1.Determination of Cobalt by Spectrophotometry Using a New Reagent, 1-(6-bromo-2-benzothiazolyl)-3-(5-bromo-8-quinolyl)-triazene

Objective To synthesize a new reagent, 1-(6-bromo-2-benzothiazolyl)-3-(5-bromo-8-quinolyl)-triazene (BBTBQT) and apply to the determination of cobalt. Methods The reagent had been synthesized by diazotization and coupling reaction. After purification and characterization, BBTBQT was tested for its color reaction conditions with cobalt in the presence of cetylpyridinium bromide (CPB). Results In the presence of borax buffer solution at pH value of 9.0, the reagent reacted with cobalt to form a blue stable complex with a molar ratio of 2∶1; the complex had a maximum absorption at 640 nm. The apparent molar absorptivity of the complex is 1.55 ?105 L/(mol?cm). Beer’s law was obeyed in the range of 0-0.28 mg/L for cobalt. The method had been applied to determine cobalt in drainage sediment and vitamin B12 injection solution with a mean relative error of 1.9%-5.8% and a standard relative deviation of 1.7%-3.8%. Conclusion The present method is selective, sensitive, accurate, convenient and suitable for determination of trace cobalt in the samples.
2.The effect of Vitamin C on the growth of cervical carcinoma HeLa cells in vivo

Aim To study the effect of Vitamin C on growth and apoptosis of human cervical cancer cell line HeLa and its mechanisms. Methods HeLa cell line was treated with VitC. Cell proliferation was evaluated with MTT assay. The distribution of cell cycle,the rate of apoptosis and the expression of HPV18 E6,p53,Bax and Bcl-2 protein of the heLa cells, were observed using flow cytometer respectively. TRAP-PCR-ELISA assay was used to evaluate telomerase activity. Results Vitamin C arrested the HeLa cell line in G_0/G_1 phase in a dose and time-dependent manner, and significantly inhibited HeLa cells growth and increased the rate of apoptosis.Expression of HPV18 E6 and Bcl-2 proteins, was down regulated expression of p53 and Bax proteins was upregulated, and the telomerase activity was reduced. Conclusion Vitamin C can significantly inhibit HeLa cells growth and proliferation,arrest them in G_0/G_1 phase and induce their apoptosis.The reason of these changes is the decrease of E6 gene′s expression, and the succedent changes of proteins′ expression in cancer cells,including the downregulated expression of HPV18 E6 and Bcl-2 proteins, upregulated expression of p53 and Bax proteins and reduced telomerae activity.
3.Effects of taurine on the change of apoptosis induced by IL-1?, TNF-? and IFN-? in rat pancreatic islet cells

AIM: To investigate the changes of apoptosis in isolated pancreatic islet cells, insulin secretion, expression of Bcl-xL and Bax induced by combination of IL-1?, TNF-? and IFN-?, and effects of taurine on them. METHODS: Isolated pancreatic islet cells from Wistar rat were incubated in monolayer in vitro. NO-2/ NO-3 production, NOS activity, insulin secretion, the protein expression of Bcl-xL and Bax, percentage of islet cell apoptosis and DNA fragmentation in pancreatic islet cells incubated with combination of IL-1?, TNF-? and IFN-? were measured, and the effects of taurine on the changes of them were further investigated. 4.Proteomic study on effect of tangcao pill on microsome CYP450.

Tangcao pill is commonly applied in adjuvant and even alternative therapy for patients with AIDS. However, the herb contains complex ingredients, but with unknown effect against anti-HIV drug and unknown function. Because CYP450 emzyme is the main metabolic enzymes of the drug, it is of important significance to study the regulation of CYP450 enzymes before and after the combined administration of Tangcao pill and EFV. Proteomics, due to its high throughout and high sensitivity, has been widely applied in CYP450 enzyme study. In this paper, liver microsomes were separated through differential centrifugation. Their proteins were separated through SDS-PAGE. The three protein bands that CYP450 enzymes were located were cut and identified by liquid chromatography tandem mass spectrometry. Totally 16 CYP450 isoenzymes were identified. Furthermore, in order to make a quantitative analysis on the effect of tang herb on CYP450 emzyme, the multiple reaction monitoring (MRM) technology based on MS was adopted. The CYP2C11 was selected based on the results of the mass spectrum identification of proteins. The characteristic polypeptides were obtained through searching Expasy blast database. The m/z of the fragment ions was less than 800. In the paper, the m/z of ion pairs of CYP2C11 were 711.5/232.1, 711.5/319.2, 711.5/466.2 and 711.5/595.3, and the m/z of ESAT-6 (internal standard, IS) were 735.5/215.3, 735.5/389.3, 735.5/460.3 and 735.5/524.3. The relative peak (analyte/IS) area was adopted for the relative quantitative analysis. Compared with the EFV single administration group, the EFV and Tangcao pill combined administration group showed a 1.6-fold increase in CYP2C11. The results of the paper indicated that Tangcao pill may affect drug metabolism by regulating metabolic enzymes such as CYP2C11, but the specific mechanism still unknown.

6.Characteristics of nobiletin-loaded nanoemulsion and its in vivo distribution in mice.

The purpose of this study was to prepare the nobiletin-loaded nanoemulsions (NOB-NE) and study its in vivo distribution in mice. The characteristics and stability of the unloaded and drug-loaded nanoemulsions were investigated. The size, apparent viscosity and pH value of NOB-NE were respectively (15.5 +/- 2.9) nm, (3.10 +/- 0.33) mPa x s and 6.56 +/- 0.05, which were all higher than those of unloaded nanoemulsions. The zeta potential of unloaded and drug-loaded nanoemulsions carried negative charge. The NOB-NE after diluted by 5% glucose solution was stable in 8 h, and there was no significant difference in the size, content and diluted stability of its preconcentrate in long-term storage. The concentration of nobiletin in plasma and tissues was determined by HPLC after intravenous administration of NOB-NE. Based on AUC(0-t), MRT and C(max), the nanoemulsions delivered more nobiletin into the brain and kidney compared to those of nobiletin solution. The brain and kidney targeting efficiency was improved. In addition, the results fitting using SAAM II software show that the higher drug concentration of the NOB-NE in the brain might be owed to the quicker transport rate from the blood to the brain, and that in the kidney relate to the probable accumulation effect. These results indicate that the in vivo distribution of NOB-NE with consistent quality in mice could be changed and its brain and kidney targeting absorption capability was enhanced comparing with nobiletin solution.

7.Microwave ablation for solitary hepatocellular carcinoma sized less than 5 cm: analysis of prognosis and its influence factors

Objective To evaluate the curative effect of microwave ablation (MWA) for solitary hepatocellular carcinoma (HCC) sized less than 5 cm,and to discuss the prognosis and its influence factors.Methods The clinical data of 603 HCC patients,who were admitted to authors' hospital during the period from 2011 to 2013 to receive MWA,were retrospectively analyzed.The patients were followed up till March 2016.Results According to the inclusion criteria,a total of 603 patients were enrolled in this study.Complete ablation of HCC lesions was obtained in 5.8% of patients (35/603).No statistically significant difference in complete ablation rate existed between each other among groups of different tumor diameter (P=0.056).The incidence of severe complications was 1.7% (10/603).The median survival time was (59.6±1.9)months,while the 1-,2-and 3-year overall survival rates were 94.7%,81.9% and 71.8% respectively.In terms of overall survival rate,no statistically significant difference existed between groups of different age as well as between groups of different tumor diameter (P=0.225 and P=0.777 respectively),but statistically significant difference existed between groups of different sex as well as between groups of different recurrence interval (P=0.029 and P<0.001 respectively).Univariate and multivariate analysis showed that gender,preoperative albumin level and recurrence interval were the factors influencing overall survival rate (P=0.035,P=0.006 and P<0.001 respectively).Conclusion For the treatment of solitary HCC sized less than 5 cm,MWA has reliable curative effect,it is a safe and minimally-invasive therapy for HCC.The overall survival rate between patients with different age or between patients with different HCC size is not obviously different.The gender,preoperative albumin level and recurrence interval are independent factors that affect the overall survival rate.

Objective To study the impacts of variation of folate metabolic component including folic acid (FA), riboflavin (RF) and MTHFR C677T polymorphism on chromosome 8 and 17 segregation. Method RPMI1640 medium was modified with different combinations of low (LF) and high (HF) concentrations of folic acid (20, 200 nmol/L) and low (LR) and high (HR) concentrations of riboflavin (1 and 500 nmol/L). Objective To establish standardized traceable management procedure for implanted high-value consumables in operating room.Methods The management model combining information-based system operation process and quality control process was designed,and management results before and after implementation were compared.Results There were statistically significant differences in error rates of information recording,bar code sticking and charging of implantable high-value consumables after the implementation of the process management mode (P<0.05).At the same time,there were statistically significant differences in improvement of traceability of high-value consumables,adverse event reporting and patient satisfaction(P<0.05).Conclusion Establishment of management model in operating room for implanted high-value consumables can ensure medical safety and increase medical quality.It was proved to improve the level of hospital management.
10.Chemical constituents of Ligularia duciformis.

Ligularia duciformis is a plant of the Ligularia genus (Compositae) which has been reported to have therapeutic effects of treating diseases such as bronchitis and coughing. In order to find out the bioactive components, the 85% ethanol extract of the root and rhizome of L. duciformis was treated by solvents extraction and column chromatography on silica gel and Sephadex LH-20. Six compounds were obtained from the ethyl acetate fraction and identified as 3-methoxy-4-hydroxyphenylpropyl caffeate (I), 6beta, 8beta-dihydroxyeremophil-7 (11)-en-12, 8alpha-olide (II), 6beta, 8alpha-dihydroxyeremophil-7 (11)-en-12, 8beta-olide (III), 6beta-hydroxyeremophil-7 (11)-en-12, 8alpha-olide (IV), stigmasterol (V) and beta-daucosterol (VI) by analysis of their physic-chemical constants and spectral data. Compound I is a new compound and compounds II - V were isolated from L. duciformis for the first time. The antitussive effect of the compounds is under evaluation.
