Objective To observe efficacy of treatment comdyloma acuminatum with CO2 laser. Methods A local infiltration anesthesia was engaged in 29 cases of females with genital pointel condyloma and 3cases of males with anus pointed condyloma,and a compression anesthesia was conducted on the penis root of 31 cases of males with genital pointed condyloma,CO2 laser with 5 ～ 15 W output power was focused on skin lesion until it was singed then to carbon or gas. Results 24 cases of females recover,29cases of males recover,the cure rate was 84%. Conclusion CO2 laser is one basic cure for condyloma acuminatum sufferers.

Objective To investigate the relationship between the expression of NF-κB and Pglycoprotein(P-gp) in cervical cancer tissue before and after neoadjuvant chemotherapy(NACT). Methods The mRNA and the protein expression levels of NF-κB and P-gp were determined by Real time-PCR and immunohistochemical assay respectively in cancer specimens from 40 cevical cancer patients, which were collected before and after NACT. Results The mRNA expression levels of NF-κB were 50. 340 ± 0. 780 and 67. 595 ± 0. 914 before and after NACT respectively, which showed significant difference statistically ( t = -73. 384 ,P ＜ 0. 01 ). We also found significant difference in the comparison between MDR1 mRNA expression before and after NACT(45. 800 ±0. 636 vs 62. 381 ±0. 743 ,t = -66. 582,P ＜0. 01 ). The positive rate of NF-κB protein expression before and after NACT were 50% ( 20/40 ), 80% ( 32/40 ) respectively, which showed significant difference statistically( χ2 = 7. 912, P = 0. 005 ). The positive rate of P-gp protein expression after NACT was significantly higher than that before NACT [ 40% ( 16/40 ) vs 70% ( 28/40 ), χ2 = 7. 273, P = 0. 007 ]The mRNA and protein expressions of NF-κB were positively correlated with those of P-gp before and after NACT( r = 0. 408,r = 0. 327 respectively, Ps ＜ 0. 05 ). Conclusion NF-κB and P-gp may play an important role in multi-drug resistance of cervical carcinoma. Chemotherapy drugs in our NACT strategy may induce the activation of NF-κB and multi-drug resistance. Activation of NF-κB may correlate with multi-drug resistance mediated by P-gp.

Objective: To study the effect of HSVtk/GCV system on ascites tumors. Methods: Using the methods of modified XTT assay, animal experiment, transmission electron microscopy (TEM ) and flow cytometric ( FCM) assay. Results: The sensitivity of P388tk cell was about 37-fold than that of their patental P388 cell. Furthermore we found P388tk cells exhibited potent bystander killing. In vivo , the growth of tumors, which were produced by injecting P388tk cells or a mixture of 50% P388tk cells and 50% P388 cells into DBA/2 mice, was inhibited , furthermore mice survival periods were prolonged contrasted to control groups. Conclusion: HSVtk/GCV suicide gene system could effectively killed the HSVtk-gene-positive P388tk cells and nearby HSVtk-gene-negative cells by the bystander effect in vitro and in vivo.

Objective To investigate the clinical significance of acute insulin resistance during surgical sepsis. Methods Forty surgical patients with sepsis in experimental group and twenty patients with similar APACHEⅡ but without sepsis in experimental control group were enrolled in our study. In the experimental group, the patients were categorized to sepsis group and septic shock and MODS(SM)group according to clinical manifestations, and were also grouped according to different causes of sepsis, i.e.severe acute pancreatitis(SAP)group, intestinal fistula group and other diseases group. Determinations of fasting blood glucose, fasting plasma insulin, clinical nutritional parameters, plasma TNF-α, APACHEⅡ, sepsis-related organ failure assessment(SOFA)and severe scores were simultaneously performed at designated time points. Insulin resistance index was calculated using homeostasis model assessment(HOMA)to assess insulin sensitivity of surgical patients with sepsis. Results (1)A significant elevation of lgHOMA-IR levels was found in 84% of tests in sepsis. Peak lgHOMA-IR values increased in all patients.(2)LgHOMA-IR was significantly higher in patients with sepsis than those without sepsis from the first 24 hours(1.22±0.23 vs 0.73±0.21, P＜0.01)to the 7th day(0.46±0.32 vs 0.30±0.13, P＜0.01).(3)There was no significant difference in lgHOMA-IR among SAP, intestinal fistula and other groups at different times(P＞0.05).(4)There was significant difference in lgHOMA-IR between sepsis group and SM group during the whole septic episode(P＜0.01).During the septic episode, there were significant correlation between lgHOMA-IR and APACHEⅡ(r=0.591, P＜0.01), lgHOMA-IR and SOFA(r=0.496, P＜0.01), lgHOMA-IR and SS(r=0.553, P＜0.01), and lgHOMA-IR and TNF-α(r=0.658, P＜0.01).(5)lgHOMA-IR was independently directly correlated with LDL, prealbumin, cholesterol and triglyceride. R2 of the equation was 0.188. Conclusion There usually is insulin resistance in surgical sepsis, regardless the primary diseases. ObjectiveTo investigate the status of an illness of endemic fluorosis in Xuchang city,and to evaluate the effect of implementation of control measures.MethodsIn 2009,Changge city and Xuchang county were chosen as survey counties(city).We investigated the surveillance counties (city) about the running state of water-inproving projects,and fluoride content of drinking water in the villages,and ten villages were chosen as survey villages,dental fluorosis and fluoride content of children aged 8 to 12,clinical skeletal fluorosis and urinary fluoride content of 16-year-old adult were investigated.ResultsIn all the villages with projects of defluoridation for drinking water of the two monitoring counties,that the projects was being used accounted for 61.39％(97/158),and the projects discarded accounted for 38.61％(61/158).In the 9 non-water-improving villages,the average water fluoride ranged 1.32 - 2.25 mg/L; in the 1 water-improving village,the mean water fluoride was 0.44 mg/L.The total rate of dental fluorosis of children aged 8 to 12 was 38.65％ (80/207),and the geometric mean of urinary fluoride was 1.65 mg/L.The rate of adult skeletal fluorosis by clinical detection was 0.30％ (9/2982),and the geometric mean of urinary fluoride was 2.09 mg/L.ConclusionsXuchang city is a mild to moderate fluorosis epidemic areas of endemic fluorosis,and condition of the disease has not yet been effectively controlled,and there is need to further intensify the prevention and control efforts.

Objective To make soluble expression of Plasmodium falciparum (FCC1/HN) glutamate dehydrogenase(GDH) in Escherichia coli, purification and immunocompetence identification of the recombinant non-fusion GDH. Methods The GDH gene was cloned into prokaryotic expression vector pET23(a) to form recombinant expression vector pET23(a)/GDH. pET23(a)/GDH was transformed into E.coli BL21(DE3). Induced by IPTG(isopropyl-beta D-thiogalactoside), GDH was highly expressed in the supernatant after sonication. The soluble recombinant GDH was purified by Source-Q and Source-S chromatography. Enzyme-linked immunosorbent assay and Western blotting were carried out to identify the immunocompetence of the purified product. Results SDS-PAGE analysis showed that the soluble GDH protein accounted for approximately 15% of the total bacterial protein. By two-step ion-exchange chromatography, the purity of GDH reached more than 90% and the GDH possessed high antigenicity. Conclusion The soluble expression of GDH results in an integral three-dimensional structure epitope with high biological activity.
