Objective To study the components of Forsythia suspensa in stalks and leaves, and compare the content between the crude-sun-cured and the steam-sun-cured. Methods The components and content in stalks and leaves to fruits of Forsythia suspensa were compared by RP-HPLC. Results There were some same active components in stalks, leaves and fruits, and the steam-sun-cured had higher content. Conclusion The steam-sun-cured stalks and leaves of Forsythia suspensa could be used to extract components and develop the tea.
4.Analysis of the plasma free fatty acid composition in T2DM by high performance liquid chromatography

Objective To analyze the plasma free fatty acid (FFA) composition in patients with T2DM. Methods All subjects were from Zhongnan hospital of Wuhan university, and they were divided into three groups: normal control ( n = 94 ), T2DM ( n = 101 ) and T2DM with hyperlipidemia ( n = 77 ). Fasting blood samples were taken from the participants, and plasma FFA were separated using a modified Doles method with the bromoacetophenone, pre-column-derivative. The quantitation of FFA was performed on were (355.63 ± 100. 35) μmol/L, (421.21 ± 200. 83 ) μ mol/L, ( 473.04 ± 213.40 ) μmol/L in healthy controls, T2DM group and T2DM with hyperlipidemia group, respectively. The significant differences were observed among the 3 groups(x2 = 13.08, P <0.01 ). However, there was no significant difference of UFA concentrations among the 3 groups [(206.29± 61.94) μ mol/L, (218.11 ± 110.28) μmol/L and ( 240.94 ± 116.79 ) μmol/L, x2 = 2.17, P > 0.05]. Compared to normal control [( 355.63 ± 100.35 )μmol/L], the FFA concentration[(421.21 ±200.83) μmol/L] in T2DM has significantly increased (x2 =FFA concentrations were higher in T2DM with hyperlipidemia [(473.04 ±213.40) μmol/L] (x2 =27.93,P <0.01 ). The RSD values for intra- and inter-day precision were less than 5%, and the minimal detection limits ranged from 0.05 μmol/L to 0.35 μmol/L The recoveries of high, intermediate and low-level materials were 96.4% -104.8%. Conclusions The total FFA concentration in T2DM has increased, most of which are saturated FFA. The unsaturated FFA has not significantly increased. They seem to be related to the development of T2DM, and might be a new biomarker for clinical monitoring of metabolic disorder of T2DM.
5.Construction and efficacy identification of a lentiviral vector harboring RNAi based on gene NSBP1

Objective To construct and identify the efficacy of a lentiviral vector harboring RNAi sequence targe-ting NSBP1 gene. Methods Three siRNA targeting the NSBP1 mRNA were designed, the pGCSIL-GFP-NSBP1 lentivirus vectors were constructed and confirmed by DNA sequencing. A total of 293T cells were co-transfected with pGCSIL-GFP-NSBP1, pHelper1.0 and pHelper2.0 for the virus stocks produced, the titer of the virus was test-ed. After lentivirus transfecting into DU145 ceils, Western-blot and MTT methods were used to determine the ex-pression and biological activity of NSBP1 gene, the cells were transplanted into nude mice, then inhibitive effect was observed. Results PCR analysis and DNA sequencing demonstrated that the RNAi sequence targeting the hu-man NSBP1 gene was successfully inserted into the lentiviral vector. The titer of the recombinant]entiviral vector was 2 × 10~8TU/mL. NSBP1 protein expression level in transfected cells was significantly decreased and growth rate of cells transfected with lentivirus was decreased by MTT assay, the downregulation of NSBP1 reduced growth rate of transplantated tumor, whereas tumorgenicity was not influenced. Conclusion The construction of the]entiviral vector of NSBP1 has been successfully prepared and NSBP1 plays an important regulatory role in androgen-inde-pendent prostate cancer cell proliferation.
6.Growth hormone with glutamine accelerate remnant intestinal adaptation after massive small bowel resection in rat

Objectives: In order to investigate effect and mechanism of solely or combined treatment of glutamine (Gln) and growth hormone (GH) on remnant intestinal adaptation after massive small bowel resection, we observed the weight of the rats, and analyzed morphology, nutrients absorptive function and gene expression of the remnant intestine. Methods: All rats undertaken 85% small bowel resection and placement of intravenous catheter and gastric tube were randomly assigned to 4 groups :①glycine (SB); ②GH+ glycine (SB/GH); ③Gln (SB/Gln); ④Gln+GH (SB/GH/Gln). All rats received isocaloric, isonitrogenous and isovolumic nutrition support. All rats were weighed daily after operation. On the POD12, crypt depth, villus height and mucosa thickness of remnant small bowel were analyzed; the \[ 14 C\] glucose and \ palmitic acid absorptive test were in vivo assayed; plasma insulin like growth factor 1(IGF 1) was measured; IGF 1, sodium glucose cotransport 1(SGLT1) and intestinal fatty acid binding protein(I FABP) mRNA were semiquatified analyzed by RT PCR. Results: The body weight, plasma IGF 1, villous height and mucosal thickness,\[ 14 C\] glucose and \ palmitic acid absorption both in SB/GH group and SB/Gln group had significantly increased than those in SB group, while those in SB/GH/Gln were significantly higher than those in any other group. The mRNA expression of IGF 1, SGLT1 and I FABP in SB/GH and SB/GH/Gln groups were significantly higher than those in SB and SB/Gln groups. Conclusions: Combined treatment of GH and Gln can enhance the intestinal trophic effect after massive small bowel resection. Gln can enlarge the mucosal absorptive surface in remnant small bowel. GH can not only enlarge the mucosal absorptive surface, but also enhance absorptive functional gene expression of enterocyte via mediation by local expression of IGF 1 .
7.Antibacterial action and sensitization by percutaneous administration of lavender essential oil

Abstract: Objective To investigate the antibacterial action and skin sensitization by percutaneous administration of lavender essential oil (LEO), providing a basis for its antibacterial application of percutaneous administration. Methods The disk diffusion method was used to evaluate the antibacterial effect of LEO on five types of bacteria, and to measure its minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC); The effect of LEO on guinea pig skin irritation was observed by topical application, and its allergic reaction and allergic rate were evaluated; the levels of IgA, IgG, and IgE in the supernatant of serum and spleen tissue sensitized with dimethylbenzene and smeared with LEO were determined by the enzyme-linked immunoassay (ELISA), and the antibacterial ability, skin sensitization, and inflammation of LEO were comprehensively evaluated. Results The antibacterial circle experiment showed that LEO had an inhibitory effect on Staphylococcus epidermidis, Escherichia coli, Candida albicans, Staphylococcus aureus, and Propionibacterium acnes; the antibacterial ability from strong to weak was Staphylococcus Epidermidis (8.25 mg/mL), Escherichia coli (15.00 mg/mL), Candida albicans (16.31 mg/mL), Staphylococcus aureus (18.00 mg/mL), Propionibacterium acnes (20.78 mg/mL). The percutaneous administration of LEO did not cause skin sensitization and inflammatory reaction in guinea pigs. Compared with the blank group, the effect of topical LEO application on the weight of the guinea pig's spleen is not statistically significant (P>0.05), and the effect on the levels of IgA, IgE, IgG in the serum and spleen tissue of guinea pigs is not statistically significant (P>0.05). Conclusions LEO has a certain antibacterial effect on five common pathogenic bacteria such as Staphylococcus epidermidis, Escherichia coli, Candida albicans, Staphylococcus aureus and Propionibacterium acnes, and is safe for percutaneous administration. The results provide some reference for the development of LEO related products and their application in the field of dermatology.
8.Investigation of the surgical safety checklist implementation in a tertiary hospital

Objective To investigate the implementation of the surgical safety checklist in the hospital.Methods The investigation covered the participants of 560 surgical operations of a tertiary hospital,including the surgeons,surgical assistants,scrub nurses and anesthetists,to learn their compliance and awareness of the content of the surgical safety checklist.Results Poor compliance and unawareness of some items of surgical safety checklist in surgical team members were found,plus insufficient understanding of some the items on the checklist.This checklist can improve the quality and safety awareness of the team.Conclusion The implementation of the surgical safety checklist is feasible and effective for avoidance of risks in selective operations,and conducive to promoting communication among the surgical team and preventing surgical errors.
9.The expression and clinical significance of the serum IL-17/LTB4/IgE in children with asthma

Objective To explore the impact of levels of serum IL-17,Leukotriene B4 and IgE on pathogenesis of childhood asthma.Methods Totally 60 children with asthma acute exacerbation ( 29 children with mild asthma,31 children with moderate-severe asthma) were selected as study group,24 healthy children were selected as control group.Serum IL-17 and LTB4 were measured with euryzemLinked immunosorbent assay,serum IgE was determined with enzyme-linked fluoroimmuneassay by pharmacia CAP Sytem,PMN was determined with automatic blood analyser,pulmonary function was measured in the study group.Results ( 1 ) The level of serum IL-17 ( 1.15 ± 0.10 μg/L,2.80 ± 2.30 μg/L,0.83 ± 0.10 μg/L),LTB4 (2.22 ± 1.01 μg/L,8.79 ± 9.36 μg/L,1.94 ± 1.13 μg/L) and IgE( 123.70 ±86.94 μg/L,322.27 ±332.28 μg/L,24.27 ±7.64 μg/L) were significantly different among mild asthma group,moderate-severe asthma group and control group( P < 0.001 ).( 2 )The N% of mild asthma group,moderate-severe asthma group and study group were( 55.06 ± 1 1.15 ) %,( 64.44± 11.87)%,(47.96 ± 13.52)%,L% were(42.20 ± 11.04)%,(33.93 ± 10.02)%,(49.65 ± 13.02)%,and there were significant differences in N% and L% between study group and control group( P < 0.05 ).( 3 ) There were significant positive correlations between the serum IL-17 levels and IgE,LTB4 and IgE,IL-17 and LTB4 in asthmatic children( P <0.05).(4) There were significant negative correlcations between the level of serum IL-17,LTB4 and FEVI,PEF( P <0.001 ).There were significant positive correlations between serum IL-17,LTB4 and N% (P <0.001 ).(5)There were not correlations between the level of serum IgE and FEV1,PEF and N%in asthmatic children( P >0.05 ).Conclusion The levels of serum IL-17,LTB4 and IgE participated in pathogenesis on asthmatic children patients.
10.Relationship between pulse wave velocity and blood pressure classification and cardiovascular risk factors stratification in patients with essential hypertension

Objective To explore the relationship between pulse wave velocity(PWV)and blood pressure classification and cardiovascular risk factors stratification in patients with essential hypertension. Methods Totally,390 patients with essential hypertension were recruited in the study and their carotid- femoral PWVs were recorded by an automatic pulse wave velocity measurement system.The patients were stratified by their cardiovascular risk factors.PWVs were compared between patients with varied blood pressures or with and without cardiovascular risk factors.Results There was no significant difference in PWV between patients of phase 1 and phase 2 hypertension,[(10.8?1.7)m/s vs.(11.9?1.2)m/s,P =0.398].PWV in patients with more than one risk factor was significantly higher than that in those with hypertension alone,[(10.4?1.4)m/s vs.(11.7?1.4)m/s,P=0.018].Conclusions PWV in hypertensive patients with one or more than one other risk factors was significantly higher than that in those without other risk factors.Cardiovascular risk factor might play a more important role in arterial stiffness.
