1.Helicobacter pylori-induced VEGF expression in human gastric cancer MKN45 cells mediated by COX-2

Background and purpose: Vascular endothelial growth factor (VEGF) is an important proangiogenic factor, and Helicobacter pylori (H. pylori) infection-induced gastric over-expression of VEGF is an important factor of gastric cancer growth and metastasis, but its expression mechanism is not clear. Cyclooxygenase-2 (COX-2) is a rapid response protein, which is closely related to the occurrence and development of gastric cancer. Our study was to investigate the effect of COX-2 on H. pylori-induced VEGF expression in human gastric cancer cells, and to reveal part of the mechanism of gastric cancer growth and metastasis promoted by H. pylori infection. Methods:The expression ofVEGF mRNA in human gastric epithelial cells (MKN45) infected by standard H. pylori NCTC 11637 and the expression of COX-2 protein were evaluated by real-time fluorogenic quantitative polymerase chain reaction (RFQ-PCR) and assayed by Western blot. After inhibiting COX-2 expression with COX-2 specific inhibitor NS398 (50 μmol/L), VEGF mRNA expression induced by H. pylori in human gastric cancer MKN45 cells was evaluated by RFQ-PCR. Results: H. pylori significantly stimulated the expression ofVEGF mRNA in MKN45 cell line. Compared with control MKN45 cells; VEGF mRNA had 2.33 fold up-regulation after 6 h (P<0.05); and had 5.69 and 5.04 fold upregulation respectively after 12 and 24 h (P<0.01).When MKN45 cells were infected with H. pylori for 24 h, COX-2 protein expression also increased significantly (P<0.01), and after inhibiting the COX-2 expression with COX-2 specific inhibitor NS398, H. pylori-induced VEGF mRNA expression was significantly reduced. Conclusion: H. pylori could induce the expression of COX-2 and VEGF in human gastric cancer cells, and could enhance VEGF expression by COX-2 pathway, which might be one of the important mechanisms of gastric cancer growth and metastasis promoted by H. pylori infection.
2.The effect of intra-articular ozone injection on the cytokines of collagen-induced arthritis

Objective To observe the effects of intra-articular ozone injection on the secretion of tumor necrosis factor (TNF)-α,Interleukin (IL)-6,IL-17A,and vascular endothelial growth factor (VEGF) in the serum of rats with collagen-induced arthritis (CIA) and explore the therapeutic mechanism of ozone in RA treatment.Methods Thrity-two Wistar rats were randomized into 4 groups,including the ozone groups that receivedintra-articularinjection of 40 μg/ml ozone (O3 group),a blank control group (normal group),a methotrexate (MTX) group (MTX group) anda collagen-induced arthritismodel (CIA group).All the rats,except for those in the blank control group,were subjected to hypodermic injection of bovine collagen Ⅱ and complete Freund's adjuvant to induce CIA.Ozone treatment was administered once weekly for 3 weeks starting at 14 days after the model were established.MTX group were treated with methotrexate 0.9 mg/kg,once a week,a total of three weeks.The swelling degree of the foot were observed,and the serum contents of TNF-oα,IL-6,IL-17A and VEGF were detected.One-way analysis of variance or Kruskal-Wallis test was used to evaluate the experimental data.Results At the end of treatment,the degree of foot swelling was reduced significantly in rats with O3 group compared with that in the CIA group [(4.21±0.14) ml vs (9.12±0.17) ml,t=8.43,P=0.023].The serum concentration of TNF-α,IL-6 and VEGF showed significant difference between the CIA group and the O3 group[91.55(86.55,98.53) pg/ml vs 14.45 (12.55,16.15) pg/ml,x2=6.216,P=0.002;145.08(37.44± 362.82) pg/ml vs 5.84(5.47,15.93) pg/ml,x2=13.136,P=0.004;51.56(46.09,74.10) pg/ml vs.36.22(32.18,41.69) pg/ml,x2=3.732,P=0.002].There was no statistically significant difference between the O3 group and MTX group [14.45(12.55,16.15) pg/ml vs [12.45(11.80,15.60) pg/ml,x2=0.243,P>0.05;5.84(5.47,15.93) pg/ml vs 7.86(5.25,15.23) pg/ml,x2=0.058,P>0.05;36.22(32.18±41.69) pg/ml vs 40.17(35.47,50.73) pg/ml,x2=0.516,P>0.05].The serum concentration of IL-17A showed no significant difference between the normal group,the CIAgroup,the MTX group and the O3 group (F=1.827,P=0.165).Conclusion Intra-articular injecfion of 40 μg/ml ozone can attenuate synovitis in rats with CIA,the mechanism may relate to the inhibition of TNF-oα,IL-6 and VEGF in the serum.
3.Intra-articular injection of ozone on serum receptor activator for nuclear factor-κB ligand and osteo-protegerin level in collagen-induced arthritis rats

Objective To evaluate the clinical efficacy of intra-articular injection of ozone in collagen-induced arthritis (CIA) rats and to assess its effects on serum receptor activator of nuclear factor κB ligand (RANKL) and osteoprotegerin (OPG) levels. Methods Forty weight age malched Wistar rats were randomly divided into normal control group (normal group), the CIA model group (CIA group), ozone (O 3 group), and methotrexate (MTX group). In addition to the normal control group, Freund's complete adjuvant and bovine type Ⅱ collagen were injected to establish the rat model of CIA. After the model was sucessfully developed, double ankle injection concentration ozone group of 40 μg/ml of O3 each 1 ml, 1 times a week, a total of injection for 3 weeks for the experimenal group. MTX group of 0.9 mg/kg was injected 1 times a week for 3 weeks for the MTX group. Degree of foot swelling was measured, and radiographic assessment of arthritis index (AI) score was performed. One week after treatment, angular vein blood was collected for the rats after the intervention, flow multi-factor detection technology was used to test each rat. T test or Wilcoxon rank test was used to compare the difference between groups. Results ① After 3 week administration with O3, dcgree of foot swelling, and AI of the O3 group was reduced significanly than the CIA group during the same period (foot swelling degree: O3 group: (4.21±0.14) ml, CIA group (9.12±0.17) ml, T=64.08, P=0.00; AI O3 group: [(2.97± 0.18) ml, CIA group: 5.76 ±0.13, T=37.24, P=0.00], and X-ray showed joint damage was alleviated. ② The serum level of RANKL in the CIA group was significantly higher than of the normal group [CIA model group 1890.70(797.03, 10571.94)], normal group [74.46(29.21, 95.37), T=43, P=0.005] during the same period; The serum level of RANKL in the O3 group was significantly lower than the CIA group [O3 group 28.09 (14.11, 207.30), CIA group 1890.70 (797.03, 10571.94), T=39, P<0.05]; RANKL level of the Ozone group when compared with MTX group, was not statistically significantly difference (T=52, P>0.05).③Serum RANKL/OPG of the CIA group was significantly higher than that of the normal group during this period, the difference was statistically significant [CIA group 250.68(42.33, 2959.78), normal group 4.32(3.16,5.30), T=36, P<0.01);The serum level of RANKL/OPG in the O3 group was significantly lower than the CIA group [O3 treatment group 5.10 (3.38, 6.64), CIA group 250.68 (42,33, 2959.78), T=36, P<0.01]; There was no statistically significant difference of the serum RANKL/OPG between the O3 group and the MTX group (T=62.5, P>0.05). Conclusion Intra-articular injection of concentration of 40 μg/ml of O3 can reduce RA rat joint swelling degree, which may relate to the mechanism that O3 can lower levels of serum RANKL and RANKL/OPG ratio, reduce osteoclast formation and activation.

5.One year evaluation of endodontic microsurgery in 54 cases with persistent apical periodontitis.

OBJECTIVETo evaluate the outcome of endodontic microsurgery in 54 cases with persistent apical periodontitis.
METHODSAll surgical procedures were performed under a dental operating microscope after root canal treatment. Surgical procedures included periapical curettage, root-end resection, retropreparation with ultrasonic microtips, and root-end filling with mineral trioxide aggregate (MTA). Healing was monitored clinically and radiographically.
RESULTSOne year later, 31 (57.41%) cases were cured, 19 (35.19%) cases were improved, 4 (7.41%) cases were failure. There were no statistical difference between sex, age, tooth location, size of the lesion and pathological features (P>0.05).
CONCLUSIONThe outcome of the present study indicates that endodontic microsurgery may result in a predictable treatment outcome in teeth with persistent periradicular lesions.

6.The expression and role of LncRNA MEG3 in human glioma cells

Objective The aim of this study was to illuminate effects of LncRNA MEG3 on the viability and apoptosis of hu-man glioma cells as well as the mechanisms involved. Methods The expressions of MEG3 and microRNA-21(miR-21) in normal human astrocytes cell line(NHAs)and human glioma cell line(U87)was detected by Real-Time quantitative polymerase chain reac-tion(RT-qPCR). U87 cells was transfected with MEG3 plasmid alone or transfected with both MEG3 plasmid and miR-21 mimic, cell viability was detected by CCK-8 assay,cell apoptosis was detected by TUNNEL,and the expression of Bcl-2/Bax and Cleaved caspase-3 in cells was detected by Western blot. Results Compared with NHAs,the expression of MEG3 in U87 was significantly decreased and the expression of miR-21 was significantly increased. Overexpression of MEG3 significantly decreased the expression of miR-21 in U87 cells,inhibited the viability of U87 cells,decreased the level of Bcl-2/Bax,increased the content of Cleaved-caspase-3 to promote apoptosis;Co-overexpression of MEG3 and miR-21 significantly increased the viability of U87 cells,elevated Bcl-2/Bax levels,and decreased the expression of Cleaved caspase-3 to inhibit apoptosis. Conclusion Overexpression of MEG3 reduces the viability and promotes apoptosis of glioma cells by inhibiting miR-21.
7.Nursing of patients who use ultrasound-guided thermal ablation to treat severe secondary hyperparathyroidism

Objective To summary the nursing methods of patients who use ultrasound-guided thermal ablation to treat severe secondary hyperparathyroidism.Methods Retrospective analysis was used to analyze the nursing process of 10 patients who use ultrasound-guided thermal ablation to treat severe secondary hyperparathyroidism. Ultrasound-guided thermal ablation was applied to patients with significant cardiopulmonary dysfunction and cannot undergo general anaesthesia operation, and patients suffering from recurrent forearm grafting after parathyroidectomy. So, in the nursing of these patients, we should pay special attention to the observation and management of specific complications in thermal ablation including dyspnea, apnea, laryngeal and recurrent laryngeal nerve injury and hemorrhage, besides the routine psychological care, diet nursing and medication nursing.Results Ten patients in the study who underwent ultrasound-guided thermal ablation, the intact serum parathyroid hormone ( intact parathyroid hormone, iPTH ) , calcium and phosphorus were all decreased, and symptoms like itching, general bone pain also were released. The frequency of precordial pain was reduced and we gain good therapeutic effect.Conclusions Rigorous nursing after ultrasound-guided thermal ablation can promote the recovery of patients with severe secondary hyperparathyroidism.
8.Application of TMS in quantitative assessment of full term neonatal brain maturation with HIE

Objective To investigate the applications of total maturation scores (TMS) in assessment of the hypoxic ischemic encephalopathy (HIE) group and matched group with MRI and to find which patterns of HIE can delay the brain maturation in neonate.Methods MRI were performed on 43 full term neonates (28 HIE newborns and 15 matched controls) who were further divided into three groups,including 37-39 weeks postmenstrualage (PMA) (13 cases),39-41 weeks PMA (16 cases),and 41-44 weeks PMA (14 cases).MRI features and TMS scores were evaluated simulataneously.Results TMS gradually increased with PMA at each stage.While TMS,cortical folding and germinal matrix were all higher in matched group (P<0.05),myelination and glial cell migration item were lower in HIE group (P>0.05).Conclusion TMS can reflect brain development in the full term neonates.HIE may delay the brain maturation due to injury of the cortical myelin and neurons.TMS can evaluate the neonatal brain development and injury easily,quantitatively and effectively.
9.The relationship and mechanism between accessory renal artery and essential hypertension by CTA

Objective To explore the relationship between accessory renal artery(ARA)and essential hypertension,and the possible mechanism using CT angiography(CTA).Methods The patients who underwent CTA examination on renal artery were reviewed retrospectively in our hospital.A variety of CTA reconstruction techniques were used to observe the type and number of ARA,as well as the diameter of ARA and the main renal artery.Results A total of 126 ARA(66 left ARA and 60 right ARA)were found in 253 patients. 10% of the patients had more than two ARAs.In 164 patients with essential hypertension,ARA detection rate was 40.2%(66/164).In the non-hypertension patients,the proportion of ARA was 31.5%(28/89).There was higher proportion of young(P=0.002)and male (P=0.022)patients in ARA hypertension group compared with ARA non-hypertension group.There were no significant differences on the prevalence of type(P=0.826)and number of ARA(P=0.501)between these two groups.In all of the patients with single ARA,no significant differences were detected on the ratios of diameter of ARA and main renal artery between the two groups(P=0.32). However,in ARA hypertension group,the diameter of main renal artery on the ARA side was significantly smaller than that on the opposite side(P=0.01).In non-hypertension ARA group,no statistical difference was found between the diameter of bilateral main renal arteries(P=0.06).Conclusion ARA is more prevalent in essential hypertensive patients,especially in young male.The decrease of the diameter of main renal artery in the ARA side may be a possible mechanism for essential hypertension.
10. The treatment of application of pedicled myocutaneous island flap of anterolateral thigh muscle for the suprapubic defect in adolescent with bladder exstrophy

Objective:
To explore the treatment effect of pedicled myocutaneous island flap of anterolateral thigh muscle on the suprapubic defect caused by bladder exstrophy in children.
Methods:
The clinical data of 3 adolescents with bladder exstrophy were analyzed. All 3 cases underwent one-stage Kelly procedure and received the treatment using pedicled myocutaneous island flap of anterolateral thigh muscle to repair the suprapubic defect. The bladder expansion was performed with ileal sarcoplasmic layer graft, and the Cohen ureteral reimplantation was performed as well, for 1 case 1, 1 year after the first operation. The others 2 patients didn′t receive the second operation.
Results:
All operations were successful. The femoral donor regions were directly closed. Postoperative follow-up period was 1-7 years. The myocutaneous island flaps were survived and grew well. All incisions of the femoral donor region were primary healing without scar hypertrophy. Urinary continence was achieved in 2 cases, and partially achieved in 1 case. Recurrent urinary tract infection, recurrent fever or abdominal pain were not observed in all patients.
Conclusions
The pedicled anterolateral thigh myocutaneous island flap is an alternative way to repair suprapubic defect caused by bladder exstrophy.
