1.Clinical observation of capecitabine and temozolomide in the treatment of advanced pancreatic neuroendocrine tumors

Objective:To observe the therapeutic effect and toxicity of capecitabine and temozolomide in the treatment of advanced pancreatic neuroendocrine tumors. Methods:A total of 14 patients with stageⅣwell-differentiated pancreatic neuroendocrine tumor (NET G1/G2/G3) were treated with oral CAPTEM regimen, and the response rate, PFS and adverse effect after treatment were analyzed. All data analyses were performed using software SPSS17.0. Results:These 14 patients were followed-up for more than 2 years. Till Oct 2016, one patient got CR, one patient got PR, four patients got SD. Median progression-free survival was 8.9 months. The two year survival rate was 85.7%. Only one patient experienced grade 3 adverse events. Conclusion:CAPTEM is an effective and well-tolerated salvage regimen for the treatment of advanced well-differentiated pNET.

3.Prospective memory deficit in acute stroke patients with cognitive impairment

Objective To explore the characteristics of prospective memory ( PM ) deficit in acute stroke patients with cognitive impairment.MethodsSixty patients suffering from acute stroke who met the inclusion criteria in screening with the neurobehavioral cognitive status examination (NCSE) were enrolled into the experimental group. Sixty healthy participants who matched the experimental group in gender distribution and average age and education level were assigned as a control group. All the participants completed several neuropsychological evaluations,including the Chinese version of the Cambridge prospective memory test ( C-CAMPROMT), the Chinese version of the Rivermead behavior memory test (C-RBMT), a digit-span test (DS), the Chinese version of Stroop's word-color test (C-SWCT) and a color trail test (CTT).ResultsTime-based PM (TBPM) scores in the control group were significantly higher than in the experimental group. At the intention initiation stage TBPM scores in the control group were also significantly better than in the experimental group. The TBPM performance of the experimental group was significantly worse than that of the controls when PM performance was compared with other cognitive functions controlled for.ConclusionsThe acute stroke patients with cognitive impairment showed greater TBPM performance deficits than the controls. This may have resulted from impairment at the intention initiation stage. TBPM deficits may exist independently. If so, they could serve as an assessment of cognitive impairment after stroke.
4.Study on relationship between fat-rich diet-induced nonalcoholic fatty liver disease and endoplasmic reticulum stress in rats

Objective To establish the nonalcoholic fatty liver disease(NAFLD) model in SD rats and to investigate if endoplasmic reticulum stress(ERS) plays a role in the pathogenesis of NAFLD. Methods A total of 36 male SD rats were randomly divided into 4 groups: normal control group 1(N1,n=8), normal control group 2 (N2,n=8), model group 1(M1,n=10) and model group 2(M2,n=10). The two control groups were fed with normal diets, whereas the two model groups were provided diets enriched in fat (10% lard oil and 2% cholesterol). The rats in N1 and M1 groups were sacrificed at the 12th week, and those in N2 and M2 groups were sacrificed at the 20th week. The serum alanine aminotransferase (ALT), aspartate aminotransferase(AST), alkaline phosphatase (ALP), total protein (TP), triglycerides (TG), total cholesterol (TC), low-density lipoprotein (LDL), high-density lipoprotein (HDL) were measured. The steatosis, immflamation and fibrosis of the liver were observed with HE and Masson staining. The expressions of GRP78, CHOP and procaspase-12 mRNA were tested using real-time PCR and the activation of procaspase-12 protein was detected by Western blot. Results At the 12th week, the liver index and the serum levels of ALT, AST,ALP,TP,TC,LDL in M1 group were significantly higher than those in N1 group (P<0.01), but HDL level was significantly lower in M1 group than that in N1 group (P<0.01). At the 20th week, the concentrations of TC and LDL in M2 group were significantly increased in comparison with N2 group. The histochemical study revealed that the hepatic steatosis and inflammation in M1 and M2 groups were more serious than those in N1 and N2 goups(P<0.01). The slight fibrosis was seen in M2 group. At the 12th and 20th weeks, the expressions of GRP78,CHOP and procaspase-12 mRNA and protein in M1 and M2 groups did not differ in N1 and N2 groups. Conclusions The fat-rich diet might successfully induce NAFLD in rats and there is no ERS observed in the study, which suggests that ERS may not be involved in the pathogenesis of NAFLD.
5.Analysis of glucose dysbolism in patients with ectopic ACTH syndrome

The characteristics of glucose dysbolism in ectopic ACTH syndrome (EAS) were investigated by analysing the clinical data of 117 patients with Cushing syndrome (CS). The patients with CS were divided into 3 groups : EAS group (n = 9) , Cushing's disease (CD) group (n = 67) and adrenocortical adenoma (AA) group (n =41). Retrospective analysis of the data covered the prevalence of secondary diabetes, blood glucose, HbAlc, blood potassium, and blood ACTH and 24 h urinary-free cortisol (UFC) levels. Compared with CD and AA groups, the prevalence of secondary diabetes and blood glucose level were higher in EAS group, while HbAlc level was lower. Blood glucose level in patients with ectopic ACTH syndrome decreased to normal after tumor removal.
6.Lentivirus-mediated NGF gene silencing inhibited differentiation of PC12 Cells

Aim To investigate the effects of lentivirus mediated nerve growth factor ( NGF) gene silencing on pheochromocytoma cells ( PC12 ) and the possible mechanisms .Methods The NGF shRNA expression vector was constructed .PC12 cells were randomly divi-ded into five groups (n=3 each) as follows: negative control group ( NC ) , control lentivirus group ( LV CON) , lentivirus NGF shRNA1 group ( LV shNGF1 ) , lentivirus NGF shRNA2 group(LV shNGF2), lentivir-us NGF shRNA3 group(LV shNGF3).The cells in NC group were cultured in DMEM/HG and polybrene me-dium, while others were cultured in DMEM/HG, poly-brene and corresponding lentivirus medium .After the treatment, the infection efficiency was determined by fluorescent microscope .Relative expression of NGF , extracellular signal-regulated kinase ( ERK1/2 ) and p-ERK1/2 were assessed by Western blot .The expres-sion of NGF mRNA was analyzed by quantitative re-verse transcription polymerase chain reaction ( qRT-PCR) .The differentiation degree was valued according to the length of neuritis and max diameter of cells .The cell viability was detected by CCK-8.Results The in-fection efficiency in PC12 cells reached over 90%. Compared with NC group , the relative expression of NGF mRNA and NGF protein was significantly down-regulated ( P<0.05 ) .There was no difference in the expression of ERK1/2 protein and cell viability .The expression of p-ERK1/2 protein was markedly down-regulated in LV shNGF3 group ( P<0.01 ) .The cells morphology was changed , and the length of neuritis and max diameter of cells were strained in LV shNGF 3 group than those in NC group ( P<0.01 ) .Conclusion Lentivirus-mediated NGF gene silencing inhibits the differentiation of PC12 cells through suppressing the activation of ERK1/2.
7.The Construction and Practice Research of Career-oriented Practical Pharmaceutical English Course in High-er Vocational Education

OBJECTIVE:To provide reference for improving the application ability of professional English for students major-ing in pharmacy in higher vocational education. METHODS:The career-oriented practical pharmaceutical English course and teach-ing system were constructed through the way of formulation of curriculum standards,establishment of teachers’team,development of school-based teaching material,application of CBI theme teaching mode and variety teaching methods,and it was carried out among students majoring in pharmacy in 2011-2014 grades in our school. Besides,experimental and survey research were used for the evaluation of Proctical Pharmaceutical English. RESULTS:There was no significant difference in the academic achievement and questionnaire evaluation between 2 class before teaching(P>0.05);however,academic achievement and questionnaire evaluation were higher than 2 class after teaching,experimemal class higher than control class,the difference was statistically significant(P<0.05). CONCLUSIONS:The career-oriented Practical Pharmaceutical English course and teaching system is feasible and effective;compared with traditional teaching,it can inspire students' interest in English learning,improve the ability to use English in the work and motivate their comprehensive qualities.

9.TRβ gene mutation in a family with thyroid hormone resistance syndrome

Objective To detect the gene mutation of thyroid hormone receptor β ( TRβ ) in a family with thyroid hormone resistance syndrome.Methods The genomic DNA was extracted from peripheral blood leukocytes of the patient and his 5 family members.The exons 1-10 ofTRβ gene were amplified by PCR.The products of PCR were sequenced directly to detect the gene mutation.Results Two members of this family were confirmed to have the C y A transition mutation at nucleotide 1642 site within exon 10 of TRβ gene,which was a missense mutation causing the substitution of Proline to Threonine (P453T).The mutation was Heterozygous.Conclusions It was confirmed that the patient has TRβ gene mutation P453T in exon 10.The mutation may lead to the occurrence of thyroid hormone resistance syndrome.
10.Relationship between Homer1a and nerve cell loss in hippocampus after severe diffuse brain injury in rats

(DBI) in rats by observation on the changes of Homer 1a expression and apoptotic nerve cells . Methods Spraque-Dawlley(SD) rats were randomly ( random number ) divided into control group and severe DBI group .DBI rat model was established according to the de-scription of Marmarou′s diffused brain injury .No injury was done on control group .The changes of neuron pathology were observed by light microscopy and electron microscope .The expression of Homer1a was observed by immunohistochemistry and western blot .The quan-tity of apoptotic cells was measured by terminal deoxynucleotidyl transfernase medicated nick end labeling ( TUNEL) method. Results The death rate of rats in severe DBI group was 49.3%.Compared with the control group , the ultrastructures in hippocampal neurons in-cluding organelle , axonal and capillary were damaged seriously after injury , the survival rate of nerve cells decreased significantly at 1 h after injury ([99.4 ±0.6]%vs [94.4 ±5.6]%, P<0.05), and peaked at 72 h ([99.2 ±0.8]%vs [54.7 ±33.8]%, P<0 .05) in DBI group.The expression of Homer1a protein increased significantly at 1 h after injury(0 .136 ±0.024 )and peaked at6 h(0.178 ± 0.028) and maintained to 24 h (0.176 ±0.027), while decreased at 48 h (0.145 ±0.02)and 72 h (0.117 ±0.012) in DBI group;the expression of Homer 1a was obviously higher at each time point in DBI group than that in control group (P <0.05).The apoptoticindex of TUNEL positive cells increased at 6 h and demonstrated significant difference at 72h in comparison to control group ([41.78 ±3 .96]%vs [1.92 ±0.22]%, P<0.05).The correlation analysis indicated that Homer1a expression from 1~24 h and 24 h~72 h was related to the survival rate of nerve cells ( r=-0.726, P<0.05; r=0.842, P<0.05) and the quantity of TUNE positive cells(r=0.738, P<0.0;5 r=-0.898, P<0.05). Conclusion The dynamic expression of Homer1a in hippocampus after severe DBI can reflect nerve cell loss.
