1.The clinical curative effect of dampness Kushen Decoction in the treatment of psoriasis vulgaris with damp heat syndrome

Objective To study and discuss the clinical curative effect of dampness Kushen Decoction in the treatment of psoriasis damp heat syndrome.Methods100 cases of psoriasis vulgaris(damp heat syndrome) patients who treated in our hospital from January 2014 to October 2016 were selected as the research object, the patients were divided into two groups by taking the single blind randomly grouping method, each group had 50 cases, the control group used Tacrolimus Ointment chemophlebitis, the observation group treated with dampness Kushen decoction on the basis of the control group, the total effective rate and skin injury score were compared between two groups.ResultsAfter 8 weeks of treatment, the total effective rate in the observation group was significantly higher than the control group (P<0.05);after treatment, the PASI scores in two groups were significantly decreased (P<0.05), but the PASI score in the observation group was significantly lower than the control group (P<0.05).ConclusionThe dampness Kushen Decoction in the treatment of psoriasis damp heat syndrome has significant curative effect, can effectively promote the skin damage and improve the prognosis better.
2.Detection of Mycobacterium Tuberculosis DNA from the Skin Lesion of Sarcoidosis by Polymerase Chain Reaction

The cause of sarcoidosis is still unknown. However,the histological similarity between the disorder and tuberculosis suggests that M. tuberculosis might contribute to the pathogenesis of sarcoidosis. In attempting to confirm the relation between M. tuberculosis and sarcoidosis,we used the polymerase chain reaction to detect M. tuberculosis DNA from formalin--fixed,paraffin--embeded skin biopsy specimens of 12 patients with sarcoidosis, M. tuberculosis DNA was found in 4 patients. This finding suggests an aetiological role for M. tuberculosis in sarcoidosis.
3.Simultaneous determination of benzophenones and xanthone in leaves of Aquilaria sinensis by RP-HPLC-UV.

This study is to develop a sensitive method by using reversed-phase high performance liquid chromatography coupled with UV detector (HPLC-UV) to simultaneously determine four bioactive compounds, iriflophenone 3-C-beta-D-glucoside, iriflophenone 3,5-C-beta-D-diglucoside, mangiferin, and iriflophenone 2-O-alpha-L-rhamnoside in the leaves of Aquilaria sinensis. An Agilent Zorbax SB-C, column (4, 6 mm x 250 mm, 5 microm) was used, and the gradient elution was performed with mobile phase of 0.1% aqueous phosphoric acid and acetonitrile at a flow rate of 1 mL x min(-1). The detection wavelength was 280 nm, and the column temperature was 25 degrees C. The four marker compounds were well separated with good linearity (R2 > 0.9990), precision, stability and repeatabili y. The-recovery rates were in the range of 98.80%-101.39%. For 15 branch of the leaves, the contents of iriflophenone 3-C-beta-D-gluoside, iriflophenone 3,5-C-beta-D-diglucoside, mangiferin, and iriflophenone 2-O-alpha-L-rhamnoside were between 0.41-14.48, 0.72-3.85, 4.30-29.07, 0.24-5.06 mg, respectivley. This method is precise, accurate and reliable, which provides an efficient way for the quality control of the leaves of A. sinensis. This will promote the comprehensive usage of this plant.

4.Long-pulsed 1064 nm laser combined with triamcinolone in treatment of keloids

Objective To evaluate the clinical efficacy of long-pulsed 1064 nm Nd ∶ YAG laser combined with intralesional injection of triamcinolone in the treatment of keloids.Methods A total of 78 cases were randomly divided into two groups.The treatment group was treated with long-pulsed 1064 nm Nd ∶ YAG laser combined with intralesional injection of triamcinolone.The control group was only given intralesional injection of triamcinolone.The efficacy and adverse reactions were determined at the end of the first and the second course,respectively,and follow-up for one year was conducted in order to observe the recurrence.Results When the first course was over,cure rate of 21.95 % and efficiency rate of 73.17 % were observed in the treatment group,while they were 10.81% and 45.95 %,respectively,in the control group; the efficacy rate was statistically significant different in these two groups (P<0.05).After two courses,cure rate of 70.73 % and efficiency rate of 100 % were observed in the treatment group while they were 45.95 % and 83.78 %,respectively,in the control group,showing that both of the rates were statistically higher in the treatment group (P<0.05).Conclusions Compared to intralesional injection of triamcinolone,long-pulsed 1064 nm Nd ∶ YAG laser combined with injection of triamcinolone has a more satisfactory effect on keloids.
5.The Screen of Plant Endophytic Actinomycetes Producing ?-1,3-Glucanase and Antifungal Activity of ?-1,3-Glucanase

Using of clearing zones on pachyman agar medium, there are 217 plant endophytic actinomycetes, producing ?-1,3-glucanase were screened. 45.6% of the strains produced ?-1,3-glucanase, in which the strains from cucumber are up to 38. The percentages of endophytic actinomycetes from different hosts produceing ?-1,3-glucanases were different. The percentage of the strains in Rhizoma Polygonatum produced ?-1,3-glucanases is the highest, up to 88.9%. The Inhibited effects of plant endophytic actinomycetes which produced extracellular ?-1,3-glucanases on mycelium growth of Sclerotinia sclerotiorum were detected in vitro. Cucumber endophytic actinomycete gCLA4 strain was screened out from 99 isolates, which can strongly inhibit the growth of S. sclerotiorum. The optimal ?-1,3-glucanases fermentation conditions of strain gCLA4 were investigated, they were pachyman 0.2%, peptone as nitrogen, pH 7~8 for 5 days. The ?-1,3-glucanases of strain gCLA4 had some inhibiting efficiency on 13 plant pathogens, in which inhibiting efficiency to Botryosphaeria dothidea was the strongest.
6.Outcomes and prognostic factors of advanced squamous cervical cancer after concurrent chemoradiotherapy

Objective To evaluate the outcomes and the prognostic factors for advanced squamous cervical cancer after concurrent chemoradiotherapy (CCRT).Methods Totally 172 patients with International Federation of Gynecology and Obstetrics stage Ⅱ b-Ⅳ who were treated in Cancer Hospital,Chinese Academy of Medical Sciences between January 2007 and December 2008 were retrospectively analyzed.Patients were received external radiotherapy,high-dose rate brachytherapy and cisplatin-based chemotherapy concurrently.Results The median follow-up period was 54.5 months.The 2-year and 5-year overall survival (OS) were separately 81.5% and 68.8%.The 2-year and 5-year progress-free survival (PFS) were separately 69.2% and 63.1%.Using univariate analysis followed with multivariate analysis,the results showed that these clinicopathological factors including stage (Ⅲ and above versus Ⅱ b; P =0.021,HR =1.95 ; P =0.020,HR =1.86),maximum diameter of local tumor size (>4 versus ≤4 cm; P =0.009,HR =2.55 ; P =0.033,HR =1.94),squamous cell carcinoma antigen (SCC) level before treatment (>3 versus ≤3 μg/L; P =0.010,HR =2.47; P =0.013,HR =2.09) and retroperitoneal lymph node status on imaging (para-aortic lymph node positive versus negative,P =0.009,HR =3.00,P=0.010,HR =2.74; pelvic lymph node positive only versus negative,P =0.044,HR =1.98,P =0.033,HR =1.92) had the significant effect on OS and PFS.Patients with no above adverse prognostic factor were assigned to Group A (n =18),those with one factor were assigned to Group B (n =43),and those with no less than two factors were assigned to Group C (n =1 11).Among three groups,the 2-year OS were separately 94.1%,97.7% and 73.1%,the 5-year OS were separately 81.4%,90.1% and 58.6%,the 2-year PFS were separately 88.2%,90.4% and 57.9%,the 5-year PFS were 82.4%,87.9% and 50.0%.The results showed that group C was significant difference from Group A or B in OS and PFS (all P < 0.05),while Group A had no significant difference from Group B in OS and PFS (P > 0.05).Conclusions Stage Ⅲ or above,maximum diameter of local tumor size > 4 cm,SCC level > 3 μg/L before treatment and positive retroperitoneal lymph nodes on imaging are four independent adverse factors for prognosis of squamous cervical cancer of advanced stage after CCRT.The treatment of patients with no less than two adverse factors should be considered to be improved.
7.Relative bioavailability and bioequivalence of azithromycin tablets in healthy volunteers.

OBJECTIVETo evaluate the bioavailability and bioequivalence of azithromycin tablets in healthy volunteers.
METHODSA single dose (500 mg) of azithromycin tablet (from 3 pharmaceutical companies) was given to 18 healthy volunteers according to a randomized cross-over design. The plasma concentrations of azithromycin were determined by liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS).
RESULTS AND CONCLUSIONThe T(max) of azithromycin products of the 3 pharmaceutical companies were 2.3-/+1.1, 3.0-/+1.1 and 2.9-/+0.8 h, C(max) were 319.2-/+176.7, 303.10-/+144.60 and 313.70-/+165.00 ng/ml, AUC(0-144) were 5073.60-/+2933.00, 4296.80-/+1896.20 and 4797.80-/+3234.00 ng.h/ml, AUC(0-max) were 5461.60-/+3236.00, 4804.40-/+2162.90 and 5163.20-/+3497.50 ng.h/ml, respectively. Statistical analysis of the above main pharmacokinetic parameters of azithromycin tablets suggests the bioequivalence of the three formulations.

8.Prenatal ultrasonographic diagnosis and prognosis of congenital cystic adenomatoid malformation

Objective To investigate the ultrasound characteristics of congenital cystic adenomatoid malformation (CCAM). Methods The ultrasonographic features of 21 cases of fetal CCAM confirmed by prenatal ultrasound were analyzed. And ultrasound ifndings were compared with the postnatal thoracic computed tomography (CT), surgical result and follow-up data. Results (1) Prenatal ultrasonographic ifndings:2 cases of CCAM typeⅠ(10%, 2/21), 13 cases of typeⅡ(62%, 13/21), and 6 cases of typeⅢ(28%, 6/21). Of them, 95%(20/21) lesions were located in unilateral thorax, while 5%(1/21) in bilateral thorax. CCAM was ifrst detected at 18th-25th gestational week with a median of 22nd week. The CVR was<1.6 and>1.6 in 16 (76%, 16/21) and 5 (24%, 5/21) cases respectively. After reaching the maximum volume, the size of mass remained stable in 8 cases and regressed in 13 cases with even complete remission in 5 cases. There were mediastinal shift in 19 fetuses (90%, 19/21), polyhydramnios in 6, and concomitant structural abnormalities in 3 cases such as pulmonary sequestration, diaphragmatic hernia and fetal hydrops. (2) Clinical outcomes:15 newborns were conifrmed as CCAM by CT, in which 9 cases underwent surgical resection. One case was conifrmed by autopsy. Nineteen cases showed good prognosis. Whereas 1 pregnancy with fetal hydrops was terminated, and 1 fetus with diaphragmatic hernia died after birth. Conclusions Dynamic prenatal ultrasound assessment is the key factor in prognosis prediction of fetal CCAM. CCAM without other structural abnormalities has a good outcome.
9.Detection of CEA negative colorectal cancer and prognostic biomarkers of colorectal cancer

ObjectiveTo study serum proteomic fingerprints of colorectal cancer during onset and progression and to screen tumor markers related to prognosis.Methods Serum from colorectal cancer patients, non-cancer patients, and healthy control were profiled using WCX ProteinChip or magnetic beads and analyzed by mass spectrometry. Results Seven protein peaks were found related to colorectal cancer. Several peaks were closely related to lymph node metastasis, distal organ metastasis and decreased after surgery. The diagnostic model composed of 3398.3、5477.1、8453.9 u can detect CEA negative colorectal cancer in 100%. Conclusion Protein fingerprinting technology (PFT) in conjunction with bioinformatics can significantly identify novel biomarkers in the serum of colorectal cancer patients with potential values for prognostic evaluation, detection of CEA negative colorectal cancer and changing its progression.
10.Intraperitoneal injection of bleomycin induces pulmonary fibrosis in mice:a long-term stability evaluation

BACKGROUND:There is no effective drug for idiopathic pulmonary fibrosis (IPF), because of a lack of the animal model imitating the complete pathogenesis of human IPF. Therefore, it is critical to establish an ideal animal IPF model used for investigating the underlying pathogenesis and developing a kind of effective drug. OBJECTIVE:To establish an animal model that can mimic more characters of human IPF. METHODS:Seventy male C57BL/6 mice were randomly divided into two groups, fol owed by subjected to the intraperitoneal injection of bleomycin (35 mg/kg) on days 1, 4, 8, 11, 15, 18, 22, and 25, twice (group A) or once (group B) a week. Mice were sacrificed at 2, 4, 6, 8, and 10 weeks after the eighth injection, and the lung tissues were moved used for hematoxylin-eosin, Masson and immunohistochemical stainings. RESULTS AND CONCLUSION:There were various degrees of alveolitis and pulmonary fibrosis in the two groups at different time points after the last injection. The scores of alveolitis and pulmonary fibrosis in the group A began to gradual y increase from the 2nd week and reached the highest level at the 6th-8th weeks until the 10th week. In contrast, the scores of alveolitis and pulmonary fibrosis in the group B peaked at the 2nd week, then fluctuately decreased, and were significantly lower than those in the group A at the 6th week (P<0.05). Immunohistochemistry showed that type I col agen deposition was mainly distributed in the subpleural region, peri-vascular region and alveolar septa, which was consistent with Masson staining findings. The expression levels of transforming growth factorβ1 (TGF-β1) andα-smooth muscle actin (α-SMA) in the regions developing alveolitis and pulmonary fibrosis were significantly increased. In the group A, the expression levels of type I col agen, TGF-β1,α-SMA, and the hydroxyproline content in the lung tissues reached the peak level at 6-8 weeks. However, in the group B, al above indicators reached the highest level at the 2nd week, but gradual y decreased thereafter. At the 4th week, the expression Levels of TGF-β1 andα-SMA in the group B were significantly lower than those in the group A (P<0.05). At the 6th week, the hydroxyproline and type I col agen levels in the group B were significantly lower than those in the group A (P<0.05). In conclusion, the mouse model of pulmonary fibrosis induced by intraperitoneal injection of 35 mg/kg bleomycin twice weekly can be used to mimic the repetitive wound healing process, pathological morphology and cytokine changes of human IPF, which is prone to administration, with better stability and repeatability.
