1.Ingredients and cytotoxicity of MTA and 3 kinds of Portland cements.

The aim of this study was to compare the compositions and cytotoxicity of white ProRoot MTA (white mineral trioxide aggregate) and 3 kinds of Portland cements. The elements, simple oxides and phase compositions of white MTA (WMTA), gray Portland cement (GPC), white Portland cement (WPC) and fast setting cement (FSC) were measured by inductively coupled plasma atomic emission spectrometry (ICP-AES), X-ray fluorescence spectrometry (XRF) and X-ray diffractometry (XRD). Agar diffusion test was carried out to evaluate the cytotoxicity of WMTA and 3 kinds of Portland cements. The results showed that WMTA and WPC contained far less magnesium (Mg), iron (Fe), manganese (Mn), and zinc (Zn) than GPC and FSC. FSC contained far more aluminum oxide (Al2O3) than WMTA, GPC, and WPC. WMTA, GPC, WPC and FSC were composed of main phases, such as tricalcicium silicate (3CaO.SiO2), dicalcium silicate (2CaO.SiO2), tricalcium aluminate (3CaO.Al2O3), and tetracalcium aluminoferrite (4CaO.Al2O3.Fe2O3). The significance of the differences in cellular response between WMTA, GPC, WPC and FSC was statistically analyzed by Kruskal-Wallis Exact test with Bonferroni's correction. The result showed no statistically significant difference (p > 0.05). WMTA, GPC, WPC and FSC showed similar compositions. However there were notable differences in the content of minor elements, such as aluminum (Al), magnesium, iron, manganese, and zinc. These differences might influence the physical properties of cements.

2.Chemical characteristics of mineral trioxide aggregate and its hydration reaction.

Mineral trioxide aggregate (MTA) was developed in early 1990s and has been successfully used for root perforation repair, root end filling, and one-visit apexification. MTA is composed mainly of tricalcium silicate and dicalcium silicate. When MTA is hydrated, calcium silicate hydrate (CSH) and calcium hydroxide is formed. Formed calcium hydroxide interacts with the phosphate ion in body fluid and form amorphous calcium phosphate (ACP) which finally transforms into calcium deficient hydroxyapatite (CDHA). These mineral precipitate were reported to form the MTA-dentin interfacial layer which enhances the sealing ability of MTA. Clinically, the use of zinc oxide euginol (ZOE) based materials may retard the setting of MTA. Also, the use of acids or contact with excessive blood should be avoided before complete set of MTA, because these conditions could adversely affect the hydration reaction of MTA. Further studies on the chemical nature of MTA hydration reaction are needed.

4.A Study on Public Nuisance in Seoul, Pusan and Daegu Cities: Part 1. Survey on Air Pollution and Noise Level.

During the period from July 1st to the end of November 1970, a survey on air pollution and noise level was made in Seoul, Pusan and Taegu, the three largest cities in Korea. Each city was divided into 4-6 areas : the industrial area, the semi-industrial area, the commercial area, the residential area, the park area and the downtown area. Thirty eight sites were selected from each area. A. Method of Measurement : Dustfall was measured by the Deposit Gauge Method, sulfur oxides by PbO2 cylinder method, suspended particles by the Digital Dust Indicator, Sulfur dioxide (SO2) and Carbon Monoxide (CO) by the MSA & Kitakawa Detector and the noise levels by Rion Sound Survey meter. B. Results: 1. The mean value of dustfall in 3 cities was 30.42 ton/km2/month, ranging from 8.69 to 95.44. 2. The mean values of dustfall by city were 33.17 ton/km2/month in Seoul, 32.11 in Pusan and 25.97 in Taegu. 3. The mean values of dustfall showed a trend of decreasing order of semi-industrial area, downtown area, industrial area, commercial area, residential area, and park area. 4. The mean value of dustfall in Seoul by area were 52.32 ton/km2/month in downtown, 50.54 in semi-industrial area, 40.37 in industrial area, 24,19 in commercial area, 16.25 in park area and 15.39 in residential area in order of concentration. 5. The mean values of dustfall in Pusan by area were 48.27 ton/km2/month in semi-industrial area, 36.68 in industrial area 25.31 in commercial area, and 18.19 in residential area. 6. The mean values of dustfall in Taegu by area were 36.46 ton/km2/month in downtown area, 33.52 in industrial area, 20.37 in commercial area and 13.55 in residential area. 7. The mean values of sulfur oxides in 3 cities were 1.52mg SO3/day/100cm2 PbO2, ranging from 0.32 to 4.72. 8. The mean values of sulfur oxides by city were 1.89mg SO3/day/100cm2 PbO2, in Pusan, 1.64 in Seoul and 1.21 in Taegu. 9. The mean values of sulfur oxides by area in 3 cities were 2.16mg SO3/day/100cm2 PbO2 in industrial area, 1.69 in semi-industrial area, 1.50 in commercial area, 1.48 in downtown area, 1.32 in residential area and 0.94 in the park area, respectively. 10. The monthly mean values of sulfur oxides contents showed a steady increase from July reaching a park in November. 11. The mean values of suspended particles was 2.89mg/m3, ranging from 1.15 to 5.27. 12. The mean values of suspended particles by sity were 3.14mg/m3 in Seoul, 2.79 in Taegu and 2.25 in Pusan. 13. The mean values of noise level in 3 cities was 71.3 phon, ranging from 49 to 99 phon. 14. The mean values of noise level by city were 73 phon in Seoul, 72 in Pusan and 69 in Taegu in that order. 15. The mean values of noise level by area in 3 cities showed a decrease in the order of the downtown area, commercial area, industrial area and semi-industrial area, park area and residental area. 16. The mean values of noise level by area in 3 cities indicated that the highest level was detected in the downtown area in Seoul and Taegu and in the industrial area in Pusan. 17. The daily average concentration o sulfur dioxides (SO2) in 3 cities was 0.081 ppm, ranging from 0.004 to 0.196. 18. The daily average concentration of sulfur dioxides by city were 0.092 ppm in Seoul, 0.089 in Pusan and 0.062 in Taegu in that order. 19. the weekly average concentration of carbon monoxides (CO) was 27.59 ppm. 20. The daily average concentration of carbon monoxides by city were 33.37 ppm. in Seoul, 25.76 in Pusan and 23.65 in Taegu in that order. 21. The concentration of SO2 and CO reaches a peak from 6 p.m. to 8 p.m. 22. About 3 times probably the daily average concentration of CO could be detected in the downtown area probably due to heavy traffic emission in comparison with that in the industrial area. 23. As for daily variation of the concentration of SO2 and CO it was found that the concentration maintains relatively higher value during weekdays in the industrial area and on the first part of the week in the downtown area.

5.Mineral trioxied aggregate and its substitutes.

Since its introduction in 1993, Mineral Trioxide Aggregate (MTA) has been shown to be superior to others in sealing, biocompatibility, and many other aspects of clinical endodontics. MTA is primarily Portland cement with bismuth oxide as a radiopacitifier. Although some studies suggested that the reasonable-priced Portland cement could be used instead of MTA, but MTAs are different from Portland cement in its composition, especially in heavy metal contents. Therefore, clinicians should be meticulous adapting the Portland cement as a MTA substitute.

6.Failure of orthograde MTA filling: MTA wash-out?.

Mineral trioxide aggregate (MTA), which was originally developed for repair of root perforations, is a biocompatible material with numerous clinical applications in endodontics. MTA must be allowed to set in the presence of moisture to optimize the material's physical and chemical properties. In the clinic, occasionally unset MTA has been detected after application of MTA on the tooth, and the reason has been unclear. This case report presents MTA washed-out for several years after placement at the root apex as an apical plug, and discusses the reason and things to consider in clinics.

7.Evaluation of the effect of blood contamination on the compressive strength of MTA modified with hydration accelerators.

OBJECTIVES: This study was performed to evaluate the effect of blood contamination on the compressive strength (CS) of Root MTA (RMTA) modified with Calcium chloride (CaCl2) and Disodium hydrogen phosphate (Na2HPO4) as setting accelerators over time. MATERIALS AND METHODS: A total of 110 cylindrical specimens of RMTA were divided into 6 experimental groups as follows: Group1, RMTA; Group 2, RMTA modified with CaCl2 (RMTA-C); Group 3, RMTA modified with Na2HPO4 (RMTA-N); Group 4, RMTA contaminated with blood; Group 5, RMTA-C contaminated with blood; Group 6, RMTA-N contaminated with blood. The CS of specimens in all groups was evaluated after 3 hr, 24 hr, and 1 wk. In the modified groups (groups 2, 3, 5, and 6) the CS of five specimens per group was also evaluated after 1 hr. RESULTS: Blood contamination significantly reduced the CS of all materials at all time intervals (p < 0.05). After 3 hr, the CS of specimens in the RMTA groups (with and without blood contamination) was significantly lower than those in the RMTA-C and RMTA-N groups (p < 0.05). The CS values were not significantly different at the other time intervals. In all groups, the CS of specimens significantly increased over time (p < 0.05). CONCLUSIONS: Blood contamination decreased the CS of both original and accelerated RMTA.

8.Physical and chemical properties of pulp capping materials based on bioactive glass.

OBJECTIVE:
To investigate the physical and chemical properties of pulp capping materials based on bioactive glass (BG).
METHODS:
Novel BG pulp capping materials were composed of powder and fluid. The powder was BG (82.36% SiO2, 15.36% CaO, and 2.28% P2O5) synthesized by using the sol-gel method combined with template technology. Two kinds of fluid were provided: (1) phosphate buffer (PB) solution and (2) phosphate buffer solution with 1% sodium alginate (SA) addition. After mixing the powder and fluid, BG-PB and BG-PB-SA were prepared. Setting time and compressive strength of the BG pulp capping materials were tested by setting time loading system and mechanical testing machine. Statistical analysis was performed using the independent sample t-test, with the significance set at 0.05. pH meters was used to test the pH of the BG pulp capping materials and mineral trioxide aggregate (MTA). The sealing ability of the BG pulp capping materials and MTA was tested by methylene blue dye leakage model. Statistical analysis was performed using one-way ANOVA analysis and LSD multiple comparison, with the significance set at 0.05.
RESULTS:
(1) Setting time: the initial and final setting time of BG-PB were (7.2±0.3) min and (12.7±0.9) min, respectively. And the initial and final setting time of BG-PB-SA was (7.5±0.3) min and (13.6±1.6) min. There was no significant difference between BG-PB and BG-PB-SA groups (P>0.05). (2) Compressive strength: the compressive strength of BG-PB was (16.5±1.8) MPa at 1 day and (14.1±3.7) MPa at the end of 28 days. However, the compressive strength of BG-PB-SA was (26.6±6.3) MPa on day 1 and (21.6±5.6) MPa on day 28, which was significantly higher than that of BG-PB (P<0.05). (3) pH: the pH of BG pulp capping materials' bulk immersed in simulated body fluid (SBF) went up to 8.06, and the highest pH of MTA was 8.47. Significant difference was observed between the BG pulp capping materials and MTA (P<0.05). (4) Sealing ability: the optical density (D) in positive control group was significantly higher than ln BG-PB, BG-PB-SA and MTA groups (P<0.05). And BG-PB and BG-PB-SA showed the similar favorable sealing ability with MTA, and no significant difference was observed among the three groups (P>0.05).
CONCLUSION
The novel BG pulp capping materials showed good physical properties, especially BG's setting time was short; BG pulp capping materials are promising.

9.Effects of mineral trioxide aggregate and ethanolic extracts of Shandong propolis on the biological properties of human dental pulp fibroblasts.

OBJECTIVE:
To evaluate the effect of mineral trioxide aggregate (MTA) and propolis from Shangdong province on the cell viability, mineralization and migration and anti-inflammatory ability of dental pulp fibroblasts.
METHODS:
The human dental pulp fibroblasts were cultured and subjected to 10 mg/L of propolis and 1:8 dilution of MTA extraction. The cell viability was evaluated with cell counting kit-8 (CCK-8) after 1, 5, 7 and 9 days. The cells in the upper inserts and the test culture media on the bottoms of 24-well plates interacted for 15 hours. Then the numbers of cells migrated through the permeable membranes were compared. The cells seeded in the 24-well plates were incubated in osteogenic medium with different materials for 21 days and stained with alizarin red S, then photographed. To evaluate the deposition of calcified matrix, the wells were destained with 100 mmol/L cetylpyridinium chloride. Finally, the cells were exposed to 1 mg/L lipopolysaccharide (LPS) to induce an inflammatory response, in the presence of propolis, MTA extraction. The cells were collected after 3 h, and the expressions of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-6 (IL-6) were determined using real-time polymerase chain reaction (real-time PCR). Statistical analysis was performed by one-way ANOVA and nonparametric tests (P<0.05).
RESULTS:
The cell viability of propolis group was significantly lower than those of MTA and control groups on days 5, 7 and 9, while MTA significantly increased the numbers of the viable cells on days 7 and 9. The migration cells of propolis group (26.67±2.52) were fewer than control group (61.33±4.93), and the cells of MTA group (80.00±2.65) were statistically more than those of the other two groups. The propolis group significantly induced more calcified matrix deposition than MTA group after 21 days of culture. Propolis significantly suppressed the expressions of IL-1β and IL-6 after LPS exposure compared with MTA and control groups.
CONCLUSION
The propolis from Shandong compared with MTA showed a certain degree of cytotoxicity, and had no significant effect on cell migration. On the other hand, propolis exhibited significant anti-inflammatory and mineralization promotion effect, suggesting that the active ingredients of propolis could be introduced as a supplement of pulp capping materials, or used as an irrigant or intracanal medicament due to its excellent anti-inflammatory effect. Propolis may have potential in vital pulp treatment of young permanent tooth suffering pulp inflammation.

10.An experimental study of expression changes of inducible nitric oxide synthase in rat dental pulp during direct pulp capping with two capping agents.

OBJECTIVETo observe the expression of inducible nitric oxide synthase (iNOS) in rat dental pulp after direct pulp capping with calcium hydroxide (CH) and mineral trioxide aggregate (MTA).
METHODSSamples from 42 Wistar rats were immediately collected at 1, 3, 7, 14, 21 and 28 days after direct pulp capping with CH and MTA. Pulp tissues were studided with immunohistochemical stain. Software for image analysis was used to test the optical density of each specimen. The data were analyzed by paired t-test and Dunnett-t test with SPSS 11.5.
RESULTSIn normal pulp, the stain of iNOS was negative. After direct pulp capping with CH or MTA, iNOS expression demonstrated weakly positive, positive, weakly positive, negative at different time, the expression was low immediately after capping but became strong after 3 days. Until 7 days the stain became less positive gradually and at 21 days the stain approached normal. The expression was mainly at the fibroblasts, odontoblasts, vascular endothelial cells, neutrophils, etc near the bottom of the pulp capping agent. The diffence of iNOS expression was significant 3 days after direct pulp capping with this two capping agents.
CONCLUSIONAfter direct pulp capping, iNOS expression increasd at first and then decreased, the diffence of capping agents will have an impact on the expression of iNOS 3 days after direct pulp capping.
