2.Progress of temsirolimus in the treatment of mantle cell lymphoma

Mantal cell lymphoma (MCL) is a rare and special type of non-Hodgkin lymphoma.MCL is classified as an aggressive variety of lymphoma and yet it sometimes behaves as an indolent lymphoma.Although the first-line therapy of MCL is highly effective,but it is incurable through conventional chemotherapy.Patients with MCL have a poor prognosis.This review focus on a new therapeutic approach,mammalian target of rapamycin (mTOR) inhibitor.Temsirolimus,a derivation of rapamycin,in MCL cell lines inhibited mTOR.Temsirolimus could inhibits tumor cell proliferation through cell cycle arrest and able to down-regulation of cyclin D1 and Ki-67 in lymphomacell.Clinical trials showed that temsirolimus induced patients with MCL response rate and a prolong progression-free survival.In addition,preclinical studies demonstrated the use of a combination of temsirolimus with monoclonal antibodies or conventional chemotherapy to improve the efficacy of single agent use.Altogether,temsirolimus shows promising results in the therapy of patients with heavily MCL,and especially responses in chemotherapy refractory patients,providing a novel approaches to treatment of MCL.
3.Neuroprotective mechanisms of curcumin in cerebral ischemia

Curcumin,a polyphenol compound,is a bioactive constituent extracted from the roots of the perennial herb Curcuma longa L.It has antioxidant,anti-inflammatory,anti-infective,heart protective and anti-tumor properties.Recent research has shown that curcumin can exert neuroprotective role in cerebral ischemia via the mechanisms of antioxidative stress,antiapoptosis,anti-cerebral edema,and anti-inflammation.This article reviews the neuroprotective mechanisms of curcumin in cerebral ischemia.
5.Protective effects of self-designed Shanghai multi-organ preservation solution on kidney cortex mitochondria function during cold storage

Objective:To study the protective effect of self-designed Shanghai multi-organ preservation solution(SMO) on mitochondria function in canine kidney cortex during cold storage.Methods: Canine kidneys were perfused and stored with cold(0-4℃) SMO,HC-A and UW solution (the later 2 as control) separately.Ultrastructure alteration of mitochondria was observed by electron microscopy.Mitochondria were isolated from cortex homogenization by differential centrifugation and their respiratory function was measured polarographically with Clark oxygen electrode.The four-state curve of mitochondria respiration was drawn and the state Ⅲ and Ⅳ oxygen consumption rate,the respiration control proportion(RCR) and phosphatasel/oxygen ratio(P/O) were calculated.Results: We found that the mitochondria swelling and membrane disruption in HC-A group were more severe than those in SMO and UW group.The rate of oxygen consumption in state Ⅲ,RCR and P/O were obviously decreased in all groups with the prolongation of cold storage time.The rate of oxygen consumption in state Ⅲ in HC-A group was lower compared to that in SMO group during 1-3 d cold storage(P
6.To enhance the therapeutic efficacy on amblyopia with classical therapies

There are many methods adopted in the treatment of amblyopia.Now many accessory therapies are enhanced instead of classical therapies,such as application of some therapeutic instruments for amblyopia,acupuncture and Chinese traditional medicine. We still suggest treating amblyopia with classical therapies:correction of refractive errors,occlusion and penalization.In this way,we may avoid attending to trifles and neglect the essentials,and miss the opportunity of treatment.(Ophthalmol CHN,2006,15:292-293)

8.RADIATION ACCIDENT IN 1963 AND 18 YEARS FOLLOW-UP OF THE 4 SURVIVED CASES

In 1963, we treated 6 cases of acute radiation sickness (200-8,000 rad) resulted from an accidental uneven 60Co ?-ray irradiation; two of them died of intestinal type within two weeks. Four cases survived and one of them recovered from septicemic shock. Loss of hair, systemic infection, high fever and bleeding occurred in 5 patients. The essential therapeutic measures were strict isolation, preventive treatment with antiinfection drugs, fresh blood transfusion and sometimes infusion of blood formed elements. Among the survivals 2 received homologous bone marrow transplantation.The general conditions of 4 patients followed up for a period of 18 years are apparently good with transparent lens, normal thyroid function and immunological reactions, except one who had low serum IgG level. Three of them showed subnormal adrenocortical activity and impairment of sex gland function. A girl patient (400 rad) gave birth to 2 children one of which has retarded intellectual development. Serial EEG changes occurred only in those received high cranial doses. In all the cases persistence of chromosome aberration in peripheral lymphocytes was observed. With high doses (above 10,000 rad) remote regional effects led to amputation of one leg in one patient and pathological fracture of the femur in another.
9.THE TOXIC EFFECTS OF STREPTOMYCIN ON THE ANIMAL NERVOUS TISSUE: A HISTOCHEMICAL STUDY

Healthy adult male animals (48 guinea pigs and 8 rats), showing allert responsesto hearing and equilibrium tests, were selected for chronic and acute experiments respec-tively. Twenty-five guinea pigs received intramuscular injections of streptomycin sulfatedaily in different doses (100 mg/kg in water, 200 mg/kg in water, or 400 mg/kg in 5%calcium gluconate). The rest 23 guinea pigs received same amount of water or calciumgluconate for control. Four rats received a single injection of streptomycin sulfate (600mg/kg in water) and the other 4 controls received water instead. Guinea pigs, admitting 100 mg/kg of streptomycin sulfate daily for one month, showedno conspicuous changes in both functional tests and histochemical examinations. Animals,receiving a daily dose of 200mg/kg or 400 mg/kg with Ca, for two months, lost equili-brium response and became dull in hearing. But these intoxicative effects were observedas early as 58 days and as late as 158 days in individual cases. Histochemical examinations of the pons and medulla oblongalta showed that, in theintoxicated guinea pigs from the chronic experiments, the chromophobe (Ⅲ-type) cellsincreased in number in the terminal nucleus of the cochlear nerve, the lateral terminalnucleus of the vestibular nerve and the primary nucleus of the trigeminal nerse. In thesenuclei, the histochemical reactions of SH-groups, protein-bound histidine, tyrosine and try-ptophan, PAS and succinic dehydrogenase showed a general fall in intensity, while thoseof allcaline and acid phosphatases had no obvious changes. The acetylcholinesterase of the end plates in the intercostal muscle of the rats into-xicated with a single heavy dose of streptomycin sulfate decreased in activity, while thosefrom the chronic experiments did not show any change. Calcium gluconate exhibited the detoxication effect in acute experiments and couldincrease the tolerance dosage in the animals. The author concluded from the experimental facts that streptomycin has a generaltoxic effect on the entire nervous system and discussed the results with the current lite-rature.
10.NOTES-transvagina endoscopic cholecystectomy:report of 31 cases

Objective:To investigate the feasibility,safety and superiority of transvagina endo- scopic cholecystectomy used in clinical application.Methods:31 patients with gall-bladder diseases were treated in transvagina endoscopic cholecystectomy.Through a 5mm cut below the umbilicus, inserted laparoscopic guide gastroscope and NOTES manipulator enter abdominal cavity through posterior fornix 15mm cut.After the gastroscope can capture stable vision,withdraw the laparoscope and insert normal laparoscopic manipulator.With the gastroscope monitoring,accomplish cholecystectomy using umbilical and pudendum manipulator.Results:All of patients were accepted this new type operation.The operation time was 60~150 minutes,and the average time was 85 minutes.No postoperative draining,bleeding and biliary fistula occurred in those patients. There was no obvious scar in belly,and all of patients felt no pains after operation.The average inpatient time was shorten,the medical cost decreased.Conclusion:Transvagina endoscopic cholecystectomy is feasible and safety.Comparing to traditional laparoscopic operation,it has obviously superiority.Transvagina endoscopic cholecystectomy should be widely applied in good condition hospitals.
