1.Simultaneous Determination of 5 Components in Xiaoer Qingyan Granules by HPLC

OBJECTIVE:To develop a method for simultaneous determination of harpagide,harpagoside,chlorogenic acid, caffeic acid and phillyrin in Xiaoer qingyan granules. METHODS:HPLC method was adopted. The determination was performed on Zorbax SB-C18 column with mobile phase consisted of acetonitrile-0.1% phosphoric acid solution (gradient elution) at a flow rate of 1.0 mL/min. The detection wavelength was set at 210 nm (0-13 min,harpagide),327 nm (13-25 min,chlorogenic acid and caffeic acid),277 nm(25-29 min,phillyrin),210 nm(29-40 min,harpagosid);the column temperature was 30℃,and sam-ple size was 10 μL. RESULTS:The linear ranges of harpagide,harpagosid,chlorogenic acid,caffeic acid and phillyrin were 8.400-168.0 ng(r=0.9996),11.30-226.0 ng(r=0.9998),128.8-257.6 ng(r=0.9993),8.110-162.2 ng(r=0.9996),29.69-593.8 ng(r=0.9994),respectively. LOQs of the 5 components were 33.39,451.2,515.2,324.5,1188 ng/mL;LODs were 8.348, 112.8,128.8,81.12,297.0 ng/mL,respectively;RSDs of precision,stability and reproducibility tests were all lower than 2.0%. The recoveries were 96.39%-98.64%(RSD=0.83%,n=6),96.60%-98.89%(RSD=0.89%,n=6),96.28%-99.22%(RSD=1.25%,n=6),96.49%-99.54%(RSD=1.16%,n=6),96.26%-99.70%(RSD=1.30%,n=6),respectively. CONCLUSIONS:The method is simple,accurate and suitable for simultaneous determination of 5 components in Xiaoer qingyan granules
2.A quantitive detection of serum Cyfra21-1 in patients with oral squamous cell carcinoma

Objective: To examine the level of serum Cyfra21-1 in patients with oral squamous cell carcinoma(OSCC) and to explore the potential application of serum Cyfra21-1 detection in the diagnosis of OSCC. Methods: The serum Cyfra21-1 concentrations were detected by ELISA. Results: The preoperative serum Cyfra21-1 concentration in patients with OSCC was (1.12±0.61) ng/ml, which was significantly higher than that in healthy persons (0.34±0.20) ng/ml and that in patients with oral benign tumor (0.50±0.27) ng/ml. The sensitivity rate and specificity rate of the test were 83.9% and 89.3%, respectively. For 10 patients with OSCC, the serum Cyfra21-1 concentrations were decreased from (1.31±0.25) ng/ml preoperatively to (0.65±0.14) ng/ml postoperatively. Conclusion: Cyfra21-1 may be a good marker in diagnosis of OSCC.
3.Polymorphisms of elastin and collagen type Ⅰ α2 genes and intracranial aneurysm rupture

Intracranial aneurysms are mainly caused by the local arterial wal defects and the increased intraluminal pressure. Usualy, the unruptured smal aneurysms are asymptomatic, and the ruptured aneurysms can cause subarachnoid hemorrhage. So far, the etiology and pathogenesis of intracranial aneurysms are not fuly understood. A lot of evidence has showed that intracranial aneurysms are a complex disease of environmental factors and multi-gene interaction. This article reviews the correlation between polymorphisms of elastin and colagen type Ⅰ α2 genes and intracranial aneurysm.
4.Comparison between ThinPrep cytologic test and colposcope pathological biopsy in screening of cervical cancer and premalignant lesion

Objective To study the application values of ThinPrep cytologic test (TCT) and colposcope pathological biopsy in screening of cervical cancer and premalignant lesion.Methods Two hundred and forty-two female patients with TCT results abnormal were selected as the research object,and they were examined in colposcope pathological biopsy.The diagnostic accordance between TCT and colposcope pathological biopsy was analyzed,and the result of colposcope pathological biopsy in cervical different parts was compared.Results In TCT,the ratio of cervical intra-epithelial neoplasia (CIN) in atypical squamous cell of undetermined significance was 56.52% (52/92),that in low-grade squamous intraepithelial lesion was 88.16%(67/76) and in high-grade squamous intraepithelial lesion was 100.00%(67/67),the test results of two methods had consistency by the Pearson rank correlation test (Pearson contingency coefficient =0.7588,x2 =328.37,P < 0.05).In colposcope pathological biopsy,the positive rate of lesions center was 90.22% (83/92),that of lesions edge was 73.17% (60/82) and that of 3,6,9,12 point was 71.74% (66/92),there were significant differences among the positive rate of the three parts by the unidirectional ordered categorical data rank and inspection (Kruskal-Wallis method)(P < 0.05),the positive rate of lesions center was higher than that of lesions edge and 3,6,9,12 point (P< 0.05),there was no significant difference in positive rate between lesions edge and with 3,6,9,12 point (P >0.05).Condusion The result of TCT and colposcope pathological biopsy is consistent,and the positive rate of lesions center of colposcope pathological biopsy is higher,which is higher than that of lesions edge and 3,6,9,12 point.
5.STUDIES OF APOPTOSIS INDUCED IN MALIGNANT LYMPHOMA CELL LINES BY ARSENIC TRIOXIDE

Objective To investigate the effects of a wide range of concentrations of As2O3 malignant lymphoma cell lines. Methods Cell apoptosis situation ,cellular DNA contents and bcl-2 protein expression were determined by cell stain, TdT-mediated dUTP Nick-end Labeling (TUNEL) and flow cytometry. Results 0.5 μmol/L~2.0 μmol/L As2O3 could inhibit Raji cell growth accompanied by the appearance of morphologic characteristics of apoptosis. Number of sub-G1 cells significantly increased and bcl-2 protein expression in Raji cells was significantly down-regulated assayed by flow cytometry. The relationship between bcl-2 protein and the concentrations of As2O3 showed a dose-and time-dependent effect; so did the number of sub-G1 cells. However, there was no effect of 0.5 μmol/L~4.0 μmol/L As2O3 on Jurkat cell.Conclusion It is found that As2O3 could significantly inhibit the growth of B lymphoma cell lines and induce their apoptosis.
6.Relationship between P2 purinoceptor subtypes and coronary heart disease

In this article we review the relation between P2 purinoceptor subtypes and coronary heart disease (CHD).P2Y_1 ,P2Y_2 ,P2Y_(11) and P2X_7 receptors are related to inflammatory reaction in atheroselerosis plaque.P2X_3 receptor plays an important role in anginal nocuity response.P2X_4 receptor is involved in vascular remodeling and enhancement of myocardial contractility in non-infarct zone.P2Y2 and P2Y6 receptors are involved in resteno-sis.A polymorphism in P2Y_(11) receptors increase the risk of myocardial infarction.P2Y_1、P2Y_(12) and P2X_1 receptors play a role in thrombotic formation.The specific agonist and antagonist of P2 purinoceptor subtypes could be poten-tial targets for new drugs to treat CHD.
7.A meta -analysis of thymosin-?_1 monotherapy for chronic hepatitis B virus infection.

Objective To investigate the efficacy of thymosin alpha-1(T?_1,Zadaxin)monotherapy for chronic HBV infection by a meta-analysis of the published data. Methods Randomized controlled trials on thymosin alpha-1 monotherapy in chronic HBV infection were searched from electronic databases such as MEDLINE、PUBMED and Blackwell. Results Meta-analysis of 7 randomized controlled studies investigating the safety and efficacy of thymosin alpha-1 monotherapy for the treatment of chronic hepatitis B showed that 6 months treatment with thymosin alpha-1(1.6 mg twice weekly)almost tripled the sustained response rate(38%)compared with controls(13%).Conclusion These results suggest that a 6-month course of thymosin alpha-1 therapy is effective and safe in patients with chronic hepatitis B;thymosin alpha-1 can effectively reduce HBV replication in CHB with patients. Compared with IFN-?,thymosin alpha-1 which has less side effects is better tolerated and seems to induce a gradual and more sustained normalization of ALT and loss of HBV DNA.
8.Simultaneous Determination of 4 Components in Fufang Kushen Shuiyangsuan Powder by RP-Ion Pair HPLC with Wavelength Switching

Objective: To develop a RP-ion pair HPLC with wavelength switching for the simultaneous determination of 4 components (marine, oxymarine, salicylic acid, benzoic acid) in Fufang Kusheng Shuiyangsuan powder.Methods: An Agilent ZORBAX SB-C18 column(150 mm× 4.6 mm, 5 μm) was used;the mobile phase was acetonitrile (A)-0.1% phosphoric acid solution (0.2 g sodium heptanesulfonate was added to 100 ml solution) at a flow rate of 1.0 ml·min-1;the detection wavelength was 220 nm in 0-12 min and 280 nm in 12-25 min;the column temperature was 30℃.Results: The linear range of marine, oxymarine, salicylic acid and benzoic acid was 0.006 030-0.120 6 μg (r=0.999 4), 0.016 56-0.331 2 μg (r=0.999 9), 0.717 1-14.34 μg (r=0.999 9) and 0.512 0-10.24 μg (r=0.999 9), respectively;the average recovery was 98.14%, 97.20%, 97.05% and 98.39% with the RSDs of 1.38%, 0.32%, 0.81% and 1.26%(n=6) , respectively.Conclusion: The method is simple and rapid, and can be applied in the simultaneous determination of 4 components in Fufang Kusheng Shuiyangsuan powder.
9.Progress of the immuno modulating effect of polysaccharides and their mechanisms

Polysaccharides definitely possess the effect of modulating immuno function by many ways and many targets, such as promoting the proliferation and differentiation of lymphocytes, secreting various lymphokines, modulating the balance of neuroendocrine immuno modulation network and so on. At present, the tendency of study on drugs in the world is to from synthetic drugs to natural drugs. In this review, a brief summarization is made on progress of the immuno modulating effect of polysaccharides and their mechanisms.
