OBJECTIVE: To observe the effects of matrine on proliferation and apoptosis of human renal cell carcinoma cell line GRC-1 in vitro, and to explore its mechanism. METHODS: The human renal cell carcinoma cell line GRC-1 was treated with matrine of different concentrations for 24, 48, 72 and 96 h respectively. The MTT assay was used to evaluate the cytotoxic effects of matrine on GRC-1 cells. The transmission electron microscope and flow cytometry were utilized to observe and detect the apoptosis of GRC-1 cells induced by matrine. The expression levels of Bcl-2 and Bax proteins were evaluated by streptavidin-biotin-peroxidase method. RESULTS: The matrine of different concentrations all have cytotoxic effects on GRC-1 cells, with obvious dose- and time-dependent effects. The apoptosis induced by matrine was confirmed in GRC-1 cells. With intervention of matrine (1.5 g/L) for 12 h, the expression level of Bcl-2 in GRC-1 cells was decreased while the expression level of Bax was increased as compared with those in the untreated group. CONCLUSION: The proliferation-inhibiting effects of matrine on human renal cell carcinoma cell line GRC-1 may be related to down-regulating the ratio of Bcl-2/Bax protein expression and promoting the apoptosis.

4.Mutation and polymorphism of the ameloblastin gene in ameloblastomas.

OBJECTIVETo investigate the presence of mutation or polymorphism of ameloblastin (AMBN) gene in ameloblastomas.
METHODSGenomic DNA was extracted from frozen tissues of 10 ameloblastomas and one malignant ameloblastoma. AMBN gene alterations were detected by PCR-direct sequencing. Restriction fragment length polymorphism (RFLP) analysis was used to further determine the nature of the changes in AMBN detected in tumor samples in comparison to 100 control samples.
RESULTSAMBN mutation was not identified in all 11 tumor samples. The 7 types of AMBN gene alteration identified in 9 cases were proven to be polymorphisms, three of which were not previously reported. The frequency of genotype and allele of the three SNPs complied with the Hardy-Weinberg equilibrium. Neither genotype nor allele frequency showed a significant association with ameloblastoms.
CONCLUSIONSAMBN gene mutation is not identified in the present group of ameloblastomas. The frequently detected AMBN alterations in ameloblastomas are polymorphisms, which appear to be unrelated to the occurrence of ameloblastomas.

5.Circulating endothelial cell injury in on-pump and off-pump coronary-artery bypass grafting.

OBJECTIVETo investigate the difference in circulating endothelial cell (CECs) injuries following on-pump and off-pump coronary-artery bypass surgery.
METHODSWe randomly assigned 48 patients scheduled for urgent or elective coronary artery bypass grafting into two groups to receive on-pump and off-pump procedures. Blood samples were obtained before anesthesia, at the end of operation, and on days 1 and 3 after the operation, and CECs were isolated and counted using dynabeads coated with the specific antibody of CD146. Single cell gel electrophoresis was used to observe the morphological changes of the CECs.
RESULTSIn the cardiopulmonary bypass (CPB) group, the number of CECs was significantly greater than that in non-CPB group (P<0.05) at the end of surgery and 1 day after the operation. On postoperative day 3, the number CECs was similar between the two groups (P>0.05). The length of the comet tail was longer in CPB group with stronger fluorescence intensity than in the non-CPB group.
CONCLUSIONCompared with of-pump coronary artery bypass grafting, on-pump coronary artery bypass grafting results in more serious CEC injury, which is closely related to the prognosis.

6.Research progress on foot and ankle injuries of occupants in frontal motor vehicle crashes

Road traffic injuries have become a major social issue. With the development of technology, legislation and safety consciousness, the fatal rate of brain and chest injuries has been declined. However, as to prevent foot and ankle injuries of occupants still no effectual protective devices have been developed. In this paper, research progress on foot and ankle injuries of occupants was reviewed, which found out that most of such injuries in the frontal motor vehicle crashes occurred more easily to the drivers, which was possibly due to the special loading on their lower extremity during braking. Although there still exists a continuing debate about taxonomy of the frontal motor vehicle crashes, researchers have approached unified understanding that foot and ankle injuries are usually caused in the real-world narrow object frontal crashes. Until now, studies on foot and ankle injury and its protective mechanisms in various types of crashes are rarely conducted. A combination of the in-vitro experiment and computational modeling would be an ideal method to solve this issue.
7.Analysis of pain knowledge, belief, behavior level and influencing factors of cancer pain patients

Objective:To understand the pain knowledge, belief and behavior level of cancer pain patients, analyze the influencing factors.Methods:A total of 176 patients with a convenient sampling method selection on November 20, 2021 in Ningxia medical institutions that have created cancer pain in the treatment of cancer pain in Ningxia as the research object of cancer pain in medical institutions, and use the "Cancer Pain Pain Incarring Credit Council Cancer Question Question" that is designed and examined by self-examination.Results:The pain knowledge dimension score of cancer pain patients in Ningxia was 5.00 (4.00, 6.00), the belief dimension score was 3.00 (-3.00, 10.00), and the behavior dimension score was 26.00 (24.00, 28.00). The results of multiple linear regression showed that financial burden and burst pain were the factors affecting the pain belief and perception of cancer pain patients ( t=-2.96, -3.00, both P<0.05). The type of medical insurance and the duration of pain were the factors affecting the behavior of pain ( t=-2.49, 3.21, both P<0.05). Conclusions:Cancer pain patients have a low level of pain knowledge, an upper-middle level of negative pain beliefs and perceptions, and a moderate level of pain behavior. It is recommended that nursing personnel use the discharge plan service as the main line to build a full-process management model of cancer pain. During the hospitalization period, pay attention to the outbreak of pain and psychological conditions, and gradually improve the quality of health education through nursing quality evaluation. After discharge, pay attention to the practicality and long-term development of continuity health education, and promote the full management of patients with cancer pain.

The accelerating growth of the public microbial genomic data imposes substantial bur-den on the research community that uses such resources. Building databases for non-redundant ref-erence sequences from massive microbial genomic data based on clustering analysis is essential. However, existing clustering algorithms perform poorly on long genomic sequences. In this article, we present Gclust, a parallel program for clustering complete or draft genomic sequences, where clustering is accelerated with a novel parallelization strategy and a fast sequence comparison algo-rithm using sparse suffix arrays (SSAs). Moreover, genome identity measures between two sequences are calculated based on their maximal exact matches (MEMs). In this paper, we demon-strate the high speed and clustering quality of Gclust by examining four genome sequence datasets. Gclust is freely available for non-commercial use at https://github.com/niu-lab/gclust. We also introduce a web server for clustering user-uploaded genomes at http://niulab.scgrid.cn/gclust.
9.Clinical Implication of the Presence of Notch Wave in ECG for Patients With Left Circumflex Artery-related Acute Myocardial Infarction

Objectives: To evaluate the clinical implication of notch (N) Wave in ECG for patients with left circumflex artery-related acute myocardial infarction. Methods: A total of 416 patients with left circumflex artery-related acute myocardial infarction hospitalized in our hospital from January 2013 to December 2016 were included in this study. According to the electrocardiogram, 156 people were divided into ST segment elevation myocardial infarction group, 108 patients in N wave non-ST segment elevation myocardial infarction group, and 152 patients in non-N wave NSTEMI group. Troponin I and creatine kinase isoenzyme, hospitalization to operation time, vascular lesion site and degree, the intraoperative and postoperative complications were compared among the 3 groups. Results: Troponin I and creatine kinase isoenzyme levels were significantly lower in non-N wave NSTEMI group than in STEMI group and N wave NSTEMI group (P<0.05). The occurrence rate of no-reflow phenomenon was significantly higher in N wave NSTEMI group than in STEMI group and non N wave NSTEMI group (P<0.05). There was no significant difference in incidence of cardiac shock, ventricular fibrillation, ventricular aneurysm and death rate among the 3 groups(P>0.05). Incidence rate of lesion located in the proximal and middle section of left circumflex artery as well as the mean vascular stenosis degree were significantly lower, while incidence rate of lesion located in the distal section of left circumflex artery and obtuse marginal branches was significantly higher in non N wave NSTEMI group than in STEMI group and N wave NSTEMI group(P<0.05). Conclusions: Presence of Notch wave in ECG is associated with higher incidence of lesion located in the proximal and middle section of left circumflex artery, larger infarct size and higher incidence of no-reflow in patients with left circumflex artery-related acute myocardial infarction.
10.An analysis on the risk factors of discontinued therapy among patients with multi -drug resistant pulmonary tuberculosis

Objective To analyze the basic characteristics of the therapy discontinued patients with multi -drug resistant pulmonary tuberculosis (MDR -TB)and to find the causes of therapy discontinuing.Methods Using case -control study method,a questionnaire survey was conducted to analyze the causes of MDR -TB in the treatment of interruption,and to compare the characteristics with the treatment completed patients,and to analyze the risk factors of discontinued therapy among MDR -TB patients.Results The study included 45 cases of therapy discontinued MDR -TB patients,and 45 cases of MDR -TB patients who had completed the treatment as the control.Multivariate logistic regression analysis results showed that patients with over 65 years old(OR =8.69,95% CI =1.12,67.57),leak medication or drug withdrawal experience (OR =53.18,95% CI =5.90,479.27),were the risk factors of discontinued therapy(P <0.05).Patients with hospitalization for over 15 days was the protective factors for discontinued therpy(OR =0.09,95% CI =0.01,0.58), (P <0.05).Conclusion advanced age≥65 years old,low income,leak medication or drug withdrawal experience short hospitalization periods were related to the risk of MDR -TB patients therapy discontinuing.were the key targets for patient management in the future.
