1.Treatment of tibial plateau fractures with arthroscopy

Objective To discuss clinical results of the minimally invasive internal fixation with arthroscopy to treat tibial plateau fractures. Methods 12 cases of tibial plateau fractures (SchazkerⅠ~Ⅳ) were treated with arthroscopic internal fixation. Results The follow ups of the patients lasted from 4 to 6 months. Anatomic reduction was achieved in all the patients of tibial plateau fractures. The outcome was satisfactory, without complications. Conclusion Arthroscopic internal fixation is easy, and can achieve good results in the treatment of tibial plateau fractures.
2.Roles of regulation of plasma cell function by Blimp1-B cell maturation antigen in the pathogenesis of systemic lupus erythematosus

Objective To detect the expression of Blimp1 gene in MRL/lpr mice,so as to provide new ideas for plasma cell-targeting therapy of systemic lupus erythematosus (SLE).Methods Electrophoretic mobility shift assay (EMSA) and chromatin immunoprecipitation analysis (ChIP) were performed to evaluate the regulatory effect of Blimp1 on B cell maturation antigen (BCMA),and fluorescence-based quantitative PCR was carried out to detect the expression of Blimp1 mRNA in spleen and lymph node tissue of MRL/lpr mice and normal control mice.The intergroup difference in Blimp l mRNA expression was assessed by rank sum test.Results Blimp1 could bind to the BCMA gene promoter.Increased Blimp1 mRNA expression was observed in both the spleen and lymph node tissue of MRL/lpr mice compared with the normal control mice (Z =2.609,3.402,respectively,both P < 0.01).Conclusions BCMA appears to be the target gene of Blimp1,and Blimp1 may plays a certain role in the maintenance of plasma cell survival and antibody production via directly regulating BCMA gene expression.
3.Clinical Analysis on Combining with Early Short Time Venous-venous Hemofiltration in Severe Acute Pancreatitis with Acute Lung Injury

Objective To explore the therapeutic effects of early short time venous-venous hemofiltration(SVVH) on severe acute pancreatitis(SAP) with acute lung injury(ALI).Methods Twenty-there patients with SAP and ALI were treated by routine project,and among them twelve patients accepted SVVH therapy.During the therapy,life sign、PaO2/FiO2 and APACHEII score were registered.Results Compared with control group,the clinical representation and organ function of SVVH group meliorated. PaO2/FiO2 were raised, APACHEII score and death risk were declined.Conclusions The early short time venous-venous hemofiltration(SVVH) on severe acute pancreatitis(SAP) with acute lung injury(ALI) could improve clinical symptom ,protect the organ dysfunction and decline the death risk.
4.Repair of trauma soft tissue defects of leg by microsuegery

Objectve To probe the effect of treating the trauma soft tissue defects of the leg by microsurgical technique. Methods Ninety-nine cases of the trauma soft tissue defects of leg were treated by used 12 kinds of microsurgical flaps.Result All cases with soft tissue defects of the leg were repaired successfully.Follow-up 6 months to 10 years,the results were satisfactory and function improvement in 99 cases. Conclusions Accoding to soft tissue defects of size,the section and the technique of the operator to select the different microsurgical flap could be suitable and resolvable problem of soft tissue defects.
5.Chemotaxis on Umbilical Vein Endothelial Cells of SMMC7721 Hepatic Carcinoma Cells with Angiopoietin Gene Expression

Objective To observe the chemotactic role on umbilical vein endothelial cells of SMMC7721 hepatic(carcinoma) cells with angiopoietin gene expression in order to study the effects of angiopoietin on hepatocellular carcinoma angiogenesis.Methods Angiopoietin gene 1(Ang-1) fragment and Ang-2 fragment was transfected into SMMC7721 liver carcinoma cell line by Lipofectamine induced gene transfection technique.The chemotactic role of SMMC7721 liver carcinoma cell line on umbilical vein endothelial cells was observed through microchemotaxis analysis.Results The chemotactic response of the Umbilical vein endothelial cells was obviously improved with Ang-1 expression (P0.05).Conclusion Ang-1 is a chemotactic factor for vascular endothelial cell and a promoter for angiogenesis,whereas Ang2 does not show obvious chemotactic role.
6.Percutaneous minimally invasive plating for tibiofibular fractures

Objective To discuss clinical results of the percutaneous minimally invasive LC DCP plating in treatment of tibiofibular fractures which was based on biological osteosynthesis and indirect reduction. Methods 28 patients with tibiofibular fractures were treated with indirect reduction and LC DCP plating through the tunnel inside tibia. Results All the patients were followed up for 10 to 18 months (averaging 15 m). X ray films showed that the callus appeared 4 to 7 weeks postoperatively (averaging 4.6 w). The fracture healing time were 12 to 18 weeks (averaging 12.8w). All the patients healed without fracture nonunion, delayed union or plate loosening. Plates were removed without refracture in 15 patients. Conclusions The percutaneous minimally invasive LC DCP plating for tibiofibular fractures can meet the major requirements of biological osteosynthesis and result in good results.
7.Surgical experience in the treatment of refractory cholelithiasis

Objective To investigate the operative techniques for refractory cholelithiasis. Methods The clinical data of 521 patients with refractory cholelithiasis who had been admitted to People's Hospital of Hunan Province from January 1990 to December 2007 were retrospectively analyzed. Results All patients received surgery. After the operation, 3 patients died of liver and kidney failure, the remaining 518 patients were cured without severe complications. The results of B ultrasound and computed tomography showed residual stones in 78 patients (15.1%). Four hundred and twenty-three patients (81.7%) were followed up for 5 months to 17 years (mean, 7.5 years), and the rate of positive effect was 90.1% (381/423). Conclusions Most of the refractory cholelithiasis can be cured radically. Individualized surgical planning, fine and standard surgical procedure are key to the treatment effect.

ObjectiveTo study the expression of angiopoietin in hepatocellular carcinoma tissue, and its relation with tumor angiogenesis and clinical significance. Methods Using immunohistochemistry and in situ hybridization, we observed the expression of angiopoietin 1(Ang 1) and angiopoietin 2(Ang 2) mRNA in 21 HCC patients and 5 normal liver tissues. The correlation between angiopoietin expression and several pathological markers was analyzed with computer statistical tools. Results Ang 1 equally expressed in tumor tissue, the adjacent noncarcinomatous tissue and normal liver. Ang 2 was highly expressed in tumor tissue, especially in tumor margin. 9.Human bone marrow mesenchymal stem cells into vascular endothelial cells before and after differentiation induced differential expression of eNOS/activity and its metabolites

Objective To investigate the differences in eNOS gene expression,activity and its metabolites before and after human bone marrow mesenchymal stem cells (hBMSCs) are induced into vascular endothelial cells.Methods hBMSCs were induced into vascular endothelial cells.The morphological changes of the cells were observed under inverted microscope.Transwell assay was used to detect the cells' migration ability.The protein expression of eNOS was detected by immunofluorescence and Western blot.The activity of eNOS was detected by ELISA and the content of NO in cell culture supernatant was determined by nitrate reduction method.Results Compared with those in the undifferentiated group,the morphological changes of the differentiated cells were obvious.Cell migration ability increased by 238.10% (73.000±7.002 vs.21.000±4.359,P<0.05).The expression of eNOS protein increased by 114.72% (0.423±0.011 vs.0.197±0.079,P<0.05).The activity of eNOS was enhanced by 157.49% (4.967±0.073 vs.1.929±±0.103,P<0.05).The synthesis and release of NO increased by 155.67% (184.909±1.853 vs.72.323±0.426,P<0.05).Conclusion After hBMSCs are induced into endothelial cells,the expression of eNOS gene increases,their activities increase,synthesis and release of the metabolite NO increase.It may provide a basis for prevention and treatment of cardiovascular diseases with stem cells.
10.Anti-tumor Effect of Curcumin on Triple-negative Breast Cancer Mouse Model and its Mechanism

Objective To observe the anti-tumor effect and mechanism of curcumin on triple-negative breast cancer mouse model.Methods Human breast cancer cells MDA-MB-231 were inoculated to the chest subcutaneous fat pad of mice(about 1 million cells in 0.1 mL cell suspension).At day 2 after tumor inoculation,nodules were seen at inoculation site (tumor formation rate being 100%).Forty successfully-modeled mice were randomly divided into low-,middle-,and high-dose curcumin groups (10,20,40 mg/kg) and model control group,10 mice in each group.The medication lasted for 30 continuous days.After medication,the blood was taken out from the orbital venous plexus,and the serum was separated for the detection of tumor necrosis factor alpha (TNF-α) and interleukin-6 (IL-6) contents.Spleen was separated for the calculation of organ coefficient,tumor mass was weighed,and the expression levels of Bcl-2 and caspase-3 were detected by Western blotting method.Results Compared with the model control group,caspase-3 expression level was significantly increased in the three curcumin groups(P < 0.05 or P < 0.01),but the contents of TNF-α and IL-6,tumor weight,spleen index and Bcl-2 expression were significantly decreased(P < 0.05 or P < 0.01).Conclusion Curcumin has inhibitory effect on triple-negative breast cancer mouse model through promoting the apoptosis of breast cancer cells.
