Objective To explore the application and effect of aggameline in the treatment of depression. Methods 150 patients with depression treated in our hospital from January 2015 to December 2016 were randomly divided into three groups: A, B and C, 50 cases in each group. Group A was treated with rosiglitamine, group B was treated with venlafaxine, group C was treated with paroxetine. The improvement of symptoms before and after treatment in the three groups was observed[ (Hamilton Depression Scale (HAMD), Hamilton Anxiety Scale (HAMA)], Sleep Quality [Pittsburgh Sleep Quality Index (PSQI), Sleep Self-Rating Scale (SRSS)] and cognition Function [repeat sets of neuropsychological state test (RBANS), connection test (TMT)], and adverse drug reactions were recorded. Results After four weeks of treatment, the scores of HAMD and HAMA in the three groups were lower than those before treatment (P<0.05). The results of HAMD score showed that group B (P<0.05), but there was no significant difference between group A and group C. After treatment for four weeks, the scores of PSQI and SRSS were lower than those before treatment (P<0.05). There was no significant difference between group A and group B (P<0.05). After treatment, the scores of RBANS were higher than those before treatment (P<0.05), and delayed memory, attention to the two groups in group B> group C (P<0.05), There was no significant difference between the two groups in immediate memory; there was no statistically significant difference between the three groups after treatment and speech and visual acuity scores. After 4 weeks of treatment, the time to connect and connect sequentially was shorter than that before treatment (P<0.05), and the order of alternating time showed that group A

To characterize the long terminal repeat (LTR) of the ALV-J strain which can induce hemangioma, fragments of provirus LTR of the three different ALV-J strains SCAU-HN06, NX0101 and JS-nt were amplified with a pair of specific primers, then cloned and subjected to sequence analysis. In comparison with the prototype ALV-J strains HPRS-103 and ADOL-7501, the LTRs of domestic strains (SCAU-HN06, NX0101, JS-nt and SD07lk1) had an 88.0%-97.2% nucleotide sequence identity; the U5 and R regions in the LTR had a high nucleotide similarity, while the U3 region in the LTR showed significant variance. The LTR fragments from the different ALV-J strains were inserted into the upstream of bacterial CAT gene of the plasmid pCAT-Basic, respectively. The resultant recombinant plasmids were transfected into DF-1 cells. The transfected cells were harvested 48 h post-transfection, and cell lysates were prepared for CAT expression detection. The CAT assay was performed using CAT-ELISA. The results showed that the promoter activity of the LTRSCAU-HNO6 was a little higher than those of LTRJS-nt and LTRNX0101, but there was no significant difference in the promoter activity among the compared LTRs.

OBJECTIVETo observe the efficacy of polyglycoside of Tripterygium wilfordii(GTW) in preventing postoperative recurrence of Crohn disease(CD).
METHODSThirty-nine post-operative CD patients in whom all of the diseased gut had been removed from January 2005 to December 2006 were enrolled in a randomized, placebo-controlled trial. The patients took GTW(21 cases) or SASP(18 cases) in two weeks after operation. Crohn disease activity index(CDAI), ESR and CRP were collected at week 0, 13, 26, 52 or at the onset of symptoms. Ileocolonoscopy was performed at the end of the trial or at the onset of symptoms.
RESULTSOne patient in GTW group and 2 patients in SASP group were lost and 2 patients in GTW were excluded from the trial for non-compliance. Clinical recurrence was ascertained in one patient (5.6%) received GTW and in four (25.0%) received SASP. Four of eighteen patients in GTW(22.2%) had endoscopic recurrence compared with nine of sixteen(56.2%) in SASP. There were significant differences between the two groups(P<0.05).
CONCLUSIONTripterygium wilfordii showed good efficacy in preventing recurrence of postoperative CD which can maintain remission and prevent recurrence.

Objective To explore the effects of growth hormone(GH)on the proliferation,cycle,inva-sion,and migration of colon cancer cells and its possible mechanism. Methods GH3 cells with growth hor-mone-type pituitary adenoma were cultured in vitro, and the secretion of growth hormone in the supernatant of GH3 cells was detected by ELISA.Colon cancer LoVo cells in logarithmic growth phase were randomly divid-ed into the control group and the experimental group.PBS was added to the control group, while high concen-trations of recombinant GH were added to the experimental group. The two groups of cells were cultured in vitro under the same conditions.CCK-8 method was used to detect the proliferation of the cells.Flow cytome-try was used to detect the cell cycle.Transwell assay was used to detect the effect of growth hormone on the invasion and migration of the cells.Western blot was used to detect the expressions levels of E-cadherin,N-cadherin,Vimentin,and Snail-1 proteins in the cells. Results The results of ELISA showed that GH3 cells could secrete a large amount of GH,and the concentration of GH in the supernatant was(1 208±9)ng/mL.GH promoted cell growth in a dose-dependent manner within a certain concentration range, and GH 200 ng/mL was the optimal intervention concentration for subsequent experiments.Compared with the control group, the cell cycle in the experimental group changed from G1 phase to S phase and G2 phase, the ratio of G1 phase cells decreased, and the ratio of S phase cells and G2 phase cells increased(P<0.05).Compared with the control group, the number of the cell invasion and migration increased in the experimental group(P<0.05),the expression levels of N-cadherin,Vimentin,and Snail-1 was up-regulated,while the expression level of E-cadherin was down-regulated(P<0.05).Conclusion High concentration of GH promotes the prolifera-tion,invasion and migration of colon cancer cells, and induces the transition of cell cycle from G1 to S and G2 phases. The mechanism may be related to the epithelial-mesenchymal transition(EMT)of colon cancer cells promoted by high concentration of GH.

This study was aimed to investigate the effect of bortezomib combined with bisphosphonates on serum levels of DKK-1 and RANKL in multiple myeloma patients, and to evaluate its role in the therapy of osteolytic lesion. Fourty-three patients with newly diagnosed and relapsed myeloma were divided into 2 groups. Twenty-three patients were treated with bortezomib combined with bisphosphonates (A group) and 20 patients were treated with bisphosphonates combined with traditional chemotherapy (B group). Serum levels of DKK-1 and RANKL were measured by ELISA before and after 4 cycles of chemotherapy. The results indicated that serum DKK-1 level significantly decreased in patients of A group (43.2 µg/L before vs 30.4 µg/L after 4 cycles of chemotherapy), and so did for serum RANKL level in A group (0.83 pmmol/L before vs 0.45 pmmol/L after 4 cycles of chemotherapy). While there was no significant differences in DKK-1 and RANKL serum level before therapy between A and B groups, but there was significant differences in DKK-1 and RANKL levels after 4 cycles of chemotherapy (P < 0.05). It is concluded that bortezomib combined with bisphosphonates obviously reduce the serum levels of DKK-1 and RANKL, thus has beneficial effect on osteolytic lesion.

OBJECTIVETo study the effect of the DA-EPOCH chemotherapy combined with G-CSF and the CTX therapy with G-CSF on mobilizing and collecting the peripheral blood hematopoietic stem cells and the later hematopoietic recovery.
METHODSForty patients accepted mobilization and collection of peripheral blood stem cells(PBSC) after treated by CTX+G-CSF and DA-EPOCH+G-CSF therapy respectively, and were treated by auto-transfusion after BEAM pre-regimen. The mobilization efficacy, adverse effects and hematopoietic recovery after autologous transplantation were analyzed retrospectively.
RESULTSDuring the CTX+G-CSF mobilization, only one patient achieved the white blood cell(WBC) at 0.8×10/L, while the others were with the lowest WBC level above 2.0×10/L. The platelet counts were all normal with the exception of 3 cases at 80×10/L. The median percentage of CD34cells in one period of collection was 0.99(0.35-1.30)%. The median MNC was (3.80±2.05)×10. The cumulative total of mononuclear cell was (5.84±2.48)×10/kg, and the median CD34cell count was 3.84(3.91-6.5)×10/kg. During the DA-EPOCH+G-CSF mobilization, the peripheral WBC count of patients were decreased to the lowest level at (0.2-1.4)×10/L. The platelet counts were all above 40×10/L except for 1 case in which the platelet count was reduced to 8×10/L. The median percentage of CD34cells in one period of collection was 0.85(0.34-1.2)%. The median MNC was (3.68±1.56)×10. The cumulative total of mononuclear cells was (6.01±2.26)×10/kg, and the median CD34cell count was 4.44(2.7-7.10)×10/kg. There were no statistical differences between the 2 groups in the median percentage of CD34cells, the median MNC, the cumulative total of mononuclear cells and the median CD34cell counts (P>0.05). The average acquired time for granulocyte engraftment was 10.00(9.00-11.00) days, and for platelet engraftment was 12.50(11.00-17.25) days, with no statistical difference(P>0.05). No death occurred during the process of transplantation.
CONCLUSIONDA-EPOCH therapy combined with G-CSF can effectively mobilize the peripheral blood hematopoietic stem cells in NHL patients with higher safety and lower price, and proves to be worth recommending in clinical use.
