ObjectiveTo obtain the soluble expression of P protein of norovirus(NV)G Ⅱ. 2[P16]and analyze its antigenic characteristics.MethodsThe recombinant plasmid GⅡ.2-NV-pET-28a was constructed by optimizing the NV GⅡ.2[P16]P protein gene,synthesizing and cloning it into pET-28a(+)vector. The recombinant plasmid was transformed into DH5α competent cells,and the expression strain was optimized to make the P protein express in soluble form. The expressed P protein was purified by Ni-NTA affinity chromatography,of which the antigenicity was detected by Western blot,the purity was detected by size exclusion high performance liquid chromatography(SEC-HPLC),and the antigenantibody binding ability was identified by ELISA.ResultsThe target protein of recombinant BL21 Star(DE3)pLySs was highly expressed in soluble form. After purification by affinity chromatography,the purity of GⅡ.2 P protein was 95. 53%,and the antigenicity was good.ConclusionSoluble NV P protein was successfully prepared by prokaryotic expression strain,and the antigenicity of P protein was confirmed,which laid a foundation of the development of NV G Ⅱ.2[P16]detection kits and the recombinant subunit vaccines.

Objective To study the effect of articular step-off on knee joint contact pressures in fractures of the medial tibial plateau. Methods Eight specimens of embalmed adult knee joint were used to establish models of articular step-off of the medial tibial plateau by 0,1,2,3 mm respectively (Schatzker Ⅳ fracture) .The technique of pressure sensitive film was used to measure the contact pressures on the weight-bearing area of the knee joint in the 3 models. Results When the step-offs of medial tibial plateau were 0,1,2,3 mm,the average contact pressures on the lateral compartment were respectively 3.34 ± 0.06 MPa,3.48 ± 0.06 MPa,4.19 ± 0.05 MPa,4.19 ± 0.04 MPa,while the average contact pressures on the medial coMPartment were 4.04±0.17 MPa,4.57±0.20 MPa,5.64±0.20 MPa,5.64±0.19 MPa.When the step-off of medial tibial plateau was 2 mm,the average contact pressures on the medial and lateral plateaus increased by 39.6％ and 25.4％ compared with the 0 mm step-off,with statistically significant differences (P ＜0.05). Conclusions If the step-off of the medial tibial plateau reaches ≥ 2mm,open reduction and internal fixation should be indicated to prevent secondary traumatic arthritis.When the step-off of the medial tibial plateau is ＜ 2 mm,conservative treatment can be a proper choice.

There were 461939 outpatient visits in Shanghai Tianping Primary Health Center from January to December 2010,the data were retrospectively analyzed based on the electronic outpatient record.The 20 most common illnesses accounted for 73.67％ of total outpatient consultations (340310/461 939),hypertension ranked the first position of all illnesses ( 21.06％ ) and followed by coronary heart disease (8.36％).Patients aged over 65y accounted for 72.35％ of all patients,the frequency of consultations was positively correlated with age of patients(P ＜0.01 ).The frequency of consultations varies according to the gender,age of the patients as well as the organ system involved (P ＜ 0.01 ).It can be concluded that the most frequently encountered illnesses in primary health center are common illnesses and chronic diseases.There are different needs for primary health service between genders and different age groups.

This article was aimed to study the pharmacokinetics of mussaenoside in rats. Gardenoside was used as the internal standard in the content determination of mussaenoside in rat plasma with HPLC. The chromatographic conditions were Diamonsil C18 (2) column (150 mm í 4.6 mm, 5 μm), mobile phase of methanol:0.5% glacial acetic acid (30:70), flow amount of 1.0 ml/min, detection wavelength at 238 nm, column temperature at 30℃. Compartment model fitting of average plasma concentration in rats - time data had been conducted after mussaenoside was given to mice through single intravenous injection with the help of DAS 2.0 pharmacokinetics intelligence analysis software. The results showed that the plasma concentration using 2-comparment model intravenous injection calculation method, which corresponded well to that of actually measured (weighting of 1), was proved superior to other models. The distribution half-life time (t1/2α) was 9.892 min and the elimination half-life time (t1/2β) was 55.384 min. It was concluded that the established method was convenient, accurate and stable, which can be used for the study on phar-macokinetics of mussaenoside. Mussaenoside was distributed rapidly in the body of rats. The main process of mus-saenoside in the body of rats was elimination. The metabolic rate was at moderate speed.

Objective To review the outcome of coronary artery bypass graft (CABG) using left internal thoracic artery grafts in the patients aged over 70 years old.Methods Two hundred and eighty-eight patients aged over 70 years old from January 2009 to January 2014 who underwent CABG were selected.All of the patients were divided into two groups on base of the graft:control group:only saphenous vein graft was used in 138 cases; observation group:the left internal thoracic artery to the left anterior descending artery was used in 150 cases,which also included total arterial grafting,facilitated by the use of the radial artery,right gastroepiploic artery was operated in 7 cases,beside these artery grafts,saphenous vein graft was used to anastomosis with other vessels.Results Operative mortality was 2.9％ (4/138) in control group and 2.0％ (3/150) in observation group,and there was no significance (P ＞ 0.05).The volume of blood drainage was larger in observation group [(796.2 ± 382.4) ml] than that in control group [(512.8 ± 387.5) ml],and there was significant difference (P ＜ 0.05).At the same time,there was no significant difference in the incidence rate of second thoractomy,malignant arrhythmia,stroke,poor wound healing,and usage of intraaortic balloon counterpulsation between two groups (P ＞ 0.05).Conclusion It does not increase the mortality and incidence of serious complications using the left internal thoracic artery to the left anterior descending artery in the patients aged over 70 years old during CABG.

Objective] To sum up Pro. Li Saimei’s clinical experience in treating mental diseases. [Method] By following the teacher clinic and sorting out the related medical materials,the author summarizes professor Li Saimei 's academic experience of treatment of mental diseases ,and for proven cases. [Result] In professor Li Saimei's opinion,mental diseases are due to dysfunction of shaoyang's turning,and phlegm blocking the spirit orifices. Professor Li usually uses Chaihu related formulas to turn the Shaoyang,resolve the phlegm,open the three Jiao,regulate Yin and Yang.The notable clinical result has been observed.[Conclusion] Professor Li Saimei's experience is effective and worthy of inheritance and promotion.

Diabetic microvascular disease is a severe complication of diabetes mellitus (DM).There are many hypoth-esis for its mechanisms.This overview aimed at effect of C-peptide,coagulant factors,platelet and D-dimer in dia-betic microvascular disease,and provided evidence for its prevention and treatment in clinic.
