancer cells is still somewhat controversial.ROS on the one hand can promote cancer development and drug resistance, and on the other hand can also cause cell injury and even cell death.To maintain cell viability and proliferation, cancer cells usually up-regulate their antioxidant capacity in adaptation to the intrinsic oxidative stress.Such adaptive mechanisms to oxidative stress are highly important in both cancer development and also play a major role in cancer cell response to therapy.Compelling evidences from recent studies have suggested that targeting the redox regulation mechanisms through proper intervention strategies may have significant therapeutic implications in cancer treatment.This article will focus on alterations of redox status in cancer cells, their adaptation to oxidative stress, and the underlying mechanisms.Potential therapeutic strategies based on such biochemical characteristics of cancer cells will also be discussed.
2.Effect of Jian Pi Bu Shen prescription on the inflammatory factors and iron metabolism in the brain of type 2 diabetes model mice

Objective To evaluate the effect of Jian Pi Bu Shen prescription on inflammatory factors and iron metabolism in the brain of T2DM model mice.Methods A total number of 30 healthy 12-week-old male mice were used in the present study.The groups were randomly assigned to one of the following experimental groups:(1) control group:n=5,the mice were maintained on a normal diet for 4 weeks;(2) model group:n=25,the mice were maintained a high fat diet for 4 weeks.Then,the mice were deprived of food for 12 hours before a single intraperitoneal injection of streptozotocin (STZ,30 mg/kg).Then,the blood glucose levels were measured randomly 3 times within 24 hours after injection.The mice whose blood glucose was less than 16.7 mmol/l would receive another single intraperitoneal injection of STZ.Finally,we got a total number of 19 mice meeting the criteria of animal model we described above.The final 19 mice were randomized to 2 groups:diabetes group (n=10)and Jian Pi Bu Shen (JPBS) group(n=9).JPBS group received gavage administration of JPBS Prescription 7.4 g/kg/d(8weeks).Diabetes group and control group were maintained treated with saline for 8 weeks.Mice were decapitated 24 hours after the last drug treatment.The mice brain tissue slices were prepared for pathological observation.To examine the effects of JPBS prescription on neuroinflammation and iron metabolism in cerebrum and hippocampi,the relative mRNA levels of IL-6,IL-1β,DMT1,FPN1 and CP were tested by RT-PCR.Results mRNA levels of IL-6,IL-1β and DMT1 in the brains of diabetes group were higher while the levels of FPN1 and CP were lower than that of the control group (P<0.01).Compared with diabetes group,in JPBS group,mRNA levels of IL-6,IL-1β and DMTl in cerebrum and hippocampi were decreased while the levels of FPN1 and CP were increased (P<0.01).The brain tissue slices of diabetes group showed neuron loss and signs of neurodegeneration.But JPBS group attenuated neurodegenerative change.Conclusion JPBS prescription can protect neuron from apoptosis,suppress neuroinflammation and attenuate iron metabolism,which may be one of the mechanisms of traditional Chinese medicine in treating cognitive dysfunction.
3.Performance and reliability of VITEK 2-Compact GN13 method and its Advanced Expert System validation for testing imipenem susceptibility of Klebsiella pneumoniae

Objective To evaluate the performance of VITEK 2-Compact GN13 methods for testing imipenem susceptibility of Klebsiella pneumoniae and assess the reliability of its Advanced Expert System (AES).Methods A retrospective study was conducted with a total of 157K. pneumoniae strains, which were isolated from blood and intra-abdominal infections in the First Affiliated Hospital of Nanchang University from 2014 to 2015. Thein vitro minimum inhibitory concentration (MIC) values of imipenem were determined by disc diffusion, VITEK 2-Compact GN13 and broth microdilution methods, respectively. Categorical agreement (CA) rates of disc diffusion and VITEK 2-Compact GN13 methods were determined using broth microdilution as reference method. The genes encoding ESBLs and carbapenemase were screened by PCR and sequencing analysis. The phenotypic confirmatory tests such as modified Hodge test, PCR and DNA sequencing were used to confirm the resistance mechanism and evaluate the reliability of AES in interpreting the imipenem susceptibility of K. pneumoniae.Results Among the 157 isolates, 64 and 8 were identified as resistant and intermediate strains by broth microdilution method, respectively; 52 and 10 were tested as resistant and intermediate strains by disc diffusion method, respectively; 54 and 13 were determined as resistant and intermediate strains by VITEK 2-Compact GN13 method, respectively, while 70 and 3 were judged as resistant and intermediate strains by VITEK 2-Compact GN13 method plus AES validation. The CA of disc diffusion and VITEK 2-Compact GN13 methods compared with broth microdilution method were all higher than 90 %. However, the major error (ME) rate was 3.8 % and very major error (VME) rates were all 0.6 % in imipenem susceptibility testing by VITEK 2-Compact GN13 and disc diffusion. The imipenem susceptibility of 16 strains were modified by the AES, which eliminated 0.6 % VME, but increased major error by 1.3 % and minor error by 1.9 %. Phenotypic confirmatory tests showed that 75 % (12/16) of these strains were validated as producers of both ESBLs and carbapenemase, which was consistent with the result of AES validation. PCR and DNA sequencing analysis proved that 62.5 % (10/16) of these strains produce IMP-4/KPC-2 /NDM-1 and ESBLs.Conclusions Both disc diffusion and VITEK 2-Compact GN13 methods can be used for testing the imipenem susceptibility of K. pneumoniae isolates with reliable and accurate results. Attention should be paid to the possibility of ME and VME when testing imipenem susceptibility. The VME can be avoided by the AES mechanism. However, AES intervention will increase ME and minor error, which may be associated with decreased expression of carbapenemase.
4.Thermosensitive hydrogel with Aliskrien influence the heart function of rats with acute myocardial infarction

BACKGROUND:Previous experiments have shown that the thermosensitive hydrogel is safe, nontoxic, biocompatible, and has a certain cardioprotective effect, which can be used as a drug carrier injected into the myocardial tissue, in order to avoid adverse reactions produced by systemic administration. <br> OBJECTIVE:To observe the influence of thermosensitive hydrogel with Aliskrien on heart function, apoptosis and ventricular remodeling of rats with acute myocardial infarction. <br> METHODS:Total y 70 Wistar rats were randomized into sham group (n=10), PBS group (n=15), PBS+Aliskrien group (n=15), hydrogel group (n=15), hydrogel+Aliskrien group (n=15). Models of acute myocardial infarction were established in the latter four groups. After modeling, PBS, PBS with Aliskrien, hydrogel, and hydrogel with Aliskrien were respectively injected into the border of infracted myocardium. After 28 days, relevant measurements were performed. <br> RESULTS AND CONCLUSION:Compared with the sham group, the heart function and expression of tissue inhibitor of metal oproteinase-1 were decreased significantly in the other four groups (P<0.05), while the number of apoptotic cells, matrix metal oproteinase-2, matrix metal oproteinase-9, transforming growth factorβ, col agen type I and III were increased (P<0.05). The levels of angiotensin I and angiotensin II were higher in the PBS, PBS+Aliskrien, hydrogel groups than the sham group (P<0.05), while they were decreased in the hydrogel+Aliskrien group than the sham group (P<0.05). The level of tissue inhibitor of metal oproteinase-1 and heart function in the hydrogel+Aliskrien group were superior to those in the PBS, PBS+Aliskrien, and hydrogel groups (P<0.05), while the number of apoptotic cells, the levels of metal oproteinase-2, matrix metal oproteinase-9, transforming growth factorβ, col agen type I and III were lower than those in the PBS, PBS+Aliskrien, and hydrogel groups (P<0.05). These findings indicate that as a new type of sustained-release formulation, thermosensitive hydroge with Aliskrien could further improve the heart function, decrease cellapoptosis and inhibit myocardial fibrosis, and delay ventricular remodeling after myocardial infarction.
5.Bone marrow stem cell transplantation for improving heart function of patients with acute myocardial infarction:a systematic review

BACKGROUND:Although increasing studies have shown that autologous bone marrow stem celltransplantation can treat myocardial infarction, but there is a lack of large-scale multi-center randomized control ed trials to il ustrate the therapeutic effectiveness of autologous bone marrow stem celltreatment for myocardial infarction. OBJECTIVE:To systematical y evaluate the improvement of heart function in patients with myocardial infarction undergoing autologous bone marrow stem celltransplantation. METHODS:Cochrane Central Register of Control ed Trials (Central), MEDLINE, EMbase, PEDro (www.pedro.org.au), OpenSIGLE, National Technical Information Service (NTIS), CNKI, VIP database (VlP), Wanfang Data and Chinese biomedical literature database (CBM) were searched for the randomized control ed trials addressing bone marrow stem celltransplantation for heart function in patients with myocardial infarction. The bibliographies of the included studies were also searched. The time span was from database establishment to November 2012. The extracted data were analyzed by RevMan5.1. RESULTS AND CONCLUSION:A total of 14 randomized control ed trials were included. Results from the meta-analysis showed that the ejection fraction and cardiac output of patients undergoing bone marrow stem celltransplantation was significantly different from those of patients without celltransplantation (WMD=5.23, 95%CI (0.73, 9.72), P<0.01;WMD=1.69, 95%CI (1.23, 2.16), P<0.000 01). Current evidence has demonstrated that bone marrow stem celltransplantation can remarkably improve the ejection fraction and cardiac output of myocardial infarction patients, which can be clinical y recommended for improvement of heart function. Due to the limitations of the included studies, more large-sample, multi-center high-quality randomized control ed trials are required to further verify the therapeutic methods and effectiveness.
6.Pancreatic and extra-pancreatic imaging of autoimmune pancreatitis

Objective To describe the imaging findings of pancreatic and extra-pancreatic lesions of autoimmune pancreatitis (AIP),to improve the imaging diagnostic accuracy of AIP.Methods From 2011 to June 2014,38 AIP patients according to 2010 international consensus diagnostic criteria for AIP were included.Thirty-six patients underwent contrast enhanced CT scan,17 patients underwent contrast enhanced MRI,15 underwent MRCP,and the imaging data were retrospectively reviewed.Results In the 38 patients,the pancreas showed diffuse enlargement in 23,focal enlargement in 9,mixed enlargement in 6.All the patients demonstrated as progressive enhancement and gradually delayed enhancement.Fourteen cases showed diffusely irregular narrowing of the main pancreatic duct.Pseudo-capsule like structure was presented around the lesions in 24 cases.Extra pancreatic lesions included biliary duct involvement in 24 cases,renal lesions in 4 cases,peri-pancreas vessels involvement in 20 cases,8 cases with enlargement of lymph nodes.Conclusions AIP presents with characteristic imaging features in pancreas and extra pancreatic organs.Imaging tests are helpful in the early diagnosis of AIP.
7.Establishment and optimization of AFLP reaction system in Asarum sieboldii

Objective To establish and optimize AFLP reaction system used in providing necessary technique basis for genetic diversity analysis in Asarum sieboldii.Methods Leaves of A.sieboldii were used as experimental materials to analyze various essential elements of the whole process,such as quality of extracted DNA,time of restriction digest and ligation,concentration of Mg2+,primers and Taq DNA polymerase during PCR amplification etc.,so that the most optimal AFLP reaction system could be built up. Results The optimal AFLP reaction system of A.sieboldii has been constructed: in the genomic DNA extraction,mercaptoethanol being utilized and samples being incubated about 30 min at 65 ℃;the genomic DNA being digested by Trul Ⅰ and Pst Ⅰ for 3 h respectively;in PCR amplification,the final concentration of Mg2+ being 1.5 mmol/L and the volume of Taq DNA polymerase being 0.2 ?L.Conclusion The present reaction system for A.sieboldii is able to gain the favorable results of AFLP analysis and can be used in the genetic diversity research of the species.
8.Effects of bevacizumab on antioxidative function in human retinal pigment epithelial cells

Objective To investigate the effects of bevacizunab on the antioxidative function of human retinal pigment epithelium (RPE),in order to explore the possible mechanism of macular atrophy induced by the application of anti-vascular epithelial growth factor (VEGF) agents in age-related macular degeneration.Methods Human RPE cells were incubated in DMEM/F12 medium containing 0.25 g · L-1 bevacizumab and divided into 5 groups according to incubation period:0 hour(control),12 hours,24 hours,48 hours and 72 hours,and then the oxidative stress was induced by adding H2O2.Cell viability was measured by the CCK8 assay.MitoSox Red was used to determine mitochondrial reactive oxygen species (mtROS) production.Mitochondrial membrane potential was measured using the JC-1 assay.The expression levels of NOX4 and HO-1 were detected by RT-PCR and Western blot,respectively.Results CCK8 assay determination showed that the above treatment had no significant effect on cell viability,the cell viability of 0 hours,12 hours,24 hours,48 hours and 72 hours were (100.2 ±3.3)%,(99.2 ±2.7)%,(102.5 ±6.4)%,(103.9 ±3.7)%,(103.6 ±3.3)%,the difference was not statistically significant (P > 0.05).Compared with the control group,the levels of mtROS increased at 12 hours,24 hours,48 hours and 72 hours,the difference was statistically significant (P < 0.05).Mitochondrial membrane potential at 12 hours,24 hours,48 hours,72 hours were lower than the control group,the difference was significant,48 hours reached the lowest,72 hours significantly increased,but still lower than the control group.RT-PCR and western blot results demonstrated that the expression of NOX4 mRNA and protein increased at 12 hours,24 hours,48 hours and 72 hours,and reached the highest at 24 hours,then decreased significantly,but still higher than the control group,the difference was statistically significant (P <0.01).Compared with the control group,the expression of HO-1 mRNA decreased at 24 hours,48 hours and 72 hours,while the expression of HO-1 protein decreased at 48 hours and 72 hours,the difference was statistically significant (P < 0.05).Conclusion The clinical concentration of bevacizumab can reduce the anti-oxidative function of RPE cells,which may be one of the causes of progressive macular atrophy after long-term anti-VEGF therapy.
9.Analysis of iodine nutrition monitoring results in areas with iodine deficiency and high iodine in water source in Anyang City, Henan Province in 2019

Objective:To understand the level of iodine nutrition in the areas with iodine deficiency and high iodine in Anyang City of Henan Province, and to provide a basis for taking targeted prevention and treatment measures and scientific adjustment of intervention strategies.Methods:In 2019, three areas with iodine deficiency (median iodine in water < 10 μg/L) and three areas with high water iodine (median iodine in water > 100 μg/L) were selected from counties (cities and districts) in Anyang City by stratified random sampling. Children aged 8-10 and pregnant women were selected in areas with high iodine and areas with iodine deficiency, respectively. Urine samples and salt samples were collected and tested, and children's thyroids were examined by B-mode ultrasound.Results:A total of 654 salt samples were collected in iodine deficiency areas, among which non-iodized salt accounted for 1.83% (12/654). A total of 628 salt samples were collected from areas with high iodine, among which non-iodized salt accounted for 6.05% (38/628). The difference in non-iodized salt rate between the two areas was statistically significant (χ 2=15.19, P < 0.05). A total of 654 urine samples were collected from children in iodine deficiency areas, with a median of 211.15 μg/L of urinary iodine, and 628 urine samples were collected from high iodine areas, with a median of 390.50 μg/L of urinary iodine. The difference was statistically significant ( U=- 18.34, P < 0.05). A total of 300 urine samples were collected from pregnant women in iodine deficiency areas, with a median of 223.95 μg/L, and 127 urine samples were collected from pregnant women in high iodine areas, with a median of 258.00 μg/L. The difference was statistically significant ( U=- 4.07, P < 0.05). The thyroid volume of 560 children in the iodine deficient areas was detected, and the swelling rate was 1.43% (8/560). The thyroid volume of 628 children in high iodine areas was detected, and the swelling rate was 2.07% (13/628). Conclusion:The iodine nutrition level of the population in the iodine deficiency areas of Anyang City is appropriate, while the iodine nutrition level of the population in the water source high iodine areas is excessive.
10.Effects of Fucoidan on Lipidomics Profiling of Juvenile Yellow Catfish (Pelteobagrus fulvidraco) during Feeding Time

The effects of fucoidan on the lipidomics profiling of juvenile yellow catfish (pelteobagrus fulvidraco) during different feeding time (week 1, week 2, week 3 and week 8) were investigated by ultra-performance liquid chromatography-quadrupole-time-of-flight-mass spectrometry (UPLC-Q-TOF-MS) combined with multivariate data analysis, e.g.principal component analysis (PCA) and partial least squares-discriminate analysis (PLS-DA).Based on VIP and p values, 11 lipid biomarkers were screened including lysophosphatidylcholine (Lyso-PC) 16∶0, phosphatidylcholine (PC) 22∶6/16∶0, phosphatidylethanolamine (PE) 22∶6/16∶0, phosphatidylinositol (PI) 18∶0/22∶6, diacylglycerol (DAG) 16∶0/16∶0,DAG 16∶0/18∶1, DAG 18∶1/18∶1, triglycerides (TAG) 20∶5/16∶0/18∶3, TAG 20∶4/16∶1/18∶2, TAG 16∶0/18∶2/20∶5 and TAG 18∶2/14∶0/18∶1.It was found that Lyso-PC, PC, PE and PI had the largest levels at the eighth week;the levels of DAG were decreased at the second and fourth weeks, and increased at the eighth week.While TAG was different: the content of TAG 18∶2/14∶0/18∶1, TAG 20∶5/16∶0/18∶3 and TAG 20∶4/16∶1/18∶2 increased in response to fucoidan, but TAG 16∶0/18∶2/20∶5 decreased little.Therefore, for juvenile yellow catfish, fucoidan can affect its lipid metabolism, which provides a theoretical basis for investigation of the influence of fucoidan on the response mechanism of lipid metabolism of juvenile yellow catfish.
