1.Application of RAPD Molecular Marker in the Phylogenetic Analysis of Soy Sauce Strains

RAPD analysis of eight soy sauce strains isolated from commercial soy sauce koji was done with random primers,using Aspergillus oryzae AS3.951 and Aspergillus sojae AS3.495 as controls.Six appropriate primers(Primer1,Primer5,Primer6,Primer7,Primer8,Primer9)for RAPD-PCR were screened from nine random primers,and repetitive experiments demonstrated that their RAPD-PCR polymorphic patterns were stable.There were usually 4~8 bands in their RADP-PCR patterns,where the number of the main bands was 1~4 and the secondary bands were abundant.The phylogenetic tree of these strains was reconstructed according to their RAPD-PCR patterns,and it basically corresponded to traditional morphological taxology,demonstrating that the application of RAPD molecular marker in the phylogenetic analysis of soy sauce strains was feasible.
2.Clinical significance of UbcH10 expression in gastric cancer

Objective To investigate the expression and significance of UbcH10 in gastric cancer.Methods The expression of UbcH10 in 117 cases of gastric carcinoma tissues,who were followed up postoperatively,were detected by using immunohistochemistry; The expression among different histological grades and clinical stages are analyzed by statistics; Real-time polymerase chain reaction (Rt PCR) and Western blot method were used to detect the expressions of UbcH10 mRNA and protein in 43 cases of gastric carcinoma tissues and the cancer adjacent normal tissues.Results In paraffin fixed blocks of gastric cancer,positive rate of UbcH10 protein expression was higher than normal gastric tissues (83% vs 55%,Rs =-0.397,P<0.01).The difference of UbcH10 expression was not significant in gender,age,tumor site and size of the tumor(Rs =0.369,-0.378,-0.276,0.421,P < 0.05),while it was significantly correlated to tumor differentiation,invasive depth,lymphatic metastasis and clinical stage (Rs =0.369,-0.378,-0.276,0.421,P <0.05).UbcH10 and Ki-67 protein and HER-2 expression were in significantly positive correlation (Rs =0.834,Rs =0.281,P < 0.01).UbcH10 mRNA and protein expression in fresh gastric carcinoma tissues was higher than that in their corresponding adjacent normal tissues(ACt =32.2±5.19,4.69 ±1.26,t =26.8,P<0.01),(0.74±0.45,0.11 ±0.04,t =8.09,P<0.01).The -5 year patient's survival rate in UbcH10 highly-expressed gastric cancer was higher compared to those with UbcH10 low-expression(x2 =13.116,P <0.01).Conclusions UbcH10 is upregulated in gastric cancer,and expression of UbcH10 is closely related to the prognosis of patients with gastric cancer.
3.Influence of miR-21 control Smad7 expression and lung cancer A549 cell line proliferation

Objective To explore miR-21 regulation of Smad7 in lung cancer A549 cell line as well as its impact on the proliferation of lung cancer A549 cell line. Methods miR-21 mimic and inhibitors in lung cancer A549 cell line were transfected by using Lipofectamine 2000 Reagent. After 48 h, Western blot and qRT-PCR were applied to assess the expression of protein and mRNA of Smad7 . MTT assay was used to determine the proliferation influence of the transfected lung cancer A549 cell line. Results Western blot and qRT-PCR showed that A549 cell trans-fected miR-21 mimic exhibited down-regulated Smad7 protein and mRNA expression, and A549 cell transfected miR-21 inhibitor exhibited up-regulated Smad7 protein and mRNA expression. The A549 cell proliferation activity decreased significantly after transfected miR-21 inhibitors. Conclusion miR-21 inhibitors can increase Smad7 pro-tein and mRNA expression, and suppress the proliferation activity of lung cancer A549 cell significantly.
5.Research on Separation Characteristics of the Extracellular Algae-lysing Components from Three Algae-lysing Bacteria

Three algae-lysing bacteria (L7、L8、L18) have been isolated. We used starch medium,which has no passive effect on growth of the Anabaena flos-aquae as cutivate medium. After being cultivated for 4d on a reciprocal shaker at 30℃ and a speed of 100 r/min,the cultivate broth were centrifugated at a speed of 4000 r/min for 20min、filtrated with 0.22 ?m membrane,and condensed by vacuum rotary evaporator at 65℃. The sterile condensed bacteria-free filtrate obtained are dialysed,sedimentated by ethanol,extracted by organic solvents,absorbed by activated carbon. The molecular weight of the extracellular algae-lysing components of L7 are less than 3.5 kD,while the molecular weight of extracellular algae-lysing components of L8 and L18 are between 3.5 kD~7 kD; ethanol can not separate extracellular algae-lysing components from other componets efficiently; carbon tetrachloride can separate the extracellular algae-lysing components of L7 into water-layer efficiently,while petroleum ether can separate the extracellular algae-lysing components of L8 and L18 into water-layer efficiently,the ex-tracellular algae-lysing
6.A Modified Method for Acute Isolation of Atrial Myocytes in Diabetic Rats

Objective To establish a stable method for acute isolation of atrial myocytes in diabetic rats,so as to provide materials for electrophysio?logical study of diabetic rats. Methods Streptozotocin(STZ)was used to establish type I diabetic model. Atrial myocytes were isolated with modi?fied perfusion buffer by Langendorff perfusion. The atrial myocytes were morphologically observed with optical microscope and identified by morphol?ogy and immunofluorescence staining. The action potential was recorded by patch clamp technique. Results STZ can establish a stable type I dia?betic model. The modified perfusion method can yield calcium tolerant and spindle shaped atrial myocytes. Immunofluorescence indicated that atrial myocytes were positive for Kv1.5 which was expressed in atrial myocytes. The atrial myocytes obtained by this method were able to generate action po?tentials. Conclusion The modified perfusion method is suitable for acute isolation of atrial myocytes in rats with diabetes mellitus,which may help to study the electrophysiology of diabetic heart.
7.Imaging presentation of pylorus obstruction caused by foreign bodies in the gastric anrum and granulomatous inflammation

Pylorus obstruction caused by foreign bodies in the gastric anrum and granulomatous inflammation is rarely seen.The clinical symptoms of this disease are unspecific.Combination of X-ray radiography,computed tomography and gastroscopy could make definite diagnosis.Differential diagnosis between pylorus obstruction and peptic ulcer,gastric cancer and duodenal obstruction should be done before operation.On April 13,2012,a patient with pylorus obstruction caused by foreign bodies in the gastric anrum and granulomatous inflammation was treated at the Second Hospital of Jiaxing,the imaging characteristics of the disease were summarized to provide referrence for the diagnosis and treatment of this disease.
8.Genistein inhibited hypoxia inducible factor-1? expression induced by cobalt chloride in human retinal pigment epithelium cells

Objective:The time-dependent changes of hypoxia-inducible factor-1? (HIF-1?) expression induced by CoCl_(2 ) treatment and the effects of genistein on the level of HIF-1? expression in human retinal pigment epithelium cells were studied. Methods:Using a confocal scanning laser microscope coupled to a computer, HIF-1? expression was determined. Results:CoCl_(2 ) treatment could significantly elevate the level of HIF-1? expression. At 0.5 hour after CoCl_(2 ) treatment, the highest level was observed. Genistein 50 ?mol/L, 100 ?mol/L, 200 ?mol/L could suppress HIF-1? expression in a concentration-dependent manner. Conclusions:These results suggested that genistein could inhibit HIF-1? expression induced by CoCl_(2).
9.Determination of Rebamipide in Rats' Plasma and Colonic Tissue by HPLC Gradient Elution

OBJECTIVE: To establish an HPLC method for the determination of rebamipide in rats' plasma and colonic tissue.METHODS: The rats were assigned to receive 4 mg rebamipide chitosan capsule or its gelatin capsule or its CMC-Na solution peros.Then the plasma and colon tissue samples were taken at different time for a determination of the concentration of rebamipide by HPLC gradient elution with Cmax computed.RESULTS: The linear range for rebamipide was from 20~2 000 ng?mL-1.The average extracting recoveries of rebamipide from the plasma and the colon were 88.4% and 90.8%,respectively.After oral administration of 4 mg rebamipide chitosan capsule or its gelatin capsule or its CMC-Na solution,the Cmax of rebamipide in the colon tissue were 5 963.9,2 190.7 and 1 185.8 ng?g-1 respectively versus 252.7,949.0 and 1 023.3 ng?mL-1 respectively in plasma.CONCLUSIONS: The established HPLC gradient elution in the study can be used to determine quantitatively the rebamipide level in plasma or colon tissue for the development of its colon specific delivery preparations.
10.Analysis of Clinical Efficacy of Azithromycin Combined Glucocorticoids in Treatment of Severe Pneumonia Mycoplasma

Objective:To analyze the influences of azithromycinantibiotic combined glucocorticoids on the levels of erythrocyte sedimentation rate (ESR),serum lactate dehydrogenase (LDH) and inflammatory cytokines levels as well as the clinical effect of severe pneumonia mycoplasma.Methods:112 children with severe pneumonia mycoplasma who were treated in our hospital were selected and randomly divided into the control group and the observation group with 56 cases in each group.The patients in the control group were treated with azithromycinantibiotic,while the patients in the observation group were treated with glucocorticoid on the basis of the control group.Then the levels of ESR,LDH,isoenzyme MB (CK-MB),creatine kinase (CK),aspertate aminotransferase (AST),c-reactive protein (CRP),interleukin 6 (IL-6),tumor necrosis factor-o (TNF-α),CD4+ and CD8+,the clinical efficacy and adverse reactions between two groups were observed and compared before and after the treatment.Results:The effective rate in the observation group was higher than that of the control group,and the difference was statistically significant (P<0.05).After treatment,the levels of ESR,LDH,CK-MB,CK,AST,CRP,IL-6,TNF-α and CD8+ in the two groups decreased,which were lower in the observation group than those of the control group (P<0.05).After treatment,the levels of CD4+ in both groups increased,which was higher in the observation group than that of the control group (P<0.05).The disappearance time of fever,cough relief,lung rale of observation group were shorter than those of the control group(P<0.05).There was no statistically significant difference in the adverse reactions between the two groups (P>0.05).Conclusion:Azithromycin combined with glucocorticoids was more effective than azithromycin alone in the treatment of severe mycoplasma pneumonia with high safety,which could obviously relieve the myocardial injury and inflammatory response,enhance the immune function.
