1.Effects of Several Factors on the Chlamydospores Production of Trichoderma aureoviride T-33

The effects of several factors on the chlamydospores production of Trichoderma aureoviride T-33 during the fermentation were researched.Based on the results above, the orthogonal test was made to screen out the best prescription.The results showed that, the best single-factor conditions for the chlamydospores production of T.aureoviride T-33 were, liquid culture of oat powder, 30℃, pH4.0, 120r/min, 24 hours oscillate incubating as well as liquid culture volume of 80mL/bottle when the 250mL size triangle bottle was used.The result of orthogonal test showed that, the best prescription for temperature, pH and oscillating speed was 30℃, pH4.0 and 140r/min.3.37?10~ 7 spore/mL chlamydospores were obtained at this combined condition.
2.Clinical study on contrast-enhanced ultrasound in traumatic brain injury

Objective To explore the value of contrast-enhanced ultrasound (CEUS) in classifying lesions,measuring the range of lesions,and the ability of manifesting the boundary and the severity of lesions after traumatic brain injury.Methods 83 patients with traumatic brain injury underwent the emergency surgery operations were enrolled in this study.Intraoperative ultrasound was applied in detecting the echo characteristics of lesions and classifying.Traumatic brain injuries which were classified indefinitely by conventional ultrasound were examined by CEUS to observe enhanced features of trauma lesions and the surrounding tissue,confirm the types of lesions,identify the border of lesions,measure the range of lesions,and compare with conventional ultrasound,preoperative CT and operative results.The severity of lesions was judged according to the parameter of time intensity curve (TIC).The results which would help the operators to adjust the method of treatment timely was informed.Results 148 lesions were detected by intraoperative ultrasound among 83 patients in this study.Thirty-two lesions which were classified indefinitely by conventional ultrasound were examined by CEUS.The results of classifying were confirmed by CT and operations,the diagnostic accuracy rate was 100%.The absolute peak intensity was changed with the injury severity,the more serious of the tissue,the lower absolute peak intensity was displayed.The boundary of lesions was manifested clearly after CEUS.Compare to the conventional ultrasonography,the range of lesions was larger.The difference between them was meaningful significally(P =0.01).Based on the results of CEUS,the way of operation was adjusted in 21 lesions.Conclusions Using the contrastenhanced ultrasound and intraoperative ultrasound,various traumatic brain injuries can be diagnosed accurately.By CEUS,the perfusion of lesions would be displayed,which would hint the severity of the brain tissue and help the operators adjust the methods of treatment timely,and provide the reliable evidence for the operation precisely.
3.Effects of inward rectifier potassium channel blockers on EPCs function.

OBJECTIVETo investigate the effects of inward rectifier potassium channel blockers (BaCl2, CsCl) on the functions of endothelial progenitor cells (EPCs).
METHODSDensity gradient centrifugation-isolated rat hone marrow mononuclear cells were cultured in vitro. EPCs were harvested and seeded on six culture dish when cells grew to 3-5 passages. Before testing the EPCs were synchronized with M199, which contain 2% fetal calf serum. In the end, EPCs were treated with different intervention. The experiment mainly included two parts: (1) BaCl2 (100 micromol/L) and free BaC2 of Tyrodes solution; (2) CsCl (1 mmol/L) and control. Cell pretreated with blockers above mentioned for 12 h, then the gene expression of stromal cell-derived factor-1 (SDF-1), epoprotenol (PGI2) were assessed, beyond that the ability of adhesion, migration were assayed with different tests. In addition, the medium was collected when EPCs were treated for 3 days. The levels of SDF-1 were measured by sandwich enzyme-linked immunosorbent assay (ELISA). Going even further, EPCs were treated with the signal pathway blockers in advance, after repeat the above steps, in order to analyze the change of SDF-1 and then discuss its mechanism.
RESULTSCompared with control group, BaCl2, CsCl could increase EPC adhesion and migration to same extent. Moreover, the gene expression of SDF-1, PGI2 was significantly up-regulated and the production of SDF-1 increased evidently. Furthermore, the mechanism of SDF-1 secretion increasing mainly was associated with eNOS signaling pathways.
CONCLUSIONBa2+ and Cs+ play important roles in increasing EPCs functions, such as adhesion, migration and secretion.

4.Study of attribution of multicomponent original medicinal materials in gegen qinlian decoction with intestinal permeability.

The complex level of constructing biopharmaceutics classification system of Chinese materia medica CMMBCS) was the study of traditional Chinese compound, on the premise of insisting that the multicomponent simultaneous determination, when carrying out the study of intestinal permeability, the primary task was to define the source of the components that was absorbed through the intestinal wall, namely, which medicinal material the components belonged to in traditional Chinese compound. The technology of chemical fingerprint and in vitro everted gut sac model were used in this research to make multicomponent an intuitive source attribution which permeated the intestine in the classic formula Gegen Qinlian decoction, and to lay the foundation for the further qualitative and quantitative research of intestinal permeability.

5.Microsurgical perioperative care of brain stem vascular cells tumor

Objective To summarize the microsurgical perioperative care of brain stem vascular cells tumor. Methods The perioperative care were summarized for twenty-one patients taken microsurgical treatment of brain cells of blood vessels, and effect of nursing were analyzed. Results When 21 patients discharged, their nervous system symptoms were improved. 18 cases were followed up for 1 ~ 42 months, average 14 months. 16cases went well and 2 cases aggravated in terms of neurological symptoms. Conclusions The complications after the operation of brain stem vascular cells is complex, proper nursing is the important guarantee for perioperative security and the best way to improve patients' rehabilitation.
6.Chemical constituents from EtOAc fraction of Sophora dunnii.

Sixteen compounds have been isolated from the EtOAc fraction of 95% ethanolic extract of Sophora dunnii through silica gel, Sephadex LH-20 and semi-prerarative HPLC column chromatographies. Their structures were identified on the basis of NMR and MS spectra data as phaseollidin (1), L-maackiain (2), 2-(2',4'-dihidroxyphenyl)-5,6-methylenedioxy benzofuran (3), 8-demethyl-farrerol (4), liquiritigenin (5), genistein (6), 6-methylgenistein (7), 5-O-methyl genistein (8), 7,2',4'-trihydroxys-5-methoxy-isoflavanone (9), 7, 3', 4'-trihydroxy-isoflavanone (10), erythribyssin D (11), calycosin (12), trans-resveratrol (13), cis-resveratrol (14), stigmasterol (15), β-sitosterol (16). Among these, compounds 1-14 and 16 were isolated from this plant for the first time.

7.Application of ligature technology in Gastrectomy

ObjectiveTo compare the safety and efficacy of ligature between traditional ligation way in gastrectomy.Methods60 patients with gastrointestinal cancioma were selected and randomly divided into two groups,30cases of ligature(treatment group),30 cases of ligation (traditional group).ResultsThe operative time,blood loss,postoperative hospital stay,complications and treatment costs was compared between the two groups.Compared with the traditional group,there were shorter treatment time,reduced bleeding,shorter hospital stay,fewer complications,reduced treatment costs in treatment group.ConclusionLigature gastrectomy is safe and effective.
8.Morphologic and functional characteristics of the immortalized human liver sinusoidal endothelial cell line.

OBJECTIVETo investigate the morphologic and functional characteristics of the immortalized human liver sinusoidal endothelial cell line (LSEC line).
METHODSImmunofluorescence staining and fluorescence microscopy were used to detect the classic endothelial cell markers in LSEC line, and flow cytometry was used to analyze the purity of the human LSEC line. The morphology (including W-P bodies and surface fenestrations) and phagocytotic capacity of the human LSEC line were observed by transmission and scanning electron microscope. The proliferation curve of the human LSEC line was analyzed by MTT assay. The functional differences between the human LSEC line and human primary LSEC in expression of ELAM-1 and ICAM-1, activities of fibrinolysis (PAI-1, t-PA, u-PA), releasing of IL-6 and IL-8 were compared respectively by enzyme linked immunosorbent assay. Comparison of the susceptibility to hypoxia-reoxygenation induced apoptosis between the human LSEC line and human primary LSEC were investigated by TUNEL.
RESULTSThe established human LSEC line maintained a high proliferative ability and has been passaged for more than 80 times in the absence of any growth factors. Immunofluorescence staining showed that the human LSEC line could express classic endothelial cell marks including von Willebrand Factor (vWF), and could take up acetylated low-density lipoproteins (Ac-LDL). The purity of the human LSEC line was confirmed over 95% by flow cytometric analysis. The W-P bodies and the phagocytosis of Dynabeads was demonstrated by transmission electron microscope. And fenestrations could be found cellular surface with scanning electron microscopy. When compared with human primary LSEC, the human LSEC line has an equivalent responsiveness to tumor necrosis factor in up-regulation of ELAM-1 and ICAM-1. The human LSEC line can also release PAI-1, t-PA, u-PA but can not release IL-6 and IL-8 to TNF-alpha. In contrast, human primary LSEC could release IL-6. The human LSEC line showed higher susceptibility to hypoxia-reoxygenation-induced apoptosis, and the percentage of apoptotic cells was as high as (38.4 +/- 6.7)%, while (28.6 +/- 4.5)% and (7.8 +/- 1.2)% respectively in primary LSEC and in human umbilical vein endothelial cells.
CONCLUSIONSThe established human LSEC line maintains the special phenotypes and the major functional characteristics, and especially maintains the high susceptibility to hypoxia-reoxygenation-induced apoptosis. Therefore it is feasible to use this cell line for the study of liver ischemia-reperfusion injury.

10.Impacts of multicomponent environment on solubility of puerarin in biopharmaceutics classification system of Chinese materia medica.

To illustrate the solubility involved in biopharmaceutics classification system of Chinese materia medica (CMMBCS) , the influences of artificial multicomponent environment on solubility were investigated in this study. Mathematical model was built to describe the variation trend of their influence on the solubility of puerarin. Carried out with progressive levels, single component environment: baicalin, berberine and glycyrrhizic acid; double-component environment: baicalin and glycyrrhizic acid, baicalin and berberine and glycyrrhizic acid and berberine; and treble-component environment: baicalin, berberin, glycyrrhizic acid were used to describe the variation tendency of their influences on the solubility of puerarin, respectively. And then, the mathematical regression equation model was established to characterize the solubility of puerarin under multicomponent environment.
