1.Determination of hydrocortisone by flow-injection chemiluminescence

Objective To establish a new flow-injection chemiluminescence(CL) method for the determination of hydrocortisone.Methods In H2SO4 solution,hydrocortisone could obviously enhance the chemiluminescence intensity of the reaction of KMnO4-Na2SO3 system.Based on this,a new flow-injection CL method for the determination of hydrocortisone was developed.Results There was a good linear relationship between CL intensity and the concentration of hydrocortisone in the range of 1.0?10-9-1.0?10-6g/mL.The detection limit was 4.0?10-10g/mL at a signal-to-noise ratio of 3∶1.The RSD of 11 assays was 2.2%.Conclusion This method can be successfully used to determine the quantity of hydrocortisone in injection.It is rapid,accurate,simple,and has high sensitivity and wide linear range.
2.Biocompatibility between SGBG/PHBV and Human PDLC

[Objective] The biocompatibility between SGBG/PHBV and human PDLC were investigated to provide a basis for the choice of the scaff material of periodontal tissue engineering.[Methods] Human PDLC were cultured using tissue-explant technique.Seeding on 96 wells plate,9 wells for one group,Four different concentrations (100%,75%,50%,25%,0%) of maceration extract of SGBG/PHBV were added into the culture plantsafter 48-h cell seeding,the grades of the cytotoxicity of SGBG/PHBV was evaluated by MTT assay.Human PDLC cultured in vitro were collected and seeded on the three-dimensional scaffolds of SGBG/PH-BV,the cellular morphology and cell growth on the scaffolds were observed and photographed by scanning electronic microscope.HumanPDLC seeded on the three-dimensional scaffolds of SGBG/PHBV in the experimental group,and human PDLC seeded by DMEM in the control group,after 12-,24-,and 48-h cell seeding,got 27 simples for each group,and the affection of the SGBG/PHBV on cell secretory function was observed by spectrophotometry which assayed the biochemical indexes ALP in supinate.[Results] The grades of the cytotoxicity of SGBG/PHBV were 0 and 1.It was displayed that human PDLC adhered and proliferated well on the scaffold of SGBG/PHBV under the scanning electronic microscope.The significant difference of ALP in supinate between the experimental group and the control group (P < 0.05).[Conclusions] SGBG/PHBV had no cytotoxicity to human PDLC.SGBG/PHBV is potential to further study as the scaffolds of periodontal tissue engineering since it displayed the satisfactory biocompatibility with human PDLC.
3.The Prognostic Value of Glycated Haemoglobin in Patients With Acute Myocardial Infarction

To evaluate the prognostic value of glycated haemoglobin (HbA1c) in patients with acute myocardial infarction (AMI). Methods: A total of 1952 AMI patients were retrospectively studied. Based on medical history and HbA1c level, the patients were divided into 4 groups: Diabetes mellitus (DM) group, the patients with known DM or taking hypoglycemic drugs, n=492, Newly diagnosed DM group, MD was diagnosed during hospital stay and HbA1c≥6.5%, n=128, Pre-DM group, HbA1c 5.7%-6.4%, n=783 and Non-DM group, HbA1c<5.7%, n=549. The patients were followed-up for 25.6 months, prognostic differences during hospital stay and follow-up period were assessed by single- and multi-factor analysis. Results: The in-hospital mortality in DM group, Newly diagnosed DM group, Pre-DM group and Non-DM group were 4.88%, 3.91%, 3.96% and 2.91% respectively, P=0.435. As HbA1c level increasing, the incidences of all-cause mortality, non-fatal MI and re-hospitalization were elevating, while the differences among groups were similar. The incidences of major adverse cardiovascular events (MACE) in above 4 groups were 39.84%, 35.94%, 33.97% and 27.87% respectively, P=0.001. Compared with Non-DM group, MACE incidences in the other 3 groups were as OR=1.33, 95% CI 1.05-1.69, OR=1.45, 95% CI 0.97-2.18 and OR=1.71, 95% CI 1.32-2.22 respectively, Ptrend<0.001; with adjusted baseline parameters, Ptrend=0.008. Conclusion: In our research, MACE incidence was increasing upon HbA1c level elevating in AMI patients and it was not related to in-hospital death. HbA1c level should be screened in AMI patients, lifestyle and drug intervention could be used as necessity.
4.Poly(hydroxybutyrate-co-hydroxyvalerate)-sol-gel bioactive glass promotes periodontal tissue regeneration

BACKGROUND:Our previous studies have shown that the poly(hydroxybutyrate- co-hydroxyvalerate) - sol-gel bioactive glass (PHBV-SGBG) has good biocompatibility and promote bone tissue repair, but its specific role in periodontal tissue regeneration has not been investigated. OBJECTIVE:To investigate the periodontal regenerative effects of a PHBV-SGBG scaffold in beagle dogs. METHODS:Alveolar bone defects (5 mm×5 mm) were surgicaly created bilateraly at the buccal side of the mandibular third and fourth premolars of four beagle dogs. PHBV-SGBG scaffold was randomly filed in the defects as experimental group and nothing was put into the contralateral as control group. Histological and scanning electron microscopy observations, cone-beam CT evaluation and the Ca/P concentration ratio analysis were processed at 2, 4, 8 and 12 weeks after surgery. RESULTS AND CONCLUSION:After surgery, the height of the regenerated tissue increased with time in both groups, and the regenerated tissue height in the experiment group was higher than that in the control group (P < 0.05). At 12 weeks after surgery, the Ca/P concentration ratio of the experiment group was close to that in the normal tissue (P > 0.05), but higher than that of the control group (P < 0.05); the histological observation showed that the regenerated tissue of the experimental group was close to the normal tissue, and the regenerated tissue of the control group tended to be mature, with a smal amount of new blood vessels. Under the scanning electron microscope, no scaffold structure was visible in the experimental group with the presence of bone lacuna at 8 weeks after surgery, while in the control group, there was no bone lacuna and obvious osteoblasts; at 12 weeks after surgery, the structure of the regenerated tissue of experimental group was more regular and close to the normal tissue with no remarkable osteoblasts, and in the control group, the regenerated tissue was disordered, with several cavity. These results show that the PHBV-SGBG scaffold can enhance periodontal bone regeneration effectively.
5.Biocompatibility of double-layer poly(L-lactic acid) electrospun nanofiber scaffold with human periodontal ligament cells

BACKGROUND:The morphological structure of nanofiber scaffold which fabricated by electrospinning technique is similar to the natural extracelular matrix, which provides a good microenvironment for cel growth and proliferation, and can also enhance cel adhesion, migration, proliferation and differentiation. OBJECTIVE: To observe the biocompatibility of double-layer poly(L-lactic acid) electrospun nanofiber scaffold and human periodontal ligament cels. METHODS: Electrospinning technique was used to prepare double layers poly(L-lactic acid) electrospun nanofiber scaffold. The toxicity of different concentrations of (100%, 75%, 50%, 25%) double-layer poly(L-lactic acid) electrospun nanofiber scaffold extracts to human periodontal ligament cels was evaluated by MTT assay. The double-layer poly(L-lactic acid) electrospun nanofiber scaffold was co-cultured with human periodontal ligament cels. The cel adhesive capacity in early stage was determined by MTT assay. The growth of cels on the scaffold was observed by scanning electron microscopy. RESULTS AND CONCLUSION: Different concentrations of double-layer poly(L-lactic acid) electrospun nanofiber scaffold extracts did not create any toxicity to human periodontal ligament cels. After co-culture for 2, 6, 24 hours, human periodontal ligament cels were poorly adherent onto the double-layer poly(L-lactic acid) electrospun nanofiber scaffold. After 7 days of co-culture, human periodontal ligament cels adhered wel on the loose surface of scaffold, maintained the original shape, stretched wel, and interconnected processes were observed; on the dense surface of the scaffold, multi-layer cels were observed. The cels showed fusiform or polygonal appearance and were connected together. These results demonstrate that the double-layer poly(L-lactic acid) electrospun nanofiber scaffold has good biocompatibility with human periodontal ligament cels.
6.Determination of acyclovir and gancyclovir using flow-injection chemiluminescence method

Objective To develop a new flow-injection chemiluminescence(FI-CL) method for the determination of acyclovir and gancyclovir by using the CL system of Ce(IV)-Rhodamine B.Methods The redoxreaction of Ce(IV) and acyclovir/gancyclovir in H2SO4 medium could generate CL signal.Rhodamine B could obviously sensitize this signal,and the CL intensity was proportional to the concentration of acyclovir and gancyclovir.Therefore,a new FI-CL method has been described for the determination of acyclovir and gancyclovir.Results For acyclovir,the determination range was 3.0?10-5g/L-7.0?10-2g/L,with 1.56?10-5g/L as its determination limit.During 11 repeated measurements for 1.0?10-3g/L acyclovir,the relative standard deviation was 2.08%.For ganciclovir,the determination range was 5.0?10-5g/L-7.0?10-2g/L,with 2.35?10-5g/L as its determination limit.The relative standard deviation was 2.83%,with 11 repeated measurements of 1.0?10-3g/L ganciclovir.Conclusion This method has broad linear range,high sensitivity,and convenience and speediness,it therefore,can be successfully used to determine the content of ganciclovir in injections.
7.Prenatal MRI diagnosis of fetal bowel obstruction

Objective To investigate the clinical value of prenatal MRI in the diagnosis of fetal bowel obstruction.Methods Pregnant women suspected to have fetal abdominal abnormalities by ultrasonography were suggested to undergo MRI examinations within two days.Scanning sequence included FIESTA,SSFSE and T1WI SPGR sequence,with field of view focused on the fetal abdomen.After the final diagnoses of the cases were obtained by induced labor pathological examination or postpartum imaging or operation,the imaging data and the clinical data were reviewed and analyzed retrospectively.Results A total of 23 cases with bowel obstruction were included in the study.Four fetuses with duodenal atresia showed low T1 signal,high T2 signal characterized by double-bubble sign on MRI.There were 10 fetuses with jejunoileal atresia,showing bowel dilatation and hyperintense micro-colon on T1WI.Five cases of them depicted expansion of the terminal ileum with high T1 meconium signal.One each fetus had colonic atresia,intestinal malrotation with double-bubble and whirl sign.Annular pancreas with double-bubble sign and pressure trace of the bracket shape was detected in 3 fetuses.Meconium peritonitis was present in 4 fetuses,with 2 of them showing dilatation of intestine,ascites and pseudocysts.Conclusions According to the signal characteristics of amniotic fluid and meconium in the gastrointestinal tract on MRI,the obstructive level and development status of the distal bowel can be determined with MRI.It can provide additional information to ultrasonography,which brings clinical significance to prenatal diagnosis and intrapartum surgical operation.
8.Biocompatibility of a novel chitosan-collagen scaffold and periodontal ligament cells

BACKGROUND:At present, col agen as a material in periodontal tissue engineering has some disadvantages such as poor mechanical strength and rapid degradation speed. Col agen combined with chitosan can improve above-mentioned problems.
OBJECTIVE:To evaluate the biocompatibility of a novel chitosan-col agen scaffold in vitro.
METHODS:Cytotoxicity of the extract of chitosan-col agen scaffold in different concentrations (100%, 75%, 50%, 25%) was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Human periodontal ligament cel s at 4-6 passages were cocultured with the chitosan-col agen scaffold. Cel growth on the scaffold was observed. Changes in alkaline phosphatase activity were detected in periodontal ligament cel s before and after coculture.
RESULTS AND CONCLUSION:The novel chitosan-col agen scaffold was made up of double layers with one dense layer and another loose layer. The grade of the cytotoxicity of the scaffold was from 0 to 1. Scanning electron microscope and histological observation demonstrated that cel s grew wel on the chitosan-col agen scaffold;the dense layer could prevent cel s to migrate into the scaffold. There were no significant differences in alkaline phosphatase activity in human periodontal ligament cel s before and 24 hours after combined culture (P>0.05). Alkaline phosphatase activity in human periodontal ligament cel s was greatly higher at 48 and 72 hours after combined culture compared with that before culture (P<0.05). Above results indicated that the novel chitosan-col agen scaffold has a good biocompatibility and barrier function, and potential as a scaffold for periodontal tissue engineering.
9.A heart failure staging model based on machine learning classification algorithms

BACKGROUND:Early detection and accurate staging diagnosis of heart failure are the basis of good clinical therapy efficacy. Due to lack of simple and effective staging model for the diagnosis of heart failure, it is difficult to diagnose heart failure in clinics, leading to poor control of heart failure. OBJECTIVE:To establish the disease staging model based on Adaboost and SVM for heart failure, and improve the accuracy of diagnosis and staging of heart failure. METHODS:A total of 194 cases were roled into this study, including heart failure patients and healthy physical examination persons. According to the stage standards formulated by American Colege of Cardiology and American Heart Association, specific clinical feature parameters closely related to heart failure were colected and selected. Based on clinical diagnosis results and using Adaboost model and SVM model, we trained the models for heart failure diagnosis and staging, thus obtaining diagnosis model. RESULTS AND CONCLUSION: The parameters included stroke volume, cardiac output, left ventricular ejection fraction, left atrial diameter, left ventricular internal diameter at end-systole, N-terminal pro-brain natriuretic peptide and heart rate variability. As for the Adaboost model, its sensitivity and specificity was 100% and 94.4%, respectively. At the same time the SVM model had good sensitivity and specificity, 86.5% and 89.4% respectively. Adaboost classification model can be accurate in the diagnosis of heart failure symptoms, the accuracy reached 89.36%. On the basis of the diagnosis of heart failure, the SVM classification model is effective in staging the severity of heart failure, staging accuracy for staging B and C was 86.49% and 81.48%, respectively. The findings indicate that, combining Adaboost and SVM machine learning models could provide an accurate diagnosis and staging model for heart failure.
10.Influence of anxiety and depression on cellular immune function of patients with breast cancer before chemotherapy

Objective To evaluate the effects of the comprehensive psychological intervention on anxiety and depression of the patients with breast cancer during chemotherapy ,and to explore the influence of depression and anxiety before and after chemother-apy on the cellular immune function .Methods A total of 81 married female patients with breast cancer were surveyed and assessed with self-designed questionnaire ,the Herth Hope Scale ,the Hamilton Depression Scale(HAMD) and the Hamilton Anxiety Scale (HAMA) .64 cases of breast cancer with depression and anxiety according with the included standards (HAMD scores ≥8 and HA-MA scores ≥7) were randomly divided into the intervention group(n=32) and the control group(n=32) .The levels of T lympho-cyte subsets(CD3+ ,CD4+ ,CD8+ ,CD4+ /CD8+ ) and the natural killing(NK) cells in peripheral venous blood were determined by the flow cytometry(FCM) .Results The Herth scores after chemotherapy in the intervention group were increased by 37 .19% than that of the control group ,while the HAMD scores were decreased by 36 .01% compared with the control group ,the differences be-tween them showing statistical significance (P<0 .01) .In the comparison between before and after chemotherapy ,the Herth scores in the intervention group were increased by 12 .57 ± 4 .57 ,while the HAMD and HAMA scores were declined by 9 .32 ± 1 .86 and 4 .35 ± 1 .15 respectively ,the differences showed the statistical significance (P< 0 .01) .Compared with the control group ,CD3+ , CD4+ ,CD4+ /CD8+ ratio and NK cells before and after chemotherapy in the intervention group were increased by 2 .10 ± 1 .63 , 1 .51 ± 1 .02 ,0 .05 ± 0 .02 and 0 .78 ± 0 .52 correspondingly ,the differences among them had statistical significance(P<0 .001) .The HAMD scores were negatively correlated with the CD4+ /CD8+ ratio(r= -0 .554) .Conclusion The cellular immune hypofunction in the patients with breast cancer is probably related with anxiety and degree of depression .The comprehensive psychological inter-vention model can improve the anxiety and depression mood during chemotherapy .
