1. Determination of the Dissolution Curves of Albendazole Tablets

OBJECTIVE: To establish a determination method of the dissolution curves of albendazole tablets with differentiation ability and investigate the similarity of dissolution curves in vitro between reference preparation and generic preparations.METHODS: Paddle method was adopted with rotation speed of 50 r•min-1 and the dissolution medium volume was 900 mL. The dissolution media were pH 1.2 hydrochloric acid solution and water. The similarity factor (f2) method was used to evaluate the comparability of dissolution curves.RESULTS: The three established dissolution curves had good distinguishing ability. As shown by the similarity factor (f2), the generic preparation from manufacture A was similar to the reference preparation, while those from the manufacture B and C were not similar.CONCLUSION: The study provides the experimental basis for the consistency evaluation of the dissolution curves of abendazole tablets.
2. Contrast-enhanced CT histogram analysis in differentiating benign and malignant thyroid nodules

Objective: To explore the value of histogram features based on plain and enhanced CT for differential diagnosis of benign and malignant thyroid nodules. Methods: A total of 132 patients with 140 thyroid nodules confirmed by postoperative pathology were retrospectively analyzed. The level with the largest axial focus was selected, and then Mazda software was used to sketch ROI along the edge of focus and perform histogram analysis to obtain 9 parameters. The parameters were compared between benign and malignant nodules, and then statistically significant gray-scale histogram parameters were used to analyze their value of differentiating benign and malignant thyroid nodules. Results: On plain CT, the mean value, the 10th, 50th and 90th percentiles of malignant nodules were higher than those of benign ones (all P<0.05), while on enhanced CT, the mean value, variance, and the first, 10th, 50th and 90th percentiles of malignant nodules were all higher than those of benign ones (all P<0.05). The average value of malignant nodules was higher than that of benign ones by the first, 10th, 50th and 90th percentage points, and the variance of benign group was higher than that of malignant group (all P>0.05). Among the parameters of CT plain and enhanced histogram, the 10th percentile AUC was the highest of 0.68, and the sensitivity of identifying benign and malignant thyroid nodules was 74.32%,and specificity was 62.12%. Conclusion: CT histogram analysis can be used as an important auxiliary means to identify benign and malignant thyroid nodules.
3.Dickkopf-1 inhibits the secretion of MUC5AC induced by Mycoplasma pneumoniae P1-C in mouse lung epithelial cells.

Mycoplasma pneumoniae is the most common pathogen of respiratory tract infection in children and adults. Clinical observation shows that M. pneumoniae infection can cause massive mucus secretion in the respiratory tract, which makes the breathing of patients difficult. Studies have shown that M. pneumoniae infection can cause massive secretion of mucin 5AC (MUC5AC). Adhesin P1 plays an important role in the pathogenesis of M. pneumoniae infection by mediating the adhesion of pathogens to host cells, and the C-terminal residues of P1 (P1-C) are immunogenic. This study investigated the molecular mechanism of Wnt/β-catenin signaling pathway inhibitor Dickkopf-1 (DKK1) in the secretion of MUC5AC in mouse airway epithelial cells (MAECs) induced by P1-C. Scanning electron microscope and hematoxylin-eosin staining were used to observe the effect of P1-C on mucus secretion of MAECs. Protein chip was used to detect the secretion of cytokines and analyse the enrichment of related signaling pathways induced by P1-C in MAECs. Periodic acid schiff stain (PAS) staining, Tunel staining and Masson staining were used to detect the damage of the lungs of mouse exposed to P1-C. Immunohistochemistry was used to detect the secretion of MUC5AC expression, and Western blotting was used to reveal the molecular mechanism of DKK1-regulated secretion of MUC5AC induced by P1-C protein in MACES. The results showed that P1-C induced the massive secretion of mucus and inflammatory factors in MAECs. During P1-C infection, DKK1 down-regulated janus kinase 2 (JAK2), phosphorylation signaling and transcription activator 1 (p-STAT1) and phosphorylation signaling and activator of transcription 3 (p-STAT3) expression. Overexpression of DKK1 significantly up-regulated the expression of MUC5AC repressor transcription factor fork-head box protein A2 (FOXA2). At the same time, the expression of MUC5AC induced by P1-C was inhibited significantly. It is speculated that DKK1 can effectively reduce the secretion of MUC5AC in MAECs induced by P1-C by inhibiting the JAK/STAT1-STAT3 signaling pathway and up-regulating the expression of FOXA2.

4. Association of mental health,job burnout and mental fatigue in mental workers

OBJECTIVE: To explore the association of mental health,job burnout and mental fatigue in mental workers.METHODS: A total of 276 mental workers in 11 research institutes in Ningxia were selected as research subjects by convenient sampling method. The questionnaire survey was carried out using the scales of Self-Reporting Inventory( SCL-90),Maslach Burnout Inventory-General Survey and Mental Fatigue subscale of Fatigue Scale-14 to investigate the workers' mental health,job burnout and mental fatigue. RESULTS: The scores of SCL-90 and job burnout in mental workers were( 142. 4 ± 42. 2) and( 41. 8 ± 10. 5). The medium( P25,P75) mental fatigue total score was [3. 0( 2. 0,4. 0) ].The correlation analysis showed that the total score of SCL-90 was positively correlated with that of mental fatigue and job burnout( P < 0. 01). The job burnout was the intermediate variable between mental fatigue and mental health( 43. 4% of the total effect). The dimension of low sense of accomplishment was the regulating variable. CONCLUSION: Job burnout plays an intermediate role between mental fatigue and mental health in mental workers. The low sense of accomplishment plays a regulatory role.

6.Clinical efficacy of oblique lumbar interbody fusion combined with unilateral pedicle screw fixation via Wiltse approach in the treatment of lumbar spinal stenosis.

OBJECTIVE:
To evaluate the efficacy of oblique lumbar interbody fusion combined with unilateral pedicle screw fixation via Wiltse approach in the treatment of lumbar spinal stenosis.
METHODS:
From July 2017 to January 2019, 90 patients with lumbar spinal stenosis, including 38 males and 52 females, aged from 43 to 75 years old with an average of(59.9±8.8) years old, and were treated with oblique lumbar interbody fusion(OLIF) combined with Wiltse unilateral pedicle screw fixation. Surgical decompression and fixation was performed in 50 cases of single segment, 32 cases of double segments and 8 cases of three segments. The distribution of responsible segments included 8 cases of L2-L3, 12 cases of L3-L4 and 30 cases of L4-L5 on single segment, 10 cases of L2-L4 and 22 cases of L3-L5 on double segments, and 8 cases of L2-L5 on three segments. The operation time, blood loss and occurrence of complications were recorded, Visual analogue scale(VAS), Oswestry Disability Index(ODI) and SF-36 scale were used to evaluate clinical efficacy. Lumbar X-ray and MRI were taken at three days after operation, interverterbral space height, intervertebral foraminal height, interverterbral foraminal area, and spinal canal area were measured, and interbody fusion was evaluated according to CT at half a year after operation.
RESULTS:
All patients were followed up from 12 to 33 months, with an average of (20.2±6.6) months. Mean operation time was (103.3±35.9) min, and mean intraoperative blood loss was (70.4±17.8) ml. VAS of low back pain leg pain, and ODI decreased from 6.2±1.1, 6.1±0.9 and (59.9±4.2)% to 2.7±0.5, 2.5±0.5 and (31.3±8.8)%. SF-36 scale significantly increased from (37.2±3.1) to (54.9±6.1) at the six months postoperation(P<0.05). The intervertebral space height, intervertebral foraminal height, intervertebral foraminal area, and spinal canal area were significantly improved at 3 days after operation(P<0.05). Six months after operation, CT scan showed well fusion in 87 cases, but 3 cases with poor fusion, including 1 case of single segment, 2 cases of multi-segments. The total fusion rate was 96.7% (87/90), the single segment fusion rate was 98.0% (49/50), and the multi-segments fusion rate was 95.0%(38/40). The overall incidence of complications was 17.8%(16/90), including transient iliopsoas muscle weakness in 5 cases (5.6%), endplate fracture in 2 cases (2.2%), peritoneal injury in 1 case (1.1%), postoperative hematoma in 1 case (1.1%), adjacent segment disease in 1 case(1.1%), and fusion cage subsidence in 6 cases (6.7%). Three patients was followed up for recurrent nerve root pain and the symptoms were relieved after revision operation. All complications were relieved or disappeared in varying degrees during the follow-up period, and there were no complications such as cage displacement and screw fracture.
CONCLUSION
OLIF combined with unilateral pedicle screw fixation via Wiltse approach is effective in treating lumbar spinal stenosis with minimally invasive advantages such as less trauma and less complications. Under the premise of strictly grasping the indications, this method could also achieve satisfactory clinical results in multi-segments oprations.

7.Current status of hospital infection management in psychiatric hospitals in the Ningxia Hui Autonomous Region

ObjectiveTo investigate current status of hospital infection management in psychiatric hospitals in the Ningxia Hui Autonomous Region, so as to provide references for improving the level of hospital infection management of psychiatric hospitals. MethodsIn December 2020, on-site supervision was conducted on hospital infection management in all 9 psychiatric hospitals in the Ningxia Hui Autonomous Region, meantime, the self-compiled questionnaire on hospital infection management status was used for investigation. ResultsAmong the selected hospitals, nine (100.00%) psychiatric hospitals had the main hospital leaders in charge of hospital infection management, five (55.56%) hospitals had established a hospital infection management committee, six (66.67%) hospitals had established an independent hospital infection management department, and one (11.11%) hospital had developed all 13 systems mentioned in the questionnaire related to hospital infection management and job responsibilities. In terms of hospital infection management staff, there were 23 staff members in the nine psychiatric hospitals, including 3 in the specialty (13.04%) and 20 in the part-time setting (86.96%). The score of the implementation of the basic system of hospital infection management in nine hospitals was (3.28±2.22). ConclusionThe system specification related to hospital infection management in the Ningxia Hui Autonomous Region psychiatric hospitals needs to be improved and further strengthened, the professionalism of hospital infection management personnel needs to be improved.
8. Effect and Mechanisms of Lycii Fructus on Hepatoma HepG2 Cell Line-induced Cachexia in Mice

Objective: To investigate the inhibitory effect and mechanism of the extracts from fresh Lycii Fructus (LBL) on hepatoma HepG2 cell-induced cachexia in mouse.Method: The human hepatoma cell line HepG2 was injected into BALB/C mice to establish the cachexia model.Then the LBL was fed to the models respectively in low dose (5 mg·kg-1) or high dose (25 mg·kg-1).After 28 days of continuous feeding,the mice's body weight was detected.The expression levels of creatine kinase (CK),interleukin-1β (IL-1β),interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) were evaluated by enzyme linked immunosorbent assay (ELISA).The muscle degradation and the expression of p38 mitogen-activated protein kinase (p38 MAPK) were detected by immunohistochemistry staining.The expression levels of phospho-p38 mitogen-activated protein kinase (p-p38 MAPK) and nuclear factor-κB (NF-κB) were determined by Western blot.Result: Compared with cachexia group,the loss of body weight and muscle decomposition were significantly inhibited both in the low dose LBL group and high dose LBL group (P<0.05,P<0.01).The level of plasma CK decreased significantly both in the low-dose LBL group and the high-dose LBL group (P<0.01).ELISA tests revealed lower expression levels of IL-1β,IL-6 and TNF-α in both the low-dose LBL group and the high-dose LBL group (P<0.05,P<0.01).Immunohistochemistry staining showed that the expression of p38 MAPK was inhibited both in the low-dose LBL group and the high-dose LBL group (P<0.05).Western blot indicated that the expressions of p-p38 MAPK and NF-κB were inhibited both in the low-dose LBL group and the high-dose LBL group (P<0.01).We found that the high-dose LBL group shows a higher inhibitory capability than the low-dose LBL group.Conclusion: LBL could inhibit the cachexia induced by hepatoma HepG2 cell line in mouse,suggesting LBL could reduce major cytokine and plasma inflammatory factors through p-p38 MAPK pathway.
9.Circ_0075960 targets the miR-202-5p/CTNND1 axis to promote the growth and migration of endometrial carcinoma cells via regulating Wnt/β-catenin signaling activity

Background:
Endometrial carcinoma (EC) is one of the most common malignant tumors of the female reproductive tract, involving multiple molecular alterations. Circular RNA (circRNA) dysregulation is frequently observed in EC tissues, suggesting the involvement of circRNA in EC development. We aimed to investigate the role of circ_0075960 in EC.
Methods:
Real-time quantitative polymerase chain reaction (RT-qPCR) and western blot assays were applied for expression analysis. CCK-8, EdU, colony formation, flow cytometry and wound healing assays were employed for functional analysis. The predicted binding relationship between miR-202-5p and circ_0075960 or CTNND1 was validated by dual-luciferase reporter experiment. In vivo animal models were constructed in nude mice to verify the role of circ_0075960 in tumor growth.
Results:
Circ_0075960 and CTNND1 were upregulated, while miR-202-5p was downregulated in EC. Knockdown of circ_0075960 induced EC cell apoptosis, suppressed cell proliferation and migration, and repressed tumor growth in animal models. MiR-202-5p was targeted by circ_0075960 and it directly bound to CTNND1 3’UTR. The inhibition of circ_0075960 knockdown or miR-202-5p enrichment on EC cell proliferation and migration was reversed by miR-202-5p depletion or CTNND1 overexpression, respectively. Circ_0075960 targeted miR-202-5p to positively regulate CTNND1 expression. Moreover, circ_0075960 knockdown weakened the activity of Wnt/β-catenin signaling via targeting the miR-202-5p/CTNND1 axis.
Conclusion
Circ_0075960 targets the miR-202-5p/CTNND1 axis to modulate Wnt/β-catenin signaling activity, thus contributing to the malignant development of EC.
10.Changes in corneal biomechanics after small incision lenticule extraction and femtosecond laser-assisted laser in situ keratomileusis evaluated by corneal visualization Scheimpflug technology

AIM:To evaluate the changes in corneal biomechanics of patients with moderate refractive error after receiving small-incision lenticule extraction(SMILE)and femtosecond laser-assisted laser in situ keratomileusis(FS-LASIK)using the corneal visualization Scheimpflug technology(Corvis ST).METHODS:Prospective cohort study. A total of 65 moderate myopia patients(65 eyes)who were scheduled to undergo refractive surgery at the Ophthalmic Refractive Surgery Center of the Ningxia Eye Hospital from November 2020 to November 2021 were included in the study, and there were 30 eyes in the SMILE group and 35 eyes in the FS-LASIK group. The changes in corneal biomechanical parameters, including integrated radius(IR), inverse concave radius(ICR), deformation amplitude ratio 2mm(DAR2), stiffness parameter at first applanation(SP-A1), ambrosio relational thickness(ARTh)and the central curvature radius at highest concavity(HC-Radius)were observed by Corvis ST between both groups preoperatively and 1 and 3mo postoperatively.RESULTS: There were no statistical significance in biomechanical parameters between two groups of patients 1 and 3mo postoperatively(P>0.05). IR, ICR and DAR2 of each groups of patients 1 and 3mo postoperatively were significantly increased than those preoperatively, and SP-A1, ARTh and HC-Radius were significantly decreased than those preoperatively(all P<0.05). The biomechanical parameters at 1mo and 3mo postoperatively showed no statistical significance(P>0.05). In addition, a positive correlation was found between central corneal thickness(CCT)and ARTh and SP-A1 of the two groups of patients at 3mo postoperatively(FS-LASIK group: r=0.727, 0.819, SMLIE group: r=0.683, 0.434, all P<0.05), while a negative correlation was found between CCT and IR and ICR at 3mo postoperatively.(FS-LASIK group: r=-0.697, -0.622, SMLIE group: r=-0.447, -0.491, all P<0.05).CONCLUSION:For patients with moderate myopia, both SMILE and FS-LASIK can reduce corneal biomechanical stability. Both surgeries showed no significant differences in the effect on biomechanical, and the biomechanical has been stabilized at 1mo postoperatively. A correlation was found between postoperative CCT and ARTh, SP-A1, IR and ICR.
