1.Establishment of porcine combined liver/small bowel transplantation model

ObjectiveTo set up the porcine combined liver/small bowel orthotopic allotransplantation model.MethodThe donors including liver, about 2 to 3?m proximal jejunum, duodenum and a sma ll part of pancreas head were transplanted to the hybridized long-white recipie nt pigs after the resection of their livers and the majority of small bowels.ResultWithout using immunosuppressive drugs, the median survival time of the animals w as 92?h with the longest survival time being 196?h, and there was about 75?% of the animals lived for more than 24?h after operation.ConclusionThe successful establishment of the combined liver/small bowel transplantation m odel will do great favor to our future clinical practice and further study of th e combined liver/small bowel transplantation.
2.Clinical and pathological analysis of seven patients with limb-girdle muscular dystrophy 2B

Objective To analyze the clinical and pathological features of limb-girdle muscular dystrophy2B(LGMD2B).Methods The clinical and pathological features of 7 patients with LGMD2B were analyzed retrospectively.Results Seven patients had a slow onset,and progressive proximal muscle weakness,muscle atrophy,progressive,and incresed serum creatine phosphokinase; muscle biopsy showed different degree of muscle fiber degeneration,necrosis; stromal and inflammatory cell infiltration in muscle fiber; monoclonal antibody immunohistoehemical staining:showed expression of Dysferlin protein was not found in muscle cell membrane,Dystrophin,Sarcoglycans protein expression was normal.Monoclonal antibody immunohistochemical staining the proteins were expressed in normal cell membrane.Conclusions LGMD2B is a slow onset,progressive proximal muscle weakness,muscle atrophy.Histochemical staining on the basis of further immunohistochemical staining,to detect the membrane protein and Dysferlin protein expression,that is a necessary means to diagnose LGMD2B and inflammatory myopathies.

Objective:To evaluate the clinical efficacy and safety ofHJ-RY-1 therapeutic pad ofsoft stone in treating osteoarthritis.Methods:Patients with osteoarthritis with syndrome ofqi stagnancy and blood stasis were enrolled according to the diagnostic criteria and the evaluation standard ofcurative effect in Instruction Principle ofClinical Research ofNew Chinese Medicines, and were randomly divided into experimental group(treating by HJ-RY-1 therapeutic pad ofsoft stone) and the control group(treating by the paster offar infrared bioactive ceramics).Put them respectively on the afflicted part for more than 12 hours a day, and the treatment course ofboth groups lasted 14 days.The main turnover was determined by the improvement of6 clinical parameter indices and the incidence rate ofwhole curative effect and side effect after a treatment of7 days and 14 days.Results:72 patients were randomly equally divided into two groups, and received predetermined interveneing therapy.There were similar baseline characteristics and comparabilities between them.Totally 68 patients finished 2 weeks follow-up observation and 4 cases were missed(33/36 in experimental group;35/36 in the control group).1 weeks later, there were no significant difference in clinical situation and overall curative effect between two groups.But after two weeks interveneing therapy, there were significant difference on activity pain(t-value was 3.3104), the joint movement range(t-value was 3.1596), self-sensation(t-value was 2.5521) and overall curative effect(t-value was 3.5694) between them(P
5.Effect of ulinastatin on renal function during perioperative period in infants undergoing open heart surgery under CPB

Objective To investigate the effect of ulinastatin on the renal function during perioperative period in infants undergoing open heart surgery under CPB.Methods Forty ASA Ⅱ infants (25 male, 15 female)aged 3-5 months weighing 5.3-6.8 kg undergoing open heart surgery under CPB were randomly divided into 2 groups (n = 20 each): control group (group C) and ulinastatin group (group U). Ulinastatin 20 000 U/kg in normal saline 20 ml was infused iv in 3 parts (1/3 was infused via CVP line when the catheter was successfully placed in the internal jugular vein; 1/3 at the beginning of CPB and 1/3 at aortic unclamping). Blood and urine samples were collected at 30 min before incision ( T1 ), 5 min before aortic clamping (T2 ), 5 min after aortic unclamping (T3 ), at the end of operation (T4) and 24 and 48 h after operation (T5, T6 ) for determination of serum urea nitrogen (BUN) and creatinine (Cr) and urinary N-acetyl-β-D-glucosaminidase (NAG) and β2-microglubin (β2-MG) levels. Results There was no significant difference in serum BUN and Cr concentrations between the two groups. The urinary β2-MG concentration and NAG activity were significantly increased at T2-6 as compared with baseline values at T1 in both groups. The urinary β2-MG concentration and NAG activity were significantly lower in group U than in group C at T3-5 . Conclusion Ulinastatin can protect the renal function during perioperative period in infants undergoing open heart surgery under CPB.
6.Effects of antidiabetic drug metformin on human lung adenocarcinoma cell A549 proliferation and apoptosis in vitro

Background and purpose:Metformin is known to be an insulin sensitization agent and is a fi rst line treatment for patients with type 2 diabetes. Recent clinical studies have revealed that metformin treatment has been associated with reduced cancer risk,which indicated that metformin may be a potential anti-neoplastic agent. We investigated the effects of antidiabetic drug metformin on proliferation and apoptosis in human lung adenocarcinoma cell line A549 in vitro and explored the possible underlying mechanisms. Methods:A549 cells were treated with 0.5 mmol/L,2 mmol/L and 8 mmol/L metformin for 48 hrs. Growth inhibition rates of the cells were measured by MTT assay. Cell apoptosis were detected by ? ow cytometery(FCM). Expressions of three genes including p53,Bcl-2 and Bax mRNA in the cells were measured by Real-Time PCR. Results:The proliferation of A549 cells was inhibited by metformin in a dose-dependent manner. The Inhibition rates of metformin at dosage of 0.5 mmol/L,2 mmol/L and 8 mmol/L group were (29?5)%,(68?3)% and (84.1?2.6)%,respectively. Apoptosis was induced when the cells were treated with moderate to high concentrations of metformin.The percentage of cells in early and late stage of apoptosis was increased from (1.1?0.3)% and (1.78?0.22)% in controlled group to (2.1?0.5)% and (9?4)% in metformin 8 mmol/ L group,respectively. The expressions of p53,Bcl-2 and Bax mRNA were all up-regulated after metformin treatment while the Bcl-2/Bax ratio was signifi cantly decreased. Conclusion:Metformin can inhibit the proliferation of human adenocarcinoma cancer cell line A549 and induce cell apoptosis with moderate to high drug concentrations in vitro,which may partly be attributed to the up-regulation of p53 and down-regulation of the Bcl-2/Bax ratio.
7.Reaction Time in Patients with Depression

ObjectiveTo investigate whether subjects with major depression display attention deficits.Methods35 antidepressant-free,non-elderly patients with unipolar depression diagnosis according to ICD-10 and CCMD-Ⅲ as well as 35 health control subjects matched for education,age and sex were administered three tests for attention(sustained attention,Selective attention and Attention Blink).ResultsOn the Continuous Performance Test(CPT),depressed participants needed more reaction time and made more mistake than nondepressed participants.There were no differences between two block in reaction time in the CPT of patients and control subjects.The performance on the Stroop Color-Word Test was impaired in depression.Compared with normal subjects,depressed participants presented increased choice reaction times(CRT).Stroop interference was higher in depressed patients than in controls.On Attention Blink(AB) paradigm,both groups performed equally well at reporting the first target appearing in a rapidly presented stream of stimuli.All groups showed an AB,and AB for depressive participants were larger and longer.Correlations were observed between the HAMD score and the reaction time in CPT.ConclusionThe depressive participants showed a decrement tendency in vigilance,a higher Stroop interference and a larger and longer AB.

ObjectiveTo investigate the characteristics of attentional blink (AB) in the patients with depression.MethodsThirty-five antidepressant-free, non-elderly patients with depression diagnosed according to CCMD-Ⅲ as well as thirty-five health control subjects matched for education, age and sex were tested with the rapid serial visual presentation (RSVP) paradigm.ResultsAll cases of two groups showed an AB, and AB for depressive participants was larger and longer.ConclusionThe patients with depression have attentional impairments.
9.Liver graft induces the immune tolerance of intestinal allograft in auxiliary en-bloc Hver-small bowel transplantation in pigs

Objective To study the effects of liver graft on the immune tolerance of intestinal allograft in auxiliary en-bloc liver-small bowel transplantation in pigs.Methods Seventy outbreed Landrace pigs were divided into 4 groups.Ten auxiliary liver-small bowel allotransplantations were performed in group A,B,C,respectively,and 5 segmental small bowel allotransplantatiom were performed in group D.Pigs were administered with routine and lower dose of cyclosporine and methylprednisolone in group B and C,respectively. No immunosuppressive agent was administered to pigs in group A and D.Results The initial time of acute rejection was obviously prolonged in group A than group D.and the acute rejection was milder in group A than group D(P<0.05).There was no significant difference upon postoperative survival time,initial time of acute rejection and degree of acute rejection between group B and C(P>0.05).Conclusions The immune tolerance of intestinal allograft Can be induced by liver graft in auxiliary en-bloc liver-small howel transplantation.
10.Effect of jianpi jiedu recipe on microvessel density and cyclooxygenase-2 expression in Heliobacter pylori induced gastric cancer.

OBJECTIVETo investigate the regulatory effect of jianpi jiedu Recipe (JJR) on the microvessel density (MVD) and cyclooxygenase-2 (COX-2) in long-term infection of Helicobacter pylori induced gastric cancer of C57BL/6 mice, thus providing experimental bases for its treatment of the H. pylori correlated gastropathy.
METHODSC57BL/6 mouse gastric cancer model induced by H. pylori infection was established by gastrogavage of H. pylori standard strain SS1. Mice were divided into the control group, the model group, low dose JJR group, and the high dose JJR group, 40 in each group. Mice were sacrificed after 72-week medication. Changes of the gastric mucosa MVD of mice in each group were detected by immunohistochemical method. Expressions of COX-2 mRNA and protein were detected by Real-time fluorescent quantitative polymerase chain reaction and immunohistochemical method.
RESULTSThe occurrence rate of gastric cancer in the control group, the model group, the low dose JJR group, and the high dose JJR group was 0, 22.2%, 11.1%, and 10.0%, respectively. The gastric mucosa MVD (number/cm2) of mice in each group was 2.50 +/- 1.54, 18.56 +/- 2.62, 14.61 +/- 3.60, and 7.39 +/- 1.75, respectively. The gastric mucosa MVD in the model group increased more obviously than that in the control group (P < 0.01). The gastric mucosa MVD significantly decreased in the low dose JJR group and the high dose JJR group (P < 0.01). Expressions of COX-2 mRNA and protein in the model group increased more obviously than those in the control group (P < 0.01). Low dose JJR and high dose JJR could decrease their expressions in a dose dependent manner.
CONCLUSIONSH. pylori infection could increase the gastric mucosa MVD of C57BL/6 mice and promote COX-2 expressions, which might play a promoting effect in the incidence of H. pylori induced gastric cancer. JJR could decrease the gastric mucosa MVD and inhibit COX-2 expressions, which might be one of its important mechanisms of preventing and treating gastric cancer.
