1.Levels of interlukin-2, interlukin-6 and its soluble receptors in the first episode depression of Uygur patients before and after treatment with venlafaxine

Objective To investigate the action of interlukin-2(IL-2),interlukin-6(IL-6) and its soluble receptors (sIL-2R,sIL-6R) in the first episode of depression in the patients of Uyghur nationality and the differences in IL-2,IL-6 and sIL-2R,sIL-6R levels between the responsive depressed patients and the refractory depressed patients treated with venlafaxine.Methods A case-control study design was conducted.57 first-episode patients with depression (patient group) and 55 healthy people matched with gender and age (control group) were recruited in the study.An intervention with sustained-releasing venlafaxine tablets at fixed dose of 150 mg/d was performed in the patient group.The severity of the illness was evaluated by using the Hamilton's depression scale (HAMD-17) before and after the therapy.And by calculating the reduction rate of HAMD-17 (≥ 50% or <50%),the patients were divided into the responsive or refractory groups.The serum levels of IL-2,IL-6,sIL-2R and sIL-6R in patients and controls were tested by ELISA,and a re-test was done with the patients after treatment.Results There were statistical significant differences of the levels of serum IL-2,IL-6 and sIL-2R,sIL-6R between the patients and the control group (P < 0.01).After treatment,the levels of serum IL-2,IL-6,sIL-2R and sIL-6R in responsive patients were significantly decreased when compared with those before the treatment(P< 0.01).The four indexes of refractory patients didn' t alter after venlafaxine treatment (P > 0.05).There were positive correlations between HAMD,serum IL-2 (r =0.677 ; P =0.000) and IL-6 (r =0.197 ; P =0.033) before treatment in all patients.Conclusion Serum IL-2 and IL-6 may play a role in the onset of the depression.The efficacy of venlafaxine is negatively correlated with the levels of serum IL-2 and IL-6.Regulating the imbalanced inflammatory cytokines and the immune system may be one of the mechanisms of drug therapy of depression.
2.Correlation between cognition need and information seeking behaviors

The effect of cognition need on information seeking behaviors of information users and the difference in behaviors of information users were investigated in 184 undergraduates from the same grade with different back-grounds of subjects using the cognition need scale and undergraduates information-seeking behaviors scale ,respec-tively.The data were analyzed by descriptive statistical analysis, correlation analysis, rank test, and multivariate linear regression analysis , respectively , showing that the cognition need is significantly correlated with the informa-tion seeking behaviors of undergraduates in terms of information evaluation, information searching, information management and information use .
3.Effects of HO-1 modified rat bon e marrow mesenchymal stem cells on lymphocytes in vitro

Objective To investigate the immunomodulatory effects of heme oxygenase-1 ( HO-1) modified rat bone marrow mesenchymal stem cells ( BM-MSCs) on lymphocytes .Methods Rat BM-MSCs were cultured and identified in vitro.HO-1 gene was transduced into BM-MSCs via recombinant adenovirus (rAdV) to construct HO-1/BM MSCs, which were then co-cultured with rat spleen lymphocytes .The cellu-lar activity of lymphocytes was detected by using MTT method .The cell cycle distributions were analyzed by propidium iodide staining and flow cytometry .ELISA was used to detect the level of cytokines in the super-natant of cultured cells .The expression of lymphocyte activation markers , CD25 and CD71, were measured by using fluorescence labeled antibodies and flow cytometry .Results Compared with the control group , the lymphocytes co-cultured with HO-1/BM MSCs showed a suppressed proliferation and cell cycle progression . HO-1/BM MSCs inhibited the secretion of IL-2, TNF-αand IFN-γ, but enhanced the secretion of IL-10 in lymphocytes.Furthermore, the expression of CD25 and CD71 were also down-regulated by HO-1/BM MSCs. Conclusion Compared with BM MSCs , HO-1/BM MSCs show higher immunosuppressive effects on lym-phocytes .
4.Associations between interleukin 1β gene polymorphism-511C/T,-31T/C and depressive disorders in Uygur population of Xinjiang

Objective To explore the distribution on-511C/T,-31T/C single nucleotide polymorphism of IL-1β gene among Uygur population in Xinjiang,and to analyze the correlation between IL-1β gene polymorphism and depressive disorders.Methods A total of 100 patients with depressive disorders and 120 control subjects were selected.Polymerase chain reaction and restriction fragment length polymorphism (RFLP-PCR),enzyme digestion and sequence reaction were used to detect the common-511C/T,-31T/C polymorphism of the IL-1β gene.The relationship between the polymorphism in the IL-1β gene and the severity of depressive disorders was analyzed.Results The frequencies of CC,CT,TT in-511 were 19.0%,58.0% and 23.0% in patient group,while those were 19.2%,55.0%,25.8% in the control group,which did not show statistically significant differences (x2=0.266,P=0.875).The frequencies of CC,CT,TT in-31 were 24.0%,58.0% and 18.0% in the patient group,while those were 24.2%,58.3%,17.5% in the control group,which did not show statistically significant differences (x2=0.0093,P=0.995).The frequencies of CC,CT,TT genotypes of the IL-1β gene polymorphism (-511 C/T,-31 T/C) were not statistically different between depressive patients and healthy controls.Conclusion The findings suggest no significant association between-511C/T,-31T/C polymorphism and depressive disorders in Uygur population of Xinjiang.
5.Construction and Application of Lentiviral Vectors Expressing microRNA-203 Tough Decoy

Objective To establish method of constructing lentiviral vectors to express microRNA (miRNA) ''tough decoy''(TuD)and to detect the effects of the TuD on cellular endogenous miRNA level and cellular phenotypes. Methods Two-step cloning strategy was utilized to first generate a universal miRNA TuD frame vector,followed by con-structing the TuD expression vector specially targeting miR-203. The package of the recombinant lentivirus was per-formed in 293T cells. Then the rat bone marrow mesenchymal stem cells(BM-MSCs)were infected by the miR-203 TuD expression lentivirus. The pSIH1-H1-copGFP vector was also packaged and the BM-MSCs infected by this lentivirus were served as control. Endogenous miR-203 level in BM-MSCs was measured by quantitative RT-PCR,and cellular vi-ability and apoptosis were detected by CCK-8 test and Annexin V-PI staining respectively. Results The miR-203 TuD expression vector was successfully constructed and inserted sequence was validated. At the 3rd,6th and 9th days after in-fected by the miR-203 TuD expression lentivirus,rat BM-MSCs exhibited a repressed endogenous miR-203 level. The miR-203 TuD also promoted viability and inhibited apoptosis of BM-MSCs. All these differences between miR-203 TuD group and control group were statistically significant. Conclusion The two-step cloning method for the construction of miRNA TuD expression vector is simple and efficient. The miRNA TuD can effectively suppress the level of the target miRNA and affect cellular phenotypes.
6.Synthesis of hyaluronic acid coated magnetic nanoparticles for highly efficient tumor CD44 cell labeling and in vitro study

Objective To explore a promising system for tumor CD 44 receptor-targeted imaging and to investigate their physic-chemical properties and targeting effect on CD 44 abundant cancer cells in vitro.Methods The superparamagnetic iron oxide ( SPIO) nanoparticles were prepared by a coprecipitation in alkaline media starting from a mixed of the ferrous and ferric solution.And then the surface of the SPIO nanoparticles were modified with APTMS by a reaction with the hydroxyl groups.Finally, the hyaluronan-modified SPIO ( SPIO-HA) nanoparticles were prepared.Control and experimental groups were established after adding SPIO or SPIO-HA as agents respectively.Transmission electron microscopy ( TEM) and particle size analyzer were used to measure these nanoparticle sizes and the hydrodynamic diameters.Thermogravimetric analysis ( TGA) was carried out to evaluate the HA-content on the surface of SPIO-HA.The MRI T2 ralaxivities (1/T2 ) of the two groups at different Fe concentrations (0.09, 0.18, 0.27, 0.36, 0.45 mmol/L ) were measured on a 3.0T MR system.HepG2 cells and HL7702 cells were used for assessment of cells viability by methyl thiazolyl tetrazolium ( MTT ) assay.Prussian blue staining , immunoassay fluorescence image and flow cytometry were carried out to determine the targeted cellular uptake of SPIO-HA nanoparticles.MRI were performed to show the MR T 2 value changes after incubating with HepG2 cancer cells by using T 2 WI sequences at a clinical 3.0 T MR system.One-way analysis of variance was performed to determine significant changes in MR T 2 values of blank control , SPIO-HA and SPIO groups.Results The SPIO-HA and SPIO NPs were fairly homogeneous with an average core size of 18.2 and 22.4 nm, hydrodynamic diameter of 91.1 and 103.2 nm, Zeta potential of (-45.00 ±0.86) mV and (-18.50 ±0.73) mV, and magnetic relaxivity of 0.212 ×106 M-1 · s-1 and 0.191 ×106 M-1 · s-1.Based on the TGA data , HA accounted for 24%weight of each SPIO-HA.The internalization of the SPIO-HA was confirmed by prussian blue staining , while the cells showed no obvious blue stains with SPIO , incubation of SPIO-HA with tumor cells led to blue color inside the cells.After that, we examined cancer cell binding of FITC-SPIO-HA by immunoassay fluorescence image and flow cytometry.The green fluorescence resulting from FITC-SPIO-HA was observed inside the cells in both the cytoplasm and the plasmalemma.Tumor cells treated with SPIO-HA exhibited higher fluorescence signals with 7.97-fold enhancement observed for HepG 2 cells over control particles.In vitro MR, mean T2 values of blank control , SPIO and SPIO-HA groups were ( 115.20 ±0.36 ), ( 115.07 ±0.81 ) and ( 21.67 ±0.21 ) ms, respectively.There was significant difference among those three groups (F=31 703.339,P<0.01), MR T2 values of HepG2 cells treated with the SPIO-HA NPs were lower than blank and SPIO group.In comparison, SPIO did not generate any MRI signal changes compared with blank group.Conclusion The tumor CD44 receptor-targeted MR molecular probe SPIO-HA had a good physic-chemical property and well targeted HepG2 cells.
7.Investigation of self-acceptance status and its influencing factors of elderly people in nursing home

Objective:To understand the present situation of self-acceptance of elderly in nursing home and explore its influencing factors, so as to provide basis for improving self-acceptance level of elderly in nursing home.Methods:Totally 415 elderly people in 3 Medical and nursing homes in Fuyang city of Anhui province were selected by Self-Acceptance Scale, and influencing factors were analyzed.Results:The total score of Self-Acceptance Scale of the elderly in nursing home was (44.11±7.25) points, the score rate was 67.86% (44.11/65), in the middle level; the results of multiple linear stepwise regression showed that the relationship with children, hobbies, number of children, length of stay in nursing home, frequency of visits and age were the main influencing factors ( F value was 48.228, P<0.001). Conclusion:The self-acceptance of the elderly in nursing home is in the middle level. The nursing home or social should pay attention to the higher age group and the newly admitted elderly in nursing home, give such people timely psychological guidance, help them to plan their later life, cultivate their interests and hobbies through recreational activities, and take corresponding measures to harmonize their relationship with their children in order to improve their self-acceptance level and promote mental health.
8.Clinical value of interventional radiology in hemodialysis patients with stuck tunnel dialysis catheter

Objective To evaluate clinical value of interventional radiology in difficult extraction of stuck tunnel hemodialysis catheter (TCC).Methods Maintenance hemodialysis patients who required removal of stuck TCC in our hospital from June 2015 to June 2017 were enrolled.Medical history and preoperative examinations were carefully evaluated.Patients were pulled rotationally the catheter under the fluoroscopy guidance and exchanged the TCC.The information and procedure-related complication were recorded.Balloon dilation or snare technique would be used when failed to remove the stuck catheter.Results A total of 22 patients were enrolled,with an average age of (67.2±18.5) years,a median dialysis time of 54(18,106) months,and catheter indwelling time of 37(14,82) months.All patients were successfully removed the stuck catheter,among which 18 cases (81.8%) had fibrin sheath and 13 cases with sheath calcification.TCC mainly stuck in the junction of internal jugular vein and innominate vein,middle and lower innominate vein and middle part of superior vena cava.No procedure-related major complication occurred.Sixteen cases (72.7%) complained of transient substernal extraction pain and 1 case occurred groin hematoma during operation.Conclusions Catheter incarceration problem should be alert to the hemodialysis patients with catheter indwelling time of more than 2-3 years.The minimally invasive interventional techniques including rotationally pull-out under fluoroscopy guidance and balloon angioplasty are safe and effective in removal of the stuck TCC.
9.Expression of visfatin and microRNAs in myocardium of acute myocardial ischemia in rats

Objective To study the expression of visfatin and related mRNAs in myocardium of acute myocardial ischemia in rats,and to explore the role of both in acute myocardial ischemia model and to propose new ideas for clinical cardiovascular diseases.Methods Wistar rats (n=20) were randomly divided into sham operation control group and acute myocardial ischemia groups.Left anterior descending (LAD) of coronary artery was ligated to establish the model of acute myocardial ischemia.The total myocardium proteins were collected 12 h after operation.Western blot was used to observe the expression changes of visfatin in myocardium.The expressions of miR-124 and miR-327 in myocardium were detected by real-time PCR.Results The levels of serum CK and LDH in the model group of myocardial ischemia rats were elevated [(46.20±7.12) vs (72.19±9.38)kU/L;(2542.23±173.54) vs (6428.76±237.81)U/L,P<0.05)].The visfatin expression levels in the rats of acute myocardial ischemia groups were higher than those in controls [(0.804±0.042) vs (1.046±0.031),P<0.05].MiR-124 was up regulated and miR-327 was down regulated in acute ischemia myocardium.Cndusion The up-regulation of miR-124,the down-regulation of miR-327 and the increase of visfatin expression may be related to acute myocardial ischemia.The changes of visfatin expression may be regulated by miR-124 and miR-327 posttranscriptional levels.
10.A family with creatine deficiency syndrome due to SLC6A8 gene novel mutation

To analyze the clinical, molecular genetic and biochemical characteristics of a family diagnosed with creatine deficiency syndrome (CDS) due to SLC6A8 gene mutation diagnosed in Department of Pediatrics, the First Affiliated Hospital of Xinxiang Medical University in September 2018.The boy was referred at the age of 13 for intermittent vomiting and symptoms were relieved after symptomatic treatment, 7 months later, the patient was diagnosed again.He ate less and constipated as an infant, and had intermittent vomiting and severe constipation from 12 years old.He could walk at the age of 1 year and 6 months, could speak his first word at 2 years old, and only spoke simple sentences at the diagnosis.He rarely communicated with his parents.His brother has severe intellectual deficiency and seizures.His mother has cognitive impairment and had repeated stillbirths and miscarriages.When visiting the doctor, his weight was 22 kg (< P3), height was 131 cm (< P3), and head circumference was 52 cm.Besides, he had severe mental retardation and sluggish expression, poor communication, broad forehead, musculoskeletal atrophy and arched foots.His brain magnetic resonance imaging showed mild enlargement of the lateral ventricles.Magnetic resonance spectroscopy showed normal signals in the bilateral hippocampuses.The boy′s exon sequencing related to intellectual deficiency revealed the hemizygous mutation of SLC6A8 gene c. 626(exon3)-c.627(exon3)delCT.His mother and brother were carriers of the variant gene, but his father and grandmother had no such mutation.The urinary creatine to creatinine ratio was elevated but guanidinoacetate was normal, it was further confirmed that CDS can be caused by the novel mutation c. 626(exon3)-c.627(exon3) delCT in SLC6A8.In clinical practice, boy with mental retardation of unknown origin and often have gastrointestinal symptoms, especially when feeding difficulties and growth retardation occur in infancy, the early diagnosis of CDS can be achieved by examination of urinary creatine to creatinine ratio and gene detection.
