Academic Journal of Second Military Medical University 2014;35(12):1387-1390
Objective To explore the correlation of preoperative guanylate cyclase C (GC-C) mRNA in peripheral blood with postoperative gastric cancer recurrence. Methods The preoperative blood samples were collected from 60 patients with gastric cancer (GC) before operation, also from 21 patients with dysplasia, 15 with intestinal metaplasia, and 20 healthy volunteers during the same period. GC-C mRNA in the peripheral blood samples was assessed by real-time fluorescent quantitative PCR (RFQ-PCR). The association of peripheral blood GC-C mRNA expression with pathological parameters and gastric cancer recurrence was analyzed. Results The positive rate of preoperative GC-C mRNA in blood samples of gastric cancer patients (48. 3% [29/60]) was significantly higher than that of patients with dysplasia (20. 0%[3/21], P
3. Study on in vitro hypoglycemic effect and mechanism of Polygonum capitatum
Chinese Traditional and Herbal Drugs 2017;48(16):3401-3407
Objective: To investigate the hypoglycemic targets of Polygonum capitatum. Methods: Human liver cancer HepG2 cells were adopted to detect the supernatant culture medium glucose content, and the effect on PPAR-α and GLUT4 gene expression was investigated by qRT-PCR after treatment of P. capitatum extracts (PCB). INS-1 cells similar to islet β cells, divided into drug protection group and repair group, were adopted to determine the cell proliferation activity by MTT; The intracellular SOD and MDA levels were measured by biochemical method; The Cyt C and Caspase-3 protein expression levels were detected by Western blotting. Adopting maltose as substrate of α-glycosidase enzyme inhibition model, the inhibitory efficiency of PCB on glycosidic enzyme was determined. Results: PCB group significantly promoted the absorption of HepG2 cells to supernatant glucose and increased the expression of PPAR-α and GLUT4 genes significantly. Aim at protection and repair of INS-1 cells, PCB group significantly increased cell vitality and SOD level, reduced MDA level compared with model group, and at the same time significantly reduced Cyt C and Caspase-3 protein expression levels. PCB had inhibitory activity to α-glycosidase enzymes, with IC50 of 11.53 mg/mL. Conclusion: PCB could significantly increase the PPAR-α and GLUT4 genes expression to promote the absorption of HepG2 cells to supernatant glucose by blocking the Cyt C-Caspase-3 pathways to reduce apoptosis of islet cells which were damaged by STZ and by raising SOD and declining MDA to improve INS-1 cell oxidative stress; What’s more it has inhibitory activity to α-glycosidase enzymes.

Objective To investigate the clinical significance of protein expression of Six1 in cervical cancer. Meth?ods The immunohistochemical (IHC) staining was applied to detect the expression of Six1 protein in normal cervical tis?sues (n=32), cervical intraepithelial neoplasia (CIN) tissues (n=49) and cervical cancer tissues (n=123). The localization of Six1 protein was detected in vitro cultured HeLa cells using immunofluorescence (IF) staining. Results The positive rate of Six1was significantly higher in cervical cancer (72.3%) than that of CIN tissues (28.6%) and normal cervical tissues (15.6%,χ2=13.118 and 10.058 respectively, P<0.01). There were significance differences in expression levels of Six1 protein be?tween different tumor sizes and metastasis of cervical cancer (P < 0.01). The Six1 protein showed positive signals in cyto?plasm and nucleoli in HeLa cells. Conclusion Six1 expression is associated with cervical cancer, which may be a potential biomarker for invasion and metastasis of cervical cancer.

Objective To investigate the safety and efficacy of low-dose recombinant tissue plasminogen activator (rtPA) in Chinese patients with acute ischemic stroke. Methods Consecutive patients with acute ischemic stroke received rtPA intravenous thrombolysis within 4. 5 h after symptom onset were enrol ed retrospectively. According to the dosage of rtPA, they were divided into either a low-dose group (0. 5-0. 8 mg/kg, the maximum dose 50 mg) or a standard dose group (0. 9 mg/kg, the maximum dose 90 mg). The incidences of symptomatic intracranial hemorrhage ( sICH) ( the National Institute of Neurological Disorders and Stroke [NINDS], and European Cooperative Acute Stroke Study Ⅱ [ECASSⅡ] , European Safe Implementation of Thrombolysis in Stroke-Monitoring Study [ SITS-MOST ] ), 90 d mortality, 90 d good outcome (the modified Rankin Scale [mRS] score 0-1), and life self-care ability (mRS score 0-2) were compared between the two groups. Results A total of 163 patients were enrol ed, including 74 patients in the low-dose group and 89 in the standard dose group. The constituent ratios of hypertension (44. 6% vs. 68. 5%; χ2 =9. 490, P=0. 002) and diabetes (5. 4% vs. 28. 1%; χ2 =14. 216, P<0. 001) in the patients of the low-dose group were significantly lower than those of the standard group, while the constituent ratios of smoking (56. 8% vs. 38. 2%; χ2 =5. 590, P=0. 018 ) and smal artery occlusive stroke (21. 6% vs. 10. 1%; χ2 =4. 122, P=0. 042 ) in patients of the low-dose group were significantly higher than those of the standard group. After adjusting for age, National Institutes of Health Stroke Scale (NIHSS) score, hypertension, diabetes, smoking, and ischemic stroke typing, multivariate binary logistic analysis showed that there were no significant differences in NINDS defined sICH (10. 8% vs. 9. 0%, odds ratio [OR] 1. 077, 95%confidence interval [CI] 0. 338-3. 436), ECASS Ⅱdefined sICH ( 9. 5% vs. 9. 0%; OR 0. 976, 95% CI 0. 296- 3. 221 ), SITS-MOST defined sICH (8. 1% vs. 4. 5%; OR 2. 269, 95% CI 0. 522-9. 852), 90 d mortality (17. 6% vs. 14. 6%; OR 1. 720, 95% CI 0. 578-5. 119), 90 d good outcome (35. 1% vs. 32. 6%;OR 0. 780, 95% CI 0. 356-1. 709), and life self-care ability (48. 6% vs. 42. 7%;OR 0. 936, 95% CI 0. 441-1. 987) between the 2 groups (al P>0. 05). Conclusions The program of intravenous thrombolysis for the treatment of Chinese patients with acute ischemic stroke with low-dose rtPA is safe and effective.
10. Research progresses in transcutaneous vagus nerve stimulation
Journal of Shanghai Jiaotong University(Medical Science) 2020;40(4):539-542
The vagus nerve is involved in the regulation of human inflammation, mood and pain. Stimulation of the vagus nerve has assured therapeutic effects on intractable epilepsy and refractory depression. On the surface of the human body, the ear is the only area in which vagal afferent fibers distribute. According to the "bottom-up" mechanism of the signal processing, the central nervous system can be modulated by stimulating the peripheral nerve. Therefore, stimulation of the vagus nerve can regulate the activities of brain stem, thalamus, cerebral cortex and other related areas to produce therapeutic effects. Transcutaneous vagus nerve stimulation is a safe, low-cost, non-invasive physical therapy modified from vagus nerve stimulation. This review focuses on the regulation of organ function by transcutaneous vagus nerve stimulation and its application to diseases treatment.
