1.Neuroprotective effect screening and the mechanism of 10 kinds of coumarin derivatives.

The study reports the detection of neuroprotective effect of 10 kinds of coumarin derivatives and explores their possible mechanism. MTT method was used to screen the neuroprotective effect of 10 coumarin derivatives on neurotoxic agents (Aβ25-35 and rotenone) or OGD (oxygen-glucose deprivation). A compound with better protective effect was obtained. Then the effect of this compound on neurotoxic agents on PC12 was detected by the morphological observation. Furthermore, the effect of compound 3 on microglia with lipopolysaccharide (LPS) induced inflammation was detected. And the inflammatory factor was tested. Finally, direct free radical scavenging ability was detected. Compound 3 was found to be the best compound through three neurons toxic models. Not only compound 3 ameliorated cell viability reduced by three neurons toxic models, but also significantly inhibited the production of inflammatory factor (TNF-α and IL-1β). And its free radical scavenging ability is very good, especially the effect on superoxide anion, which is comparable with vitamin C. The significant scavenging effect of compound 3 on superoxide anion might be the mechanism of the neuroprotection. Compound 3 as a potential neural cell protective agent merits further investigation.

2.Effect of Small Knife Needle on β-enorpin and Enkehalin Contents of Tansverse Process Syndrome of the Third Vertebra.

OBJECTIVETo explore the analgesic mechanism of small knife needle for treating transverse process syndrome of the third vertebra (TPSTV) by observing peripheral and central changesof β-endorphin (β-EP) and enkephalin (ENK) contents.
METHODSTotally 30 Japanese white big-ear rabbits of clean grade were divided into 5 groups according to random digit table, i.e., the normal control group, the model group, the small knife needle group, the electroacupunture (EA) group, and the small knife needle plus EA group, 6 in each group. The TPSTV model was established by inserting a piece of gelatin sponge into the left transverse process of 3rd lumbar vertebrae. Rabbits in the small knife needlegroup were intervened by small knife needle. Those in the EA group were intervened by EA at bilateralWeizhong (BL40). Those in the small knife needle plus EA group were intervened by small knife needleand EA at bilateral Weizhong (BL40). Contents of β-EP and ENK in plasma, muscle, spinal cord, and hypothalamus were determined after sample collection at day 28 after modeling.
RESULTSCompared with the normal control group, contents of β-EP and ENK in plasma and muscle increased significantly, and contents of β-EP and ENK in spinal cord and hypothalamus decreased significantly in the model group (P < 0.05, P < 0.01). Contents of β-EP and ENK approximated normal levels in the three treatment groups after respective treatment. Compared with the model group, the content of β-EP in muscle decreased, and contents of β-EP and ENK in hypothalamus increased in the three treatment groups after respective treatment (P < 0.05). There were no significant difference among the three treatment groups (P > 0.05).
CONCLUSIONSSmall knife needle treatment and EA had benign regulation on peripheral and central β-EP and ENK in TPSTV rabbits. Small knife needle treatment showed better effect than that of EA.

3.Mutation analysis of PAX6 gene in three Chinese families with aniridia

Background Human paired box gene 6 (PAX6)encodes a transcriptional regulator.It is essential for eye and brain morphogenesis.Mutation of PAX6 gene isresponsible for many congenital ocular malformations,such as aniridia.Aniridia is a autosomal dominant inheritance mode.Objective In this study,PAX6 gene mutation was analyzed in three Chinese families with aniridia through polymerase chain reaction (PCR) and sequencing.Methods The blood specimens were collected from 5 suffers and normal individuals of 3 aniridia families to extract DNA.The sequences of extron 4-13 were designed based on PAX6 gene.The primer was amplified by PCR and sequenced and compared with the known PAX6 gene sequence.This study complied with Declaration of Helsinki and approved by ethic committee of Sichuan University.Written informed consent was obtained from each individual before any medial examination.ResultsThere were 5 suffers in the 3 families.A heterozygous mutation (c.718 C>T) in PAX6 gene was identified in 2 patients of family A.This mutation caused an amino acid substitution of arginine to termination codon at position 240 ( p.Arg240X) of PAX6 protein.No similar change in the normal families.No any the alteration of PAX6 gene was detected in family B whatever suffers and normal individuals.In family C,a deletion mutation of c.331 delG ( p.Val111 SerfsX13 ) in PAX6 gene was found.The deletion of one base caused frame shift mutation of PAX6 protein,and no such mutation was seen in other families.Conclusions Mutation of PAX6 gene appeares to be causative mutations of the disease in family A and C.
4.Regulation on intercross effect between Cdk5 mediated apoptosis and autophagy after cerebral ischemia

Cyclin-dependent kinase 5 (Cdk5),as a key neuro-nal regulator,has received extensively attention. In our previous experiments, we have preliminary confirmed the importance of the regulation on neuronal apoptosis after cerebral ischemia by Cdk5 signal path. According to the documents reported, we found that there were signal transductions between apoptosis and autophagy, which acted the"rapier"position after cerebral is-chemia. Furthermore,Cdk5 could mediate protein kinase B(Akt or PKB) to play bidirectional regulation on the intercross be-tween apoptosis and autophagy. So,there would be of great sig-nificance to reveal the signal transduction relationship between apoptosis and autophagy mediated by Cdk5. Owing to the impor-tance of the intercross-effect between the two programmed cell death paths,we aimed at the imparity viewpoints between apop-tosis and autophagy after cerebral ischemia, raising the sugges-tions as follows:it is appropriate to reveal the effects of Cdk5 on Akt kinase dynamically, and discuss the double regulation mechanism of co-regulators between apoptosis and autophagy af-ter cerebral ischemia, which would provide references for the following researches.
5.Correlation between immunochemical level and patient with caries.

OBJECTIVETo explore the relationship between immunochemical level of salivary and caries in children aged 4-6 years old.
METHODSTwo groups were assorted as patients with caries and without caries. Every group included 45 people. Measurements of salivary secretory immunoglobulin A (SIgA)were performed by using radio-immunoassay and single agar diffusion assay. The levels of lysozyme (LZ), alkaline phosphatase (ALP) and lactic dehydrogenase (LDH) were studied with colorimetry and turbidimentry.
RESULTSThe levels of LDH, SigA, ALP, LZ had significant difference between the two groups (P < 0.05). The level of LDH between patients and peoples without caries had little difference (P > 0.05).
CONCLUSIONThe incidence of caries is associated with age, and it may have association with immunochemical levels of salivary.

6.Expression of proline-rich tyrosine kinase-2 in human primary colorectal carcinoma and its prognostic implication.

OBJECTIVETo investigate the expression of proline-rich tyrosine kinase-2 (Pyk2) in human primary colorectal carcinoma (CRC) and it's prognostic significance.
METHODSThe expression of Pyk2 was retrospectively examined with immunohistochemistry (IHC) in 108 tissues of primary CRC. The correlation of Pyk2 expression to prognosis and relevant clinical factors were analyzed.
RESULTSThe rate of Pyk2 low-expression in CRC was 56.5% (61/108). The expression of Pyk2 correlated significantly to the histological grade (P < 0.05) and the TNM stage (P < 0.05), while no correlation between Pyk2 expression and age, tumor size (P > 0.05). Patients with Pyk2 over-expression had significantly higher 5-year survival rate (66.0%) than those with Pyk2 low-expression (31.4%). Pyk2 expression, together with carcinoma histologic grade and TNM stage were prognostic factors to CRC on the multivariate analysis.
CONCLUSIONSPyk2 expression can be a prognostic factor to the CRC patients together with other predictors.

7.Effects of low concentrations of di-(2-ethylhexyl) and mono-(2-ethylhexyl) phthalate on steroidogenesis pathways and apoptosis in the murine leydig tumor cell line MLTC-1.

The aim of this study was to evaluate the effects of low concentrations of DEHP and MEHP on steroidogenesis in a murine Leydig tumor cell line (MLTC-1) in vitro. The result of flow cytometry analysis revealed that the proportion of apoptotic cells was significantly increased after the exposure to DEHP. All three genes (P450scc, P450c17, and 3βHSD) under study showed an increased expression following exposure to DEHP or MEHP, although some insignificant inhibitory effects appeared in the 10 μmol/L treatment group as compared with the controls. It was also found that compared with the controls. It was also found that DEHP or MEHP stimulated INSL3 mRNA and protein especially in the 0.001 μmol/L treatment group. Testosterone secretions were stimulated after the exposure to DEHP or MEHP. Alternations of steroidogenic enzymes and INSL3 in MLTC-1 cells might be involved in the biphasic effects of DEHP/MEHP on androgen production.

8.The complications of nerve injury after the Le Fort I osteotomy.

OBJECTIVETo investigate the incidence of the temporary and permanent sensory disturbance of the inferorbital nerve (ION) after Le Fort I osteotomy.
METHODSThirty patients with 60 inferorbital nerves were examined preoperatively, 1 week, 1 month, 3 months and 6-months after the operation of the Le Fort I osteotomy by using sharp-blunt testing, 2-point discrimination, electric pain response test (ZGK-1 electrometer).
RESULTSThe incidence of the temporary nerve impairment was 75% (45 of 60) and the obvious recovery was found after 1 to 3 months after the operation. No permanent sensory disturbance was found in the patients.
CONCLUSIONSThe inferorbital nerve injury after Le Fort I osteotomy is usually temporary and the sensory recovery was in 1 to 3 months after the operation.

9.Influential factors on psychosocial health of the migrant workers in Guangzhou.

OBJECTIVETo study the influential factors on psychosocial health of the migrant workers in Guangzhou.
METHODSThe Symptom Checklist 90 (SCL-90) and Eysenck Personality Questionnaire (EPQ) were used to investigate 518 migrant workers in Guangzhou.
RESULTSThe rate of migrant workers with psychosocial problems was 36.5%. The scores of SCL-90 and positive rates in migrant workers with the different personality types had significant difference (P < 0.01). The results of binary logistic regression analysis indicated that the working years, drinking, sex, P scores, E scores and N scores of EPQ were main predictors of the poor physical fitness status. The vocations, working years, P scores and N scores of EPQ were strong predictors of the somatization. he vocations, P scores and N scores of EPQ were strong predictors of the obsessive compulsive symptom. The smoking, P scores and N scores of EPQ were strong predictors of the interpersonal sensitivity. The working years, P scores of EPQ were strong predictors of the depression. P scores of EPQ was strong predictors of the anxiety. P scores, E scores and N scores of EPQ were strong predictors of the hostility. The working years, smoking, P scores, E scores and N scores of EPQ were strong predictors of the phobic anxiety. The working years, P scores of EPQ were strong predictors of the paranoid ideation. The working years, P scores and N scores of EPQ were strong predictors of the psychosis.
CONCLUSIONThe level of mental health of the migrant workers was significantly associated with the personality. The results of present study indicated that different vocation, sex, working years, smoking and drinking might interfere with the psychological states. The migrant workers with the personality of psychoticism, neuroticism and introversion may have unhealthy mental reaction.

10.Comparison of the inhibitory activities of salvianolic acid B and Ginkgo biloba extract EGb 761 on neurotoxicity of beta-amyloid peptide.

AIMTo compare the effects of salvianolic acid B (Sal B) and Ginkgo biloba extract EGb 761 on beta-amyloid peptide (beta-AP) fibril formation and cytotoxicity to PC12 cells.
METHODSThe inhibitory effects of Sal B and EGb 761 on beta-AP1-40 fibril formation were determined by using fluorescence analysis with Thioflavin T (ThT) and electron microscopic image. beta-AP25-35 was aged by incubating at 37 degrees C for 7 d, then the protein was incubated with PC12 cells. The protective effects of Sal B and EGb 761 against cytotoxicity induced by aged beta-AP25-35 in PC12 cells were evaluated by MTT reduction assay and flow cytometric analysis. beta-AP25-35-induced accumulation of intracellular reactive oxygen species (ROS) was determined by fluorescence analysis.
RESULTSBoth Sal B and EGb 761 inhibited the formation of amyloid fibrils, protected PC12 cells from beta-AP25-35-induced cytotoxicity, and decreased ROS accumulation caused by beta-AP25-35. The effective doses of Sal B were far lower than those of EGb 761.
CONCLUSIONSal B was much more efficient than EGb 761 in inhibiting beta-AP aggregation and in protecting PC12 cells from beta-AP-induced cytotoxicity.
