1.Embryonic stem cell differentiation towards cardiomyocytes and its application in cardiac tissue engineering

Embryonic stem cells (ESCs) show potential prospect of wide application in establishing cardiac pharmacological model system, forming three-dimensional cardiac tissues and constructing biological pacemaker. The process of ESCs differentiating into cardiomyocytes is to simulate heart growth and development in vitro. The cases of transplanting ESCs into animal models confirm that the cardiac function is improved. Moreover, the research and exploitation of tissue engineered cardiac muscle tissue with vessel system brings hope for clinical cardiac muscle restoration. ESC-derived pacemaker can functionally integrate with quiescent ventricular muscle cells to induce rhythmic electrical and contractile activities in vitro. However, the immunological rejection limits its clinical application. Although ESCs have strong differentiation potential, the problems such as the differentiation process towards cardiomyocytes, ESC induction and signal regulatory pathway, and whether induced cardiomyocytes can substitute in vivo cardiomyocytes still need exploration.
2.Regard and Perfection of Radiation and Health Protection for Radio-subjects

Objective To explore in our country how to do a good job of radiation health protection for radio-subjects.Methods The people are divided into physical examination group and in & out-patient group,the radiation health protection status of the current domestic and foreign subjects are contrastively analyzed.Results The western developed countries radiology examinations have stronger sense of patient protection,but the prevalent problems are neglected and abused in domestic hospital.Conclusion The radiation health protection in healthy people and medical patients can be regarded and consummated,which is people-centered and can be strengthen radiological protection awareness,strictly implement radiation protection standards of hygiene and put an end to the indiscriminate seizure.
3.Study of event-related potentials N400 in children with Tic disorders

Objective To investigate the change of event-related potentials(ERPs)N400 and its significances in children with Tic disorders.Methods N400 of 35 children with Tic disorders(case group)and 20 healthy children(control group)were taken.The results of case group and control group were compared with each other.Results N400 latency of the case group was(706.87?198.99)ms,and the amplitude was(26.12?13.64)?V;N400 latency of the control group was(485.75?85.76)ms,the amplitude was(22.35?19.76)?V.There was obvious difference in latency between two groups(P0.05).Conclusion ERPs N400 latency of children with Tic disorders prolong obviously which shows cognitive dysfunction in the patients.

Objective To investigate the effect of midazolam on inflammatory response in patients with moderate and severe craniocerebral injury and its protective effect on the brain and mechanism.Methods A prospective study was conducted.One hundred and twenty patients with moderate and severe craniocerebral injury admitted to the Intensive Care Unit (ICU) of Jiangsu Subei Peoples' Hospital from April 2013 to July 2016 were enrolled,and they were divided into a conventional treatment group (58 cases) and a midazolam group (62 cases) according to the random number table method.Both groups were given conventional western medicine treatment,and in cases with surgical indications,operations were performed;in midazolam group,additionally intravenous injection of midazolam 2-3 mg was given firstly,and then continuous intravenous infusion of the drug 0.05-0.10 mg· kg-1· h-1 was applied by a pump,and in operative patients,the above management was given 3 hours after operation.The mean arterial pressure (MAP),heart rate (HR),Riker sedation agitation score (SAS) and electroencephalogram bispectral index (BIS) were measured before and after treatment for 24,48 and 72 hours,respectively.The levels of interleukin-6 (IL-6) and soluble nerve chemotactic protein (sFkn) in plasm and cerebrospinal fluid (CSF) were measured by double antibody sandwich enzyme linked immunosorbent assay (ELISA) at each time point;the incidence of epilepsy and 28-day mortality were recorded.Results Before and after treatment,the MAP and HR in the two groups of patients were stable,the difference being not statistically significant (both P > 0.05).Compared with those before treatment,after treatment the SAS score and BIS in two groups of patients were gradually decreased,and at 72 hours reached the lowest levels (SAS score:conventional treatment group was 3.8 ± 1.0 vs.5.7 ± 2.0,midazolam group was 3.6 ± 0.9 vs.5.8 ± 1.7;BIS:conventional treatment group was 69± 12 vs.82± 12,midazolam group was 72± 15 vs.82± 12,all P < 0.05),but there were no significant differences between the two groups (all P > 0.05),suggesting that the two groups had achieved the desired goal of sedation.ELISA results showed:compared with those before treatment,after treatment for 24 hours,the CSF IL-6,sFkn and plasm sFkn levels were temporarily increased in short term,and then showed a tendency of gradually decreasing,and the plasm IL-6 presented persistently descending in the conventional treatment group,while in the midazolam group,since 24 hours after treatment,each index showed a trend of decrease and continued to 72 hours.After treatment at each time point,the CSF and plasm levels of IL-6 and sFkn were significantly lower in midazolam group than those of the conventional treatment group,and reached to the minimal levels at 72 hours [CSF:IL-6 (ng/L) was 251.6 ± 145.7 vs.347.3 ± 146.4,sFkn (ng/L):289.7 ± 79.3 vs.423.6 ± 132.8;plasm:IL-6 (ng/L) was 54.4± 27.3 vs.85.6 ± 41.8,SFkn (ng/L):919.9±426.3 vs.1 199.4 ± 414.8,all P < 0.05].The incidence of epilepsy in the midazolam group was obviously lower than that in the conventional treatment group [1.61% (1/62) vs.10.34% (6/58),P < 0.05],but there was no significant difference between midazolam group and the conventional treatment group in the 28-day mortality [11.29% (7/62) vs.10.34% (6/58),P > 0.05].Conclusion Midazolam can reduce the incidence of epilepsy in patients with moderate and severe traumatic brain injury,and its brain protective effect may be related to the decrease of CSF and plasm IL-6 and sFkn levels.
8.Clinical study on inlay butterfly cartilage myringoplasty

Objective To evaluate the application of the inlay butterfly cartilage graft technique in myringoplasty.Methods Thirty--eight ears in 38 patients (experiment group) with dry central drum perforations were treated with inlay butterfly cartilage myringoplasty,and 46 ears in 46 patients with underlay temporalis fascia myringoplasty (control group).Results After a mean follow-up of 6 months,the healing rate did not differ between two groups (92.1% in experiment group,91.3% in control group,P> 0.05).Average pure-tone hearing threshold improved than 10 dB and closure of the air-bone gap (ABG) within 10 dB were not different (P > 0.05).The cured patients were followed up for 12-38 months,there was no ear perforation of drum in experiment group,but 4 ears in control group.Condusion Inlay butterfly cartilage myringoplasty is the reliable and ideal method for tzeating dry central drum pedoration with high success rate.
9.Study on immunogenicity of B cell epitopes of Epstein-Barr virus latent membrane protein-2

Objective To analyze the immunogenicity of selected B-cell epitopes of Epstein-Barr virus (EBV) latent membrane protein-2 (LMP2). Methods Three potential dominant B-cell epitopes of LMP2199-209, LMP2318-322 and LMP2381-391 from EBV LMP2 had been predicted using bioinformaties methods. The gene fragments of three epitopes were cloned respectively into pET32a(+) vector and transformed into E. coli strain BL21 (DE3). After identification by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting, the expression products were purified by Ni-NTA agarose affinity chromatography. BALB/c mice in immunized groups were immunized by multi-point intracutaneous injection with the three purified epitope proteins,respectively; and mice in control groups were injected with pET32a (+) protein or phosphate buffered saline(PBS), respectively. The sera from mice at week O, week 3 and week 6 of injection were collected for determination of epitope-specific antibody IgG by enzyme linked immunosorbent assay (ELISA) using epitope proteins as coating antigens. The ability of serum antibody recognizing nature EBV antigen was determined at week 6 of immunization. Results Three epitope proteins of LMP2199-209 ,LMP2318-322 and LMP2381-391 were successfully expressed in prokaryotic system. Epitopespecific antibodies IgG could be detected respectively in the sera of all immunized mice, and the levels of antibodies increased with immunized time increasing. The antibody levels in LMP2318-322 immunized group at week 3 and week 6 were significantly higher than that of pET32a (+) protein control group (F= 493.85 and 773.99, respectively; both P<0. 05), and the antibody levels in LMP2381-391 immunized group at week 3 and week 6 were also significantly higher than that of pET32a (+) protein control group (F= 926.33 and 309.14, respectively; both P<0.05). Antibody level in LMP2199-209 immunized group at week 6 was significantly higher than that of pET32a ( + ) protein control group (F=87.27, P<0.05). The antibody IgG in serum from immunized mice with three epitope proteins could all recognize nature EBV antigens, especially LMP2199-209 and LMP2381-391 immunized groups.Conclusions Three possible dominant epitopes of LMP2199-209, LMP2318-322 and LMP2381-391 from EBV LMP2 are prepared by prokaryotic expression system and exhibit obvious immunogenicity, which could be used for further research of EBV infection and related tumor vaccine.
10.The analaysis of excessive daytime sleepiness and relevant factors in the elderly

Objective To investigate the prevalence rate and related factors of excessive daytime sleepiness (EDS) in the elderly. Methods Epworth Sleep Scales (ESS) was used to make a randomized questionnaire among the retired elderly. Results 1 000 questionnaires were released and totally 768 qualified answers were returned. The mean score of ESS was 4.22?0.10 and the score ≥8 was used as an abmormal value. The prevlence rate of EDS in the elderly group was 10.9%.
