1.Expression of miR-106 a in fresh and paraffin-embedded tissues and in precancerous lesions of gastric cancer

Purpose To investigate the comparison of miR-106a in fresh and paraffin-embedded tissues and its expression in precan-cerous 1esions of gastric cancer. Methods Human gastric cancer tissues inc1uding 30 coup1es of fresh and 40 coup1es of paraffin-em-bedded samp1es were co11ected,quantitative rea1-time PCR was used to detect the expression of miR-106a in these two samp1es. Anoth-er paraffin-embedded samp1es inc1uding 20 cases of precancerous 1esions and 40 cases of gastric adenocarcinoma were a1so co11ected,in situ hybridization was used to assess the expression of miR-106a in these stages. Results The re1ative expression of miR-106a in fresh tissues was 3. 25 ± 1. 99,in paraffin-embedded tissues was 3. 18 ± 2. 14,indicating that the expression of miR-106a in these two sam-p1es has no statistica1 difference(P>0. 05),corre1ation ana1ysis showed that there was a significant corre1ation of miR-106a expression in these two samp1es(rs =0. 998,P<0. 001). The expression of miR-106a was detected in the stage of precancerous 1esions with the positive rate was 70%. The positive signa1s were 1ocated in dysp1astic epithe1ia1 ce11s and appeared as dark b1ue fine granu1es. The positive rate of miR-106a in gastric cancer tissues was 87. 5% and the frequency and extent increased and enhanced compared with pre-cancerous 1esions. Conclusion miR-106a has significant corre1ation in gastric cancer fresh tissues and paraffin-embedded tissues. Detection of miR-106a with paraffin tissues and its ear1y changes in prema1ignant 1esions can provide va1uab1e information for the ear1y diagnosis of gastric cancer.
2. Blood glucose lowering effect of ophiopogonis tuber extract and mechanism of anti-insulin-resistance

Objective To study the hypoglycemic effect and insulin sensitization mechanism of ophiopogonis tuber extracts on the 3T3-L1-induced adipocytes, and also in rats with reproduction of type 2 diabetes mellitus (T2DM). Methods 3T3-L1 cells were induced and differentiated into adipocytes. After the intervention with ophiopogonpolysaccharide (OPSR) and ophiopogonin (OPG), glucose consuming rate was detected for screening the extracts which may have effective hypoglycemic effects. The insulin resistance (IR) adipocyte model was established by dexamethasone induction, and then it was treated with OPSR. The protein expression levels of leptin, adiponectin and resistin were detected by Western blotting. The T2DM rat model was reproduced and then treated with OPSR for 4 weeks. Body weight (BW), triglyeride (TG), fasting blood glucose (FBG) and fasting insulin (FINs) of the rats were measured respectively. Results OPSR in dosage of 0.5-50mg/L promoted glucose consumption of adipocytes in a dose-dependent manner, the glucose consumption ratios were 32.27%, 75.14% and 90.47% respectively. OPG of 50mg/L showed very weak activity with glucose consumption ratio of only 8.49%. OPSR could significantly promote the protein expression of leptin and adiponectin, and showed an inhibitory effect on the protein expression of resistin (P<0.05). After treatment with OPSR for 4 weeks, the BW of rats increased obviously, while TG, FBG and HOMA-IR decreased significantly (P<0.05 or P<0.01). Conclusions OPSR may promote glucose transport and utilization of adipocytes, decrease the level of FBG and TG, and improve the condition of IR in T2DM rats. The mechanism of blood glucose lowering effect may be attributed to secretion of adipokines, such as leptin, adiponectin and resistin by IR adipocytes.
3.Hepatocyte growth factor gene C57488A polymorphism and essential hypertension

AIM: The impaired function of vascular endothelial cells occurs popularly in hypertensive patients. Hepatocyte growth factor (HGF) plays a crucial role on impaired function, and is related to prevalence, severity and target organ damages of hypertension. This study explored the relationship between HGF gene C57488A polymorphism and essential hypertension (EH). METHODS: ①Between June and December in 2006, totally 107 EH patients with an average age of (61?9) years, in them 47 cases with family history of hypertension, were recruited in this study. Informed consents were obtained from the patients who were all the Han residents of Hefei area, and the experiment was approved by the hospital ethical committee. The hypertension criteria formulated by Chinese Medical Association included the systolic pressure ≥140 mm Hg or diastolic pressure ≥ 90 mm Hg (1 mm Hg=0.133 kPa), or having received drug therapy for hypertension over one year. The patients were administrated with hypotensive drugs with one month. Meanwhile 110 normotensives without family history of hypertension and drug therapy were served as controls, with an average age of (60?8) years. Inclusion criteria: systolic pressure 0.05).②The distributions of HGF gene C57488A polymorphism were in agreement with Hardy-Weinberg equilibrium, indicating population representativeness. The frequencies of three genotypes CC, CA, AA were 0.809, 0.170, 0.021 and C, A alleles were 0.894, 0.106 in the EH group with family history respectively. Correspondingly, the frequencies were 0.733, 0.233, 0.033 and 0.850, 0.150 respectively in the EH patients without family history. Meanwhile, the frequencies were 0.527, 0.355, 0.060 and 0.705, 0.295 respectively in the controls.③The frequencies of genotype CC and allele C were both significant higher in EH with or without family history compared with the controls (P 0.05). CONCLUSION: HGF gene C57488A polymorphism is possibly associated with EH, and genotype CC and allele C may be the factors of genetic predisposition of EH in Han population of Hefei area.
4.Comparative analysis of Han and Uygur college students emmetropia biology measurement

AIM: To observe the differences in central cornea thickness, anterior chamber depth, corneal anterior curvature and corneal posterior curvature between the Uygur and the Han nationality college emmetropia students, and analyze the relationship of corneal thickness and corneal curvature.
METHODS: More than 500 students in grade one in Xinjiang Medical University were underwent screening, 56 emmetropia eyes in Han nationality students and 51 in Uygur students were selected. Sirius corneal topography was applied to the measurement.
RESULTS: Uygur and the Han nationality college emmetropia students central cornea thickness ( Uygur:0.53±0. 03mm, Han: 0. 54±0. 03mm), anterior chamber depth ( Uygur: 2. 91±0. 26mm, Han: 3. 14±0. 25mm), corneal posterior curvature K2 ( Uygur: -6. 30±0. 23D, Han: -6. 38±0. 28D) had significant difference (P<0. 05). Corneal anterior curvature ( K1, K2 ) before and corneal posterior curvature K1 there was no significant difference (P>0. 05). Han had no significant relationship in corneal thickness and corneal anterior curvature ( r<0 ) , or in corneal posterior curvature ( r < 1 ) . Uygur has no significant relationship in corneal thickness and corneal anterior curvature (r<0), or in corneal posterior curvature (r<1).
CONCLUSION: There are significant differences between the Uygur and the Han nationality college emmetropia students in cornea thickness, anterior chamber depth and corneal posterior curvature K2. There is no significant relationship between corneal thickness and corneal curvature in Uygur and the Han nationality college students.
5.Expression and significance of NF-κB and P-glycoprotein in cervical cancer tissue before and after neoadjuvant chemotherapy

Objective To investigate the relationship between the expression of NF-κB and Pglycoprotein(P-gp) in cervical cancer tissue before and after neoadjuvant chemotherapy(NACT). Methods The mRNA and the protein expression levels of NF-κB and P-gp were determined by Real time-PCR and immunohistochemical assay respectively in cancer specimens from 40 cevical cancer patients, which were collected before and after NACT. Results The mRNA expression levels of NF-κB were 50. 340 ± 0. 780 and 67. 595 ± 0. 914 before and after NACT respectively, which showed significant difference statistically ( t = -73. 384 ,P < 0. 01 ). We also found significant difference in the comparison between MDR1 mRNA expression before and after NACT(45. 800 ±0. 636 vs 62. 381 ±0. 743 ,t = -66. 582,P <0. 01 ). The positive rate of NF-κB protein expression before and after NACT were 50% ( 20/40 ), 80% ( 32/40 ) respectively, which showed significant difference statistically( χ2 = 7. 912, P = 0. 005 ). The positive rate of P-gp protein expression after NACT was significantly higher than that before NACT [ 40% ( 16/40 ) vs 70% ( 28/40 ), χ2 = 7. 273, P = 0. 007 ]The mRNA and protein expressions of NF-κB were positively correlated with those of P-gp before and after NACT( r = 0. 408,r = 0. 327 respectively, Ps < 0. 05 ). Conclusion NF-κB and P-gp may play an important role in multi-drug resistance of cervical carcinoma. Chemotherapy drugs in our NACT strategy may induce the activation of NF-κB and multi-drug resistance. Activation of NF-κB may correlate with multi-drug resistance mediated by P-gp.
6.Application of minimal invasive radical hysterectomy with laparoscope in uterine malignancy

Objective To investigate and analyze the clinical value of minimal invasive surgery with laparoscope for radical hysterectomy and pelvic lymphadenectomy for the treatment of cervical and endometrial carcinomas in early stage. Methods A retrospective analysis was carried out based on the clinical data of 64 patients with FIGO stage Ⅰ-Ⅱ malignant uterine tumor, among them 34 were operated with the aid of laparoscope and 30 were operated with laparotomy for radical hysterectomy and/or pelvic lymphadenectomy. Comparison was done between the two groups on operative time, blood loss, complications and postoperative recovery. Results There was no significant difference in the incidence of complications and operation time between laparoscopic radical hysterectomy and pelvic lymphadenectomy for the treatment of the patients diagnosed with early stage of cervical and endometrial carcinomas compared with those with traditional radical hysterectomy and pelvic lymphadenectomy through laparatomy. Objective To study the antitumour, immunomodulating and enterobacteria regulating effects of exopolysaccharides (EPS). Method The suppressive rate of growth of sarcoma 180, the index of immunity, and the intestinal microflora were determined. Results (1) EPS inhibited the growth of sarcoma 180 by oral for BALB/c mice significantly. The suppressive rate at the dose of 120mg/kg bw?d was 42.7%. (2) At the dose of 120mg/kg bw?d the index of thymus and IL-2 in serum increased significantly. There was increasing trend in respect of TNF-? and killing activation of NK cell. (3) The amounts of Lactobacillus in groups treated with EPS increased significantly, and the amounts of Enterobacteriacea and Enterococcus droped significantly. Conclusion EPS had the antitumour effect in vivo, and could modulate the immune function, improve the microbial balance in the intestinal tract of tumour bearing mice to some extent. So the antitumour mechanism was probably correlated with the immunomodulating effect.

OBJECTIVE:To evaluate the relative bioavailability of two products(CLMC 1 ,CLMC 2 )of Clarithromycin in man.METHODS:A single oral500mg dose of CLMC 1 or CLMC 2 was given to20healthy male volunteers in an open randomized crossover study.The plasma concentrations of CLMC 1 and CLMC 2 were measured by microbial assay.The pharmacokinetic parameters were calculated with3p97pharmacokinetic program and the bioequivalence was evaluated.RESULTS:The con?centration-time curves of CLMC 1 and CLMC 2 fitted to a two-compartent open model.C max were(2.23?0.83)?g/ml and(2.14?0.70)?g/ml;T max were(1.95?0.39)h,(1.78?0.41)h;AUC (0~T) were(9.50?2.52)(?g?h)/ml,(9.35?2.54)(?g?h)/ml respectively.The relative bioavailability of CLMC 1 was(101.60?9.35)%.CONCLUSION:The results of two and one-side t tests suggest that the CLMC 1 is bioequivalent with the CLMC 2 .
