2.Study of effect of extracting time and solvent on the quality of extract of Salvia Miltiorrhiza Bunge

Objective:To study the effect of the extracting time and solvent on the concentration of main active components in Salvia Miltiorrhiza Bunge, and to provide basic data and theoretical guide to GEP(general extraction practice). Methods: Salvianolic acid B and Tanshinone ⅡA were extracted by 90% alcohol and water respectively from Salvia Miltiorrhiza Bunge and their contents were etermined by means of HPLC at regular intervals. Results: Extracting time and solvent were significant factors in the prcess of concentrating Salvianolic acid B and Tanshinone ⅡA from extract of Salvia Miltiorrhiza Bunge. Conclusion: Water is suitable for extracting Salvianolic acids, the extracting time is 40~50 minutes; alcohol is suitable for extracting Tanshinones, the extracting time is 20~30 minutes.
3.Theoretical simulation and experimental research on constructing different compositions of ginsenoside

AIM: To Structure a series of diversifying combination of ginsenoside in order to provide the different samples for the pharmacological and pharmacodynamics experiments. METHODS: UNIFAC group contribution method is used for simulating the liquid liquid extraction, the proper extractants are chosen to carry out the experiments, and then the contents of the ginsenosides obtained with different extractants are detected through HPLC. RESULTS: The contents of the main ginsenoside varied with the change of the extractants' ratio, as a result a series of diversifying combination of ginsenoside are constructured. Within the ginsenoside which has greater amount, ginsenoside Rb 1 can be easily separated from Re, while Rd is hard to be separated, the experimental data were accordant to the theoretical prediction. CONCLUSION: Theoretical calculation combined with the optimizing of the extractants can structure a series of distinguished ginsenoside.

Objective: To study the change of placental corticotropin releasing hormone level in pregnant women entering spontaneous labor at term and preterm or posterm,and to investigate the value of CRH as an indicator of preterm labor. Methods: Subjects( n =200) were recruited to the study at their antenatal visits,and plasma samples were obtained at their routine antental clinic visits from 28 weeks to parturition at 2 weeks intervals.CRH were measured by radioimmunoassay. Objective To investigate the application value of transcranial Doppler ultrasonography(TCD) in critically ill children with cerebral dysfunction.Methods We detected the cerebral blood flow parameters[systolic velocity(Vs),mean velocity(Vm),end of diastolic velocity(Vd),pulsatility index(PI),resistance index(RI)] of middle cerebral artery in cerebral dysfunction group(n=20) and control group(no cerebral dysfunction,n=20),and observed the differences of those parameters between two groups.Children in cerebral dysfunction group were also divided into groups according to Glasgow Coma Score(GCS) and prognosis.The differences of each TCD parameter in diverse groups divided by GCS and by prognosis were analysed.Children in cerebral dysfunction group were checked TCD everyday until TCD parameters were normal.The relation between the days of TCD parameters′ reaching normal and GCS,duration of disorder of consciousness with correlation analysis were analysed.Results Vs,PI,RI in cerebral dysfunction group were higher than those in control group,while Vd was lower (P<0.05).(2)PI in the group of GCS less than 6 points(0.91±0.21) was higher than that in group of GCS 7-13 points(0.83±0.14)(P<0.05).The other parameters between two groups of GCS less than 6 points and GCS 7-13 points were not significantly indistinctive.Between the good prognosis group and the bad prognosis group,there were no significant differences in all parameters.(3) The days of TCD parameters′ reaching normal were negatively correlated with GCS(r=-0.653,P<0.01),but positively correlated with the duration of disorder of consciousness(r=0.923,P<0.01).Conclusion The cerebral hemodynamics of children with cerebral dysfunction is abnormal,the more serious cerebral dysfunction,the higher PI and the longer time for regaining normal TCD parameters.Constantly monitoring TCD could reflect the changes of cerebral hemodynamics and is valuable to assess the state of illness and prognosis.

Objective To evaluate the clinical application value of dual color fluorescence in-situ hybridization (FISH) in detecting urothelial carcinoma of the urinary bladder.MethodsThe probes of chromosome 3,7,17centromeres and chromosome 9p21 region (p16) were labeled by random primer method.FISH was performed on interphase nuclei of 80 urine specimens of cancer of the urinary bladder and 20 cases of healthy persons served as normal controls.Threshold value was established.The pathological diagnosis was the golden standard.Chromosome aberration was counted.The correlations between chromosome aberration and pathologic grading and staging and their sensibility of diagnosis for urothelial carcinoma of the urinary bladder were analyzed. Results The rate of numerical aberration of chromosome 3,7,17,9p21 was 47.5%(38/80),60.7% (49/80),51.3% (41/80) and 58.8% (47/80) respectively.The positive rate of the combined use of 3,7,17 and 9p21 chromosome probes for detecting urothelial carcinoma of urinary bladder was 76.3% (61/80).The aberrations had no correlation to tumor stage.The aberration of chromosome 3,7 and 17 were correlated to pathologic grade significantly (P < 0.05).ConclusionThe progression of urothelial carcinoma of the urinary bladder is related to the aberrations of chromosome.FISH is believed to be a very important method in diagnosis of urothelial carcinoma of the urinary bladder,which may have important clinical significance for the postoperative recurrence detection and prognosis.
8.The application of multi-slice CT angiography in the diagnosis and treatment of intracranial aneurysm

The development of the imaging technology enables diagnostic methods of detect intracranial aneurysms diversified. In recent years, the diagnosis of aneurysms was confirmed by digital subtraction angiography (DSA) which was regarded as the "gold standards". However, CT angiography (CTA), which appears as a fast, non-invasive, and easily achievable examination, is becoming as a new option in clinic. With the development of the CT facility and upgrade of the workstation software, CTA is more widely used in both diagnosing and treating intracranial aneurysm. This article reviews the sensitivity and specificity of CTA in the detection of intracranial aneurysms, and systematically analyzes its advantages and disadvantages as well as its development, imaging methods.
9.The detection of herpes simplex virus infection rate by jointly using chemiluminescence assay and PCR

Objective Detect the infection rate of herpes simplex virus (HSV ) by jointly using chemiluminescence assay and PCR ,and provides reference for clinical diagnosis .Methods The serum samples were collected from the pregnant women who had routine examination records in the hospital .Chemiluminescence assay was used to detect HSV IgM and IgG in those samples .Cervi‐cal secretions were collected from pregnant women with positive results and qualitatively tested for HSV DNA .Results The posi‐tive rate of HSV1 DNA was 0 .5% (7/1 422) ,the positive rate of HSV2 DNA was 1 .1% (16/1 422) .For pregnant women whose HSV IgM and IgG were both positive ,positive rate of HSV1 DNA was 0 .4% (4/1 008) and that of HSV2 DNA was 0 .6%(6/1 008);for those who only had HSV IgM positive ,the positive rate of HSV1 DNA was 0 .8% (1/130) ,and that of HSV2 DNA was 3 .1% (4/130);for those who only had HSV IgG positive ,the positive rate of HSV1 DNA was 0 .7% (2/284) ,that of HSV2 DNA was 2 .1% (6/284) .Among those three HSV antibody positive cases ,the difference in HSV1 DNA positive rate was not sta‐tistically significant(P>0 .05) ,while the difference in HSV2 DNA positive rate was statistically significant(P<0 .05) .Conclusion The test of HSV antibodies during pregnancy can be used as a routine test ,and HSV DNA test can be used as further test for those with HSV antibody positive ,which could improve the accuracy of diagnosis .Early screening ,detection ,and treatment are im‐portant for pregnant women with HSV infection .
10.Effects of ionizing radiation on expression of P21 protein in Jurkat cell line and p21 gene in thymocytes and splenocytes of mice

Objective To investigate the effects of ionizing radiation on the expression of P21 protein in Jurkat cell line and p21 gene in thymocytes and splenocytes of mice.Methods Flow cytometry (FCM)was used to analyze the expression of P21 protein in Jurkat cells at 12 and 24 h after irradiation to 0,0.5,1.0,2.0,4.0,and 6.0 Gy.Real-time PCR was used to detect the expression of p21 gene in thymocytes and splenocytes of mice at4 and 24 h after irradiation to 0,0.5,1.0,2.0,4.0,and 6.0 Gy.Multi-staining was used to analyze the micronucleus rates of Rct in bone marrow.Results The expressions of P21 protein were increased in a dose-dependent manner during 0.5-4.0 Gy(t=-24.23--3.96,P<0.05),but decreased at 6.0 Gy at 12 and 24 h post-irradiation(t=-11.19,-14.50,P<0.05).The expressions of p2 1 gene in both thymocytes and splenocytes of mice were increased in dose-dependent manner in the range of 0-6.0 Gy(including 6.0 Gy)(t=-29.96-8.80,P<0.05),and reached to the peak at 6.0 Gy at 4 and 24 h post-irradiation(t=-11.84--3.42,P<0.05),except thymocytes at 4 h and 1.0 Gy post-irradiation(t=-3.42,P>0.05).Conclusions The expressions of P21 protein and p21 gene could be increased by X-ray irradiation.which shows good dosedependent manners in certain range of dose.
