1.Determination of Ritodrine in Human Plasma by LC-MS

OBJECTIVE:To establish an LC-MS method for the determination of the ritodrine content in human plasma.METHODS:The determination was performed on Agilent Eclipse Plus C18 column with the column temperature kept at 40 ℃.The mobile phase consisted of methanol-water(95∶5,containing 0.02 mol?L-1 ammonium formate)with a flow rate of 0.5 mL?min-1.The extraction agent was acetic ether-dichloromethane(4∶1).A triple quad mass spectrometer system equipped with an electro spray ionization ion-trap source was used as the detector and operated in the positive ion model.Selected reaction monitoring(SRM)was employed to detect ritodrine(m/z 288.2→121.1)and the internal standard dobutamine(m/z 302.1→107.0).RESULTS:The linear range of ritodrine was 0.25~100 ng?mL-1(r=0.999 1),with the lower limit of quantification at 0.25 ng?mL-1.Both the intra-day RSD and inter-day RSD were less than 15%.The methodological recovery was 98.00%~103.44%.CONCLUSION:The method is sensitive,accurate,reproducible,specific and suitable for the pharmaceutical study of ritodrine.
2.Intraoperative reduction and internal fixation for atlanto-axial dislocation

Atlanto-axial complex is the most important functional unit of the cervical spine.Atlanto-axial instability or dislocation,one of the most serious pathologies in spinal surgery,is often managed by posterior atlantoaxial fixation and fusion.Several posterior fixation techniques have been used to stabilize the atlanto-axial complex,including wire/cable-bone fixation,Halifax clamps,transarticular screw fixation,screw-plate fixation and screw-rod fixation.For atlanto-axial dislocation that cannot be reduced with skull traction before operation,intraoperative reduction is often needed.Current intraoperative reduction techniques can be classified into transoropharyngeal surgical release followed by posterior reduction and fixation,transoropharyngeal reduction and fixation,and posterior reduction and fixation using screw-rod system according to approaches.Furthermore,posterior reduction can be performed using elevating-pull,leverages,and cantilever techniques.This study reviews the intraoperative reduction and fixation techniques mentioned above.
4.ESTIMATION OF POSTMORTEM INTERVAL BASED ON COMPLEMENT 3(C3) CLEAVAGE AND RECTAL TEMPERATURE

The C3 cleavages of human blood kept at different temperatures in vitro were studied. In 14 autopsy cases, the postmortem intervals(PMI) were estimated based on the C3 cleavage rates and the rectal temperatures. The 95% confidence of predicting PMI was?11 hours. The phenotyping of C3 could be achieved while the C3 cleavage rate was measured. It is suggested that it is useful for the personal identification of unknown corpses.
5. Determination of 10 phenolic compounds in extract from Bidens bipinnata by HPCE

Objective: To develop a novel method using high-performance capillary electrophoresis coupled with DAD (HPCE-DAD) for the simultaneous determination of 10 phenolic compounds (luteolin-7-O-glucoside, rutin, quercetin-3-glucoside, quercetin-3- rhamnoside, salicylic acid, luteolin, kaemferol, quercetin, gallic acid, and protocatechuic acid) in Bidens bipinnata. The extraction efficiency of above mentioned compounds was compared using six different ethanol/water solutions. Methods: The effects of pH value and concentration of buffer, applied voltage, and temperature on the separation were researched. The 10 compounds were baseline separated within 16 min in a 60 cm length capillary at a separation voltage of 25 kV with a running buffer consisting of 25 mmol/L borate (pH 9.5) at wavelength of 214 nm. Results: Excellent linearities of luteolin-7-O-glucoside, rutin, quercetin-3-glucoside, quercetin-3-rhamnoside, salicylic acid, luteolin, kaemferol, quercetin, gallic acid, and protocatechuic acid were observed at 5.0-120, 5.0-120, 5.0-120, 5.0-120, 1.0-120, 2.5-120, 5.0-120, 2.5-120, 2.5-120, and 2.5-120 mg/L, respectively. The detection limits were at 2.5, 2.5, 2.5, 2.5, 0.5, 1.0, 2.5, 1.0, 1.0, and 1.0 mg/L, respectively. The relative standard deviations (RSD) of precision were below 5.17% (n = 6). The mean recoveries for 10 phenolic compounds in B. bipinnata ranged from 94.4% to 105.8%, with RSD values of 0.32%-4.33%. Conclusion: The contents of luteolin-7-O-glucoside, rutin, quercetin-3- glucoside, quercetin-3-rhamnoside, salicylic acid, luteolin, kaemferol, quercetin, gallic acid, and protocatechuic acid in B. bipinnata are 95.04-105.84, 78.03-110.45, 96.45-177.96, 178.78-300.00, 62.06-66.54, 71.08-72.85, 77.39-95.30, 68.27-77.43, 68.47- 88.47, and 107.24-142.43 μg/g, respectively.

The proliferation of splenic lymphocytes, the humoral immune response to sheep red blood cells and the phagocytotic function of intraperitoneal macrophages of mice were all stimulated by ig the flesh of Meretrix meretrixL. (FM) 20 g/kg and 10 g/kg for 7 days. FM 20 g/kg and 10 g/kg ig for 15 days also enhanced the activity of serum T-SOD, and decreased the level of serum MDA in old mice. The anti -stress effect of FM, such as increasing the swimming time of mice in 20癈 water by ig FM 20 g/kg and 10 g/kg for 7 days, was also achieved.

Objective To observe the distribution of protein kinase C? subunit(PKC?) in the rat nervous system. Methods Immunocytochemical staining for PKC? by using specific antibody against PKC? was performed. Results Intensely and densely stained PKC? immunoreactive neurons were mainly observed in the cerebral cortex,hippocampus,amygdaloid complex,cerebellar cortex, ventral and dorsal cochlear nuclei, caudal spinal trigeminal nucleus and the superficial layers of the spinal dorsal horn.Conclusion\ PKC?\|containing neurons are widely located in the rat nervous system.These results provide morphological evidence for the functional roles of PKC? in the signaling transduction of the nervous system.\;[
10.Influence of Heating Method on the Quality of Extracts from Radix Salviae Miltiorrhizae

Objective To observe the influence of heating method (or temperature control strategy) on the concentration of main active components in extracts from Radix Salviae Miltiorrhizae. Methods Common chromatographic peak area and the contents of Salvianolic acid B in three kinds of extracts obtained by 3 kinds of heating methods were compared. Results With the content of extracts extracted by the first heating method as the baseline, content of salvianolic acid B in extracts extracted by the second heating method was decreased 21.73 percent and that by the third method decreased 55.44 percent. Conclusion Quality of Salvianolic acid extracts can be affected greatly by temperature range and temperature control strategy. It is important to strictly control the heating method for the exaction of heat- sensitive active compounds from Chinese herbs.
