1.Effects of Semen Litchi on the Expressions of S180 and EAC Tumor Cells and Bax and Bcl-2 Proteins in Rats

OBJECTIVE: To study the effects of Semen Litchi on the expressions of S180 and EAC tumor cells and Bax and Bcl-2 proteins in rats. METHODS: EAC, S180 and hepatic carcinoma tumor-bearing mice models were established in mice by inoculating them with tumor strains. EAC, S180 and hepatic carcinoma suspensions were diluted respectively with normal saline in a ratio of 1∶1 for the preparation of tumor-bearing ascites suspension. Each rat was subcutaneously inoculated with the mixture 0.2 mL via axilla 24 hours before medication, with the whole sterile operation finished within 60 minutes. After treated with Semen Litchi for 10 consecutive days, the growth of EAC, S180 and hepatic carcinoma in mice was observed. RESULTS: The growth of EAC and S180 and hepatic carcinoma cells in mice was significantly inhibited by Semen Litchi. CONCLUSION: Semen Litchi can inhibit the growth of S180 and EAC tumor cells in vitro and in vivo, and its antitumor effect is related to its ability to promote apoptotic of tumor cells.
2.Effects of Extracts from the Seed of Litchi on the Expression of ER and PR in Rat Mammary Tissue and Apoptosis of Mammary Glandular Cell

OBJECTIVE: To study the effects of extracts of the seed of Litchi (ESL) on the expression of estradiol receptor (ER) and progestogen receptor (PR) in mammary tissue and apoptosis of mammary glandular cells in rats with hyperplasia of mammary glands (HMG). METHODS: The HMG model was induced by intramuscularly injecting with 0.5 mg?kg-1 estradiol benzoate for 25 days, subsequently intramuscularly injecting with 4 mg?kg-1 progesterone for 5 days. 60 healthy nonfertile SD rats were divided into normal group, model group, rujieping group and high-dose, medium-dose and low-dose ESL groups, treating with distilled water, distilled water, 3.3 g?kg-1 Rujieping and 44, 22, 11 g?kg-1 ESL via i.g. gtt once a day for 30 days, respectively. Immunohistochemical method was applied to observe the expression of ER and PR in rat mammary tissue and apoptosis of mammary glandular cells. Uterus index was also calculated. 5.Determination of Ritodrine in Human Plasma by LC-MS

OBJECTIVE:To establish an LC-MS method for the determination of the ritodrine content in human plasma.METHODS:The determination was performed on Agilent Eclipse Plus C18 column with the column temperature kept at 40 ℃.The mobile phase consisted of methanol-water(95∶5,containing 0.02 mol?L-1 ammonium formate)with a flow rate of 0.5 mL?min-1.The extraction agent was acetic ether-dichloromethane(4∶1).A triple quad mass spectrometer system equipped with an electro spray ionization ion-trap source was used as the detector and operated in the positive ion model.Selected reaction monitoring(SRM)was employed to detect ritodrine(m/z 288.2→121.1)and the internal standard dobutamine(m/z 302.1→107.0).RESULTS:The linear range of ritodrine was 0.25~100 ng?mL-1(r=0.999 1),with the lower limit of quantification at 0.25 ng?mL-1.Both the intra-day RSD and inter-day RSD were less than 15%.The methodological recovery was 98.00%~103.44%.CONCLUSION:The method is sensitive,accurate,reproducible,specific and suitable for the pharmaceutical study of ritodrine.

7.Clinical analysis of fungal rhino-sinusitis in 189 cases.

OBJECTIVE:
To investigate the clinical features of fungal rhino-sinusitis.
METHOD:
Clinical data of 189 patients suffering from fungal rhino-sinusitis treated by functional endoscopic sinus surgery was analyzed retrospectively. The analyzed data included clinical type, age of onset, predilectionsite, reason, and surgical outcome.
RESULT:
Among the 189 patients with fungal rhino-sinusitis,181 cases were fungal ball,6 cases were allergic fungal rhino-sinusitis, 1 case was acute invasive fungnal rhino-sinusitis, 1 case was chronic invasive fungnal rhino-sinusitis. One hundred and twenty-eight cases were in the maxillary sinus (123 cases were unilateral, 5 cases were bilateral). Nineteen cases were in the ethmoid sinus. 31 cases were in the sphenoid sinus. Two cases were both in the maxillary sinus and ethmoid sinus, 1 case was both in the maxillary sinus and sphenoid sinus. Two cases invasive fungnal rhino-sinusitis had diabetes history. All the patients treated by functional endoscopic sinus surgery, 184 cases without recurrence, 5 cases suffered re-operation.
CONCLUSION
The incidence of fungal rhinosinusitis is showing a rising trend, fungal ball is the highest. The sinusitis patients whom we highly doubt for fungal infection should be confirmed by using sinonasal secretion smear, cultivation and histopathological examination. Surgical treatment should completely remove the fungal masses, to avoid reoperation.

8.Separation and Preparation of Ginkgolic Acid ?_1 by RP-HPLC

Objective:To develope a method for the separation and preparation ginkgolic acid ? 1. Methods: Ginkgolic acids were extracted by alcohol, and adsorbed by silica gel to remove most of the polar impurities. It was preparated with RP HPLC. Results: The extraction ratio of ginkgolic acid ? 1 by this extraction process was above 84%, and the purity of ginkgolic acid ? 1 reached 98%. Conclusion: This procedure is simple and relatively effective.
9.Additive effects of the variants in the peroxisome proliferator activated receptor γ2 and uncoupling protein 2 genes on type 2 diabetes

Objective To investigate the additive effects of uncoupling protein 2(UCP2) gene 3′-untranslated region(3′-UTR) 45-base pair insertion/deletion( I/D) variation and peroxisome proliferator activated receptor( PPAR)γ2 gene Pro12Ala variation on type 2 diabetes(T2DM) in Chinese Han popula-tion.Methods The UCP2 gene 3′-UTR I/D variation and PPARγ2 gene Pro12Ala variation were exam-ined by polymerase chain reaction-restriction fragment length polymorphism ( PCR-RFLP) in 490 type 2 dia-betes subjects and in 585 control subjects .The additive effects of the two gene mutations were analyzed . Results ⑴The frequency of PPARγ2 gene Pro12Ala variation in type 2 diabetes was not significantly dif-ferent from that in control subjects.(χ2 =0.058, P =0.809).In T2DM group, A12 allele carriers had larger waist circumference than Pro12Pro genotype carriers;Glucose stimulated insulin secretion by oral glu-cose tolerance test (OGTT) was significantly lower in carriers of the Ala12Ala or Pro12Ala genotype com-pared with Pro12Pro genotype.( Z =2.222, P =0.026; Z =2.051, P =0.040; Z =2.079, P =0.038 ) .⑵There wasn't significant difference among 3 genotypes with 3′-UTR I/D variation in UCP2 gene (χ2 =2.311 , P =0.315 ) .In control group , Glucose stimulated insulin secretion by oral glucose tolerance test ( OGTT) was significantly higher in carriers of the I/D or I/I genotype compared with D/D genotype ( Z =1.997 , P =0.046 ) .⑶The genotype carriers with Pro/Ala +Del/Del were the greatest relation to T2DM (OR=1.22, 95%CI 1.078~1.386).Conclusion Though the UCP2 gene mutation alone or the PPARγ2 gene mutation alone is not associated with T 2DM, the possible additive effects of the two micro genes increase the occurring of T 2DM.
10.The inhibitory effect of endostatin on human gastric carcinoma cells in an athymic mouse model and the impact on expression levels of Bcl-2 and Bax

Objective To investigate the inhibitory effect and the mechanism of endostatin on human gastric carcinoma model in nude mice. Methods Athymic mice were injected by in vitro-cultured SGC-7901 human carcinoma cell strain to observe the inhibitory effect of endostatin. Real time fluorescent quantitative PCR and immunohistochemistry stain were used to quantify the expression levels of Bcl-2 and Bax genes both on mRNA and protein levels. Results The average tumor volume in endostatin group was statistically smaller than control group (P<0.05). The inhibitory rate on the 7~(th), 10~(th), 13~(th), 16~(th) and 19th day was 26.2%, 47.7%, 38.9%, 45.8% and 51.7%, respectively. The results of real time fluorescent quantitative PCR demonstrated that mRNA expression of Bcl-2 in endostatin group was obviously lower than it in control group (P<0.05), however, the expression level of Bax had no statistical difference between these two groups (P>0.05). The results of immonohistochemistry were consistent with PCR. Conclusions Endostatin can inhibit the proliferation of SGC-7901 human gastric carcinoma in athymic mouse model, and the mechanism involves the suppression of Bcl-2 expression.
