1.Effect of Jiuqiang Naoliqing on the microcirculation of the cheek pouch of golden hamsters

ObjectiveTo observe the effects of Jiuqiang Naoliqing(JNQ) on the microcirculation of the cheek pouch of golden hamsters. MethodsAutomatic measuring device was used to evaluate the changes of microcirculation. ResultsAfter the disorder of microcirculation of cheek pouch made by noradrenaline(NA), the JNQ group recovered better and more rapidly than other groups. ConclusionJNQ can prevent and reverse the disorder of microcirculation made by NA, and do better than NQ.
3.Causes analysis of early postoperative high intraocular pressure of angle - closure glaucoma underwent compound trabeculectomy

To analyze the causes and treatments of high intraocular pressure ( > 21mmHg ) of angle - closure glaucoma underwent compound trabeculectomy 1mo after surgery.
●METHODS: This was a retrospective study of our hospital, from March 2010 to March 2013. Thirty-four (38 eyes) of angle-closure glaucoma patients were collected, who underwent compound trabeculectomy with high intraocular pressure ( > 21mmHg) 1mo after operation. We analyzed the causes and summarized the treatments.
●RESULTS:The causes which lead to early postoperative high intraocular pressure included malignant glaucoma (9 eyes of 8 cases), blood clot and connective tissue block under the scleral flap (15 eyes of 13 cases), hyphema after surgery(5 eyes of 5 cases), sclera flap incision was incarcerated with iris tissue ( 3 eyes of 3 cases ), preoperative high intraocular pressure for a long time(5 eyes of 4 cases), 1 eye of 1 case for unknown reason. After proper treatments, intraocular pressures of all patients were bellowed 21mmHg.
● CONCLUSlON: Early postoperative high intraocular pressure of angle - closure glaucoma underwent compound trabeculectomy is caused by various factors, and the early prevention and timely treatment are key points of a successful operation.
4.Effect of Jiuqiang Naoliqing on the behavior of Kunming mice

ObjectiveTo observe the effect of Jiuqiang Naoliqing (JNQ) on the behavior of Kunming mice.MethodsSpontaneous movement, Morris Water Maze, Rotarod, anti caffeine test, sleeping time of pentobarbital sodium, subthreshold dose of pentobarbital sodium, and anti pentylene tetrazol test were adopted to evaluate the behavioral changes. ResultsCompared with the control group, the low dose of JNQ can increase spontaneous movement of the mice, the middle and high dose of JNQ can increase time on the rotating rods. JNQ can also increase sleeping time of pentobarbital sodium test and percent of falling asleep in subthreshold dose of pentobarbital sodium test, as well as antagonize caffeine's effect on mice. ConclusionJNQ can also do sedative and hypnotic effect on Kunming mice as well as improve their ability of balance and coordination.
5.The impact of diabetes mellitus and insulin interference on cortical SCF/KIT of mice.

OBJECTIVETo investigate the expression of cortical SCF/KIT system at different blood glucose level in mice.
METHODS27 male C57 mice were randomly divided into control group, diabetes group, and diabetes plus insulin group. The diabetic mice were induced by streptozotocin. Western-blot and double-immunofluorescence histochemistry were used to detect the expression of SCF and KIT.
RESULTSBoth methods indicate that the level of S-SCF and M-SCF were decreased significantly in the diabetes group, and this trend can be reversed effectively when the insulin was utilized.
CONCLUSIONThe decline of SCF might be one of underlying mechanisms of diabetic encephalopathy.

6.Forensic Application of SiFaSTRTM 23plex DNA ID System in Han Population of Eastern China

Objective To investigate the genetic polymorphism of 21 autosomal STR loci and DY S391 locus of SiFaSTRTM 23plex DNA ID system in Han population of eastern China and to evaluate its ap-plication value in forensic science. Methods Typing test of 2000 unrelated individuals was performed using SiFaSTRTM 23plex DNA ID system. The population genetic parameters of STR loci were statistically analysed. A total of 3198 parentage confirmed cases were detected with that system and the mutation conditions were observed in 21 autosomal STR loci. Results All the 21 autosomal STR loci showed no significant departure from Hardy-Weinberg equilibrium (P>0.05). The Ho ranged from 0.6175 to 0.9270. The DP ranged from 0.7964 to 0.9869, as well as the PIC distributed from 0.5611 to 0.9123. The CDP was 0.999999999999999. The CPEduo was 0.999997431701961, while CPEtrio was 0.999999999654865. Five alleles were detected in DY S391 locus, with the allele frequency from 0.0040 to 0.7290, and GD was 0.4189. Except D13S317 and D10S1248, seventy-six mutation events were observed at the rest nineteen autosomal STR loci. Among them, seventy-five (98.68%) were one step mutation, and only one (1.32%) was three steps mutation. The mutation rate ranged from 0.2465×10-3 to 2.7114×10-3, and the averaged mutation rate was 0.8921×10-3 (95% CI: 0.70×10-3-1.10×10-3). In 33 trio mutation cases, the proportion of the paternal mutation and the maternal mutation was 2.09:1. Conclusion The involved STRs are highly polymorphic in Eastern Han population with acceptable mutation rates by the SiFaSTRTM 23plex DNA ID system, which is suitable for paternity testing and individual identification.

8.Guar gum/ethylcellulose coated pellets for colon-specific drug delivery.

The aim of this work was to investigate guar gum/ethylcellulose mix coated pellets for potential colon-specific drug delivery. The coated pellets, containing 5-fluorouracil as a model drug, were prepared in a fluidized bed coater by spraying the aqueous/ethanol dispersion mixture of guar gum and ethylcellulose. The lag time of drug release and release rate were adjustable by changing the ratio of guar gum to ethylcellulose and coat weight gain. In order to find the optimal coating formulation that was able to achieve drug targeting to the colon, the effect of two independent variables (the ratio of guar gum to ethylcellulose and the coat weight gain) on drug release characteristics was studied using 3 x 4 factorial design and response surface methodology. Results indicated that drug release rate decreased as the proportion of ethylcellulose in the hybrid coat and the coat weight gain increased. When the ratio of guar gum to ethylcellulose was kept in the range of 0.2 to 0.7, and the coat weight gain in the range of 250% to 500%, the coated pellets can keep intact for about 5 h in upper gastrointestine and achieve colon-specific drug delivery. The pellets prepared under optimal conditions resulted in delayed-release sigmoidal patterns with T(5%) (time for 5% drug release) of 5.1 - 7.8 h and T(90%) (time for 90% drug release) of 9.8 - 16.3 h. Further more, drug release was accelerated and T(90%) of the optimum formulation pellets decreased to 9.0 - 14.5 h in pH 6.5 phosphate buffer with hydrolase. It is concluded that mixed coating of guar gum and ethylcellulose is able to provide protection of the drug load in the upper gastrointestinal tract, while allowing enzymatic breakdown of the hybrid coat to release the drug load in the colon.

10.Relationship between gene polymorphism of CYP2E1, CYP1A1, IL-4 and medicamentosa-like dermatitis induced by trichloroethylene.

OBJECTIVETo investigate the relationship between gene polymorphism of CYP2E1, CYP1A1, IL-4 and susceptibility of medicamentosa-like dermatitis induced by trichloroethylene (TCE).
METHODS35 patients with medicamentosa-like dermatitis induced by TCE were chosen as the patient group, and 35 healthy workers as control group. The real-time quantitative polymerase chain reaction (PCR) with TaqMan minor groove binding (MGB) probes was used to test single nucleotide polymorphisms (SNP) of CYP2E1, CYP1A1 and IL-4 in the patients with medicamentosa-like dermatitis as well as in the control. The genotypes and the frequency of genotype or allele were compared between the patients and control with statistical analysis.
RESULTSThe frequency of allele G within CYP1A1 gene (rs1048943) was significantly higher in TCE patients (37.1%) than that in control (P<0.05); the frequency of allele T within CYP2E1-1053 C/T was significantly higher in TCE patients (41.4%) than that in control (P<0.01); the frequency of T/T within IL-4-588 C/T (rs2243250) was significantly higher in TCE patients (75.0%) than that in control (P<0.01), and the frequency of allele T within IL-4-588 C/T (rs2243250) was also significantly higher in TCE patients (87.5%) than that in control (P<0.01).
CONCLUSIONThe gene polymorphism of CYP2E1, CYP1A1, IL-4 is probably associated with hypersensitivity for the TCE patients with medicamentosa-like dermatitis, and could be one of the genetic factors related to the individual susceptibility to TCE exposure.
