1.Evaluation of germplasm resource of Ophiopogon japonicus in Sichuan basin based on principal component and cluster analysis.

OBJECTIVETo compare and appraise the quality of germplasm resource of Ophiopogon japonicus in Sichuan basin.
METHODAccording to the main contents and yield traits, 24 wild germplasm resources of O. japonicus from different areas of Sichuan basin were comprehensively compared by the SPSS 17.0 software with principal component analysis and cluster analysis.
RESULTThe six samples of Ziyang, Jianyang, Leshan, Yibin, Chongqing, Mianyang, their comprehensive evaluation value of quality were higher than the others, and the sample of Ziyang had the best quality, the sample of Dazhou had the least quality, the results of the cluster analysis to raw data were also shown a similar results as principal component analysis.
CONCLUSIONThe wild resources of O. japonicus in Sichuan basin is rich, there are much differences among their quality; the method, through principal component analysis to study the comprehensive evaluation of the O. japonicus quality, is reliability and the results of cluster analysis is also support the conclusions, it could be able to provide a reference to select high O. japonicus quality resources.

2.Comprehensive comparison between erective and creepy Ophiopogon japonicus of Sichuan.

OBJECTIVETo compare the agronomic characters, HPLC fingerprints, the content of main component and amino acid between erective and creepy Ophiopogon japonicus of Sichuan.
METHODAgronomic characters were measured by conventional methods; HPLC was applied on a C18 column with CH3OH-CH3CN-2% CH2COOH solution by gradient elution, quercetin was used as the internal standard reference, the contents of total saponins, flavone and polysaccharide were determined by UV spectrophotometry and amino acid was determined by automatic amino acid analyzer.
RESULTThere were extremely significant differences in the most agronomic characters between erective and creepy O. japonicus of Sichuan. The yield per plant was closely relevant to the roots number and the fresh weight of aerial part. The differences were not significant both in 69 common peaks and 23 uncommon peaks in HLPC fingerprints and the content of main component between erective and creepy O. japonicus.
CONCLUSIONThere is no obvious difference in chemistry component between the two types of O. japonicus. The yield per plant was closely relevant to the roots number and the fresh weight of aerial part. In cultivation it is appropriate to choose the creepy O. japonicus.

3.Effects of soil factor on active components of Radix Ophiopogonis.

OBJECTIVETo study the effects of soil factors on the active components in Radix Ophiopogonis.
METHODThe content of polysaccharide, flavonoids, saponins, water-soluble extract and inorganic elements in Radix Ophiopogonis gathered from 7 different places were compared, physical and chemical properties and inorganic elements of soil were analyzed. The path and grey connection analysis were applied for studying the effects on the active components of Radix Ophiopogonis.
RESULT AND CONCLUSIONThe concentrations of mineral elements in plant were mainly adjusted by active absorption. The active components in Radix Ophiopogonis was primarily effected by soil enzyme activity, the other important factors were potassium, pH, and organic matter. K, Fe, Mn, B, Ba, Zn of soil also had much influence on it then others inorganic elements.

4.Simultaneous determination of three homoisoflavonoids in Ophiopogon japonicus by HPLC.

Three homoisoflavonoids, 6-aldehydo-3-ophiopogonanone A, methyl ophiopogonanone A and ophiopogonanone A from Ophiopogon japonicus were analyzed simultaneously by HPLC with acetonitrile-water containing 0.5% H3 PO4 (58:42) as the mobile phase, and the detection wavelength was set at 296 nm (0-14 min) and 275 nm (14-22 min). The mean recoveries of three homoisoflavonoids were 99.41%-99.86% (RSD 0.82%-1.05%). The linear response ranges of. 6-aldehydo-3-ophiopogonanone A, methyl ophiopogonanone A and ophiopogonanone A were 0.165-0.990 microg (r = 0.999 9), 0.153-0.918 microg (r = 0.999 9), and 0.270-1.620 microg (r = 0.999 9), respectively. This method was certified to be accurate and reliable and can be used for quality control of O. japonicus.

Dengzhan Shengmai Capsules( DZSMC),a well-known traditional Chinese medicine( TCM) formula,is comprised of the main drug of Erigeron breviscapus,and supplemented with Panax ginseng,Ophiopogon japonicus and Schisandra chinensis,with functions of supplementing Qi and nourishing Yin,promoting blood circulation and strengthening brain. DZSMC is the only Chinese patent drug with A-level evidence-based medicine in secondary prevention for stroke and ranks first among TCMs for neurological treatment. Modern studies indicate that the chemical constituents of DZSMC mainly include flavonoids,phenolic acids,lignans,saponins and so on. Pharmacological experimental studies have shown that DZSMC has such pharmacological effects as anti-oxidation,anti-inflammatory and anti-myocardial ischemia. DZSMC is mainly used in the convalescent care of ischemic cardiovascular and cerebrovascular diseases,and is often used in combination with various conventional therapeutic drugs to exert clinical efficacy through brain protection,neuroprotection,etc.,and improve clinical symptoms in patients. In this review,according to domestic and international related literature combined with research results obtained by our project,the research advances in the chemical constituents,pharmacological effects and clinical application of DZSMC have been systematically reviewed and summarized,providing reference and support for further study and secondary development of the formula.

7.Studies on using spray drying technique in preparing Ophiopogon japonicus saponin enteric microsphere by orthogonal experimental design.

OBJECTIVETo explore the best technique parameters on preparing ophiopogon japonicus saponin enteric microsphere by spray drying technique.
METHODThe best technique parameters were investigated by orthogonal experimental design and by the target, such as surface appearances, encapsulated efficiency, etc.
RESULTThe best technique parameters included the inlet temperature (90 degrees C ), the feeding speed (10 mL x min(- 1)), and the rotate speed of atomizer (50 r x min(-1)).
CONCLUSIONOphiopogon japonicus saponin enteric microsphere accorded with the expecting demand. The main influencing factor was inlet temperature . It is suitable to industrialize in preparing Traditional Chinese Medicine microsphere.

8.Determination of diosgenin and ruscogenin in Radix Ophiopogonis by nonaqueous capillary electrophoresis.

Nonaqueous capillary electrophoresis is used for the determination of the contents of diosgenin and ruscogenin in Radix Ophiopogonis. The operating buffer was composed of 20 mmol x L(-1) Na2B4O7-HCl (pH 7.61) in 70% methanol. The applied voltage was 25 kV and detection potential was at +0.70 V. With these conditions, the components were successfully separated. The content of diosgenin in Radix Ophiopogonis was 0.018 mg x g(-1) and ruscogenin was 0.008 mg x g(-1). The average recoveries of diosgenin and ruscogenin were 102% and 99.2%, respectively. A new method of the quality control of diosgenin and ruscogenin in Radix Ophiopogonis is provided.

9.Isolation, purification and structural analysis of a polysaccharide MDG-1 from Ophiopogon japonicus.

AIMTo separate and purify the anti-myocardial ischemic polysaccharide fraction with a homogenous molecular weight from Ophiopogon japonicus, then study the chemical structure of the parts.
METHODSCrude polysaccharides were prepared by extracting the tube root fraction of Ophiopogon japonicus with water, then precipitation with ethanol. From the crude polysaccharides, the polysaccharide of MDG-1 was separated and purified using ultrafiltration, DEAE Sepharose FF and Sephadex G-25 column chromatography. Its structure was studied by complete hydrolysis, periodate oxidation, Smith degradation, methylation analysis, 1H NMR and 13C NMR analysis etc.
RESULTS AND CONCLUSIONMDG-1 was a water-soluble beta-D-fructosan, containing a backbone composed of Fruf (2 --> 1), and a branch of Fruf (2 --> 6) Fruf (2 --> per average 2. 8 of main chain residues. Mn, Mw and Mp of MDG-1 were 3 400, 4 800 and 5 000, respectively. MDG-1 contains trace of Glc, which maybe connect to its reducing terminal. Molar ratio of Fru and Glc is approximately 35: 1.

10.HPLC simultaneous determination of contents of 5 saponin constituents in Ophiopogonis Radix.

This research is to establish an HPLC method for the simultaneous determination of ophiopogonin D, ophiopogonin D', ophiopogonin C, deacetylophiopojaponin A and ophiogenin-3-O-α-L-rhamnosyl-(1-->2)-β-D-glucoside in Ophiopogonis Radix. HPLC-ELSD analysis was performed on a Kromasil 100-5 C₁₈ column (4.6 mm x 250 mm, 5 µm), with the mobile phase of acetonitrile (A) -water (B) in gradient elution mode (0-45 min, 35%-55% A), at a flow rate of 1 mL · min⁻¹. The column temperature was 35 °C and the drift tube temperature was 100 °C in a gas flow rate of 3.0 L · min⁻¹. The result showed that baseline of all the 5 constituents was well separated, and every constituent had wide linearity range and good linear relation (r > 0.999). The recovery rate was between 95.75% and 103.1%. The new established method for simultaneous determination of saponin constituents in Ophiopogonis Radix was sensitive and has good, repeatability. It could be applied to quality evaluation of Ophiopogonis Radix.
