1.The expressions of B7-H1 and PD-1 signal pathway in patients with chronic hepatitis B

Objective To study the expressions of B7-H1 and PD-1 which are costimulatory molecules in the pathogenesis of chronic hepatitis B (CHB). Methods The heparin anticoagulant blood samples from 11 CHB patients and 16 healthy controls were collected. The expressions of B7-H1 and PD-1 on myloid dendritic cells (mDC) and T lymphocytes in the peripheral blood were analyzed by flow cytometry. The data comparison was done by t test. Results The expressions of B7-H1 on CD3~+ ,CD4~+ , CD8~+ T lymphocytes and mDC in CHB patients were (40. 69± 14. 49)%, (42. 84 ±11. 19)%,(33. 48±14.07)% and (16. 60±4. 04) % , respectively, and those were (14. 66±10. 11) % , (4. 62±3.84)%, (1.89±2.31)% and (0. 49±0. 37)% , respectively in healthy controls. The expressions of B7-H1 on T-lymphocytes and mDC in CHB patients were significantly higher than those in healthy controls, and the differences were significant (t=-2.884,t=-10.894, t=-7.378, t=-13. 182,respectively; all P<0. 05). The expressions of PD-1 on CD3~+ , CD4~+ and CD8~+ T lymphocytes in CHB patients were (12. 45±6. 36)%, (11. 42 ± 6. 20)% and (13. 03± 6. 71) %, respectively, and those were (7. 80±3. 53)%, (7. 12±2. 60)% and (7. 88±3. 74)% , respectively in healthy controls.The expressions of PD-1 on T lymphocytes in CHB patients were significantly higher than those of the healthy controls, the differences were significant (t = -2.323, t =- 2. 355, t =- 2. 439,respectively) all P<0. 05). Conclusion The expressions of B7-H1 and PD-1 on T lymphocytes and mDC in CHB patients are higher than the healthy people.

3.X-linked inhibitor of apoptotic protein modified adipose-derived stem cells transplantation promotes the regeneration of myocardium and capillaries after myocardial infarction

Objective To investigate the mechanism of transplantation of autologous adiposederived stem cells (ADSCs) and X-linked inhibitor of apoptosis protein (XIAP) modified ADSCs for the treatment of myocardial infarction (MI) by the histological detection method.Methods Totally 27 male SD rats (aged 10 weeks,weighted 200-300 g) were randomly divided into the control group,ADSCs group and XIAP ADSCs group (n=9 each).ADSCs were isolated from inguinal adipose tissue in rats and were cultivated in Dulbecco' s modified Eagle' s medium.X-linked inhibitor of apoptotic protein expression plasmid was electrotransfected into ADSCs.Myocardial infarction model was conducted by ligating the left anterior descending artery.The control group was not given any operation.Rats in different groups received direct epicardial injections of normal saline,or ADSCs cell suspension or XIAP modified ADSCs cell suspensions respectively at five sites in the central and border zones of myocardial infarction.Results Compared with control group,the infarct areas were decreased in ADSCs and XIAP ADSCs groups,and some myocardial cells and blood capillaries could be found in the myocardial infarction zone.The capillary number was much more in XIAP-ADSCs group than in ADSCs and control groups [(18.65±1.12) /mm2 vs.(8.68±0.90) /mm2,(18.65±1.12) /mm2 vs.(7.64 ± 0.60)/mm2,t=-22.49,29.91,both P<0.05].Conclusions Autologus ADSCs and XIAP-ADSCs transplantation may reduce infarction area and inhibit left ventricular remodeling by promoting the regeneration of myocardial cells and blood capillaries,which restores the cardiac function.
4.Electrocardiographically triggered CT angiography of the whole aorta and coronary arteries with high-pitch dual-source CT

Objective To investigate the feasibility of comprehensive assessment of the whole aorta and coronary arteries (CA) simultaneously with high-pitch 128-slice dual-source CT ECG-gated FLASH protocol.Methods A total of 48 consecutive patients with suspected aortic diseases underwent CTA examination of the whole aorta and CA using a ECG-gated FLASH CT protocol (pitch =3.2) without heart rate (HR) control.Aorta,aortic valves and CA were shown with different post-processing modalities.The image quality of the aorta,aortic valves and CA was evaluated and compared according to HR (low HR group ≤ 65 bpm,high HR group > 65 bpm).The scan time,effective dose and contrast medium volume were recorded.Inter-observer differences were calculated by Kappa test.Differences between groups were analyzed by Mann-Whitney test with count data and variance test with measurement data.Results All examinations were completed successfully.The image quality was acceptable in the aorta,aortic valve (100%,48/48) and CA (94.0%,551/586).Percentage of diagnostic quality images was slightly higher in low HR group (94.7%,232/245,93.5%,319/341,Z =-2.504,P < 0.05).Inter-reader reproducibility was 95.8% (46/48) in the aorta and aortic valve,96.6% (566/586) in CA,yielded good agreement (Kappa =0.81 and 0.89).The mean attenuation of aorta and CA were higher than 300 HU,especially in low HR group (357.0-446.0) HU.Except proximal segment of right CA,the CNR of whole aorta and coronary arteries were significantly higher in the low HR group (24.5-29.0) than that in the high HR group (20.0-23.1,P <0.05).SNR of the whole aorta was higher in the low HR group (13.7-17.9) than the high HR group (11.5-13.9,P < 0.05).The mean scan time was (1.56 ± 0.08) s,effective dose was (4.12 ± 1.23) mSv (2.77-6.77 mSv),and contrast medium volume was (72.8 ± 2.1) ml.Conclusions CT angiography of whole aorta and coronary arteries could be performed simultaueously within 2 seconds with CT ECG-gated FLASH protocol.The images were of diagnostic quality for aortic and CA disease with low dose of radiation and contrast media.However,high HR could decrease the image quality of CA.
5.Determination of 1-deoxynojirimycin from mulberry resources and analysis of their bioactivities

Objective To determine 1-deoxynojirimycin(DNJ) from mulberry resources and analyse their bioactivities.Methods Determination of DNJ by derivatization with 9-fluorenylmethyl chloroformate(FMOC-Cl) and followed by reversed-phase high-performance liquid chromatography(RP-HPLC) and a fluorescence detector;the inhibitory curves on ?glucosidase at the enzymatic level and the activities to(?-)glucosidase in vitro were observed in the everted sac experiment.Results A reliable quantitative method suitable for assays of DNJ from mulberry resources has been developed.The inhibitory curves of the extracts on ?-glucosidase were parallel to those of DNJ.The extracts can inhibit hydrolyzation of starch and absorption of glucose.Conclusion The medicinal parts from mulberry resources can be used for diabetic therapy as ?-glucosidase inhibitor.
6.Clinical value of transvaginal ultrasound, MRI and hysteroscopy in the assessment of endometrial cancer lesion size

Objective To investigate the clinical significance of transvaginal ultrasound, hysteroscopy and MRI in the assessment of endometrial cancer lesions size. Methods Data from 56 patients who successively underwent transvaginal ultrasound, MRI and hysteroscopy inspection preoperative endometrial carcinoma were retrospectively analyzed to assess the accuracy of lesions size. Results The pathologic lesions size measured mean maximum diameter of 56 cases was (3.05 ± 0.23) cm, while the mean maximum diameter measured by vaginal ultrasound, MRI and hysteroscopy were respectivelly (2.46±0.31) cm, (3.12± 0.08) cm, and (3.18 ± 0.21) cm. Compared with the pathologic measured values, the compliance rates of transvaginal ultrasound, hysteroscopy and MRI were respectively 54%(30/56), 71%(40/56)and 75%(42/56), which vaginal ultrasound measurement value was significantly different than that by pathologic measured ( P=0.031), while there were significant difference between the hysteroscopy measured lesion size and pathologic measured, or between MRI measured values and pathologic measured (all P>0.05). Conclusion Preoperative assess the endometrial cancer lesions size, significance of vaginal ultrasound examination is limited, and MRI and hysteroscopy examination is accurate, but easy to over-estimated lesion size.
7.Quantitive changes of circulating myeloid and plasmacytoid dendritic cell subsets in HBV infected patients during different periods of their infection.

OBJECTIVETo study the quantities of myeloid and plasmacytoid dendritic cell (mDC, pDC) subsets associated with different blood virus loads (HBV DNA) in patients in different periods of HBV infection (immune tolerance, immune activities and non-replicating periods).
METHODSThirty HBV infected patients in different stages of infection were enrolled. They were divided into three groups: an immune tolerance group (10 cases), an immunization activities group (10 cases), and a non-replication group (10 cases). Ten healthy people were enrolled and served as controls. Blood (10 ml) from the patients and the controls were collected and the numbers of dendritic cells (DC) in percentage were counted using flow cytometry. The numbers of mDC and pDC were counted and the relevance of them with their blood virus loads (HBV DNA) was analyzed using statistical methods.
RESULTS(1) Absolute values of mDC, pDC percentage and pDC absolute value of the HBV infected patients were lower than those of the healthy control group (P less than 0.05). mDC percentage of the HBV infected patients was a little lower than that of the healthy controls but of no statistical significance (P more than 0.05). (2) The percentage of mDC in the healthy controls was higher than that in the tolerance group (P less than 0.05) and its absolute value in the healthy control group was higher than those in the immune tolerance group and the immune activities group (P less than 0.05). pDC percentage of the healthy control group was higher than those of the immune activities and non-reproduction groups (P less than 0.05). pDC absolute value of the healthy control group was higher than those of the immune tolerance, immune activities and non-reproduction groups (P less than 0.05). (3) The pDC counts in the tolerance group were negatively correlated with HBV DNA levels (r = -0.686, P less than 0.05).
CONCLUSIONS(1) DC frequency and counts of HBV infected patients in different periods decreased. The immune active group decreased significantly more than the healthy control group (P less than 0.05). (2) pDC counts show a negative correlation with HBV DNA levels in only the tolerance group.

8.Association of FGF23 gene polymorphism with Kawasaki disease and coronary artery lesions.

OBJECTIVETo study the distribution of polymorphism of c.212-37insC (rs3832879) in intron 1 of fibroblast growth factor 23 (FGF23) gene and its association with Kawasaki disease (KD) and coronary artery lesions (CAL).
METHODSForty children with KD were enrolled in this study, among whom 16 children had concurrent CAL. Twenty-six age-matched healthy children were enrolled as controls. PCR and gene sequencing were applied to explore the distribution of polymorphism of c.212-37insC (rs3832879) in FGF23 gene in KD patients and controls.
RESULTSAmong 40 children with KD, 14 (35%) carried the polymorphism of c.212-37insC (rs3832879) in FGF23 gene; among 26 controls, 6 (23%) carried such polymorphism. There was no significant difference in genotype distribution at this locus between the two groups (P=0.30). Among 16 children with CAL, 9 (56%) carried the polymorphism at this locus; among 24 children without CAL, 5 (21%) carried such polymorphism. As for the comparison of two subgroups with and without CAL, the difference in genotype distribution at this locus had statistical significance (P=0.02, OR=4.89, 95% CI: 1.21-19.71).
CONCLUSIONSThe polymorphism of c.212-37insC (rs3832879) in FGF23 gene may not be associated with the pathogenesis of childhood KD, but it may be associated with the development of CAL in children with KD.

Objective To study the relationship of proliferation and activation of T lymphocyte subsets and disease progression in antiretroviral-naive human immunodeficiency virus(HIV)-1-infected individuals.Methods Forty-nine antiretroviral-naive,chronically HIV-1 infected patients and 16 healthy,HIV-1 negative controls were enrolled in this study.The patients were divided into 3 groups according to their CD4+T cell counts:<200×106/L,(200-350)×106/L and>350×106/L.Peripheral blood mononuclear cells(PBMC)were isolated.T cell proliferation index was measured by Ki-67 staining.T cell activation was detected by CD38 staining.The samples were analyzed by flow cytometry.The data were compared by one-way ANOVA.Results The percentage of Ki-67+cells in CIM+T ceils was 7.92%±4.37%in CD4+T cell<200×106/L group,which was significantly higher than those 0.39%d:0.24%in control group,2.61%±2.12%in(200-350)×106/k group and 2.65%±2.13%in>350 X106/L group(F=21.961,P<0.01).The percentage of Ki-67+cells in CD8+T ceils in CD4+T cells<200×106/L group was 2.87%±1.13%,which was also much higher than those in other 3 groups(0.15%±0.90%,1.40%±1.17%,1.22%±0.80%,respectively F=19.203,P<0.01).The Ki-67'CD4'T cells and Ki-67+CD8+T cells were inversely correlated with CD4+T cell counts(r=-0.654,r=-0.539,respectively;P
