1.Application of the 18F-FDG SPECT-CT combined with the images of CT in the diagnosis of lung cancer

Objective To investigate the clinical value of the 18F-FDG SPECT-CT combined with the image of CT in the diagnosis of lung cancer.Methods Retrospectively analysis images of 18F-FDG SPECT-CT and CT of 162 patients pulmonary tumor-like lesions from January 2013 to October 2013 were conducted.All the patients have been confirmed by the pathological mechanism or cytologic examination.Each patient has been firstly diagnosed by the 18F-FDG SPECT-CT along and then combined with the images of CT.The sensitive rate and the accurate rate of two methods were compared by x2 test.Results There were 40 benign lesions and 114 malignant ones in 162 patients.The sensitive rates of the two methods were 86.0% and 96.5% (x2 =6.63,P < 0.05).The accurate rates of the two ways were 79% and 96.3% (x2 =7.76,P < 0.05).The differences were statistically significant.Conclusion 18F-FDG SPECT-CT combined with the images of CT can increase the diagnostic sensitivity and accuracy.It has important clinical value.
2.Study of recombinant GRA5 protein onimmuno-diagnosis of Toxoplasma infection

Objective To investigate the immunodiagnostic valueofrecombinant granulocvte protein 5(rGRA5) protein in prokaryotic Toxoplasma gondii.Methods The PCR was used to ampliify GRA5 gene,then insert the target gene into the prokaryotic expression vector pET28a,and identified by double digestion and sequencing, then transfecte the correct targert gene into BL21 competent cells, SDS-PAGE and Western blot were used to detect the expression of the recombinant protein in BL21.ELISA was used to detect the serum of 100 cases of serologically Toxoplasma-positive individuals.Results The products of GRA5 gene of 363 bp in length was successfully amplified by PCR,the prokaryotic expression vector pET28a-GRA5 was constructed successfully, the result of double digestion showed that the insert fragment size was correct, and DNA sequencing results showed that the homology of GRA5 gene with GenBank was 100%.Also the expression of GRA5 protein was successfully detected in BL21 by Western blot(about 14 ku).ELISA method was used to detect 100 cases of patients with 73 cases showed positive results, the positive diagnosis rate was 73.0%.Among them, the positive detection rate of IgG positive samples was 72.5% in 40 cases,the positive detection rate of IgM positive samples was 53.3% in 30 cases,the positive detection rate of IgG and IgM positive samples was 93.3% in 30 cases.Conclusion The prokaryotic expression vector pET28a-GRA5 is constructed successfully, and the recombinant protein has potential for immunological diagnosis of toxoplasmosis.
3.Comparison of periphery capillary whole blood glucose using four kinds of blood glucose meters and venous plasma glucose using laboratory auto-analyzer

Objective To evaluate the precision of the ACCU-CHEK Performa glucose monitor,the Optium Xceed glucose monitor,the Breeze glucose monitor and the Contour glucose monitor.Methods A total of 102 patients (diabetic patients or un-diabetic patients) who visited our endocrinology outpatient department were randomly selected.The venous blood samples were collected using the laboratory auto-analyzer,and the synchronous finger tip capillary blood were collected using the ACCU-CHEK Performa glucose monitor and the Optium Xceed glucose monitor,or using the Breeze glucose monitor and the Contour glucose monitor.Results The correlation coefficients between VPG by the laboratory auto-analyzer and CBG by the four kinds of glucose monitors was good,and R values were 0.990,0.985,0.963,0.952,and there was no significant difference between the CBG of Breeze glucose monitor and the same VPG(P > 0.05).When the blood glucose concentration was higher than 4.2mmol/L,and compared with the VPG detected by synchronous measurement laboratory,95% of the CBG test results detected by four kinds of blood glucose meters were in the range of ± 20%.Conclusion The four kinds of glucose monitors can provide high accurate.
4.Expressions of LKN-1,CCR1 and CCR3 in Placentas of Pregnant Women with Hypertensive Disorder

Objective:To investigate the role of Leukotactin-1(LKN-1),CC receptor 1(CCR1)and CC receptor 3(CCR3)in the pathogenesis of hypertensive disorder complicating pregnancy(HDCP),the expressions of these factors in placentas were measured.Methods:The levels of LKN-1,CCR1 and CCR3 in placentas were measured from 32 patients with HDCP(study group)and 32 normal pregnant women(control group)by western blot(WB).The localization of these factorsin placentas were detected by immunohistochemistry(IHC).Results:LKN-1 protein expression was negative in placentas from the two groups by WB and IHC,while both CCR1 and CCR3 in the HDCP group were significantly lower than those in the control group(CCR1 0.66±0.06 vs 0.88±0.03.CCR3 0.17±0.01 vs 0.27±0.01,P<0.01).CCR1 was localized in vascular endothelial cells and stroma cells in both groups,but rare in the trophoblast layer.CCR3 was localized in vascular endothelial cells and trophoblast cells in both groups.Conclusions:There is no LKN-1expression in placentas from the two groups.Down regulation of CCR1 and CCR3 may associate with the pathogenesis of HDCP.
6.Toxic effect of formaldehyde on reproduction and heredity in male mice

Objective To examine the effects of formaldehyde on the process of sperm development and reproductive capacity in male mice and the micronucleus rate of liver cells from offspring.Methods The mice were randomly divided into negative control group(NC),formaldehyde exposed group and positive control group (PC).The exposed dose was 21 mg?m-3 (1/24 LC50),42 mg?m-3 (1/12 LC50) and 84 mg?m-3 (1/6 LC50),respectively. 12 male mice were included in each group.Male mice from different formaldehyde exposed groups were inhaled with formaldehyde for 2 h one day,6 d each week,lasted for 13 weeks.The aberration rate of sperm in male mice and the micronucleus rate of liver cells from offspring were observed,and the reproductive capacity of male mice was detected by dominant lethal test.Results The aberration rates of sperm in mice from different formaldehyde exposed groups were significantly higher than that in negative control (P

By feeding rats with high-fat diet insulin resistant models were induced.Some of the models were treated with intragastric administration of curcumin,then the effects were evaluated by hyperinsulinaemic-euglycaemic clamp technique.Curcumin reduced the level of serum resistin and the expression of resistin mRNA in adipose tissue,and thus ameliorated insulin resistance in rats.
9.Pathway for Transformation of Scientific Research Achievement on Traditional Chinese Medicine into Standard

It is an urgent task to transform scientific research achievements on traditional Chinese medicine (TCM) into standard for the science and technology workers of TCM. The transformation not only improves the level of TCM standard, but also promotes the scientific research achievement on TCM into productivity. The Medium and Long-Term Strategic Development Plan for TCM Standardization issued by the State Administration of TCM of the People's Republic of China put forward reinforcement of study on method for transformation of TCM scientific research achievement into standard. Thus, this paper presented seven steps for the transformation on the basis of the TCM Standardization Regulation (trial) in order to provide some references.
10.Determination of hydrocortisone by flow-injection chemiluminescence

Objective To establish a new flow-injection chemiluminescence(CL) method for the determination of hydrocortisone.Methods In H2SO4 solution,hydrocortisone could obviously enhance the chemiluminescence intensity of the reaction of KMnO4-Na2SO3 system.Based on this,a new flow-injection CL method for the determination of hydrocortisone was developed.Results There was a good linear relationship between CL intensity and the concentration of hydrocortisone in the range of 1.0?10-9-1.0?10-6g/mL.The detection limit was 4.0?10-10g/mL at a signal-to-noise ratio of 3∶1.The RSD of 11 assays was 2.2%.Conclusion This method can be successfully used to determine the quantity of hydrocortisone in injection.It is rapid,accurate,simple,and has high sensitivity and wide linear range.
