1.Thyroid dysfunction induced by interferon α therapy in chronic hepatitis

Objective To investigate the prevalence and clinical characteristics of thyroid dysfunction induced by IFNα therapy in patients with chronic hepatitis.Methods Thyroid function of 310 patients with chronic hepatitis receiving IFNα therapy were evaluated.Results Serum free tri-iodothymnine (F13),free thyroxine(FT4)and sensitive thyroid-stimulating hormone(sTSH)of all patients were normal and TGAb was negative before IFNα therapy.After the treatment,10 patients(10/3 10,3.22%)had thyroid dysfunction,in which 7 were of hypothyroidism and 3 were of hyperthyroidism.The thyroid function of all patients recovered within 1-year follow-up.Conclusions IFNα therapy may induce thyroid dysfunction in patients with chronic hepatitis,in which prognosis can be good when appropriate treatment is given.

Objective To investigate the percentages of Th17 and CD4+CD25+FoxP3+ regulatory T(Tr) cells and the levels of related cytokines IL-6,IL-23,IL-17 and TGF-β in serum of patients with anlylosing spondylitis(AS).Methods Forty patients with AS and 37 age-matched healthy donors were studied.Flow cytometry Was used to analyze the percentages of blood Th17 and CD4+CD25+FoxP3+Tr cells.The levels of serum IL-6,IL-23,IL-17 and TGF-β were assayed by enzyme-linked immunosorbent assay ( ELISA).Results The proportion of Th17 cells in AS group was significantly higher than those in normal group [ (1.02±0.34)% vs (0.68±0.29)%,P<0.05) ],and the proportion of CD4+CD25+FoxP3+ cells was lower in AS group comparing with normal group [(3.77±0.81)% vs (4.69±1.23)%,P<0.05)].Meanwhile,serum levels of IL-6,IL-23 and IL-17 were significantly higher in AS group than those in normal group [ (6,15±2.71) ng/L vs (3.31±1.65) ng/L; (9.44±3.12) ng/ml vs (5.82±2.61) ng/ml;(10.53±4.97) ng/L vs (6.78±3.26) ng/L,all P<0.01 ].In contrast,TGF-β level was decreased in AS group compares with the normal group [ ( 4.76±2.15) ng/ml vs (5.16±2.02) ng/ml,P>0.05 ],but the difference was not significant.No associations of serum eytokine levels with clinical and laboratory parameters were found in AS.Conclusion The abnormality Th17 cells and Tr cells and their related cytokines IL-6,IL-23,IL-17 and TGF-β changes in patients with AS,which may be involved in immunological pathogenesis of AS.
5.Effect of Panax notoginseng saponins on human kidney fibroblast.

OBJECTIVETo study the effect of Panax notoginseng saponins (PNS) on cell proliferation, type I collagen secretion and integrin beta 1 expression in human kidney fibroblast (KFB).
METHODSKFB were cultured and stimulated by PNS in vitro. The cell proliferation, type I collagen secretion and integrin beta 1 expression in KFB were measured by methyl thiazolyl tetrazolium (MTT), enzyme-linked immunoabsorbent assay (ELISA) and flowcytometry respectively.
RESULTSWithin its optimal concentration range and effective time range, PNS could obviously inhibit the proliferation, type I collagen secretion and integrin beta 1 expression (all P < 0.05) in human KFB.
CONCLUSIONPNS would possibly be an effective drug for renal interstitial fibrosis prevention and treatment.

7.A study of dynamic changes of NO,NOS in serum and CSF in patients with CNS infection

Objective To observe the dynamic changes of NO,NOS in serum and CSF in patients with CNS infection and lay an experimental basis for discrimination of CNS infection due to different agents.Methods The method of nitrate redutase and the method of chromometry were employed to measure NO,NOS in serum and CSF at different time.The dynamic changes of NO,NOS in serum were observed on admission,on 3,5,9,14 day after admission.The dynamic changes of NO,NOS in CSF before treatment and two weeks after treatment were ob- served,too.Results There were no difference between the the concentration of NO and the vigor of total NOS in serum and in CSF of viral meningitis,bacterial meningitis and tubercular meningitis patients due to different agents. Conclusion The changes of the concentration of NO in serum and CSF,the vigor of total NOS in serum and CSF could not he seen as laboratory basis for discrimination of CNS infection due to different agents.
8.Imageologic study and clinical treatment of pelvic fractures combined with artery injuries

Objective To explore the imageologic characteristics of pelvic fractures with artery injuries and the treatment methods for embolization of arteries.Methods From January 1999 to June 2005,60 cases(42 males and 18 females)aged 21-52 years(average 34.5 years)with pelvic fractures and unsteady blood dynamics were admitted into our hospital.There were 32 cases with traffic injury,13 with crushing injury,nine with fall injury and six with other injuries.The mean injury severity score was 39?16(16-66).All cases were hypetensive with systolic blood pressure less than 90 mm Hg on the arrival.Routine X-ray examination of dorsaventral,debouch and porch of pelvis was performed.The aver- age amount of blood transfusion was 2 886 ml.All cases underwent iliac artery angiography and pelvic ar- teriography.Results X-ray examination of pelvic fractures showed posterior pelvic fracture in 25 ca- ses,with 64 branches of blood vessels injured;anterior pelvic fracture in 13,with 17 branches of blood vessels injured;acetabular fracture in six,with 12 branches of blood vessels injured;and combined pel- vic fracture in 16,with 36 branches of blood vessels injured.Three cases died,with mortality rate of 5%.One case with common arterial thrombosis was treated with artificial blood vessel transplantation, four cases with external iliac artery injuries including one with artery rupture were treated with prosthesis, and among the three cases with external iliac artery thrombosis,one was treated with dislodgment of thrombosis,one treated with recanalization of thrombolysis and one did not give any treatment.Fifty cases with injury and bleeding of internal iliac artery and its branches were treated with arterial embolization. Five cases showed no obvious injury.Conclusions The types of artery injuries can be predicted through X-ray of pelvic fracture.Posterior pelvic fracture may easily cause injury to superior gluteal arter- ies,iliac lumber arteries,and lateral sacral arteries.While anterior pelvic fracture will cause injury to obturator arteries.Superior gluteal artery is susceptible to injury.Embolization of injured arteries and an- astomosis are preferred treatment for pelvic arterial disruptions.
9.Study on tremor peak frequency, power and harmonics between Parkinson's disease and multiple system atrophy-parkinsonism

Objective To summarize the characteristics of tremor peak frequency,power and harmonics occurrence rate between Parkinson's disease (PD) and multiple system atrophy-parkinsonism (MSA-P).Methods From January to December in 2016,48 PD patients and 22 MSA-P patients were enrolled in Beijing Tiantan Hospital.Upper limb tremor patients got tremor analysis test in rest,posture and holding 1 000 g states,while lower limb tremor patients in rest and posture states.Peak frequency,power and harmonics occurrence rate of tremor were compared between PD and MSA-P patients in different states.Results Peak frequency of PD and MSA-P patients in upper rest ((4.76± 0.82) Hz vs (6.03 ± 0.25)Hz,t=-2.161,P=0.037),posture ((5.25 ±0.88) Hz vs (7.66 ±1.90) Hz,t=-4.266,P=0.001),holding 1 000 g ((5.80 ± 1.28) Hz vs (8.32 ± 1.51) Hz,t =-6.436,P =0.000) and lower posture ((4.85 ±0.78) Hz vs (6.42 ± 1.13) Hz,t =-3.001,P =0.012) showed statistically significant difference.Half-width power of PD and MSA-P patients in upper rest (extensor:15.70 (5.82,32.96)mg2/μV2 vs 3.14 (1.69,4.60) mg2/μV2,Z =-2.008,P =0.037),in posture (extensor:33.14(20.46,64.27) mg2/μV2 vs 10.02 (7.11,14.60) mg2/μV2,Z =-3.627,P =0.000;flexor:27.49(9.48,41.74) mg2/μV2 vs 5.97 (4.41,15.59) mg2/μV2,Z =-2.336,P =0.019),in holding 1 000g (flexor:17.80 (9.40,36.60) mg2/μV2vs8.15 (5.32,17.65) mg2/μV2,Z=-2.494,P=0.013),and in lower posture (flexor:26.63(7.09,85.84) mg2/μV2 vs 3.78 (2.51,7.12) mg2/μμV2,Z=-2.049,P =0.045) showed statistically significant difference.Harmonics occurrence rate of PD and MSAP patients in rest (82.5% vs 5.9%,χ2 =29.096,P =0.000),posture (80.0% vs 5.9%,χ2 =26.884,P=0.000) and holding 1 000 g (65.0% vs 0,χ2 =20.318,P=0.000) had statistically significant difference.Conclusions Tremor peak frequency in PD patients was lower than that in MSA-P patients,while power and harmonics occurrence rate in PD patients were higher than in MSA-P patients.

S-adenosylmethionine-dependent uroporphyrinogen III methyltransferase (SUMT) is a novel red fluorescence indicator. However, the production of SUMT in Escherichia coli is restricted by its relatively low solubility, and little is known about the red fluorescent materials that are associate with SUMT. Two individual SUMT mutations, L166A and L88R/L89G double mutant were produced by site-directed mutagenesis. Both mutants were overexpressed in E. coli and purified by Ni-NTA chromatography. The reddish mixtures isolated from the purified L88R/L89G double mutant were analyzed by UV-visible spectra scanning and mass analysis(MS). The L88R/L89G double mutant has enzymatic activity in vivo, whereas L166A mutant loses the activity. Trimethylpyrrocorphin is identified as the main constituent in the isolated pigments. The purified L88R/L89G mutant increases protein solubility, which is applied potentially as the fluorescent indicator denoting the solubility of protein fusion partner.
