5.Curative Effect of Vinpocetine Combined with Danhong Injections in the Treatment of Posterior Circulation Ischemia Syndrome

Objective:To observe the curative effect of vinpocetine combined with Danhong injections in the treatment of posterior circulation ischemia syndrome. Methods:Totally 120 patients with posterior circulation ischemia syndrome were selected and randomly divided into the treatment group and the control group with 60 cases in each. Both groups were treated by the conventional therapy, meanwhile, the treatment group was given vinpocetine and Danhong injections, and the control group was given vinpocetine alone. The treatment course was 14 days. The curative effect was observed, blood rheology and hemodynamics were also studied. Results:The to-tal effective rate of the treatment group was 95. 0%, which was significantly higher than that of the control group (75. 0%) with statis-tically significant difference(P<0. 05). After the treatment, the blood rheology (viscosity, low shear viscosity, plasma viscosity and reduced viscosity) and TCD ( LVA, RVA, BA and PI) in the two groups were significantly improved, and the treatment group showed more significant improvement(P<0. 05). Conclusion:Vinpocetine combined with Danhong injections in the treatment of posterior cir-culation ischemia syndrome has significant effect in the effective improvement of blood rheology and hemodynamics, which is worthy of clinical promotion and application.
6.Probiotics in the treatment of experimental colitis in rats and its influence on T cell subsets.

Objective To evaluate the effects of probiotics in experimental colitis of rats.Methods Models of experimental colitis were established.Several groups of rats were set up as probiotics group,negative group and positive group.Histologic scoring was used to estimate effects of drugs;the expression of CD~+_4 andCD~+_8 on T cell surface in peripheral blood,spleen mononuclear cells and intraepithelial lymphocytes of colon were detected by flow cytometry.Results Histopathology shows that probiotics of larger dosage was curative for experimental colitis in rats.Proportion of CD~+_4 and CD~+_8 T cells increased in colon of colitis,which was decreased by treatment of larger dose of probiotics.CD~+_4/CD~+_8 ratio and T cell subsets in systemic immune system were not influenced by probiotics.Conclusion Probiotics of large dosage are effective in the treatment of experimental colitis of rats incuced by TNBS,possibly associated with immune regulation and tends to be localized in mucosal immune system in colon.
7.The influence of T regulatory cells on experimental colitis of rate after treatment by probiotics

Objective To evaluate the effects of probiotics on experimental colitis of rats and to explore systemic and local changes of T regulatory cells(Tr) after the treatment. Methods The models of experimental colitis were established by enema with 2,4,6-trinitrobenzenesulphonic acid (TNBS). Live probiotics of combined bifidobacterium,lactobacillus and enterococcus (Bifico) were used. Four groups of rats were set up as negative group, TNBS alone group, probiotics group (high dosage and low dosage, with dose of 150 and 300 mg?kg-1?d-1, respectively), and combination group (probiotics plus prednisolone or sulfasalazine). Histologic scoring was used to evaluate the effect of medications; the proportions of CD4+CD25+ and CD8+CD28- Tr in peripheral blood, spleen mononuclear cells and intraepithelial lymphocytes of colon were detected by flow cytometry. Objective To investigate whether nuclear transcription factor κB(NF-κB) through Toll-like receptors 2(TLR2) was activated by lipid-associated membrane proteins(LAMPs) of Mycoplasma genitalium.Methods LAMPs were extractded and THP-1 cells were stimulated.The activation of NF-κBp65 was detected by ELISA and the expression of TLR2 mRNA was detected by RT-PCR.Effects of TLR2 neutralizing antibody on LAMPs induced the activation of NF-κBp65 was analyzed by ELISA.After LAMPs stimulated 293T cells with the co-transfection pFLAG-TLR2,pNF-κB-luc,pRL-TK,the activity of NF-κB firefly luciferase and pRL-TK Renilla luciferase were detected by the dual-luciferase reporter gene,to analyzed the role of TLR2-mediated NF-κB activation by LAMPs in 293T cells.Results The activation of NF-κBp65 was mediated in LAMPs induced THP-1 cells and was significantly increased by LAMPs in a dose dependent manner.when LAMPs was 4.0 μg/ml,the activation of NF-κBp65 was the highest level.TLR2 mRNA expression was up-regulated by LAMPs in THP-1 cells.TLR2 neutralizing antibody could inhibit the activation of NF-κB by 60% in LAMPs stimulated THP-1.NF-κB fluorescence was significantly increased by co-transfection pFLAG-TLR2 in a dose-dependent manner. ConclusionMycoplasma genitalium-derived lipid-associated membrane proteins activate NF-κB via TLR2 and the activation of TLR2-mediated play an important role in pathogenic process of LAMPs.
9.An epidemiological investigation on behavioral factors of antibiotics app lication in population

Objective To find out the influencing behavioral factors of antibiotics usage in population in Lanzhou. Methods Status of antibiotics usage was investigated in four hospitals in Lanzhou. Results The self-medicine pro portion among patients with respiratory system infection, infectious di arrhea, urinary system infection and choleic system infection were 44.22%, 31. 76%, 22.07% and 6.61% respectively. Patients having higher income used some antibiotics at a middle price, while patients having lower income used antibiot ics at a lower price. The educate d person applied some antibiotics at a lower price, but the illiterate person us ed some antibiotics at a higher price. The combined usage of antibio tics was affected by the disease program and distance of seeing doctor. The fart her distance away from hospital, the more proportion of antibiotics(two or thre e) used combinedly. Conclusions The antibiotics usage was affe cted by the disease variety, disease program, distance to hospital, average inco me and educ ated level.
10.Icariin and its pharmaceutical efficacy: research progress of molecular mechanism.

Icariin is one of the key active components of Epimedium species, which is most widely applied to supplement the kidney in traditional Chinese medicine. Scientific research has found that icariin possesses extensive therapeutic effects such as protecting neurons from injury, promoting growth of neuronal synapse, improving sexual dysfunction and bone morphogenesis, as well as anti-inflammation, anti-tumor and anti-depression functions. Considering that molecular mechanism is the fundamental basis for pharmaceutical efficacy of icariin, in this article, the authors retrospectively retrieved 122 scientific papers recorded in the PubMed database with "icariin" in the title from January 1, 1995 to January 5, 2011. It was found that icariin has been closely highlighted in the intervention of p38 mitogen-activated protein kinases and phosphatidylinositol 3-kinase/Akt signal pathways, inhibition of phosphodiesterase 5, and regulation of nuclear receptors. Besides, the authors also discussed the main orientation for molecular mechanism of icariin in future research.
