3.Characterization of purified coconut oil bodies as an encapsulating agent for Doxorubicin and Paclitaxel

Introduction:
Doxorubicin (DOX) and paclitaxel (PTX) are both widely used anticancer drugs with a broad spectrum of antitumor activity, commonly against breast, ovarian, and lung cancers. Currently, these drugs are commercially available in liposomal formulations for their use in chemotherapy. This study generally proposed coconut oil bodies (COB) obtained from Cocos nucifera L. as an alternative carrier for DOX and PTX rather than the currently used liposome.
Objectives:
This study aimed to compare standard liposome and coconut oil bodies as drug carriers in terms of their microencapsulation efficiencies, lipid profiles, in vitro drug release and stability, as well as their cholesterol levels.
Methods:
Coconut oil bodies (COB) were isolated and purified from Cocos nucifera L. by modified sucrose
gradient method followed by microencapsulation of standard drugs (doxorubicin and paclitaxel) through selfassembly and freeze-thaw method. The two standard drugs were encapsulated using COB and standard liposome. Encapsulation efficiency of both materials were determined. Lipid profiles of both encapsulating materials were analyzed by Fourier-transform infrared spectroscopy, gas chromatography-flame ionization detector, and cholesterol level determination. In vitro drug release and pH stability of both encapsulated drugs were analyzed.
Results:
Doxorubicin (DOX) and paclitaxel (PTX) were successfully incorporated in COB. Lauric acid was mainly
abundant in COB and was able to lower cholesterol levels (5 mg/dL). COB incorporated with DOX and PTX
showed stability at acidic and neutral pH. Drug release profile showed a rapid outburst within 3 hours compared to liposome encapsulated DOX and PTX.
Conclusion
Our study showed the encouraging potentials of using COB as wall materials that will make them
attractive candidates for the formulation of pharmaceuticals for optimized drug delivery of cancer chemotherapeutics DOX and PTX

Classic Kaposi sarcoma (KS) is a rare human herpes virus 8-associated angioproliferative disease, and the disseminated classic type of KS in Korea is even rarer. The treatment options for classic KS vary and range from surgical excision to ionizing irradiation or chemotherapy. Recently, there have been a few reports of treating classic KS with paclitaxel, which has been used to treat AIDS-associated KS and post-transplant KS. We herein report a case of disseminated classic type KS in a 78-year-old Korean male patient who showed dramatic response after only two cycles of paclitaxel treatment.

7.Carboplatin and paclitaxel as an initial treatment in patients with stage IVb cervical cancer: a report of 7 cases and a review of the literature.

OBJECTIVE: The aim of this study is to evaluate the efficacy of carboplatin-paclitaxel (TC) as an initial treatment in patients with the International Federation of Gynecology and Obstetrics (FIGO) stage IVb cervical cancer. METHODS: We retrospectively reviewed seven patients with stage IVb cervical cancer who have been primarily treated with TC. The activity and the toxicity were evaluated. Response rate was the main endpoint. RESULTS: Overall, the treatment of TC was well tolerated. The overall response rate was 71.4% (2 complete response, 3 partial response). Although grade 3-4 hematologic toxicities were observed in 3 out of 7 patients (42.8%), no patients experienced grade 3-4 non-hematologic toxicities. When we combined our present results with the previous reports, the overall response rate of TC is 63.6%. CONCLUSION: TC is active and well tolerated in patients FIGO stage IVb cervical cancer. This combination may be considered as an initial treatment regimen in this patient population.

8.Advances of taxol combinatorial biosynthesis.

Taxol is a kind of isoprenoid with strong anticancer activity. It is difficult to be obtained because of its low concentration in nature, which hinders its application in cancer treatment. Recently, biosynthesis methods for taxol production have attracted more attentions. Several systems including Escherichia coli, Saccharomyces cerevisiae, Physcomitrella patens, Arabidopsis, tomato and ginseng were explored. This review focuses on the advance in biosynthesis of taxol in different systems and features the bottleneck of scale fermentation for producing the intermediates. At the same time some advices for the further were given. At last, the future and character of Physcomitrella patens system used in taxol combinatorial biosynthesis were analyzed based on our lab's research.

9.Stabilizing Microtubular Network Facilitates the Intracellular Growth of Orientia tsutsugamushi.

Microtubule network provides many intracellular microbes with an efficient way to move within host cells. Orientia tsutsugamushi move from the cell periphery to the microtubule organizing center (MTOC) by dynein-dependent mechanism. In this study, we investigated the role of microtubule on the growth of O. tsutsugamushi. The treatment of infected cells with taxol as well as daunomycin enhanced the bacterial growth in contrast to colchicine. Immunofluorescent (IF) staining of taxol-treated cells exhibited that O. tsutsugamushi clustered tightly near the nucleus with thick bundles of microtubules, whereas dispersed in the cytoplasm in colchicine-treated cells. These results suggest that microtubule network facilitate the growth of O. tsutsugamushi.

10.The Hidden Culprit: A Case of Repeated Anaphylaxis to Cremophor.

Drug-induced anaphylaxis is a big pitfall in patients receiving antineoplastic chemotherapy. We report a case of lung cancer patient who experienced two near-fatal anaphylactic reactions that resulted from paclitaxel and multivitamin, seperately. Recurrent severe reactions to different agents led to further investigation to which material the patient was hypersensitive. The skin prick test revealed sensitization to cremophor, which is a commonly used emulsifying agent. This case emphasizes the importance of correctly identifying the culprit drug of anaphylaxis to avoid potentially fatal reaction.
