<p>OBJECTIVETo investigate the changes of adhesion molecules, cyclic adenosine monophosphate(cAMP) and cyclic guanosine monophosphate (cGMP) in divers post 480 heliox saturation diving.p><p>METHODSFour divers were compressed within 96 hours to depths of 480 m with heliox-oxygen and held at the designated depth for 49 hours, excursion to 493 m during their saturation stay, then decompressed within 302 hours to the surface. The blood samples were collected before compression and post decompression, the expression level of intercellular adhesion molecule-1(ICAM-1), E-selectin, P-selectin, cAMP, cGMP were detected with ELISA analysis box.p><p>RESULTSCompared with the levels of CAMs before compression, the levels of ICAM-1, E-selectin, P-selectin and cGMP in the serum were changed post decompression (P > 0.05). The levels of cAMP were significantly elevated post decompression (629.91 +/- 75.01) nmol/L vs (66.72 +/- 83.15) (P < 0.05).p><p>CONCLUSIONThe decompression schedule in this heliox saturation diving is safe, the decompression sickness pathology in this diving has not been induced. But the stress response of divers are enhanced by this great depth saturation diving.p>

PURPOSE: CD24 is a small, heavily glycosylated glycosylphosphatidylinositol-linked cell surface protein that is expressed in hematologic malignancies and in a large variety of solid tumors. It appears to function as a ligand of P-selectin, an adhesion molecule that is present in activated platelets and endothelial cells. We aimed to evaluate CD24 protein expression in adenomas and adenocarcinomas of the colon and to correlate it to clinicopathological data. METHODS: Adenomas and adenocarcinomas of the colon were stained for CD24 immunohistochemically. For statistical analysis, the staining was categorized according to stainability (negative, weakly, moderately, strongly positive) and staining patterns (membranous vs. intracytoplasmic). RESULTS: The present study clearly demonstrated that CD24 was much more abundantly expressed for adenocarcinomas than for adenomas in the colon (P <0.05). A higher significant association of cytoplasmic CD24 expression was observed with adenocarcinomas of the colon than with adenomas of the colon (P <0.05) and with positive nodal status of the colonic adenocarcinoma than with negative nodal status of the colonic adenocarcinoma (P <0.001). CONCLUSIONS: The stainability and the staining pattern of CD24 is an important molecular marker for colonic epithelial neoplasms and may help to define malignant transformation and to predict lymph-node metastasis.

The authors aimed to evaluate the expression of CD24 in adenomas and adenocarcinomas of the stomach and its correlation to clinicopathological data. METHODS: A series of 48 adenocarcinoma cases (advanced gastric carcinoma: 24 cases, early gastric carcinoma: 24 cases) and 12 adenoma cases of the stomach were immunohistochemically stained for correlation with the clinicopathological data. The staining was evaluated as stainability (negative, weak-, moderate-, strong-positive) and staining patterns (membranous vs. intracytoplasmic), which were used for statistical analyses. RESULTS: The present study clearly demonstrated that the stainability of CD24 expression increased with positive nodal status in advanced gastric carcinomas (P<0.005). The stainability of CD24 in the adenocarcinoma group was much higher than in the adenoma group, but this was not statistically significant. Intracytoplasmic CD24 expression was demonstrated only in the adenocarcinoma group, with the exception of the adenoma group, but this also was not statistically significant. CONCLUSION: The authors conclude that the stainability of CD24 could be a molecular marker for gastric adenocarcinomas which could help to predict lymphogenous metastasis.

OBJECTIVE: To investigate the predictive effect of platelet activation index expression before and after adenosine bisphosphate activation on bleeding risk in patients with primary immune thrombocytopenia (ITP). METHODS: Eighty-nine patients with ITP admitted in our hospital from January 2017 to October 2018 were selected and inrolled in ITP group, the bleeding scoreing and grading were performed by using the ITP-BAT for ITP patients, then 89 ITP patients were divided into 4 subgroups: nothing bleeding symptom group, mild bleeding symprom group, mode rate bleeding symptom group and severe bleeding symptom group according to bleeding scores and grades obtained from ITP-BAT detection. At the same time, 22 persons underwent the health physical examination were selected and enrolled in control group. The adenosine diphosphate (ADP) was used as activator for all patients and controls. The flow cytonetry was used to analyze the expression of platelet membranc glyco protein (GPⅠb, GPⅡb /Ⅲ a) and P-selectin before and after ADP activation, the multiple linear person's correlation analysis was used to analyze the correlation of bleeding degree of ITP patients before and after ADP acbivation with the expression levels of GPⅠb, GPⅡb/Ⅲa and P-selectin. RESULTS: After the ADP activation, the expression level of GPⅠb significantly decreased, while the expression levels of GPⅠb, GPⅡb/Ⅲ a and P-selectin significantly increased in control group, nothing bleeding symptom group and mild bleeding symptom group; but the expression level of GPⅠb significantly increased, while the expression level of GPⅡb/Ⅲ a significantly decreased in moderate and severe bleeding symptom group, the both differences were statistically significant (P＜0.05). however, the expression level of P-selectin in moderate and severe bleeding symptom groups before and after ADP activation was not statistivally significant (P＞0.05). Before ADP activation, the expression level of GPⅠb in ITP subgroups was lower than that in control group, the expression level of GPⅡb/Ⅲ a in ITP subgroups was higher than that in control group, the expression level of P-selectin in moderate and severe bleeding symptom groups was higher than that in control group (P＜0.05). After ADP activation, the expression levels of GPⅠb and P-selectin in ITP subgroups both were lower than those in control group, the expression level of GPⅡb/Ⅲa in ITP subgroups was higher than that in control group (P＜0.05). The comparison among ITP subgroups showed that before ADP activation, the expression level of GPⅠb in moderate and severe bleeding symptom groups was lower than that in nothing bleeding symotom and mild bleeding symptom groups, while the expression levels of GPⅡb/Ⅲa and P-selectin were higher than those in nothing bleeding symptom and mild bleeding symptom groups (P＜0.05), however, after ADP activation, the expression level of GPⅠb in moderate and severe bleeding symptom groups was higher than that in nothing bleeding symptom and mild bleeding symptom groups, while the expression levels of GPⅡb/Ⅲ a and P-selection in moderate and severe bleeding symptom groups were lower than those in nothing and mild bleeding symptom groups (P＜0.05). The correlation analysis showed that before ADP activation, the expression levels of GPⅠb and GPⅡb/Ⅲa positivdy correlated with the bleeding risk (r=0.483, 0.504), and the P-selectin not correlated with the bleeding risk (r=0.000); however, after ADP activation, the expression level of GPⅠb and GPⅡb/Ⅲ a negatively correlated with the bleeding risk (r=-0.627, -0.406, -0.108). CONCLUSION The expression level of platelet activation indicators before and after ADP activation is of certain value for prevention of bleeding risk in ITP patients and can be used as a reference indicator for the treatment and efficacy evaluation.

The staining was evaluated as stainability (negative, weak-, moderate-, strong-positive) and staining patterns (membranous vs. intracytoplasmic) for statistical analysis. RESULTS: A highly significant association of cytoplasmic CD24 expression with adenocarcinoma of the ovary compared to the adenoma group of this organ. The stainability and positive rate of CD24 in adenocarcinoma group was much higher than in adenoma group, but it was not statistically significant. CONCLUSION: The intracytoplasmic staining pattern of CD24 was an important molecular marker for ovarian epithelial neoplasm which could help to define malignant transformation.

BACKGROUND: The purpose of this study was to evaluate the time course of PAC-1 and the P-selectin expression after the initiation of antiplatelets in acute ischemic stroke. METHODS: We serially measured PAC-1 and the P-selectin expression level using flow cytometry immediately before and at: one day, seven days, and one month after starting antiplatelets in 25 patients with acute cerebral infarctions. RESULTS: The P-selectin expression increased initially (p<0.05) and then from the seventh day, it began to decrease continually (post-hoc analysis, p<0.05). However, the PAC-1 expression did not reveal significant alterations during the follow-up period of one month. CONCLUSIONS: Although the P-selectin expression rapidly declined, the PAC-1 expression remained elevated for one month. Our results suggest that the P-selectin may be useful in monitoring platelet inhibition during the early period after cerebral infarction.

Lichen planus (LP) is a common, pruritic and inflammatory disease of the skin, hair follicles and mucous membranes. Immunologic mechanisms, especially cell-mediated immunity, play a major role in triggering the clinical expression of the disease. P-selectin is an adhesion molecule present within endothelial cells and mediates endothelial-leukocyte interactions. Therefore, it is considered that P-selectin plays an important role in LP. The aim of our study is to research the relation between P-selectin and LP. Serum P-selectin levels were determined with the enzyme- linked immunosorbent sandwich assay method in sera from 40 LP patients and 40 healthy controls. The serum levels of P-selectin were statistically significantly higher in the patients than in healthy controls (p < 0.05), in female patients (39.32 +/- 11.34 pg/ml) than in male patients (31.93 +/- 9.83 pg/ml) (p < 0.01), and in the patients with eruptive form (40.27 +/- 9.32 pg/ml) than in those with the localised (32.83 +/- 9.93 pg/ml) and hypertrophic (31.72 +/- 8.39 pg/ml) forms (both p < 0.01). In conclusion, we found a meaningful relation between LP and serum P-selectin levels.

In order to explore the factors that affect CD62p expression in platelet during the whole course of cryopreservated platelets preparation, CD62p expression of platelet was evaluated by flow cytometry assay. The whole course of cryopreservated platelets preparation in order included whole blood collection, centrifugation for fresh platelet-rich plasma preparation, addition of dimethyl sulfoxide, and freeze in -80 degrees C refrigerator and thaw in 38 degrees C water bath. Result showed that the CD62p expressed slowly from whole blood collection to addition of dimethyl sulfoxide, but expressed abruptly after freeze and thaw and it occupied 82 per cent of the whole expression. It was concluded that the whole blood collection, centrifugation for fresh platelet enriched plasma preparation and addition of dimethyl sulfoxide were optimized in the whole course, but the damage to platelets in the whole course of cryopreservation could not be avoided. It suggests that improved techniques are needed to reduce the damage to cryopreservated platelets.

CD62p expression was an important monitoring parameter for preserved platelets quality. To setup an optimized flow cytometry assay for preserved platelets based on the CD62p expression on platelets, the platelet samples were collected, 0.1 mmol/L persantine and 1.1 mmol/L EDTA were added into the modified TB S used to replace PBS dilution; the methodological evaluation were carried out. Results showed that 0.1 mmol/L persantine and 1.1 mmol/L EDTA achieved to prevent platelets activation during the test procedure. The favorable negative or positive samples were prepared for check of fluorescence antibody's quality to ensure the validity of results. CD61 was used to identify platelets for assay and improve veracity of assay. The special injector was also replaced by special big syringe needle for blood collection to reduce in vitro artifacts, and the prepared sample can be steady-going for 48 hours at 4 degrees C after fixed by 1% paraformaldehyde. It is concluded that this flow cytometry assay for CD62p positive platelets is simple and efficient.
