2.Advances in organoids of the digestive system.

Organoid is a newly developed cellular there-dimensional culture system in recent years. Organoids have a three-dimensional structure, which is similar to that of the real organs. Together with the characteristics of self-renewal and reproduction of tissue origin, organoids can better simulate the function of real organs. Organoids provide a new platform for the study of organogenesis, regeneration, disease pathogenesis, and drug screening. The digestive system is an essential part of the human body and performs important functions. To date, organoid models of various digestive organs have been successfully established. This review summarizes the latest research progress of organoids of taste buds, esophagi, stomachs, livers and intestines, and prospects future application of organoids.

Fibrous hamartoma of infancy (FHI) is a rare benign soft tissue tumor that is composed of an organoid pattern of fibrous tissue and primitive mesenchyme and adipose tissue. The majority of cases of fibrous hamartoma of infancy occur within the first year of life as a single painless subcutaneous nodule. We experienced and report on an uncommon case of protruding fibrous hamartoma of infancy in an 1-year-old girl.

6.Research progress on vascularization of organoids.

Organoids are three-dimensional structures formed by self-organizing growth of cells in vitro, which own many structures and functions similar with those of corresponding in vivo organs. Although the organoid culture technologies are rapidly developed and the original cells are abundant, the organoid cultured by current technologies are rather different with the real organs, which limits their application. The major challenges of organoid cultures are the immature tissue structure and restricted growth, both of which are caused by poor functional vasculature. Therefore, how to develop the vascularization of organoids has become an urgent problem. We presently reviewed the progresses on the original cells of organoids and the current methods to develop organoids vascularization, which provide clues to solve the above-mentioned problems.

Organoid models are used to study kidney physiology, such as the assessment of nephrotoxicity and underlying disease processes. Personalized human pluripotent stem cell-derived kidney organoids are ideal models for compound toxicity studies, but there is a need to accelerate basic and translational research in the field. Here, we developed an automated continuous imaging setup with the "read-on-ski" law of control to maximize temporal resolution with minimum culture plate vibration. High-accuracy performance was achieved: organoid screening and imaging were performed at a spatial resolution of 1.1 μm for the entire multi-well plate under 3 min. We used the in-house developed multi-well spinning device and cisplatin-induced nephrotoxicity model to evaluate the toxicity in kidney organoids using this system. The acquired images were processed via machine learning-based classification and segmentation algorithms, and the toxicity in kidney organoids was determined with 95% accuracy. The results obtained by the automated "read-on-ski" imaging device, combined with label-free and non-invasive algorithms for detection, were verified using conventional biological procedures. Taking advantage of the close-to-in vivo-kidney organoid model, this new development opens the door for further application of scaled-up screening using organoids in basic research and drug discovery.

8.A Case of Sebaceous Adenoma.

Sebaceous adenoma is a benign organoid tumor of proliferating, incornpletely differentiated sebaceous glands. We report a case of sebaceous adenoma in a 15-year-old male. He had a 1.7 x 2.2 cm sized, asymptomatic, srnooth, round, raised, firm, reddish tumor on the left quadrant of abdomen. Histopathologically, the tumor was composed of lobules of irregular size and shape, and sharply demarcated from the surrounding tissue, The most lobules showed undifferentiated germinative and mature sebaceous cells in about equal number. Sebaceous cells in lobules were stained for prekeratin by means of avidin-biotin-peroxidase complex method. Ten months after surgical excision there was no evidence of recurrence.

In about 20 to 30 percent of cases with organoid nevus (nevus sebaceus), benign and/or ma-al lignant tumors of the epidermis and adnexae may develop. We describe a 71-year-old woman with sebaceous carcinoma and basal cell epithelioma developed in organoid nevus.

10.A Case of Verruca Vulgaris Assocaited with Nevus Sebaceus of Jadassohn.

Nevus sebaceus of Jadassohn is organoid nevus that tends to develop secondary tumors on the lesion in adulthood. We report a case of verruca vulgaris associated with nevus sebaceus in a 8-year-old girl. Histologic findings showed typical findings of verruca vulgaris such as papillomatosis, hyperkeratosis, and acanthosis with vaculoated cells as well as the findings of nevus sebaceus. To our knowledge, this is the first case report of verruca vulgaris associated with nevus sebaceus in the Korean dermatologic literatures.
